技术领域
本发明公开了一种涉及抗菌抗微生物活性物质领域。除了根据本发明所述的独特抗菌抗微生物混合物的新用途外,本发明还涉及含有此类混合物的制剂,尤其是用于营养或增香的制剂或药物制剂,以及制备此类制剂的方法和用于营养或增香的制剂或药物制剂的抗菌抗微生物处理方法。本发明的其他方面见随后的说明书,尤其是所附的专利权利要求书。
背景技术
食品,特别是含有动物源性成分的食品(例如蛋黄酱、肉馅和香肠产品)以及含糖食品(例如甜味饮料、糖浆、番茄酱和调味料),由于营养成分丰富且水分活度系数(aw值)高,因此容易发生微生物腐烂。
为了稳定此类产品,通常采用栅栏的理念[J.,Alexander,P.(2017).Lebensmittel-Mikrobiologie.斯图加特,出版商Eugen Ulmer.7,完全审核版。]:如有可能,在配制产品和加工产品时,选择初始发芽数较低的原材料,减少aw值、氧化还原电位和pH值,尤其要注意食品卫生并遵守良好生产规范。采用诸如巴氏灭菌法或使用防腐剂等保存方法作为最后的栅栏。但是,消费者越来越认为经典的保存方法很重要。例如,采用巴氏灭菌法会减少营养素的含量,但会形成煮熟的风味而影响口感。诸如山梨酸和苯甲酸的防腐剂是合成获得的,需要作为添加剂声明。诸如酸性酸、芥菜籽、香精油及其提取物的天然防腐剂只能在一定的限度内使用,因为它们会在感官上严重改变产品。

发明内容
因此,本发明的目标及主要目的是开发可延长易腐食品存放时间的替代配方,而无需采取过多的技术手段,优先选择通过添加天然植物成分,从而优选不会对食品口感产生负面影响,还能使口味浓厚或改善口味。
另外,要提供的制剂在毒理学上应该是无害的,与其他物质高度相容,性质稳定(特别是在典型的制剂中),并且/或者可以廉价地生产。
更理想的是,在各种制剂中使用尽可能少的抗微生物活性物质来达到一定的抗微生物作用。
对于本领域技术人员而言,由于以下原因,使得要寻找具有足够数量的具备一种或多种上述特征的合适物质变得更加复杂:一方面,一种物质的化学结构与其对某些微生物(细菌)的生物活性及其稳定性之间没有明确的依赖关系;另一方面,抗微生物作用、毒理学无害性和物质稳定性之间也没有可预测的联系。
因此,本发明的首要目的是提供满足上述几种或优选为所有标准的制剂、或具有上述特征的理想组合的制剂。
本发明通过使用包含以下或由以下组成的混合物来实现主要目的:
(A)一种或多种具有抗微生物作用的植物提取物和/或其中一种或多种具有
抗微生物作用的馏分,
选自由以下科的植物的提取物组成的组:
天门冬科、大麻科、杜鹃花科、千屈菜科、拉坦尼科、蔷薇科和葡萄科,优选为以下植物:啤酒花、草莓、拉坦尼、石榴、狭叶越桔、大果越桔、欧洲越桔、红豆越桔、葡萄,
及其馏分;
以及
(B)一种或多种具有抗微生物作用和/或提高成分(A)的抗微生物作用和/或减少、掩盖或改变成分(A)的难闻的味道的印象和/或增加或改变成分(A)的好闻味道的印象的调味剂,
选自由以下组成的组:阿布鲁索苷和巴兰辛(优选如WO 2012 164,062中所述的)、苹果酸、苯甲醛、γ-癸内酯、δ-癸内酯、3,7'-二羟基-4'-甲氧基异黄烷(优选如EP 2,253,226中所述的)、圣草酚、乙基-2-甲基丁酸乙酯、丁酸乙酯、己酸乙酯、香叶醛、橙皮素(优选如WO2007/014879、EP 2,368,442-B1或EP 1,909,599-B1中所述的),或者以下物质的提取物:甜叶悬钩子(优选如美国临时申请61/333,435及基于此临时申请的专利申请中所述的)、橙皮素二氢链烷酮、橙皮苷二氢链烷酮、羟基苯甲酰胺(优选如WO 2006/024587中所述的)例如2,4-二羟基苯甲酸香草基酰胺、4-羟基二氢查耳酮(优选如US 2008/0227867 A1和WO 2007/107596中所述的)例如根皮素和二氢异甘草素、对羟基苯甲醛、对羟基苯甲酸、高圣草酚、柠檬苦素、乳酸、薄荷呋喃内酯、罗汉果苷、柚皮苷、柚皮苷二氢查尔酮、新异黄酮类化合物(优选如EP 2,570,036B1中所述的)、新橙皮苷二氢查尔酮、橙花醛、δ-丁位辛内酯、墙草碱(优选如EP 2 008 530 A1中所述的,或以其中所述的和由其衍生出的香气组合物的形式)、胡椒碱、根皮苷、叶绿素异构体(优选如EP2,298,084 B1中所述的)、甜茶苷(优选如EP 2,386,211中所述的)、以下物质的混合物:甜茶苷异构体和同系物、1-(2,4-二羟基-苯基)-3-(3-羟基-4-甲氧基-苯基)-丙烷-1-酮(优选如EP 2,353,403-B1中所述的)、甜菊糖苷和甜叶菊苷(优选不同甜叶菊苷的混合物、如WO 2015062,998中所述的)、三叶苷、香草醛、香草酸(优选如序列号为EP2,517,574的欧洲专利申请中所述的)、香草醛木脂、酒石酸和肉桂醛、作为抗微生物混合物、尤其是协同抗微生物混合物。
根据本发明的一个实施方案,优先选择使用植物的根和/或结实器官或其部分、优选为头状花序的内生、中生、外果皮和/或部分,制备如(A)所述的具有抗微生物作用的一种或多种提取物和/或其一种或多种馏分。
根据本发明的一个实施方案,如(B)所述的一种或多种物质或其部分可以以提取物的形式存在/存在于一种提取物中,或者以其馏分的形式存在/存在于一种馏分中,优选选自由以下植物的提取物(或其馏分)组成的组:黄蒿、龙蒿、紫荆花、藜、一枝黄花属、柑橘属(尤其是甜橙、葡萄柚、香柠檬)、龙血树、北美圣草、甘茶、甜过江藤、黄花石蒜、苹果、滨藜、罗汉果、胡椒属、甜菊属、香草属、红叶花椒。
本发明的优选用途是用作抗微生物混合物,其中成分(B)具有如下作用:
提高成分(A)的抗微生物作用(协同抗微生物作用,即抗微生物作用超过混合物中各种具有抗微生物作用的物质的抗微生物作用之和),有利地允许少量(较少地)使用混合物中具有抗微生物作用的物质,和/或
减少、掩盖或改变成分(A)的难闻味道的印象,和/或
增加或改变成分(A)的好闻味道的印象。
因此,根据本发明,优选根据本发明在混合物中使用的成分(B)的总量足以提高成分(A)的抗微生物作用和/或减少、掩盖或改变成分(A)的难闻味道的印象和/或增加或改变成分(A)的好闻味道的印象。
在此,要减少、掩盖或改变的味道的印象优选为选自由涩味、苦味、草味、金属味、酸味组成的组。
要增加或改变的味道优选为选自由甜味、多汁的口感和浓郁的味道组成的组。
根据本发明使用的混合物非常适合作为抗微生物活性成分混合物,以延长易腐烂成分的存放时间。
有利的是,根据本发明使用的混合物也能满足最初提到的标准。
尽管专家们已经部分地考虑了在此描述的成分的抗微生物特性,但是迄今为止没有迹象表明尤其是根据本发明使用的混合物具有在此描述的作用和优点。特别令人惊讶的是,抗微生物作用明显提高(特别是针对在此所述的细菌)。
本发明基于上述令人惊讶的认识,即根据本发明的混合物显示出特别强的抗微生物作用,特别是针对真菌或酵母,尤其是一种或多种选自由以下组成的组的细菌:

根据本发明,成分(A)的一种或多种提取物优选选自由以下组成的组:优选为红莓提取物的浆果提取物、石榴提取物和香草提取物。
成分(A)的提取物可以使用本领域技术人员已知的生产方法从整株植物、植物部分、经生物技术改造或未改造的植物材料中提取,优选采用固液萃取。此外,如果需要的话,在提取之后还可以对所选物质进行富集和浓缩。
例如,可以用以下纯净的或溶剂混合液提取干燥的植物部分:丙酮、丁烷、丁烷-1-醇、丁烷-2-醇、环己烷、一氧化二氮、乙醚、乙酸乙酯、乙醇、乙基甲基酮、己烷、二氧化碳、甲醇、乙酸甲酯、植物油、丙烷、丙烷-1-醇、丙烷-2-醇和/或水。
可以将诸如表面活性物质、吸湿性物质和/或盐等赋形剂加入萃取溶剂中以提高收率。
湿的植物部分可以借助微波辐射、超声波和脉冲电场提取。
提取物分馏(在此所述的馏分的生产)优选通过分离不良的副成分和/或浓缩选定物质来进行,例如,通过吸附蒸馏色谱分离、发酵、过滤、液-液萃取、膜过滤和/或离心,特别是通过色谱分离、液-液萃取和/或膜过滤。
在提取物分馏之后,可以在适当的情况下通过已知方法测定各个馏分的成分。化合物测定的优选方法是质谱法,尤其是液质联用(LC-MS)。
根据本发明,成分(A)特别优选包含至少一种、两种、三种或更多种选自由以下组成的组的化合物:儿茶素、绿原酸、对香豆酸、表儿茶素、鞣花酸、没食子酸、咖啡酸、芥子酸和肉桂酸。
根据不同的分馏方法,尤其是儿茶素,绿原酸、对香豆酸、表儿茶素、鞣花酸、没食子酸、咖啡酸、芥子酸和/或肉桂酸与其他成分的浓度比可以通过有针对性的方式的分馏进行更改。由此,优选以能提高抗微生物作用的方式进行更改。
在另一个优选的实施方案中,成分(B)包含至少一种、两种、三种或更多种选自由以下组成的组的调味剂:圣草酚、橙皮素、高圣草酚、柚皮素、墙草碱和香草酸。
对于根据本发明使用的混合物,成分(A)的总量与成分(B)的总量之比优选在1000:1至1:10的范围内,更优选100:1至1:10,进一步优选100:1至1:1,更进一步优选10:1至1:1。关于该特征,优选将可以分配给成分(A)也可以分配给成分(B)的潜在成分量分配给成分(A)的总量。
有利的是,在此所述的混合物特别适用于制备用于营养或增香的制剂或药物制剂。
因此,本发明还涉及一种用于营养或增香的制剂或药物制剂,该制剂包含在此所述的混合物,其中,所述混合物的含量足以达到抗微生物作用、优选作为协同抗微生物作用。

由此,不限制根据本发明使用的混合物在本发明所述的制剂中的使用。因此,可以考虑用于营养或增香的多种不同制剂或药物制剂。根据本发明生产的制剂优选用在食品中,尤其是具有动物来源成分的食品(例如蛋黄酱、肉末和香肠产品)以及含糖食品(例如甜味饮料、糖浆、番茄酱和调味料)。
因此,根据本发明的一个优选实施方案,将根据本发明使用的混合物用在选自由食品组成的组的制剂中,尤其是诸如蛋黄酱、肉末和香肠产品的具有动物来源成分的食品,以及诸如甜味饮料、糖浆、番茄酱、色拉酱和调味料的含糖食品。

有利的载体例如是二氧化硅(二氧化硅、硅胶)、碳水化合物和/或碳水化合物聚合物(多糖)、环糊精、淀粉、降解的淀粉(淀粉水解产物)、化学或物理改性的淀粉、改性的纤维素、阿拉伯树胶、印度树胶、黄芪胶、梧桐树胶、角叉菜胶、瓜尔豆胶、刺槐豆胶、藻酸盐、果胶、菊粉或黄原胶;优选的淀粉水解产物是麦芽糊精和糊精,其中特别优选DE值在5至20范围内的麦芽糊精。最初由哪种植物提供淀粉来生产淀粉水解物无关紧要。合适且容易获得的有玉米基淀粉以及木薯、大米、小麦或马铃薯淀粉。载体也可以用作流动助剂,例如诸如二氧化硅。

1.石榴提取物(GRA):

替代地或附加地,可以例如通过色谱方法获得极性和非极性馏分。
为了产生极性和单极性馏分,将100mg葡萄籽提取物放入1mL去离子水中,制备溶液,并将其浇注到用水/乙醇(90/10)以10毫升/分钟的流速进行了预处理的制备型HPLC柱(PRP-1柱250×21.5mm;粒径10μm)上。在烘箱温度(等温60℃)下,使用去离子水和乙醇(99.5%)作为溶剂。首先,将提取物用水/乙醇(90/10)洗脱25分钟。之后,将乙醇的比例在十分钟之内增加10%,至20%,并保持在恒定的水平十分钟。随后,将乙醇比例在五分钟内增加到100%,并保持五分钟。在10到36分钟的运行时间内记录极性馏分,在37到55分钟的运行时间内记录单极性馏分。极性馏分的收率为48%,单极性馏分的收率为28%。
随后通过质谱检查所获得的馏分的成分。获得的馏分主要包含以下非易失性成分:

4.不同提取物的抗微生物作用的检查:
对于单独的不同提取物,即不与根据本发明的成分(B)组合的提取物的抗微生物作用的检查,使用了诸如实施例1至3中所述的示例性提取物。为此,将提取物溶于pH值为3、500mg/kg的柠檬酸盐缓冲液中。将样品用5.63×104CFU/mL的酿酒酵母、1.89×104拜耳接合酵母和3.75×104白色念珠菌接种。

酿酒酵母在培养24小时后观察到少量发芽。7天后,停止了生长。
拜耳接合酵母在7天后观察到少量发芽,从14天开始,没有新的菌落形成。
提取物在21天内抑制了白色念珠菌。
提取物在28天内可以减少出芽短梗霉菌。28天后可观察到石榴提取物将出芽短梗霉菌减少100%。
28天后,巴西曲霉可以检测到少量发芽。
如以下结果所示(参见实施例7),通过与在此所述的成分(B)组合可以提高提取物的抗微生物作用。
5.进一步检查所选提取物及其馏分的抗微生物作用:
对于单独的不同提取物,即不与根据本发明的成分(B)或其馏分组合的提取物的抗微生物作用的进一步检查,使用了诸如实施例1至4中所述的示例性提取物或其馏分。将提取物或其馏分溶于pH值为5、如表中所示的剂量的柠檬酸盐缓冲液中。将样品用3.8×104CFU/mL的酿酒酵母接种。

阴性对照没有导致发芽数明显减少。
但是,添加375mg/kg山梨酸钾的阳性对照也未显示出明显的发芽数减少。根本原因在于,一方面,与应用实施例表2中的pH3相比,山梨酸仅以其有效的未离解形式少量存在,酿酒酵母最易在pH4至6之间生长,并且在pH5时确实出现生长。
使用石榴提取物(以及葡萄提取物)时,未观察到酿酒酵母的发芽数明显减少。更令人惊讶的是,通过分馏提取物,可以观察到对抗微生物生长的抑制作用,即可以观察到发芽数减少了log2。因此,通过添加200mg/kg的葡萄提取物的非极性馏分和添加200mg/kg的石榴膜滤液>2500Da,发芽数大大减少。
如以下结果所示(参见实施例7),提取物与在此所述的成分(B)组合可以提高抗微生物作用。
6.提取物与调味剂的相互作用:

上述实施例表明,通过添加含有上述提取物的独特的调味剂,产品的感官印象得到了提高,同时还保持了产品特有的气味和口味。已经发现,植物提取物与调味剂结合后,诸如苦味、涩味、金属味的原本不好的口味可以转变成诸如更多汁、更浓郁、更甜、更复杂的正面的口感。这一结论同样适用于将在此所述的提取物的馏分与调味剂结合使用的情况。为了优化所需的作用,技术人员可以根据基质来测试和使用不同的剂量。
进一步测试中使用了由60mg/kg的安赛蜜、60mg/kg的三氯蔗糖、0.15g/100g的柠檬酸、200mg/kg的石榴提取物(实施例1)和0.1g/100g樱桃调味剂组成的静态水。添加了具有下述作用的调味剂和调味剂混合物,并与无添加的基础样品的感官特性进行比较:

在进一步测试的范围内,检查了芳香物质和提取物组合的抗微生物作用。下表显示了细菌负荷测试的结果,测试与实施例5和6的实验类似,不同之处在于混合物是用pH为7的纯水制备的。例如,葡萄籽提取物和调味剂香草酸对酵母菌作用不同。然而,在该组合中,还观察到了对真菌的作用和对酵母的最佳作用。与pH3相比,石榴和红莓提取物在pH为7时作用较弱(参见实施例5),石榴提取物在1000mg/kg的剂量下在不到14天的时间内消除了酿酒酵母。但是,在与含柠檬烯的柠檬油组合使用时,较高的pH值效果更好。
