Abstract
本发明公开了一种秸秆固态酸解的预处理方法,其预处理的步骤依次为:含湿量不超过75%的秸秆在15‑100℃固态酸解,然后,加水将可溶性糖分次浸提、挤压过滤,再用加有适量亚硫酸钠等还原性盐的稀碱于15‑100℃下将木质素溶解剥离,余下纤维素固体。由本预处理方法分离获得三大组分半纤维素混糖、纤维素和木质素的含量分别为≥90%(w/w)、≥95%(w/w)和≥90%(w/w),三者折百收率均不低于90%(w/w)。本方法不仅能达到秸秆三大组分的高效精密分离的目的,而且该过程为非高温高压、操作简单、速度快、耗水量低、分离产物含量和收率高,可实现秸秆等木质纤维素的清洁化和低成本的高值化利用。

技术领域
本发明属于生物质炼制技术领域,尤其涉及一种秸秆固态酸解的预处理方法。
背景技术
秸秆等木质纤维素由纤维素、半纤维素和木质素三大主要成分组成,秸秆的表面由木质素覆盖,且部分木质素与半纤维素相互缠绕,并将纤维素包裹起来。这种复杂的空间结构使木质纤维素可以有效抵抗外部环境,但也因此阻碍了它的生物转化以及材料化加工利用。为此,必须对秸秆等木质纤维素进行预处理。
目前,秸秆预处理技术主要有稀酸(蒸煮)水解、稀碱蒸煮、高压汽爆和离子液体萃取和SO3微热爆法等。在稀酸(蒸煮所用液固比高达10-20:1,或更高)水解处理秸秆等木质纤维素的过程中,木质素是不会溶出的,所得糖不含木质素;包括经SO3微热爆处理的秸秆在稀碱处理过程中,木质素被溶解剥离同时有部分半纤维素溶出,所得木质素含有半纤维素。可见,现有秸秆等木质纤维素的预处理技术离理想的三大组分精细分离的要求相差较大,并且成本依然较高【化工进展,2017,36(3):872-879】。
在稀酸的作用下,秸秆细胞壁会发生轻微润胀,半纤维素因酸催化水解糖化而遭破坏并溶解,并由此将纤维素-半纤维素-木质素之间的化学键破坏,从而破坏秸秆原有的结构。但稀酸水解需要在高温下蒸煮,不仅废水量大,而且会产生大量的抑制物,使得后续微生物发酵受抑制,且戊糖回收率不高。YAN等【Bioresource Technology,2009,100(5):1803-1808】对传统酸法进行改进,采用稀硫酸喷雾技术预处理玉米秸秆,戊糖回收率为90-93%,葡萄糖得率90-95%,剥离70-75%木质素。但中和耗用碱量大,若使用廉价氢氧化钙对含糖水解液进行中和时会产生水不溶的石膏。
虽然还可以采用1-2%(w/w)稀硫酸催化乙醇等有机溶剂法以及添加适量硫酸、SO2和CO2催化的高温气爆同步酸解法进行秸秆的预处理,大幅度减少了发酵抑制物的产生量,但均没有从根本上解决预处理成本高的技术问题。
我们采用SO3微热爆法【ZL200910116615.X】在常压和非高温下进行秸秆的预处理,秸秆细胞壁因SO3气体与其中水分子进行水合反应放热至原位气体急剧膨胀而破裂,从而有助于稀碱液渗入秸秆内部使木质素的剥离率提高,得低木质素含量的棕纤维素(半纤维素和纤维素的复合体)和碱木质素溶液。因半纤维素能在碱水溶液中溶胀溶解,而为了提高木质素的除去率,必然有一定量的半纤维素溶入碱木质素溶液中,而无法得到(折百)含量高的碱木质素,使得木质素的分离成本高。
发明内容
本发明目的就是为了弥补已有技术的缺陷,提供一种秸秆固态酸解的预处理方法。
为了实现上述的目的,本发明采用可气化的酸催化含湿秸秆进行固态水解,以断开木质素和半纤维素之间的酯键等链接键并将半纤维素降解为寡木聚糖和少量的木糖等;再利用木质素不溶于非强碱性水溶液以及纤维素因结晶结构不溶于水的特性,通过挤压水洗将半纤维素酸解产物从木质素和纤维素之间抽出,并由此留下镂空的木质素/纤维素复合体;然后,加稀碱液将木质素溶解过滤分离,得高浓度高纯度的碱木质素溶液和纤维素固体。从而实现秸秆三大组分的高效精密分离,具体技术方案如下:
一种秸秆固态酸解的预处理方法,包括以下步骤:
(1)秸秆的固态酸解:含水量≤75%(w/w)的秸秆与少量的可气化酸在密闭的容器中于15-100℃进行固态酸解0.25-3h,半纤维素被酸解原位生成木糖和寡木聚糖等的可溶性混糖,其中纤维素二糖不超过混糖的2%(w/w);
(2)可溶性糖的浸提与过滤分离:向经固态酸解的秸秆加适量的碱进行中和,然后,加水洗涤压滤分离,得半纤维素剥离的含湿秸秆(木质素/纤维素复合体),其中半纤维素残留量≤5%(w/w),滤液为可溶性混糖液;
(3)木质素的剥离:将半纤维素剥离的含湿秸秆与加有适量还原性盐的1-10%(w/v)NaOH或KOH水溶液按固液比1:4-1:8(w/v)混合,于15-100℃、搅拌处理1-3.5h,木质素生成为碱木质素而溶于稀碱水溶液中,然后,经挤压过滤洗涤分离,得含湿的秸秆纤维素固体物,其干品中纤维素含量≥95%(w/w),纤维素折百收率≥95%(w/w),滤液为碱木质素的稀碱水溶液;
(4)滤液分离纯化:来自步骤2的可溶性糖化液,经离子交换树脂脱盐脱色,得寡木聚糖和/或木糖混糖液,其干品中半纤维素糖含量≥90%(w/w),半纤维素混糖折百收率≥90%(w/w);来自步骤3的滤液经膜分离、浓相酸沉、分离得木质素,其干品中木质素含量≥90%(w/w),木质素折百收率≥95%(w/w);膜滤液(即稀相)循环回至步骤3配制稀碱套用。
进一步的,所述步骤1中可气化的酸包括卤化氢类无机酸、低级有机酸以及它们之间按任意比例构成的混合物,其中,无机酸既可以是气体,也可以是其水溶液,酸用量为100g干秸秆重的0.01-0.2mol。
优选的,所述步骤1中可气化的酸选自氟化氢、氯化氢、溴化氢、甲酸、乙酸中的任意一种或几种任意比例的组合。
进一步的,所述步骤1中含湿秸秆的含湿量包含来自秸秆自身保有水量,外加水量以及通过稀酸带入水量,总量不大于秸秆自身重量的3倍,优选不超过1倍。
进一步的,所述步骤1中秸秆包括:稻草、麦秸秆、玉米秸秆、高粱杆和棉秆等农作物秸秆,农作物的根皮,农产品加工过程产生的果壳,以及牧草和碎木屑等林木生物质,或者是在增湿处理之前,经过SO3微热爆处理的上述生物质。
进一步的,所述秸秆含湿量中外加水量,可以是通过通入蒸汽、喷液等外加方式控制,也可以用水溶液浸泡后挤压或加热蒸发脱除多余的水,以满足固态酸解的含湿要求。
进一步的,所述步骤2中固态酸解后秸秆的中和采用的是弱酸强碱盐,可根据工艺要求投加其固体或水溶液,弱酸强碱盐的添加量以pH达6-7.5为准。
优选的,所述弱酸强碱盐包括K2CO3、Na2CO3、KHCO3、NaHCO3、K2SO3、Na2SO3、KHSO3、NaHSO3。
进一步的,所述步骤3中在液碱中添加的还原性盐包括无机盐、有机酸盐以及抗氧剂,添加量为原干秸秆重的0-1%(w/w)。
优选的,所述无机盐包括K2SO3、Na2SO3、KHSO3、NaHSO3、K2S、Na2S、KHS、NaHS,所述有机酸盐包括甲酸盐、草酸盐,所述抗氧剂包括2,4-二叔丁基苯酚、2,6-二叔丁基-4-甲基苯酚。
本发明的优点是:
本发明提出的是利用低浓度可气化的酸催化含湿秸秆固态水解的秸秆预处理方法,秸秆经固态酸解后,用水溶出半纤维素的降解物-寡木聚糖和/或木糖等,由于该过程使用较低的酸浓度和水含量,且在固态水解不超过3h时,仅半纤维素有降解,不会产生糠醛等发酵抑制物,然后用稀碱溶出木质素,余下则为纤维素固体。该方法能达到秸秆三大组分的高效精密分离的目的,且操作过程简单、速度快、耗水量低、分离产物浓度和纯度高,可实现秸秆等木质纤维素的清洁化和低成本的高值化利用。
具体实施方式
以下结合具体的实例对本发明的技术方案做进一步说明:
实施例1
将自然晾干的稻草秸秆【纤维素32.3%、半纤维素37.33%、木质素15.47%、水6 %、灰分7.07%、水溶性木质素等物质2.83%】段切至不超过10cm,取100g,按固液比1:1加入1mol/L的稀盐酸,投入到500mL的烧瓶中,在80℃下固态酸解反应2h,然后,加11%(w/v)Na2CO3水溶液50mL中和/挤压过滤,再加去离子水100mL×2洗涤/挤压过滤,得含湿固体110g(干重50.56g,其中,纤维素63.49%、半纤维素3.64%、木质素30.6%、灰分2.27%);滤液总量约300mL。最后,利用每100mL加有0.5g Na2SO3的稀碱液将木质素溶解剥离,含湿固体与稀碱液比为1:3(w/v),80℃搅拌处理2h,挤压过滤,加去离子水50mL×2洗涤/挤压过滤洗涤过滤,得含湿秸秆纤维素45g(干重33.4g,其纤维素含量95.65%,纤维素折百收率98.9%);滤液总量约440mL。
固态酸解所得半纤维素糖化水溶液,经串联阳阴离子交换树脂处理脱盐、蒸发浓缩、干燥,得类白色粉末36g,其半纤维素混糖含量95.56%(w/w);半纤维素混糖收率92.15%(w/w)。稀碱溶解剥离所得碱木质素,经膜分离、酸沉、过滤干燥得黄褐色木质素干粉16.1g,其木质素含量91.92%(w/w);水不溶性木质素收率95.67%(w/w)。
实施例2
将自然晾干的稻草秸秆【组成含量同实施例1】段切至不超过10cm,取100g,按固液比1:1加入去离子水,投入到500mL的烧瓶中,通入约5g氯化氢气体,在80℃下固态酸解反应2h;然后,加11%(w/v)Na2CO3水溶液60mL中和/挤压过滤,再加去离子水100mL×2洗涤/挤压过滤,得含湿固体100g(干重48.8g,其中,纤维素65.43%、半纤维素2.19%、木质素31.7%、灰分0.68%),滤液合并约310mL; 最后,利用每100mL加有0.5g NaHSO3的稀碱液将木质素溶解剥离,含湿固体与稀碱液比为1:3(w/v),80℃搅拌处理2h,挤压过滤,加去离子水50mL×2洗涤/挤压过滤洗涤过滤得含湿秸秆纤维素 44g(干重33g,纤维素95.94%,纤维素折百收率98%),滤液合并约380mL。
固态酸解所得半纤维素糖化水溶液,经串联阳阴离子交换树脂处理脱盐、蒸发浓缩、干燥,得类白色粉末36.5g,其中半纤维素糖含量94.8%(w/w)、纤维素二糖0.99%(w/w)、灰分4.21%(w/w);半纤维素混糖收率92.7%(w/w)。稀碱溶解剥离所得碱木质素,经膜分离、酸沉、过滤干燥得黄褐色木质素干粉16.17g,其中木质素含量为92.76%(w/w);水不溶性木质素收率97%(w/w)。
以上所述仅为本发明的优选实施例而已,并不用于限制本发明,对于本领域的技术人员来说,本发明可以有各种更改和变化。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (10)
1.一种秸秆固态酸解的预处理方法,其特征在于,包括以下步骤:
(1)秸秆的固态酸解:含水量≤75%(w/w)的秸秆与少量的可气化酸在密闭的容器中于15-100℃进行固态酸解0.25-3h,半纤维素被酸解原位生成木糖和寡木聚糖等的可溶性混糖,其中纤维素二糖不超过混糖的2%(w/w);
(2)可溶性糖的浸提与过滤分离:向经固态酸解的秸秆加适量的碱进行中和,然后,加水洗涤压滤分离,得半纤维素剥离的含湿秸秆,其中半纤维素残留量≤5%(w/w),滤液为可溶性混糖液;
(3)木质素的剥离:将半纤维素剥离的含湿秸秆与加有适量还原性盐的1-10%(w/v)NaOH或KOH水溶液按固液比1:4-1:8(w/v)混合,于15-100℃、搅拌处理1-3.5h,木质素生成为碱木质素而溶于稀碱水溶液中,然后,经挤压过滤洗涤分离,得含湿的秸秆纤维素固体物,其干品中纤维素含量≥95%(w/w),纤维素折百收率≥95%(w/w),滤液为碱木质素的稀碱水溶液;
(4)滤液分离纯化:来自步骤2的可溶性糖化液,经离子交换树脂脱盐脱色,得寡木聚糖和/或木糖混糖液,其干品中半纤维素糖含量≥90%(w/w),半纤维素混糖折百收率≥90%(w/w);来自步骤3的滤液经膜分离、浓相酸沉、分离得木质素,其干品中木质素含量≥90%(w/w),木质素折百收率≥95%(w/w);膜滤液循环回至步骤3配制稀碱套用。
2.根据权利要求1所述的秸秆固态酸解的预处理方法,其特征在于,所述步骤1中可气化的酸包括卤化氢类无机酸、低级有机酸以及它们之间按任意比例构成的混合物,其中,无机酸既可以是气体,也可以是其水溶液,酸用量为100g干秸秆重的0.01-0.2mol。
3.根据权利要求1所述的秸秆固态酸解的预处理方法,其特征在于,所述步骤1中可气化的酸选自氟化氢、氯化氢、溴化氢、甲酸、乙酸中的任意一种或几种任意比例的组合。
4.根据权利要求1所述的秸秆固态酸解的预处理方法,其特征在于,所述步骤1中,含湿秸秆的含湿量包含来自秸秆自身保有水量,外加水量以及通过稀酸带入水量,总量不大于秸秆自身重量的3倍。
5.根据权利要求1所述的秸秆固态酸解的预处理方法,其特征在于,所述步骤1中秸秆包括:稻草、麦秸秆、玉米秸秆、高粱杆和棉秆等农作物秸秆,农作物的根皮,农产品加工过程产生的果壳,以及牧草和碎木屑等林木生物质,或者是在增湿处理之前,经过SO3微热爆处理的上述生物质。
6.根据权利要求4所述的秸秆固态酸解的预处理方法,其特征在于,所述秸秆含湿量中外加水量,可以是通过通入蒸汽、喷液等外加方式控制,也可以用水溶液浸泡后挤压或加热蒸发脱除多余的水,以满足固态酸解的含湿要求。
7.根据权利要求1所述的秸秆固态酸解的预处理方法,其特征在于,所述步骤2中固态酸解后秸秆的中和采用的是弱酸强碱盐,可根据工艺要求投加其固体或水溶液,弱酸强碱盐的添加量以pH达6-7.5为准。
8.根据权利要求7所述的秸秆固态酸解的预处理方法,其特征在于,所述弱酸强碱盐包括K2CO3、Na2CO3、KHCO3、NaHCO3、K2SO3、Na2SO3、KHSO3、NaHSO3。
9.根据权利要求1所述的秸秆固态酸解的预处理方法,其特征在于,所述步骤3中在液碱中添加的还原性盐包括无机盐、有机酸盐以及抗氧剂,添加量为原干秸秆重的0-1%(w/w)。
10.根据权利要求9所述的秸秆固态酸解的预处理方法,其特征在于,所述无机盐包括K2SO3、Na2SO3、KHSO3、NaHSO3、K2S、Na2S、KHS、NaHS,所述有机酸盐包括甲酸盐、草酸盐,所述抗氧剂包括2,4-二叔丁基苯酚、2,6-二叔丁基-4-甲基苯酚。
