Abstract
本发明涉及涉及基因工程技术领域,尤其涉及小麦TaPRR95‑B蛋白或其编码基因在调控植物株高中的应用。本发明研究发现小麦TaPRR95‑B蛋白在调控植物株高存在关键作用,当敲除植物中小麦TaPRR95‑B蛋白的编码基因时,植物的株高显著下降。本发明进一步对比TaPRR95‑B基因野生型和其四倍体突变体的表型,证明敲除该基因能够显著降低小麦的株高,为研究该基因影响株高的分子机理和应用提供了基础。
Description
技术领域
本发明涉及基因工程技术领域,尤其涉及小麦TaPRR95-B蛋白或其编码基因在调控植物株高中的应用。
背景技术
小麦是世界重要的粮食作物,也是全世界很多人口的主粮,因此小麦的产量与国家的粮食安全密切相关。随着人口的增加,耕地面积的减少,生产成本的增加,如何增加小麦产量来保障粮食安全成为亟待解决的问题。20世纪70年代,利用小麦的半矮秆基因主导的绿色革命给全球小麦产量带来了很大的增长,说明小麦株高对产量的形成有重要的影响,研究株高发育遗传规律有助于了解产量形成的作用机制,对利用株高基因选育抗倒伏和高产的品种,进而提高小麦的产量具有重要意义。
PRRs(Pseudo-Response Regulators)家族基因,具有保守的氨基端的PRR/RLD(Receiverlike domain)结构域和一个羧基端的CCT结构域,这2个结构域被1个不太保守的“可变”域分隔,N端响应调节接收结构域与参与其ASP磷酸化信号转导途径的接收域高度相似,C端的CCT结构域与酵母血红素激活蛋白(HAP2)的NF-YA2 DNA结合区相似。PRRs家族基因影响植物的生长发育,包括对开花时间的调控、对光合作用的反应、热休克反应、氧化应激反应、气孔导度的调节、线粒体的新陈代谢以及对冷逆境的响应等。
PRRs基因在被子植物中是保守的,并且在其基因组中至少保留了5个PRR基因拷贝,被子植物PRR基因在单子叶植物和双子叶植物分化之前就分为3个进化枝:PRR1/TOC1(TIMING OF CAB2EXPRESSION1)、PRR3和PRR7、PRR5和PRR9,尽管这些基因的氨基酸序列在PRR结构域和CCT基序中高度保守,但在3个进化枝的PRR结构域中存在几个氨基酸变化,能导致其功能差异。目前,在拟南芥和番木瓜中已经鉴定出5个PRR基因,分别为PRR1(TOC1)、PRR3、PRR5、PRR7和PRR9,水稻中也有5个直系同源的PRR基因,分别为OsPRR1/OsTOC1、OsPRR37、OsPRR59、OsPRR73和OsPRR95。在这些基因中,OsPRR37由于影响水稻开花期、株高和每穗小穗数而显得更为重要。但是关于小麦中PRR基因家族结构和功能的分析还是知之甚少,对该基因在小麦中的生物学功能和分子机制尚不清楚。
发明内容
为了解决现有技术存在的问题,本发明提供小麦TaPRR95-B蛋白或其编码基因在调控植物株高中的应用。本发明研究发现TaPRR95基因的B同源染色体基因与株高的关联,可以通过敲除该基因,显著降低植物株高。
第一方面,本发明提供小麦TaPRR95-B蛋白或其编码基因在调控植物株高中的应用。
本发明进一步提供小麦TaPRR95-B蛋白或其编码基因在培育制备转基因植物或植物种质改良中的应用。
进一步地,所述小麦TaPRR95-B蛋白选自SEQ ID NO:1-3任一所示氨基酸序列。
进一步地,所述小麦TaPRR95-B蛋白的编码基因选自SEQ ID NO:4-6任一所示核苷酸序列。
进一步地,所述植物为单子叶植物或双子叶植物,优选为小麦。
第二方面,本发明提供一种与小麦TaPRR95-B蛋白的编码基因连锁的SNP位点,所述SNP位点位于5号染色体第505806276位。
本发明进一步提供所述SNP位点在鉴定小麦TaPRR95-B蛋白的编码基因中的应用,所述小麦TaPRR95-B蛋白的编码基因选自SEQ ID NO:4-6任一所示核苷酸序列。
第三方面,本发明提供一种调控植物株高的方法,包括:
调控植物中小麦TaPRR95-B蛋白的编码基因的表达水平,所述小麦TaPRR95-B蛋白具有SEQ ID NO:1-3任一所示氨基酸序列。
进一步地,通过降低所述植物中所述小麦TaPRR95-B蛋白的编码基因的表达水平,降低所述植物的株高;或,通过将所述小麦TaPRR95-B蛋白的编码基因敲除的株系与其他株系杂交,培育较矮的植物株系。
进一步地,所述降低所述植物中所述小麦TaPRR95-B蛋白的编码基因的表达水平,具体为:
通过CRISP/CAS9技术敲除所述植物中的所述小麦TaPRR95-B蛋白的编码基因。
本发明具备如下有益效果:
本发明研究发现TaPRR95基因的B同源染色体基因与植物的株高相关,并且进一步通过对比其四倍体突变体和野生型的表型,证明敲除该基因能够显著降低小麦的株高,为研究该基因影响株高的分子机理和应用提供了基础。

具体实施方式
以下实施例用于说明本发明,但不用来限制本发明的范围。
实施例1 TaPRR95-B基因在核心种质材料中的基因分型以及株高关联性分析
1、选择分布于我国不同麦区的小麦微核心种质材料(105份育成品种和157份地方品种)作为多态性SNP位点的发掘材料。
2、外显子捕获获得微核心种质材料的SNP
利用罗氏外显子捕获试剂盒(SeqCap Pure Capture Bead Kit,06977952001,Roche)对262份材料的DNA进行建库测序,具体方法参见试剂盒说明书。
3、通过BWA软件进行全基因组序列比对,使用GATK软件进行变异检测和基因分型,获得该基因在262材料中的基因型。
4、利用262份微核心种质材料三年三点(2002年于河南洛阳,2005年于河南洛阳,2006年于北京)的株高表型数据,进行关联分析。TaPRR95-B基因与株高的关联结果如表1所示,可以看出,TaPRR95-B基因的SNP位点与小麦材料的株高性状显著相关。

5、根据TraesCS5B01G320500基因号,在小麦基因组数据库网站比对,获得其同源基因号TraesCS5A01G320300和TraesCS5D01G326200,得到TaPRR95基因在A、B、D同源染色体上的基因序列,分别如SEQ ID NO.4-6所示,该基因定位于第五染色体同源群上。
根据基因号,在小麦转录组数据库中查询其表达情况,发现该基因在茎、穗和花药中的表达情况如图1所示。
实施例2 PRR95-B基因突变体表型统计
为验证TaPRR95基因的功能与株高相关,本实施例利用四倍体小麦的EMS诱变库,获得了该基因B拷贝的突变体,具体信息见下表:

本实施例对比野生型和突变体的株高,得到如图2所示结果:和野生型Kronos株高相比,突变体株系kronos1181和kronos2031的株高显著降低,说明PRR95-B基因能够显著降低小麦的株高。
虽然,上文中已经用一般性说明及具体实施方案对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。

2.小麦TaPRR95-B蛋白或其编码基因在培育制备转基因植物或植物种质改良中的应用;所述小麦TaPRR95-B蛋白选自SEQ ID NO:1-3任一所示氨基酸序列;所述植物为小麦。
3.根据权利要求1或2所述的应用,其特征在于,所述小麦TaPRR95-B蛋白的编码基因选自SEQ ID NO:4-6任一所示核苷酸序列。
4.一种SNP位点在鉴定小麦TaPRR95-B蛋白的编码基因中的应用,其特征在于,所述小麦TaPRR95-B蛋白的编码基因选自SEQ ID NO:4-6任一所示核苷酸序列;
所述SNP位点和小麦TaPRR95-B蛋白的编码基因连锁,所述SNP位点位于5号染色体第505806276位。
5.一种调控植物株高的方法,其特征在于,包括:
调控植物中小麦TaPRR95-B蛋白的编码基因的表达水平,所述小麦TaPRR95-B蛋白选自如SEQ ID NO:1-3任一所示氨基酸序列;
所述植物为小麦。
6.根据权利要求5所述的方法,其特征在于,通过降低所述植物中所述小麦TaPRR95-B蛋白的编码基因的表达水平,降低所述植物的株高;或,通过将所述小麦TaPRR95-B蛋白的编码基因敲除的株系与其他株系杂交,培育较矮的植物株系。
7.根据权利要求6所述的方法,其特征在于,所述降低所述植物中所述小麦TaPRR95-B蛋白的编码基因的表达水平,具体为:
通过CRISP/CAS9技术敲除所述植物中的所述小麦TaPRR95-B蛋白的编码基因。
