Abstract
The invention belongs to the technical field of traditional Chinese medicine extraction, and particularly relates to an icariin extraction method, which comprises the following steps: mixing fresh epimedium raw material with weak base-containing alcohol solution, heating and extracting; the weak base-containing alcohol solution is prepared by mixing an alcohol solution with the mass fraction of 50-95% and an ammonia water solution with the mass fraction of 25-28% according to the volume ratio of 90-99: 1-10, mixing; the alcoholic solution is methanol solution or ethanol solution. The method for extracting icariin provided by the invention can obviously improve the content of icariin in epimedium raw materials, especially can enable the content of icariin in epimedium raw materials in a Bashu region to reach more than 0.5%, and obviously improves the utilization rate of the epimedium raw materials.

Background
Herba Epimedii is dry leaf of Epimedium brevicornum Maxim, Epimedium sagittatum Maxim, Epimedium pubescens Maxim or Epimedium koreanum nakai of berberidaceae, has effects of invigorating kidney yang, strengthening tendons and bones, and dispelling pathogenic wind and dampness, and contains flavonoids as main ingredient, wherein the higher content of ingredients include icariin, epimedin c, icarisid and icariin.
In recent years, with the increasing demand of epimedium herb extracts in the market, a large number of epimedium herb raw materials are dug with roots, so that wild raw material data are deficient, and the raw material yield is reduced year by year. However, the epimedium raw materials in the regions of the Chinese and Sichuan provinces and other Pacholymus have more raw materials, but the content of icariin in the raw materials of the epimedium in the regions is generally low, so that the utilization rate of the raw materials in the market is not high, and the waste of raw material resources is caused. Therefore, the method for efficiently extracting the icariin is provided, and particularly has important value for improving the utilization rate of the epimedium raw material in the Bashu region.

the weak base-containing alcoholic solution is prepared from 50-95% by mass of alcoholic solution and 25-28% by mass of ammonia water solution according to the volume ratio of 90-99: 1-10, mixing;
wherein the alcohol solution is a methanol solution or an ethanol solution.
Preferably, the icariin is extracted by mixing alcohol solution containing weak base with fresh epimedium herb raw material and heating for extraction;
the weak base-containing alcoholic solution is prepared from 50-95% by mass of alcoholic solution and 25-28% by mass of ammonia water solution according to the volume ratio of 90-97: 3-10, mixing;
wherein the alcohol solution is a methanol solution or an ethanol solution.
Preferably, the heating extraction is reflux extraction or ultrasonic extraction.
More preferably, the reflux extraction is carried out for 3 times at 80 ℃ for 0.5-2.5 hours, the first feed-liquid ratio is 1kg: 7L, the second feed-liquid ratio is 1kg: 5L, and the third feed-liquid ratio is 1kg: 4L.
More preferably, the ultrasonic extraction is carried out for 1 hour at the temperature of 50-60 ℃, and the material-liquid ratio is 1kg: 20L.
Compared with the prior art, the invention has the following effects:
1. according to the extraction method of the icariin, the fresh epimedium raw material is heated and extracted by using an alcohol solution containing weak base as a solvent, and as a plurality of substances with similar structures to the icariin exist in the epimedium raw material, partial acyl substituted derivatives or other unknown derivatives can be deacylated and converted into the icariin under the condition of weak base, so that the content of the icariin in the epimedium extract is obviously increased;
2. the original enzyme system can be destroyed by treating the icariin with the weak base-containing alcohol solution, so that the icariin is prevented from being further degraded, and the loss of the icariin in the extraction process is reduced;
3. the content of the icariin in the sample obtained by the method is detected by adopting an icariin detection method provided in 'Chinese pharmacopoeia' of 2015 edition, so that the content of the icariin is remarkably improved, and particularly, the content of the icariin in epimedium raw materials in a Bashu region is improved to more than 0.5 percent from 0.01 to 0.1 percent, so that the utilization rate of the epimedium raw materials is remarkably improved;
4. the weak base alcohol solution is used as a solvent for extraction, compared with direct extraction by using a dilute ammonia solution, the subsequent purification treatment is more convenient, no solvent residue exists in the obtained extract, and the energy consumption can be saved; and the recovered alcohol can be recycled after being regulated into salt by hydrochloric acid, so that the alcohol loss is saved, compared with the extraction by inorganic salt, the reaction condition is more mild, simple and controllable, and the quality of the obtained extract is more stable.

wherein the methanol solution containing weak base is prepared by mixing a methanol solution with the mass fraction of 50% and an ammonia water solution with the mass fraction of 25% according to the volume ratio of 99: 1.
Through detection, the content of icariin is 0.52 percent based on dry epimedium raw material.
Example 2

Compared with the comparative examples 1-8 and the comparative examples 1-6, the extraction method provided by the application can obviously improve the content of icariin in the epimedium raw materials, and can improve the content of icariin in epimedium in a Bashu region to more than 0.5%, so that the utilization rate of the epimedium raw materials is obviously improved.
It should be noted that when the following claims refer to numerical ranges, it should be understood that both ends of each numerical range and any value between the two ends can be selected, and since the steps and methods used are the same as those of the embodiments, the preferred embodiments of the present invention have been described for the purpose of preventing redundancy, but once the basic inventive concept is known, those skilled in the art may make other variations and modifications to the embodiments. Claims (5)
1. A method for extracting icariin is characterized in that a weak base-containing alcohol solution is used as a solvent, and the solvent is mixed with a fresh epimedium herb raw material and then is heated and extracted;
the weak base-containing alcoholic solution is prepared from 50-95% by mass of alcoholic solution and 25-28% by mass of ammonia water solution according to the volume ratio of 90-99: 1-10, mixing;
wherein the alcohol solution is a methanol solution or an ethanol solution.
2. The method for extracting icariin according to claim 1, wherein the icariin is extracted by heating after mixing an alcohol solution containing weak base with a fresh epimedium raw material in the region of Bashu;
the weak base-containing alcoholic solution is prepared from 50-95% by mass of alcoholic solution and 25-28% by mass of ammonia water solution according to the volume ratio of 90-97: 3-10, mixing;
wherein the alcohol solution is a methanol solution or an ethanol solution.
3. The method for extracting icariin according to claim 1 or 2, wherein the heating extraction is reflux extraction or ultrasonic extraction.
4. The icariin extraction method according to claim 3, wherein the reflux extraction is carried out for 3 times at 80 ℃ for 0.5-2.5 hours each time, and the first feed-liquid ratio is 1kg: 7L, the second feed-liquid ratio is 1kg: 5L, and the third feed-liquid ratio is 1kg: 4L.
5. The method for extracting icariin according to claim 3, wherein the ultrasonic extraction is carried out for 1 hour at 50-60 ℃, and the material-liquid ratio is 1kg: 20L.
