第一和第二种溶液可以与任何适当的染色剂相容,包括有机染料和量子点,因为丙烯酸介质由于脱水过程而损坏有机染料,而水性介质淬灭量子点。换句话说,第一和第二种溶液不会显著降解或影响任何染色剂,无论是否至少部分地归因于第一和第二种溶液的成分或加入第一和第二种溶液的方法。例如,当前述冷冻时,染色剂在与本文公开的溶液接触24小时后可保持至少75%的发射,或者在与本文公开的溶液接触72小时后可保留至少50%的发射。与后续发射相比较的100%的发射是当样品在没有溶液的情况下成像时相应染色剂的发射。
方法
为了获得样品,可以直接从受试者取出样品,或者可以从悬浮液中富集和/或分离样品。可以通过任何适当的富集过程富集样品,包括但不限于基于密度的分离,顺序密度分级分离,磁激活细胞分选,荧光激活细胞分选,差异裂解,耗尽过滤器等。顺序密度分级分离是通过逐步或顺序过程将悬浮液分成级分或将一部分悬浮液分成亚级分的过程,使得每个步骤或顺序导致不同级分或亚级分从上述和随后步骤或顺序中采集或分离。通过用拾取装置例如细胞挑选器、移液器、注射器等选择样品可以从其他悬浮液成分中获得样品。
在获得样品后,可以将样品置于基底上或基底中。此外,可以将样品粘附到基底上,例如用粘附溶液。例如,可以将样品重新悬浮在容器中的粘附溶液中。或者,可以将粘附溶液添加到样品中或与样品混合。将包括至少一部分粘附溶液的重新悬浮样品通过分配器(例如移液器或重复移液器)分配到基底上或分配到基底中,并铺展在基底上。然后,如果适合,可以将样品通过涂布器例如刮板、移液器尖端、刀片包括刀片和基部的两件式涂布器铺展在基底上。或者,可以通过离心、润湿或下垂基底将样品铺展在基底上。将重新悬浮的样品固化以将重新悬浮的样品粘附到基底上。或者,可以将重新悬浮的样品分配到基底上并固化而不铺展在基底上。固化可以在空气中进行,例如在室温下进行;在环境控制室中进行,例如在37℃等。此外,在完成固化步骤之后,样品可以经历额外的固定步骤,例如在福尔马林或任何适合的固定剂中。然后可以对样品进行染色,例如在自动染色器内或通过手动染色。
目标分析物和非目标分析物可具有位于表面上或分析物内的许多不同类型的受体分子。每种类型的受体是能够连接特定连接分子的分子。结果,连接分子能够用于对目标分析物和/或非目标分析物进行分类,并通过将连接到具有一种特殊荧光探针的特定受体的连接分子与之结合来确定悬浮液中存在的目标分析物和/或非目标分析物的特定类型。可以基于各种标记物对分析物进行分类,包括核标记物,细胞内标记物,细胞外标记物(例如膜标记物)和排除标记物。
染色剂是用于标记包括荧光探针和连接分子的生物材料的化合物。荧光探针是在经受光或电磁辐射激发后发光的化合物。荧光探针可以用作示踪剂,用作某些结构的标记,用作生物标记物的标记等。荧光探针可以是分布式的,或者可以通过以下方式标记合适的结构或生物标记物,包括但不限于摄取,选择性摄取,扩散和与连接分子的连接。连接分子可以是结合抗原的抗体,生物标记物,化学引诱物分子,蛋白质等。在适当的情况下,连接分子可以允许荧光探针与目标分析物或甚至非目标分析物的缀合。键和相关的吸引力可以是共价的或非共价的,包括键合,例如离子,偶极-偶极相互作用,伦敦分散力,范德华力,疏水相互作用和氢键。例如,荧光探针可以连接至EpCAM抗体,从而形成EpCAM抗体-荧光探针复合物。当EpCAM抗体-荧光探针复合物与EpCAM生物标记物一起被导入目标材料时,EpCAM生物标记物和EpCAM抗体可以彼此吸引和结合,从而将荧光探针连接到目标材料上。在另一个实例中,荧光探针可以连接到抗生物素蛋白,例如链霉抗生物素蛋白或中性抗生物素蛋白,从而形成抗生物素蛋白-荧光探针复合物。当将抗生物素蛋白-荧光探针复合物引入外表面上具有生物素分子的目标材料时,生物素和抗生物素蛋白彼此吸引并结合,从而将荧光探针连接到目标材料上。
包括至少两种染色剂并且可以包括至多4、6、8、10或以上种的染色剂组可以用于多重目标材料,使得每种染色剂结合目标材料上或内的不同抗原。例如,当施加适当的刺激(例如具有较短波长的光)时,每种类型的荧光探针发射电磁波谱的窄波长范围内的光。发射具有绿色通道中的波长的光的第一荧光探针可以连接至第一连接分子上,该第一连接分子特异性地结合第一类型的受体,而发射具有红色通道中的波长的光的第二荧光探针可以连接至第二连接分子,该第二连接分子特异性地结合第二类型的受体。作为刺激相应荧光探针的结果,基于所发射的波长的观察到的颜色鉴定受体的类型,并且因为受体对于特定目标分析物和/或非目标分析物是独特的,所以颜色也能够用于鉴定目标颗粒。这允许在任何给定时间检测目标材料上的多个受体。

Claims (20)
