Abstract
本发明公开了一种生物催化合成莱鲍迪苷M的方法,首先,利用番茄来源UDP‑糖基转移酶和马铃薯来源蔗糖合成酶,以甜菊甙为原料糖基化反应合成莱鲍迪苷E;随后,利用甜叶菊来源UDP‑糖基转移酶和马铃薯来源蔗糖合成酶,以莱鲍迪苷E为原料进一步糖基化反应合成莱鲍迪苷M。本发明的方法利用分子克隆技术,获得异源表达UDP‑糖基转移酶和蔗糖合成酶的大肠杆菌基因工程菌,经发酵产酶后以细胞粗提液直接进行催化反应,利用粗提液中UDP及额外添加的蔗糖在蔗糖合成酶分解作用下获得UDP‑葡萄糖作为糖基化反应的原材料,建立双酶循环反应体系,有效催化甜菊甙生产莱鲍迪苷M。原料成本更为低廉,工艺步骤简单,具有重要的应用价值。

技术领域
本发明属于生物工程技术领域,具体涉及一种生物催化合成莱鲍迪苷M的方法。
背景技术
甜叶菊作为一种原产于南美洲的多年本草生植物,其茎叶中可提取出丰富的甜菊糖甙,几个世纪以来,一直被南美洲人们用作为天然甜味剂[1]。自1976年我国从日本引进甜叶菊种植成功后,80年代后开始向全国推广种植。目前福建、安微、江苏等地已有大面积种植,总面积达100万亩,已成为全球最大的甜菊糖生产国和出口国[2, 3]。甜菊糖作为目前已知的最甜的甜味剂,其甜度是蔗糖的250~450倍,而热量只有蔗糖的1/300,略带有轻微涩味。其溶解度不低,且在酸和盐的溶液中能稳定存在,保存时间长、不会结块。除此之外,甜菊糖还具备对高血压、肥胖症、糖尿病等辅助药理作用[4, 5],因此,越来越受到人们的关注。
甜菊糖是甜菊糖甙的总称,目前已从甜叶菊中分离鉴定出超过35种甜菊糖甙[6]。甜菊糖甙结构围绕一个双萜甜菊醇为骨架,在C13位羟基和C19位羧基分别连接了不等的葡萄糖基团,其中,骨架上C16-C17之间的碳碳双键是提供甜菊糖甙甜味及功能的药理性基团[6, 7]。莱鲍迪苷M,于2009年报道首次从甜叶菊新品种的叶子中被提取鉴定,其甜度为蔗糖的400倍,优于现市场上在售的莱鲍迪苷A,口感也更为干净。尽管研究人员培育了高产莱鲍迪苷M的甜叶菊新品种,但其在干叶片中的含量仍低(~0.4-0.5%),单纯使用物理手段直接从叶片中提取制备高纯度的莱鲍迪苷M,经济效益是及其低下的。
专利申请CN 201310353500.9(一种酶法制备瑞鲍迪甙M的方法)与专利申请CN201410019981.4(一种酶法制备瑞鲍迪甙M的方法)中采用的是以莱鲍迪苷A或者莱鲍迪苷D为底物原料,经UDP-葡萄糖基转移酶(来自甜菊的UGT-A和/或来自水稻的UGT-B)及蔗糖合成酶偶联催化实现的,其底物莱鲍迪苷A和莱鲍迪苷D的成本均远远高于甜菊甙。
专利申请CN 201410553617.6(莱鲍迪苷M的合成方法及其中间产物和合成方法)和专利申请CN 201410314371.7(一种制备莱鲍迪苷M的工艺方法)中均涉及到部分化学试剂或化学反应,且合成步骤烦琐。本方法通过生物酶催化手段,以甜菊甙纯品为原料,搭建新的莱鲍迪苷M合成路径,经两步糖基化反应制备莱鲍迪苷M,原料成本更为低廉,工艺步骤简单,具有重要的应用价值。

发明内容
本发明的目的是针对目前制备莱鲍迪苷M经济效率低下的问题,提供一种生物催化合成莱鲍迪苷M的新路线,首先利用糖基转移酶UGTSL2糖基化作用,催化甜菊甙合成莱鲍迪苷E,随后利用糖基转移酶UGT76G1催化莱鲍迪苷E合成莱鲍迪苷M,为莱鲍迪苷M的制备及精制提供有力基础。
为了实现上述目的,本发明采用的技术方案为:
一种生物催化合成莱鲍迪苷M的方法,利用分子克隆技术,获得异源表达UDP-糖基转移酶和蔗糖合成酶的大肠杆菌基因工程菌,经自诱导发酵产酶后以细胞粗提液直接进行催化反应,无需添加UDP-葡萄糖和UDP,利用粗提液中UDP及额外添加的蔗糖,建立循环反应体系,有效催化甜菊甙生产莱鲍迪苷M。
所述自诱导发酵中诱导剂为乳糖,乳糖在发酵培养液中浓度为0.01~0.5%。
该方法具体分为两步进行,如图2所示,(1)首先,原料甜菊甙经UDP-糖基转移酶UGTSL2和蔗糖合成酶StSUS1双酶催化生成中间产物莱鲍迪苷E;(2)中间产物莱鲍迪苷E经UDP-糖基转移酶UGT76G1和蔗糖合成酶StSUS1双酶催化生成最终产物莱鲍迪苷M。
所述UDP-糖基转移酶UGT-A为番茄来源糖基转移酶UGTSL2,为NCBI ReferenceSequence: NC_015448. 3,基因序列见SEQ.No.1所示,氨基酸序列(Uniprot number:K4D508)见SEQ.No.4所示;所述UDP-糖基转移酶UGT-B为甜叶菊来源糖基转移酶UGT76G1,为NCBI Reference Sequence: AY345974. 1,基因序列见SEQ.No.2所示,氨基酸序列(Uniprot number: Q6VAB4)见SEQ.No.5所示;所述蔗糖合成酶SUS为马铃薯来源蔗糖合成酶StSUS1,为NCBI Reference Sequence: M18745,基因序列见SEQ.No.3所示,氨基酸序列(Uniprot number: P10691)见SEQ.No.6所示。
步骤1中的基因工程菌为含有UDP-糖基转移酶UGTSL2和蔗糖合成酶StSUS1共表达载体的重组大肠杆菌,UGTSL2基因片段插入在质粒载体的Nde I和Xho I酶切位点之间,StSUS1基因片段插入在质粒载体的Nco I和EcoR I酶切位点之间,最终提供了可外源共表达糖基转移酶UGTSL2和蔗糖合成酶StSUS1的重组质粒,pRSFDuet-UGTSL2-StSUS1。
步骤2中的基因工程菌为含有UDP-糖基转移酶UGT76G1和蔗糖合成酶StSUS1共表达载体的重组大肠杆菌,UGT76G1基因片段插入在质粒载体的Nde I和Xho I酶切位点之间,StSUS1基因片段插入在质粒载体的Nco I和EcoR I酶切位点之间,最终提供了可外源共表达糖基转移酶UGT76G1和蔗糖合成酶StSUS1的重组质粒,pRSFDuet-UGT76G1-StSUS1;
将重组质粒热击转化至大肠杆菌BL21(DE3)感受态细胞中,转化产物涂布于含有50mg/L的卡那霉素抗性的平板上,37℃过夜培养,获得共表达的重组大肠杆菌基因工程菌。
步骤1)和步骤2)构建得到的重组大肠杆菌基因工程菌诱导表达条件为:将重组大肠杆菌接种到LB培养基中,于16~37℃、200rpm振荡培养8~10 h,再将培养菌液按2%接种量接入TB培养基中,其中诱导剂乳糖在TB培养液中浓度为0.01~0.5%,于16~37℃诱导培养16~36 h,离心收集菌体。
所述细胞粗提液为发酵得到的菌体经超声或低温高压破碎所获得的含有UDP-糖基转移酶和蔗糖合成酶的粗酶液,粗提液中含有微量的UDP。
所述循环反应催化甜菊甙生产莱鲍迪苷M的体系为:底物甜菊甙的起始反应浓度为2~60 g/L,蔗糖与甜菊甙的质量比为1~10,由共表达UGTSL2和StSUS1的基因工程菌制备的粗酶液在反应体系中的总蛋白浓度为1~25 mg/mL,加水定容至1~100 mL,pH值为6~8,于20~50℃反应1~48 h;随后添加共表达UGT76G1和StSUS1的基因工程菌制备的总蛋白浓度为1~25 mg/mL粗酶液,继续反应1~48 h。
有益效果:
本发明采用生物酶法,通过糖基转移酶UGTSL2糖基化反应催化甜菊甙首先合成中间产物莱鲍迪苷E,再利用糖基转移酶UGT76G1转糖基作用催化莱鲍迪苷E合成莱鲍迪苷M。其中,莱鲍迪苷E经甜菊甙催化合成产率可达65%;莱鲍迪苷M经莱鲍迪苷E催化合成产率可达65%以上。

实施例1:重组大肠杆菌基因工程菌的构建
1)、共表达UGTSL2和StSUS1的重组大肠杆菌基因工程菌的获得
UGTSL2和StSUS1基因全序列由南京金斯瑞公司合成,并构建到质粒pRSFDuet-1上,其中,UGTSL2基因片段插入在质粒载体RSFDuet-1(Novagen公司)的Nde I和Xho I酶切位点之间,StSUS1基因片段插入在质粒载体pRSFDuet-1(Novagen公司)的Nco I和EcoR I酶切位点之间,最终提供了可外源共表达糖基转移酶UGTSL2和蔗糖合成酶StSUS1的重组质粒,pRSFDuet-UGTSL2-StSUS1。
添加40 uL ddH2O至重组质粒pRSFDuet-UGTSL2-StSUS1干粉中,充分溶解,取5 ul热击转化至大肠杆菌BL21(DE3)感受态细胞中,转化产物涂布于含有50 mg/L的卡那霉素抗性的平板上,37℃过夜培养,获得重组大肠杆菌基因工程菌。
2)、共表达UGT76G1和StSUS1的重组大肠杆菌基因工程菌的获得
UGT76G1和StSUS1基因全序列由南京金斯瑞公司合成,并构建到质粒pRSFDuet-1上,其中,UGT76G1基因片段插入在质粒载体pRSFDuet-1(Novagen公司)的Nde I和Xho I酶切位点之间,StSUS1基因片段插入在质粒载体pRSFDuet-1(Novagen公司)的Nco I和EcoR I酶切位点之间,最终提供了可外源共表达糖基转移酶UGT76G1和蔗糖合成酶StSUS1的重组质粒,pRSFDuet-UGT76G1-StSUS1。

实施例2:基因工程菌双酶的共表达
将含有质粒pRSFDuet-UGTSL2-StSUS1(或pRSFDuet-UGT76G1-StSUS1)的重组基因工程菌接种到含有50 mg/L卡那霉素抗性的LB培养基(0.5 g/L酵母粉,1 g/L氯化钠,1 g/L胰蛋白胨),于37℃、200 rpm振荡过夜培养,再将培养菌按2%接种量接入含有100 mL TB培养基(2.5 g/L酵母粉,1 g/L氯化钠,1.5 g/L胰蛋白胨,0.2 g/L葡萄糖,0.05 g/L乳糖)的500mL摇瓶中,于200 rpm,37℃培养2 h,随后转移至25℃继续培养24 h,离心收集菌体。超声破碎菌体,离心取上清液即为细胞粗提液,置于4℃保存待用。
实施例3:酶法催化甜菊甙合成莱鲍迪苷E
1)、催化反应体系
添加20 g/L甜菊甙、60 g/L蔗糖、3 mM Mg2+及适量共表达UGTSL2和StSUS1的细胞粗提液(~6 mg/mL总蛋白)于50 mM磷酸钾缓冲(pH 7.2)中,总体积定量20 mL。30℃,200 rpm反应24 h,定时取样500 uL,置于95℃水浴加热15 min,12000 rpm室温离心1 min,分离上清液至新1.5 mL EP管中,4℃保存,待HPCL检测。就结果来看(表1),糖基转移酶UGTSL2偶联蔗糖合成酶StSUS1催化甜菊甙合成莱鲍迪苷E是可行的,且于反应24 h时,甜菊甙的转化率已达到94.12%,莱鲍迪苷E的产率为66.32%。
2)、PHLC检测方法
色谱柱:Agilent TC-C18(4.6*250mm,5um,120A);紫外检测波长:210 nm;柱温:55℃;流动相A为含0.1%甲酸的乙腈,流动相B为含0.1%甲酸的娃哈哈水,液相检测条件:0.000min (25% A, 75% B), 15.000 min (47% A, 53% B), 20.000 min (100% A, 0% B),20.010-25.000 min (100% A, 0% B), 25.010 min (25% A, 75% B), 25.010-30.000min (25% A, 75% B);样品进样10 uL,流速1 mL/min。

实施例4:酶法催化莱鲍迪苷E合成莱鲍迪苷M
1)、催化反应体系
添加18 g/L蔗糖及适量共表达UGTSL2和StSUS1的细胞粗提液(~5 mg/mL总蛋白)于含有3 g/L或6 g/L莱鲍迪苷E的实施例3所述的终止反应后的溶液中。30℃,200 rpm反应24h,定时取样500 uL,置于95℃水浴加热15 min,12000 rpm室温离心1 min,分离上清液至新1.5 mL EP管中,4℃保存,待HPCL检测。就结果来看(表2),不同底物浓度下,糖基转移酶UGT76G1偶联蔗糖合成酶StSUS1催化莱鲍迪苷E合成莱鲍迪苷M反应12 h时,莱鲍迪苷E的转化率均已达到90%以上,24h时,莱鲍迪苷E已经全部转化完全。催化3 g/L莱鲍迪苷E 24h时,莱鲍迪苷M的产率可达到80%;催化6 g/L莱鲍迪苷E 24h时,莱鲍迪苷M的产率为36%。
2)、PHLC检测方法

上述反应12h后,在反应液中直接继续添加20 mL共表达UGT76G1和StSUS1的细胞粗提液(~6 mg/mL总蛋白),于30℃,200 rpm继续反应12 h,定时取样500 uL,置于95℃水浴加热15 min,12000 rpm室温离心1 min,分离上清液至新1.5 mL EP管中,4℃保存,待HPCL检测。
结果见表3,甜菊甙经糖基转移酶和蔗糖合成酶共同催化作用下合成莱鲍迪苷M是可行的,该两步法中,第一步甜菊甙经UGTSL2和StSUS1催化作用下转化可达到95%,中间产物莱鲍迪苷E的产率达到65%;第二步中间产物莱鲍迪苷E经UGT76G1和StSUS1催化作用下转化率达90%以上,产物莱鲍迪苷M的产率为67%。最终,以10 g/L甜菊甙为起始原料酶法合成莱鲍迪苷M实验中,莱鲍迪苷M的产率为43.5%。

色谱柱:Agilent TC-C18(4.6*250mm,5um,120A);紫外检测波长:210 nm;柱温:55℃;流动相A为含0.1%甲酸的乙腈,流动相B为含0.1%甲酸的娃哈哈水,液相检测条件:0.000min (25% A, 75% B), 15.000 min (47% A, 53% B), 20.000 min (100% A, 0% B),20.010-25.000 min (100% A, 0% B), 25.010 min (25% A, 75% B), 25.010-30.000min (25% A, 75% B);样品进样10 uL,流速1 mL/min。

Claims (10)
1.一种生物催化合成莱鲍迪苷M的方法,其特征在于,利用分子克隆技术,获得异源表达UDP-糖基转移酶和蔗糖合成酶的基因工程菌,经自诱导发酵产酶后以细胞粗提液直接进行催化反应,利用粗提液中本身含有的微量UDP及额外添加的蔗糖,建立双酶循环反应体系,有效催化甜菊甙生产莱鲍迪苷M。
2.根据权利要求1所述的一种生物催化合成莱鲍迪苷M的方法,其特征在于,催化甜菊甙生产莱鲍迪苷M包括如下两步骤:
(1)UDP-糖基转移酶UGT-A和蔗糖合成酶SUS共表达双酶催化甜菊甙合成中间产物莱鲍迪苷E;
(2)UDP-糖基转移酶UGT-B和蔗糖合成酶SUS共表达双酶催化莱鲍迪苷E合成最终产物莱鲍迪苷M。
3.根据权利要求2所述一种生物催化合成莱鲍迪苷M的方法,其特征在于,所述UDP-糖基转移酶UGT-A为番茄来源糖基转移酶UGTSL2,基因序列见SEQ.No.1所示,氨基酸序列见SEQ.No.4所示;所述UDP-糖基转移酶UGT-B为甜叶菊来源糖基转移酶UGT76G1,基因序列见SEQ.No.2所示,氨基酸序列见SEQ.No.5所示;所述蔗糖合成酶SUS为马铃薯来源蔗糖合成酶StSUS1,基因序列见SEQ.No.3所示,氨基酸序列见SEQ.No.6所示。
4.根据权利要求1所述的一种生物催化合成莱鲍迪苷M的方法,其特征在于,所述细胞粗提液为发酵得到的菌体经超声或低温高压破碎所获得的含有UDP-糖基转移酶和蔗糖合成酶的粗酶液。
5.根据权利要求1所述的一种生物催化合成莱鲍迪苷M的方法,其特征在于,所述循环反应体系为: UDP-葡萄糖与甜菊甙经UGTSL2和StSUS1催化生成莱鲍迪苷E; UDP-葡萄糖与莱鲍迪苷E经UGT76G1和StSUS1催化生成莱鲍迪苷M。
6.根据权利要求1所述的一种生物催化合成莱鲍迪苷M的方法,其特征在于,所述基因工程菌诱导表达条件为:将重组菌接种到LB培养基中,于16~37℃、200rpm振荡培养8~10 h,再将培养菌液接入TB培养基中,其中诱导剂在TB培养液中浓度为0.01~0.5%,于16~37℃诱导培养16~36 h,离心收集菌体。
7.根据权利要求2所述的一种生物催化合成莱鲍迪苷M的方法,其特征在于,所述的两步催化法为:在含有底物甜菊糖、蔗糖的反应液中,首先添加共表达UGTSL2和StSUS1的粗酶液,催化合成中间产物莱鲍迪苷E;反应一段时间后再向其中添加共表达UGT76G1和StSUS1的粗酶液,直接催化莱鲍迪苷E合成莱鲍迪苷M。
8.根据权利要求2所述的一种生物催化合成莱鲍迪苷M的方法,其特征在于,步骤1)中,底物甜菊甙的起始反应浓度为2~60 g/L,蔗糖与甜菊甙的质量比为1~10,两种粗酶液在反应体系中的总蛋白浓度均为1~25 mg/mL。
9.根据权利要求2所述的一种生物催化合成莱鲍迪苷M的方法,其特征在于,所述催化反应采用水相反应体系,水溶液pH值为6~8。
10.根据权利要求2所述的一种生物催化合成莱鲍迪苷M的方法,其特征在于,所述催化反应的反应温度为20~50℃,前后两阶段反应时间均为1~48 h。
