Abstract
本发明提供水稻基因OsGRF4在提高植物耐冷性中的应用。本发明涉及的水稻基因OsGRF4为穗部性状调控基因,来源于籼稻品种川大粒,该基因表达量的升高导致水稻耐冷性的增强。实验结果表明,在冷胁迫下OsGRF4超表达转基因株系的存活率显著高于野生型,而OsGRF4抑制表达转基因株系的存活率显著低于野生型,本发明首次证明OsGRF4基因具有增强水稻耐冷性的功能。OsGRF4基因对于培育耐冷水稻品种具有较大的应用前景,本发明为植物抗寒育种,特别是水稻抗寒育种提供了新的基因资源。
Description
技术领域
本发明属于植物基因工程领域,具体地说,涉及水稻基因OsGRF4在提高植物耐冷性中的应用。
背景技术
据世界实时统计数据库公布,2019年1月,全世界的人口达到了77.15亿。按照每年1亿的增长趋势,到2050年,全球人口总数将接近100亿。据预测,到本世纪中叶,全世界的农业水平要比2005年提高60%-120%才能养活这100亿人口。水稻是最重要的粮食作物之一,全球有一半以上的人口以稻米作为主食,稻谷的产量关系到世界的粮食安全。水稻起源于热带、亚热带,是喜温植物,对低温很敏感。低温冷害严重制约着水稻的播种面积和播种范围,同时严重影响水稻的高产与稳产。仅以我国为例,每年因冷害导致稻谷减产50-100亿公斤(赵正武等,西南农业学报,2006,19:330-335)。因此,当前迫切需要挖掘更多的耐冷功能基因、培育耐冷品种以减少冷害对水稻产量造成的损失。
根据水稻的生长阶段,可以将水稻冷害分为芽期冷害、苗期冷害和生殖期冷害。水稻的耐冷性多由数量性状位点控制,目前,研究者利用籼/粳(少数为粳/粳或野/栽)遗传群体或者利用自然群体进行连锁分析或者关联分析,定位到较多的耐冷QTL(Lv et al.,Plant Cell Environ,2016,39(3):556-570;Wang et al.,Rice,2016,9(1):61)。然而,已克隆的耐冷QTL甚少,严重制约了水稻耐冷性的分子育种进程。因此,迫切需要加快水稻耐冷资源的引进、创制与评价,挖掘更多新的水稻耐冷基因,为明晰水稻耐冷分子机理和调控网络奠定基础,也为更好更快地培育耐冷性增强的水稻新品种提供基因资源和技术支持,对保障我国粮食安全有着十分重要的意义。
发明内容
本发明的目的是提供水稻基因OsGRF4在提高植物耐冷性中的应用。
为了实现本发明目的,第一方面,本发明提供水稻基因OsGRF4在提高植物耐冷性中的应用。其中,所述水稻基因OsGRF4为水稻穗部性状调控基因,来源于籼稻品种川大粒。OsGRF4为编码如下蛋白质(a)或(b)的基因:
(a)由SEQ ID NO:3所示的氨基酸序列组成的蛋白质;
(b)SEQ ID NO:3所示序列经取代、缺失或添加一个或几个氨基酸且具有同等功能的由(a)衍生的蛋白质。
可选地,所述水稻基因OsGRF4的核苷酸序列包含从下组核苷酸序列(1)-(7)中选择的核苷酸序列:
(1)SEQ ID NO:1或SEQ ID NO:2所示的核苷酸序列;
(2)在严格条件下与(1)的核苷酸杂交且编码植物耐冷相关蛋白的核苷酸序列;
(3)与(1)的核苷酸序列具有至少70%、优选至少80%、更优选至少90%、尤其是至少95%或98%或99%同一性的核苷酸序列;
(4)与(1)的核苷酸序列编码相同氨基酸序列的蛋白质、但因遗传密码子的简并性而在序列上不同的核苷酸序列;
(5)编码如下氨基酸序列之一的核苷酸序列:SEQ ID NO:3所示的氨基酸序列,或者,由于一个或多个(例如1-25个、1-20个,1-15个,1-10个,1-5个,1-3个)氨基酸残基的替代、缺失和/或插入而与SEQ ID NO:3所示的氨基酸序列不同的氨基酸序列,或者,与SEQ IDNO:3所示的氨基酸序列具有至少70%、优选至少80%、更优选至少90%、尤其是至少95%或98%同一性的氨基酸序列;
(6)(1)-(5)任一核苷酸序列的活性片段;或
(7)与(1)-(5)任一核苷酸序列互补的核苷酸序列。
其中,(2)-(7)中定义的核苷酸序列具有增强植物耐冷性的功能。
本发明中,所述严格条件可为用6×SSC,0.5%SDS的溶液,在65℃下杂交,然后用2×SSC,0.1%SDS和1×SSC,0.1%SDS各洗膜一次。
本发明中,所述植物包括但不限于水稻、玉米、高粱、小麦和大麦。
第二方面,本发明提供一种提高植物耐冷性的方法,所述方法包括:
1)使植物包含水稻基因OsGRF4;或
2)使植物表达水稻基因OsGRF4。
前述的方法,所述方法包括但不限于转基因、杂交、回交、自交或无性繁殖。
进一步地,所述方法包括利用Ti质粒、植物病毒载体、直接DNA转化、显微注射、基因枪、电导、农杆菌介导等常规生物学方法将包含水稻基因OsGRF4的重组表达载体导入植物细胞,获得转基因植株;或者,将含有水稻基因OsGRF4的植株与另一植株杂交,获得杂交后代。所获得的后代植株其耐冷性得到改良。
第三方面,本发明提供水稻基因OsGRF4的启动子,所述启动子为:
i)SEQ ID NO:4所示的核苷酸序列;
ii)SEQ ID NO:4所示的核苷酸序列经取代、缺失和/或增加一个或多个核苷酸且具有启动子活性的核苷酸序列;
iii)在严格条件下与SEQ ID NO:4所示序列杂交且具有启动子活性的核苷酸序列;
iv)与i)、ii)或iii)的核苷酸序列具有至少70%、优选至少80%、更优选至少90%、尤其是至少95%或98%或99%同一性的核苷酸序列;
v)i)~iv)任一核苷酸序列的活性片段;或
vi)与i)~iv)任一核苷酸序列互补的核苷酸序列。
本发明还提供包含所述水稻基因OsGRF4启动子的生物材料。
借由上述技术方案,本发明至少具有下列优点及有益效果:
(一)本发明首次揭示了水稻OsGRF4基因具有增强植物耐冷性的功能,OsGRF4基因对进一步阐明植物耐冷调控机理和遗传网络有着重要的意义。
(二)实验表明,在冷胁迫下OsGRF4超表达转基因水稻株系的存活率显著高于野生型,可将OsGRF4基因用于植物遗传改良,以获得耐冷性增强的植物。
(三)本发明为植物抗寒育种,特别是水稻抗寒育种提供了新的基因资源,对培育耐冷作物新品种、保障作物生产安全具有重要的应用价值。

具体实施方式
以下实施例用于说明本发明,但不用来限制本发明的范围。若未特别指明,实施例均按照常规实验条件,如Sambrook等分子克隆实验手册(Sambrook J&Russell DW,Molecular Cloning:a Laboratory Manual,2001),或按照制造厂商说明书建议的条件。
实施例1水稻基因OsGRF4增强植物耐冷性新功能的发现
本发明人最先克隆了水稻穗部性状调控基因OsGRF4,http://rice.plantbiology.msu.edu/数据库编号:LOC_Os02g47280。本发明人在后续的研究过程中,通过查阅文献发现超表达osa-MIR396c基因可以降低水稻对盐和碱胁迫的耐受性,OsGRF4作为osa-MIR396c的一个靶基因,在生理盐水、缺氧、脱落酸和赤霉素处理时水稻OsGRF4的表达量上调,而在砷酸盐处理时OsGRF4的表达量表现为下调(Gao et al.,Planta,2010,231:991–1001)。此外,Kyndt等通过基因差异表达研究发现,水稻在根结线虫侵染前后OsGRF4的表达量有显著差异(Kyndt et al.,New Phytologist,2012,196:887–900)。因此,本发明人推测OsGRF4可能与植物抗逆相关,并以水稻品种川大粒和巨穗稻为材料通过杂交、自交和回交构建了近等基因系NIL-OsGRF4与NIL-Osgrf4(Sun et al.,JIPB,2016,58:836–847),其中所述OsGRF4为突变优异等位基因,Osgrf4为功能丧失型等位基因。通过冷胁迫对NIL-OsGRF4与NIL-Osgrf4的耐冷性进行比较分析研究,结果显示冷处理后NIL-OsGRF4植株的存活率显著高于NIL-Osgrf4的存活率,增加约50%的存活率(图1),表明含有基因OsGRF4的植株明显表现出耐冷性。由此可见,水稻基因OsGRF4具有增强植物耐冷的新功能。上述冷处理方法为:50株生长7天并且健康的水稻幼苗经过8℃低温处理6天,然后恢复正常温度生长5天(白天28℃,晚上25℃),最后调查其存活率,上述每个材料设置3个重复。
上述能提高耐冷性的优异等位基因OsGRF4的cDNA序列如SEQ ID NO:1所示,其基因组核苷酸序列如SEQ ID NO:2所示,其编码蛋白质的氨基酸序列如SEQ ID NO:3所示,其启动子核苷酸序列如SEQ ID NO:4所示。
实施例2基因OsGRF4超表达载体的构建及水稻的遗传转化
超表达载体的构建:根据目的基因OsGRF4的基因组序列设计带有BamH I酶切位点的引物GRF-OX(F:5′-gcaggtcgacggatcctactacgccacgagataacgaga-3′,R:5′-gaattcccggggatcctgatgttgacggcactgttga-3′),以亲本川大粒的基因组DNA为模板扩增4073bp的OsGRF4全长DNA(不包含启动子序列),扩增产物通过琼脂糖电泳鉴定,大小符合目标之后切胶回收。然后将目的片段与全式金公司的克隆T载体(pEASY-Blunt Cloning Kit)连接:1μLPCR回收产物加1μL T载体和3μL的水,室温反应5min。将连接产物全部加入50μL的感受态细胞,轻轻混均匀,冰上放置30min。然后42℃水浴热激30s,立刻放在冰上2min,然后加入250μL的LB培养基,200rpm在37℃培养1h。然后2000rpm将LB培养基离心1min,去掉一部分上清液,保留100μL左右,轻轻悬浮菌液,全部涂于加有抗生素的LB固体培养基平板上,在37℃培养箱中过夜培养。第二天长好单个菌斑之后,分别挑取20个菌斑接种入加有LB液体培养基的试管中,37℃培养箱中培养12h,等菌液浑浊时,分别做菌液PCR,检测目标片段是否正确连入T载体,引物用T载体上的M13引物(M13F:5′-GTAAAACGACGGCCAGT-3′,M13R:5′-CAGGAAACAGCTATGAC-3′)。对菌液PCR正确的克隆进行测序,分析目标基因的序列是否有突变,序列完全正确的克隆用于后续的表达载体连接。用BamH I分别单酶切连有目标基因的T载体和由Ubiquitin启动子驱动的pCUbi1390表达载体(图2A),利用Clontech Infusion试剂盒将目的片段与线性化的载体进行融合,转化感受态细胞,挑阳性单克隆质粒并测序。
水稻遗传转化以及表型鉴定:将上述构建的序列正确的质粒转化农杆菌感受态,然后通过农杆菌介导法转化水稻材料NIL-Osgrf4。农杆菌介导的遗传转化方法主要过程:水稻材料经过愈伤组织诱导、愈伤组织继代、预培育、农杆菌培养、农杆菌侵染、愈伤组织洗涤和选择培养、愈伤组织的分化、生根、练苗、移栽,最终得到转基因水稻植株(林拥军等,作物学报,2002,28:294-300)。总共得到28株过表达转基因T0代水稻植株,对T0代转基因苗进行潮霉素基因PCR检测(F:5′-acggtgtcgtccatcacagtttgcc-3′;R:5′-ggaagtgcttgacattggggagttt-3′),发现转基因阳性单株能扩出目标条带,而转基因阴性单株、水和非转基因阴性对照均扩不出目标条带(图2B)。此外,对转基因阳性苗的OsGRF4基因进行测序分析,结果显示转基因阴性单株在目标位点的碱基为TC,而转基因阳性单株为AA和TC的杂合型双峰(图2C)。测序结果与潮霉素基因PCR检测结果完全一致,最终获得18株超表达阳性转基因单株。对OsGRF4超表达转基因水稻株系和NIL-Osgrf4植株进行冷处理,结果显示冷处理后OsGRF4超表达转基因株系的存活率显著高于野生型NIL-Osgrf4的存活率,增加约44%的存活率(图3),再次证明基因OsGRF4具有增强植物耐冷的功能。上述冷处理方法参照实施例1。
实施例3 MicroRNA干涉载体的构建及水稻遗传转化
以pNW55载体为模板利用下列引物扩增获得大小为290bp的MicroRNA片段:

IV miR*a:5′-aaTAAAACGTTGAGATCACCCTTagagaggcaaaagtgaa-3′。
将290bp的MicroRNA片段添加PstI和SpeI酶切位点,双酶切目的片段和载体pCUbi1390,然后进行连接反应,转化感受态细胞,挑阳性单克隆测序,将得到的正确克隆质粒通过农杆菌介导的水稻遗传转化体系导入到水稻材料NIL-OsGRF4中。共得到18株T0代植株,对转基因苗进行潮霉素基因PCR检测和测序分析,最后得到10株干涉阳性单株。对OsGRF4抑制表达转基因水稻株系和NIL-OsGRF4植株进行冷处理,结果显示冷处理后OsGRF4抑制表达转基因株系的存活率显著低于野生型NIL-OsGRF4的存活率,减少约44%的存活率(图4),同样证明基因OsGRF4能调节植物的耐冷性。上述农杆菌介导的水稻遗传转化参照实施例2的方法,上述冷处理方法参照实施例1。
实施例4基因OsGRF4在培育耐冷作物新品种中的应用
在培育作物新品种的过程中,可以采用转基因方法,利用包含本发明基因OsGRF4的植物表达载体转化受体作物而获得转基因作物植株,进而可以提高受体作物的耐冷性。也可以将含有本发明提供的基因OsGRF4的作物植株与另一作物植株杂交获得后代作物植株,所获得的后代作物植株其耐冷性得到改良。所述作物包括水稻、玉米、高粱、小麦和大麦等。
虽然,上文中已经用一般性说明及具体实施方案对本发明作了详尽的描述,但在本发明基础上,可以对之做一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。

Claims (8)
1.水稻基因OsGRF4在提高植物耐冷性中的应用,其特征在于,所述水稻基因OsGRF4为编码如下蛋白质(a)或(b)的基因:
(a)由SEQ ID NO:3所示的氨基酸序列组成的蛋白质;
(b)SEQ ID NO:3所示序列经取代、缺失或添加一个或几个氨基酸且具有同等功能的由(a)衍生的蛋白质。
2.根据权利要求1所述的应用,其特征在于,所述植物包括水稻、玉米、高粱、小麦和大麦。
3.一种提高植物耐冷性的方法,其特征在于,所述方法包括:
1)使植物包含水稻基因OsGRF4;或
2)使植物表达水稻基因OsGRF4;
其中,水稻基因OsGRF4的定义同权利要求1中所述。
4.根据权利要求3所述的方法,其特征在于,所述方法包括转基因、杂交、回交、自交或无性繁殖。
5.根据权利要求3所述的方法,其特征在于,所述方法包括利用Ti质粒、植物病毒载体、直接DNA转化、显微注射、基因枪、电导、农杆菌介导的方法将包含水稻基因OsGRF4的重组表达载体导入植物细胞,获得转基因植株。
6.根据权利要求3所述的方法,其特征在于,所述方法包括:将含有水稻基因OsGRF4的植株与另一植株杂交,获得杂交后代。
7.根据权利要求3-6任一项所述的方法,其特征在于,所述植物包括水稻、玉米、高粱、小麦和大麦。
8.水稻基因OsGRF4的启动子,其特征在于,所述启动子为:
i)SEQ ID NO:4所示的核苷酸序列;
ii)SEQ ID NO:4所示的核苷酸序列经取代、缺失和/或增加一个或多个核苷酸且具有启动子活性的核苷酸序列;
iii)在严格条件下与SEQ ID NO:4所示序列杂交且具有启动子活性的核苷酸序列,所述严格条件为在含0.5%SDS的6×SSC溶液中,在65℃下杂交,然后用含0.1%SDS的2×SSC溶液,含0.1%SDS的1×SSC溶液各洗膜一次;或
iv)与i)、ii)或iii)的核苷酸序列具有70%以上,优选80%以上,更优选90%以上同源性且具有启动子活性的核苷酸序列。
