Abstract
The invention relates to the technical field of cosmetics, and particularly relates to an essence containing a water shield extract and a preparation method thereof. The essence disclosed by the invention comprises the following components in percentage by mass: 5-15% of water shield extract, 2-5% of licorice extract, 1-2% of centella asiatica extract, 0.1-0.3% of turmeric extract, 1-5% of propylene glycol, 1-5% of butanediol, 0.1-0.5% of sodium lauryl sulfate, 0.1-0.5% of triethanolamine, 0.1-0.5% of hyaluronic acid, 0.2-1.0% of hydroxymethyl cellulose, 0.2-1.0% of glutathione, 0.05-0.3% of carbomer, 0.05-0.3% of allantoin, 0.05-0.3% of essence and the balance of deionized water. The essence disclosed by the invention can effectively inhibit activities of tyrosinase, collagenase, hyaluronidase, elastase and the like, and can effectively increase skin elasticity, tighten skin and delay skin aging.

Technical Field
The invention relates to the technical field of cosmetics, and particularly relates to an essence containing a water shield extract and a preparation method thereof.
Background
Water shield (water shield, Brasenia schrebei), also known as hoof grass, lake vegetable, water sunflower or dew sunflower, grows in ponds, lakes and marshes, is a perennial freshwater aquatic herbaceous plant of the water shield genus of the Nymphaeaceae family, is fresh and tender, and is one of the precious vegetables.
The water shield has high nutritive value and medicinal value, contains rich acidic heteropolysaccharide which is an immunopotentiator, can enhance the immunity of organisms, prevents diseases and has strong inhibiting effect on a plurality of tumors, and is rich in zinc element which is 'zinc king' in plants, can promote the intelligence development of children and prevent and treat the hyperactivity of children.
At present, water shield is primarily processed to be prepared into canned food, which is sold to southeast Asia countries such as Japan, Korea and the like and is deeply favored by southeast Asia people. The tender buds and petioles of the water shield are wrapped with transparent colloid, the appearance is crystal clear, and the taste is smooth. The modern clinical pharmacological test shows that the water shield extract has the activity of inhibiting tyrosinase, elastase, collagenase, hyaluronidase and the like, and can be used for preparing cosmetics with high additional value.
Some patents relate to the application of water shield in cosmetics, but the formula is complex and the antioxidant effect is poor, and the antioxidant effect of the prepared cosmetics still needs to be further improved.

The essence containing the water shield extract consists of the following components in percentage by mass: 5-15% of water shield extract, 2-5% of licorice extract, 1-2% of centella asiatica extract, 0.1-0.3% of turmeric extract, 1-5% of propylene glycol, 1-5% of butanediol, 0.1-0.5% of sodium lauryl sulfate, 0.1-0.5% of triethanolamine, 0.1-0.5% of hyaluronic acid, 0.2-1.0% of hydroxymethyl cellulose, 0.2-1.0% of glutathione, 0.05-0.3% of carbomer, 0.05-0.3% of allantoin, 0.05-0.3% of essence and 60-80% of deionized water.
The above essence containing water shield extract can be used as facial mask essence.
The water shield extract is water shield extract.
The preparation method of the water shield extract-containing essence comprises the following steps: weighing water shield extract, licorice extract, centella extract, turmeric extract, propylene glycol, butanediol, sodium lauryl sulfate, triethanolamine, hyaluronic acid, hydroxymethyl cellulose, glutathione, carbomer, allantoin and essence according to the weight percentage, and then uniformly mixing with deionized water to obtain the water shield.
The water shield extract is prepared by a preparation method comprising the following steps:

The mass fraction of the ethanol in the step 1) is 65-75%.
In the step 1), the volume ratio of ethanol to (the mixture of water shield and water) for soaking is 1: 1.
Preferably, the licorice extract is an alcohol-water extract of licorice, contains active substances such as glycyrrhizic acid, and is provided by Jiangsu Huibo Biotech limited.
Preferably, the centella asiatica extract contains active substances such as asiaticoside and madecassoside, and is provided by Jiangsu Huibobo Biotech limited.
Preferably, the turmeric extract is an alcohol-water extract of turmeric, contains active substances such as curcumin, and is provided by Jiangsu Huibo Biotech Co.
The water shield can inhibit activity of collagenase, and with increasing of collagenase in skin layers, the generation amount of collagen cannot keep up with enzymolysis speed, so that the collagen in the skin is reduced, and the skin is aged and loses vitality. The water shield can inhibit activity of collagenase, and can reduce collagen attenuation, thereby delaying aging. Researches find that the water shield extract can inhibit the activity of collagen enzyme, inhibit the hydrolysis of collagen and effectively prevent skin aging and vitality loss.
Thirdly, the water shield extract has the function of inhibiting hyaluronidase. The hyaluronidase is a specific lyase of hyaluronic acid, and the hyaluronic acid has good moisturizing effect and is a good transdermal enhancer. Glycyrrhrizae radix is dried root and rhizome of Glycyrrhiza uralensis Fisch of Leguminosae. Has the effects of invigorating spleen and replenishing qi, clearing away heat and toxic materials, eliminating phlegm and stopping cough, relieving spasm and pain, and harmonizing the medicines. Modern clinical pharmacology tests show that the licorice extract can inhibit the activity of tyrosinase and dopachrome tautomerase (TRP-2) and inhibit the polymerization of 5, 6-dihydroxyindole, so as to prevent the formation of melanin and achieve the effect of whitening skin.

The preparation method of the water shield extract-containing essence comprises the following steps:
A. preparing water shield extracting solution
1) Washing fresh water shield with cold water, soaking for 10 hours, soaking with 70 ℃ water for 2 hours, then crushing to obtain a mixture of water shield and water, and then soaking with 65% ethanol for 8 hours to obtain a crude extract; wherein the ratio of the volume of 65% ethanol used to the volume of the above mixture is 1: 1;
2) centrifuging the crude extract in the step 1) at 10000rpm for 10min, collecting filtrate, and then removing ethanol in the filtrate in vacuum to obtain the product;
B. preparation of essence

15 parts of water shield extract, 5 parts of licorice extract, 2 parts of centella extract, 0.1 part of turmeric extract, 1 part of propylene glycol, 5 parts of butanediol, 0.5 part of sodium lauryl sulfate, 0.5 part of triethanolamine, 0.5 part of hyaluronic acid, 0.2 part of hydroxymethyl cellulose, 0.2 part of glutathione, 0.3 part of carbomer, 0.3 part of allantoin, 0.3 part of essence and 69.1 parts of deionized water.
The preparation method of the water shield extract-containing essence comprises the following steps:
A. preparing water shield extracting solution
1) Washing fresh water shield with cold water, soaking for 8 hours, soaking with 60 deg.C water for 4 hours, pulverizing to obtain water shield and water mixture, and soaking with 75% ethanol for 6 hours to obtain crude extract; wherein the ratio of the volume of 75% ethanol used to the volume of the mixture is 1: 1;
2) centrifuging the crude extract in the step 1) at 10000rpm for 12min, collecting filtrate, and then removing ethanol in the filtrate in vacuum to obtain the product;
B. preparation of essence
The skin care cream is prepared by stirring and uniformly mixing 15 parts of water shield extract, 5 parts of licorice extract, 2 parts of centella extract, 0.1 part of turmeric extract, 1 part of propylene glycol, 5 parts of butanediol, 0.5 part of sodium lauryl sulfate, 0.5 part of triethanolamine, 0.5 part of hyaluronic acid, 0.2 part of hydroxymethyl cellulose, 0.2 part of glutathione, 0.3 part of carbomer, 0.3 part of allantoin, 0.3 part of essence and 69.1 parts of deionized water.
Example 3
The essence containing the water shield extract in the embodiment is composed of the following components in parts by mass:
10 parts of water shield extract, 3.5 parts of licorice extract, 1 part of centella extract, 0.2 part of turmeric extract, 3 parts of propylene glycol, 3 parts of butanediol, 0.2 part of lauryl sodium sulfate, 0.3 part of triethanolamine, 0.3 part of hyaluronic acid, 0.6 part of hydroxymethyl cellulose, 0.6 part of glutathione, 0.15 part of carbomer, 0.15 part of allantoin, 0.15 part of essence and 76.85 parts of deionized water.
The preparation method of the water shield extract-containing essence comprises the following steps:
A. preparing water shield extracting solution
1) Washing fresh water shield with cold water, soaking for 8 hours, soaking with 60 deg.C water for 4 hours, pulverizing to obtain water shield and water mixture, and soaking with 75% ethanol for 6 hours to obtain crude extract; wherein the ratio of the volume of 75% ethanol used to the volume of the mixture is 1: 1;
2) centrifuging the crude extract in the step 1) at 10000rpm for 12min, collecting filtrate, and then removing ethanol in the filtrate in vacuum to obtain the product;
B. preparation of essence
The skin care cream is prepared by uniformly stirring and mixing 10 parts of water shield extract, 3.5 parts of licorice extract, 1 part of centella extract, 0.2 part of turmeric extract, 3 parts of propylene glycol, 3 parts of butanediol, 0.2 part of lauryl sodium sulfate, 0.3 part of triethanolamine, 0.3 part of hyaluronic acid, 0.6 part of hydroxymethyl cellulose, 0.6 part of glutathione, 0.15 part of carbomer, 0.15 part of allantoin, 0.15 part of essence and 76.85 parts of deionized water.
Experimental example 1
The extracts of examples 1 to 3 containing water shield extracts were tested for their antioxidant properties, i.e. their DPPH radical scavenging activity.
The activity of scavenging DPPH free radicals is determined by the following method:
1, 1-diphenyl-2-trinitrophenylhydrazine (DPPH) (15.0mg, accurate to 0.1mg) is accurately weighed, placed in a 500mL volumetric flask, and the volume is determined to 500mL by absolute ethyl alcohol to obtain DPPH solution.
1) Precisely remove DPPH solution (2mL) from the above-described water shield-containing water shield of examples 1-3Placing the extract essence (2mL) in a test tube, mixing well, standing for 35min, taking corresponding solvent (ethanol 4mL) as control sample, and measuring absorbance A at 517nm with spectrophotometer1。
2) Precisely transferring the water shield extract-containing essence (2mL) and absolute ethanol (2mL) of examples 1-3 into test tubes, mixing, standing for 35min, and measuring absorbance A at 517nm with spectrophotometer using corresponding solvent (ethanol 4mL) as control2。
3) Accurately transferring DPPH solution (2mL) and anhydrous ethanol (2mL) into test tube, mixing well, standing for 35min, determining absorbance A at 517nm with corresponding solvent (ethanol 4mL) as control sample, respectively using spectrophotometer3。
Clearance rate Y ═ 1- (A)1-A2)/A3]×100%
And (3) comparison test: replacing the water shield extract-containing essence in the steps 1) to 3) with commercially available water shield essence and commercially available licorice essence in a commercially available water shield mask respectively, and obtaining DPPH free radical scavenging rate by adopting the same method. The specific data are shown in table 1, and it can be seen from table 1 that the antioxidant performance of the essence prepared by the embodiment of the invention is obviously superior to that of commercially available water shield masks and liquorice masks.

Experimental example 2
Tyrosinase inhibitory Activity assay
1) Experimental group
Sample group, 2mL of the water shield extract-containing essence of examples 1 to 3 was added to a test tube, Phosphate Buffered Saline (PBS) having pH6.8 (1.8mL) and 0.03% L-tyrosine solution (2mL) were added, the mixture was stirred at 37 ℃ for 10min, aqueous tyrosinase solution (350units/mL, 0.2mL) was added, the mixture was incubated at 37 ℃ for 25min, and absorbance at 475nm (B) was measured with an ultraviolet-visible spectrophotometer.
Blank group, the procedure is the same as that of the sample group, and the L-tyrosine solution (B) is replaced by the phosphate buffer solution with the same volume0)。
And in the control group, the steps are the same as those of the sample group, deionized water with the same volume is used for replacing the essence containing the water shield extract, and the absorbance A at 475nm is measured by an ultraviolet-visible spectrophotometer.
The total blank group comprises the same steps as the sample group, the equal volume of deionized water is used for replacing the essence containing the water shield extract, the equal volume of phosphate buffer is used for replacing the L-tyrosine solution, and the absorbance A at 475nm is measured by an ultraviolet visible light spectrophotometer0。
Tyrosinase activity inhibition rate ═ 1- (B-B)0)/(A-A0)]×100%。
2) Comparison group: the serum in the commercially available water shield mask and the serum in the commercially available licorice mask are respectively adopted to replace the serum containing the water shield extract in the experimental group, and the tyrosinase activity inhibition rate is obtained by adopting the same method.

As is clear from the data in tables 1 and 2, the water shield extract-containing essence solutions in examples 1 to 3 of the present invention, which contain water shield extract, turmeric extract, centella asiatica extract, licorice extract, and hyaluronic acid as main components and auxiliary agents such as allantoin and glutathione, have significant advantages in terms of oxidation resistance and tyrosinase activity inhibition, as compared to commercially available water shield masks and commercially available licorice masks.
In light of the foregoing description of preferred embodiments in accordance with the invention, it is to be understood that numerous changes and modifications may be made by those skilled in the art without departing from the scope of the invention. The technical scope of the present invention is not limited to the contents of the specification, and must be determined according to the scope of the claims.
Claims (6)
1. The essence containing the water shield extract is characterized by comprising the following components in percentage by mass: 5-15% of water shield extract, 2-5% of licorice extract, 1-2% of centella asiatica extract, 0.1-0.3% of turmeric extract, 1-5% of propylene glycol, 1-5% of butanediol, 0.1-0.5% of sodium lauryl sulfate, 0.1-0.5% of triethanolamine, 0.1-0.5% of hyaluronic acid, 0.2-1.0% of hydroxymethyl cellulose, 0.2-1.0% of glutathione, 0.05-0.3% of carbomer, 0.05-0.3% of allantoin, 0.05-0.3% of essence and the balance of deionized water.
2. The brasenia schreberi extract-containing essence of claim 1, which is characterized by comprising the following components in percentage by mass: 5-15% of water shield extract, 2-5% of licorice extract, 1-2% of centella asiatica extract, 0.1-0.3% of turmeric extract, 1-5% of propylene glycol, 1-5% of butanediol, 0.1-0.5% of sodium lauryl sulfate, 0.1-0.5% of triethanolamine, 0.1-0.5% of hyaluronic acid, 0.2-1.0% of hydroxymethyl cellulose, 0.2-1.0% of glutathione, 0.05-0.3% of carbomer, 0.05-0.3% of allantoin, 0.05-0.3% of essence and 60-80% of deionized water.
3. The water shield extract-containing essence of claim 1, wherein the water shield extract is a water shield extract.
4. The method for preparing the water shield extract-containing essence of any one of claims 1 to 3, comprising the following steps: weighing water shield extract, licorice extract, centella extract, turmeric extract, propylene glycol, butanediol, sodium lauryl sulfate, triethanolamine, hyaluronic acid, hydroxymethyl cellulose, glutathione, carbomer, allantoin and essence according to the weight percentage, and then uniformly mixing with deionized water to obtain the water shield.
5. The method of claim 4, wherein the water shield extract is prepared by a method comprising the steps of:
1) washing water shield with cold water, soaking for 8-10 hr, soaking with 60-70 deg.C water for 2-4 hr, pulverizing, soaking with ethanol for 6-8 hr to obtain crude extract;
2) centrifuging the crude extract in the step 1), collecting filtrate, and removing ethanol in the filtrate to obtain the product.
6. The method according to claim 5, wherein the mass fraction of ethanol in step 1) is 65 to 75%.
