Abstract
本发明公开了一种微藻捕光蛋白HLR1基因及其应用,属于抗逆生理学领域,该基因为捕光蛋白家族基因NoHLR1,其序列如SEQIDNO:1所示,微拟球藻突变体的功能分析表明敲除该基因的突变子的耐高光能力和油脂产量得到显著提高,可用于微藻、植物和作物等的遗传改良。

技术领域
本发明涉及基因工程技术领域,具体地说,涉及微藻捕光蛋白NoHLR1基因及其应用。
背景技术
高等植物中,PSII位于类囊体膜内侧,主要功能是水的光解和放氧。PSII复合物包含25个以上不同蛋白亚基和50多种辅因子,包括PSII反应中心(主要是D1、D2蛋白)和PSII的捕光天线系统LHCII。LHCII可分为内周捕光天线和外周捕光天线,内周捕光天线(核心天线蛋白)主要为CP43和CP47,外周捕光天线主要有四类:LHCIIa(Lhcb4,又称CP29),LHCIIb(Lhcb1+2+3),LHCIIc(Lhcb5,又称CP26)和LHCIId(Lhcb6,又称CP24)。它们均含三个跨膜螺旋,分子量在22-29kDa之间。LHCIIb是外周天线的主要复合物,称为主体捕光天线(Lhcbl+2+3),距离反应中心较远。距离反应中心较近的外周天线系统是微量捕光天线(Lhcb4+5+6)。主体捕光天线主要以三聚体形式存在,微量捕光天线以单体形式存在。主体捕光天线在光合作用中主要是吸收光能,通过次要复合物CP29、CP26、CP24传递光能到内周天线CP43或CP47中,再分别传递能量到PSII反应中心。
Lhcb1、Lhcb2和Lhcb3三个基因编码LHCIIb三种不同形式的色素蛋白复合体脱辅基蛋白。每个基因同时存在多种形式,在拟南芥发现五种Lhcb1基因,通过EST标签发现其它的Lhcb1基因均可以归为上述五种Lhcb1基因之一,但Lhcb1基因的表达水平差异很大。编码的蛋白也会有所差异,Lhcb1.1、Lhcb1.2和Lhcb1.3编码的成熟蛋白是一样的,Lhcb1.4和Lhcb1.5两种蛋白差异较大,且集中在N端。Lhcb2的四种基因差异很小,Lhcb2.1、Lhcb2.2和Lhcb2.3编码的蛋白是一样的,与Lhcb2.4.编码的蛋白仅存在一个氨基酸的差异。Lhcb3在拟南芥中为单拷贝。Lhcb4、Lhcb5和Lhcb6三个基因编码LHCb4、LHCb5和LHCb6三种蛋白在体内主要以单体形式存在,且每个PSII核心至少包括一个LHCb4、LHCb5和LHCb6。在拟南芥中发现共有三个LHCb4基因,LHCb5和LHCb6均为单拷贝基因。表达水平上,LHCb4、LHCb5和LHCb6非常相似。
微藻中,除了存在上述形式的捕光蛋白外,还存在一种捕光蛋白,可以通过调节光化学淬灭(non-photochemicalquenching,NPQ),参与了细胞的强光胁迫适应过程。如衣藻Chlamydomonasreinhardtii中的胁迫相关的捕光复合体(light-harvestingcomplexstress-relatedproteins,LHCSR)和硅藻Phaeodactylumtricornutum中的非胁迫相关的捕光复合体(light-harvestingcomplexX,LHCX)。
强光引发的能量损失是微藻养殖过程中面临的重要瓶颈之一。高密度养殖的直接影响是降低了光线的透射深度,提高光照强度是增强透光性的主要手段。而高光强带来的光胁迫往往对细胞造成伤害。采用分子生物学技术,研究微藻强光响应和适应的分子机制将有助于研发与抗逆能力相关的分子标记辅助微藻或作物抗逆育种和研发栽培技术。
最近从海洋微拟球藻突变体文库中筛选到一种捕光蛋白NoHLR1基因的缺失株,并证明该突变株具备较强的高光适应能力。进一步的研究表明,NoHLR1基因受高光诱导而上调表达,除参与了微藻的光能捕获外,还参与了光合电子链和组成型能量淬灭(constitutivenon-regulatedenergydissipation,NO),对细胞适应高光具有重要的作用。
因此,研究HLR1类捕光蛋白在高光环境中的分子调控机制,尤其是其在电子传递、能量淬灭过程中的作用将有助于阐明捕光蛋白在光合作用过程中的作用机制。NoHLR1是在海洋微拟球藻Nannochloropsisoceanica中克隆的捕光蛋白,其功能和序列由申请人首次发现合成,尚未见报道。
发明内容
本发明的目的是提供一种捕光蛋白NoHLR1基因及其编码蛋白。
本发明的另一目的是提供的捕光蛋白NoHLR1基因在提高植物抗高光能力中的应用。
为了实现本发明目的,本发明从海洋微拟球藻中克隆获得捕光蛋白NoHLR1基因,NoHLR1基因的cDNA序列为:
i)SEQIDNO:1所示的核苷酸序列;或
ii)SEQIDNO:1所示的核苷酸序列经取代、缺失和/或增加一个或多个核苷酸且表达相同功能蛋白质的核苷酸序列;或
iii)在严格条件下与SEQIDNO:1所示序列杂交且表达相同功能蛋白质的核苷酸序列,所述严格条件为在含0.1%SDS的0.1×SSPE或含0.1%SDS的0.1×SSC溶液中,在65℃下杂交,并用该溶液洗膜;或
iv)与i)、ii)或iii)的核苷酸序列具有90%以上同源性且表达相同功能蛋白质的核苷酸序列。
测序结果表明,NoHLR1为捕光蛋白家族成员,NoHLR1基因开放阅读框全长为3819bp,编码由1272个氨基酸组成的蛋白(SEQIDNO:2)。通过对NoHLR1基因的生物信息学分析和受高光等胁迫时的表达模式判断,该基因可能参与微藻等的抗逆途径。
本发明还提供含有所述捕光蛋白NoHLR1基因的表达盒。
本发明还提供含有所述NoHLR1基因的RNAi或者CRISP/Cas9敲低或敲除载体。
携带有所述目的基因的RNAi或者CRISP/Cas9敲低或敲除载体可通过使用Ti质粒、植物病毒载体、直接DNA转化、微注射、电穿孔等常规生物技术方法导入植物细胞中或微藻细胞中。
本发明还提供捕光蛋白NoHLR1基因编码区插入了抗性片段的微藻突变株。
本发明还提供所述捕光蛋白NoHLR1基因在提高微藻(如微拟球藻等)抗高光能力中的应用。
本发明所述的捕光蛋白NoHLR1基因可以引用到微藻和植物遗传改良中,尤其是在提高微藻和植物抗强光能力中的应用。本发明所述的微藻包括但不限于绿藻、硅藻、红藻、金藻、褐藻等,所述的植物包括但不限于拟南芥、小麦、水稻、玉米、棉花等。
本发明还提供所述捕光蛋白NoHLR1基因在制备转基因微藻中的应用。
本发明还提供一种转基因微藻的构建方法。包括以下步骤:
(1)提取微藻的总RNA,反转录得到cDNA第一链;
(2)以cDNA第一链为模板,NoHLR1-F和NoHLR1-R为引物,通过PCR扩增反应获得捕光蛋白NoHLR1基因的cDNA序列;
(3)构建NoHLR1基因的RNAi或者CRISP/Cas9敲低或敲除载体;
(4)利用转基因技术将带有NoHLR1基因的敲低或敲除载体转化目标藻株,获得转基因稳定遗传藻株;
其中,步骤(2)所述引物F和R的序列如下:

本发明进一步提供NoHLR1基因的荧光定量PCR检测引物,引物序列如下:

可用于检测微拟球藻经过高光处理后NoHLR1基因的表达情况,结果表明,NoHLR1基因受高光处理诱导表达。
可用于检测RNAi载体转化后获得的微拟球藻工程细胞中NoHLR1基因的表达情况,结果表明,NoHLR1基因在工程细胞中的表达量下降。
本发明的有益效果:
本发明通过筛选随机插入突变体库,获得NoHLR1基因被敲出的突变子,对该基因进行功能研究,验证了微拟球藻NoHLR1基因突变子的高光耐受能力得到提高。
微拟球藻突变体的功能分析表明敲除该基因的突变子的耐高光能力和油脂产量得到显著提高,可用于微藻、植物和作物等的遗传改良。

具体实施方式
以下实施例用于说明本发明,但不用来限制本发明的范围。若未特别指明,实施例均按照常规实验条件,如Sambrook等分子克隆实验手册(SambrookJ&RussellDW,MolecularCloning:aLaboratoryManual,2001),或按照制造厂商说明书建议的条件。
实施例1捕光蛋白NoHLR1敲除的微拟球藻突变子制备
1.菌株及其培养:微拟球藻在F2固体培养基和F2液体培养基生长。
2.抗性基因标签法构建微拟球藻的饱和突变体库,采用潮霉素抗性标签、博来霉素抗性标签等。按照本领域技术人员熟悉的分子生物学技术,扩增抗性标签,采用适当的限制性内切酶切割含有筛选标记的片段,转化微拟球藻。
3.电转化微拟球藻细胞,将含有抗性标签的片段导入细胞。即0.5微克线性化的质粒和600万个藻细胞放在0.2cm电转化槽(Bio-Rad),2.2kV脉冲(Bio-Rad电转化仪,50μF)。
4.根据下述NoHLR1基因特异的引物鉴定在NoHLR1基因区域插入抗性标签的突变子细胞(以下简称hlr1)。

5.高光耐受型突变子细胞的表型鉴定。将野生型细胞和突变子细胞置于高光强下,测定两者的最大潜在光化学效率(Fv/Fm值),根据Fv/Fm值的大小确定高光耐受能力。
结果表明,hlr1株高光耐受能力提高(图1),最高可达20%。
实施例2捕光蛋白NoHLR1基因克隆
1、提取微拟球藻的总RNA,反转录得到cDNA第一链。
2、以cDNA第一链为模板,NoHLR1-F和NoHLR1-R为引物,通过PCR扩增反应获得NoHLR1基因的cDNA序列。
引物序列如下:

NoHLR1-R:5’-CTGGGATCCCCCGGGCATATGTTAAGAGAAAAGGGGAACACCG-3’
使用2×TaqPCRMasterMix(Takara)进行PCR扩增,反应体系为:cDNA模板2μL,引物F、R(10μmol/L)各1μL,2×PCRMasterMix25μL,加ddH2O至50μL。
PCR扩增程序为:94℃变性3min;94℃变性30s,55℃退火90s,72℃延伸2min,共35个循环;最后72℃延伸10min。
3、扩增产物经1%琼脂糖凝胶电泳后,切下目的片段,凝胶回收纯化,并克隆到pMD18-T载体上,经菌落PCR检测,送上海生工生物技术有限公司进行测序验证,测序验证正确的质粒命名为NoHLR1-18T。NoHLR1基因cDNA序列(SEQIDNO:1)为3819bp,编码由1272个氨基酸组成的蛋白NoHLR1(SEQIDNO:2)。
NoHLR1与衣藻Chlamydomonasreinhardtii的LHCSR和硅藻Phaeodactylumtricornutum的LHCX保守区的氨基酸序列同源性比对结果见图2。
实施例3NoHLR1基因受高光胁迫时的表达分析
采用荧光定量PCR方法检测微拟球藻经过高光处理后NoHLR1基因的表达情况。
高光处理:先将藻细胞过夜暗适应,然后放在光照培养箱进行高光处理,连续采集48小时样品。
根据NoHLR1基因序列设计如下荧光定量PCR检测引物:
NoHLR1-QF:5’-GGACCAGGTCGCCAACCTCAAAT-3’
NoHLR1-QR:5’-TCGGACTTGCCCTTGCTAAA-3’。

以Actin为内参基因。使用QuantStudio™qPCR荧光定量PCR仪进行荧光定量PCR,反应程序为:95℃预变性30s;94℃5s,60℃20s,72℃20s,进行45个循环。扩增结束后绘制熔解曲线,从50℃逐渐升温至95℃,升温速度0.2℃/s,全过程检测荧光信号。NoHLR1基因的表达量采用公式2−ΔΔCT计算,CT表示每个反应管内的荧光信号到达设定的域值时所经历的循环数。
结果表明,受高光处理后,微拟球藻NoHLR1基因诱导表达(图3)。
实施例4采用捕光蛋白NoHLR1基因被敲除的微拟球藻突变子在高光下生产油脂
利用实施例1中获得的捕光蛋白NoHLR1基因敲除的微拟球藻突变子hlr1,用于油脂生产,具体方法如下:
1、活化微拟球藻野生型和hlr1藻株,分别接种于含有新鲜F2培养基的光生物反应器中(PBR;直径33mm,高60cm)。
2、将PBR置于25°C和300μmolphotonsm-2s-1光照下培养。
3、八天后收集细胞,测定干重和总脂含量。
结果表明,高光处理下,与野生型对比,微拟球藻hlr1株的油脂含量和产量均有所提高(图4),分别提高了15%和65%。
实施例5捕光蛋白NoHLR1基因的RNAi沉默株制备
1.菌株及其培养:微拟球藻在F2固体培养基和F2液体培养基生长。
2.以实施例2中构建的NoHLR1-18T质粒为模板,NoHLR1i-F:5’-CACCACCACCACTAAACTAGTGATGGAAGCCTTGCAGGAGA-3’,NoHLR1i-R:5’-GTAAAGGGTCTGGGCAATCAA-3’,rcNoHLR1i-F:5’-TGCCCAGACCCTTTACCGCCGTGTAGGTCTGGTCTAATGT,rcNoHLR1i-R:5’-TCAGCACAAACAAACCCGCGGGATGGAAGCCTTGCAGGAGA为引物,扩增NoHLR1基因的ORF片段,扩增产物经In-fusionkit连接至表达载体,构建获得RNAi载体。
3.电转化微拟球藻细胞,将NoHLR1基因的RNAi载体的片段导入细胞。即0.5微克线性化的质粒和600万个藻细胞放在0.2cm电转化槽(Bio-Rad),2.2kV脉冲(Bio-Rad电转化仪,50μF)。
4.根据下述NoHLR1基因的荧光定量PCR检测引物鉴定在NoHLR1基因在工程细胞中的转录水平,获得NoHLR1基因转录量被敲低的工程细胞(以下简称HLR1i)。

结果表明,HLR1i株高光耐受能力提高(图1)。

Claims (10)
1. 微藻捕光蛋白NoHLR1基因,其特征在于,NoHLR1基因的cDNA序列为:
i)SEQIDNO:1所示的核苷酸序列;或
ii)SEQIDNO:1所示的核苷酸序列经取代、缺失和/或增加一个或多个核苷酸且表达相同功能蛋白质的核苷酸序列;或
iii)在严格条件下与SEQIDNO:1所示序列杂交且表达相同功能蛋白质的核苷酸序列,所述严格条件为在含0.1%SDS的0.1×SSPE或含0.1%SDS的0.1×SSC溶液中,在65℃下杂交,并用该溶液洗膜;或
iv)与i)、ii)或iii)的核苷酸序列具有90%以上同源性且表达相同功能蛋白质的核苷酸序列。
2.捕光蛋白NoHLR1基因编码的蛋白,其特征在于,其氨基酸序列如SEQIDNO:2所示。
3.含有权利要求1所述捕光蛋白NoHLR1基因的表达盒。
4.含有权利要求1所述捕光蛋白NoHLR1基因的载体。
5.含有权利要求1所述捕光蛋白NoHLR1基因的工程菌。
6.权利要求1所述捕光蛋白NoHLR1基因在微藻和植物遗传改良中的应用。
7.权利要求1所述捕光蛋白NoHLR1基因在提高微藻和植物抗强光能力中的应用。
8.权利要求1所述捕光蛋白NoHLR1基因在制备转基因植物中的应用。
9.一种转基因微藻的构建方法,其特征在于,包括以下步骤:
(1)提取微藻的总RNA,反转录得到cDNA第一链;
(2)以cDNA第一链为模板,NoHLR1-F和NoHLR1-R为引物,通过PCR扩增反应获得捕光蛋白NoHLR1基因的cDNA序列;
(3)构建NoHLR1基因的RNAi或者CRISP/Cas9敲低或敲除载体;
(4)利用转基因技术将带有NoHLR1基因的敲低或敲除载体转化目标藻株,获得转基因稳定遗传藻株;
其中,步骤(2)所述引物F和R的序列如下:
