技术领域
本发明属微生物领域,具体是涉及一株具有缓解便秘功效的鼠李糖乳杆菌及其应用。
背景技术
鼠李糖乳杆菌是近年来国外研究较多的一种益生乳酸细菌,其广泛存在于传统的发酵乳制品和人体胃肠道中。已有大量资料表明,鼠李糖乳杆菌在益生菌领域有很广阔的开发及应用前景。目前研究结果已经证实鼠李糖乳杆菌具有以下益生功能:(1)抗菌作用。报道称,鼠李糖乳杆菌对食物及肠道中的一些致病菌具有抑制作用,其发酵所产生的抑菌物质对革兰氏阳性、阴性菌及多种致病菌均有抑制作用;(2)抗氧化作用。研究表明,鼠李糖乳杆菌的菌悬液、无细胞提取液及胞外多糖均具有较强的抗氧化活性;(3)免疫调节作用。鼠李糖乳杆菌能够增强小鼠肠道及系统的免疫应答,并且对小鼠细胞的迟发型变态反应、脾淋巴细胞增值能力以及NK细胞活性也具有显著效果;(4)治疗肠道疾病。

本发明的目的是提供一株具有缓解便秘功效的鼠李糖乳杆菌。

一株具有缓解便秘功效的鼠李糖乳杆菌及其应用,菌株为鼠李糖乳杆菌Lactobacillus rhamnosus NCU2217已于2018年4月9日保藏在中国微生物菌种保藏管理委员会普通微生物中心,地址:北京市朝阳区北辰西路1号院3号,中国科学院微生物研究所,其简称为CGMCC,保藏编号为CGMCC NO15573。一株具有缓解便秘功效的鼠李糖乳杆菌及其应用:所述的鼠李糖乳杆菌NCU2217为活菌或灭活菌或该菌株的衍生物。
一株具有缓解便秘功效的鼠李糖乳杆菌及其应用:所述应用为应用于食品领域,所述的食品可以是发酵果蔬、发酵乳、乳酪、含乳饮料、乳粉或其他含有该菌株或该菌株衍生物的食品中的任何一种。
一株具有缓解便秘功效的鼠李糖乳杆菌及其应用:本发明所提供的菌株及其组合物具有缓解便秘的功效,具体包括:对洛哌丁胺诱导的便秘小鼠的粪便指标具有显著的缓解作用,可明显提高便秘小鼠的肠道蠕动水平,并促进结肠上皮细胞与黏膜损伤的修复。
一株具有缓解便秘功效的鼠李糖乳杆菌及其应用:所述应用为:鼠李糖乳杆菌以食品的形式组成组合物,发酵大宗果蔬益生菌发酵果蔬产品,包括:泡菜、饮料、浆料,无需添加香精香料、色素、酸味剂、稳定剂、防腐剂食品添加剂,所有风味由益生菌发酵果蔬原料产生。本发明的有益效果:
鼠李糖乳杆菌CGMCC NO15573能够加快小肠蠕动并具有部分通便效果,便秘小鼠在摄入鼠李糖乳杆菌CGMCC NO15573后,乙酸、丙酸、正丁酸和总短链脂肪酸水平均有明显提高,短链脂肪酸总体含量增加能够降低肠道环境的pH值,结肠组织染色切片结果显示鼠李糖乳杆菌CGMCC NO15573具有修复结肠粘膜损伤的特性。

具体实施方式
实验例
下面结合附图和实施例、测试数据,对本发明进行具体描述:
选取造模成功的40只小鼠随机分成模型组(Constipation)、鼠李糖乳杆菌CGMCCNO15573低剂量组(1.0×107CFU/mL)、鼠李糖乳杆菌CGMCC NO15573中剂量组(1.0×108CFU/mL)组和鼠李糖乳杆菌CGMCC NO15573高剂量组(1.0×109CFU/mL),按照10mL/kg的剂量给药,正常组(Normal)和模型组灌胃同剂量生理盐水,连续灌胃15天,实验过程中,称量体重,并测定采食量、饮水量,第15天给药后,6h内观察粪便性状,收集粪便,计数并称重,收集的粪便置于真空冷冻干燥机内冻干,计算粪便含水量,(计算公式:含水量(%)=[粪便湿重(g)–粪便干重(g)]÷粪便湿重(g)×100)最后一次给药24h后,灌胃墨汁,30min后脱锥处死,打开腹腔,分离小肠肠管,将小肠在无张力情况下拉成直线进行测量肠管长度与墨汁推进长度。墨汁推进率计算公式:墨汁推进率(%)=墨汁推进长度(cm)÷小肠总长度(cm)×10 0另外,取结肠组织并置于1 0%福尔马林固定液中固定,采用苏木素伊红(hematoxylinand eosin,H&E)染色进行病理学观察,实验结果见表1、2和说明书附图1。

结果显示,鼠李糖乳杆菌CGMCC NO15573各组小肠墨汁推进长度能够比便秘模型组至少快47%,提示鼠李糖乳杆菌CGMCC NO15573能够加快小肠蠕动并具有部分通便效果,此外,实验结果还显示便秘小鼠在摄入鼠李糖乳杆菌CGMCC NO15573后,乙酸、丙酸、正丁酸和总短链脂肪酸水平均有明显提高,短链脂肪酸总体含量增加能够降低肠道环境的pH值。此外,结肠组织染色切片结果还显示鼠李糖乳杆菌CGMCC NO15573具有修复结肠粘膜损伤的特性。
