Abstract
本发明属于基因工程技术领域,具体涉及水稻矮化脆杆突变体DBC1的调控基因及其用途。本发明通过对水稻矮化脆杆突变体DBC1进行克隆、测序,挖掘出水稻矮化脆杆的调控基因,并实验验证了所述基因具有调控水稻矮化脆杆的作用,为水稻转基因研究提供了有力的工具,可促进高产优质水稻的育种研究。
Description
技术领域
本发明属于基因工程技术领域,具体涉及水稻矮化脆杆突变体DBC1的调控基因及其用途。
背景技术
水稻是我国最重要的粮食作物,约一半以上的人口以其为主食;同时,水稻因基因组较小、具有较高的同源性、是最早测序的禾谷类粮食作物,使其成为了单子叶植物分子生物学研究的模式植物。“绿色革命”和杂交水稻的应用极大提高了水稻单量,对解决我国粮食安全产生了重要意义。秸秆还田是当今世界上普遍重视的一项培肥地力的增产措施,在杜绝了秸秆焚烧所造成的大气污染的同时还有增肥增产作用。此外,秸秆还田能增加土壤有机质,改良土壤结构,使土壤疏松,孔隙度增加,容量减轻,促进微生物活力和作物根系的发育。秸秆还田增肥增产作用显著,一般可增产5%-10%。由于秸秆有韧度,粉碎还田不是件易事,秸秆处理一直是棘手问题。脆杆突变体茎秆适度变脆,有利于秸秆还田,是解决这一矛盾的重要技术措施之一。水稻脆茎突变体一般表现为细胞壁中纤维素和木质素的含量变化,它们常常会改变茎秆机械强度,进而影响水稻抗倒伏性和环境适应性。因此,鉴定茎秆适度变脆、农艺性状不受影响或受影响较小的突变体,对其进行分子水平研究,不仅有助于阐明水稻细胞壁的生物合成机理,而且对选育“脆而不倒”的环保型水稻新品种具有重要的意义。
发明内容
本发明以水稻矮化脆杆突变体为研究对象,通过对水稻矮化脆杆突变体DBC1进行克隆、测序,挖掘出水稻矮化脆杆的调控基因,并实验验证了所述基因具有调控水稻矮化脆杆的作用。
具体的,本发明包括几个方面:
本发明的一个方面涉及一种水稻矮化脆杆突变体DBC1的调控基因,具有如SEQ IDNo.1所示的核苷酸序列,编码的蛋白质具有如SEQ ID No.2所示的氨基酸序列。
本发明的另一个方面涉及上述水稻矮化脆杆突变体DBC1的调控基因在环保型水稻品种选育中的用途。
本发明的再一方面涉及一种环保型水稻品种选育方法,所述方法包括将上述水稻矮化脆杆突变体DBC1的调控基因转化入水稻。
本发明的再一方面涉及一种调控水稻株高的试剂,其主要活性成分为上述水稻矮化脆杆突变体DBC1的调控基因编码的蛋白质、包含上述水稻矮化脆杆突变体DBC1的调控基因的载体、或包含上述水稻矮化脆杆突变体DBC1的调控基因的宿主细胞。
本发明的有益效果为:通过对水稻矮化脆杆突变体DBC1进行克隆、测序,挖掘出水稻矮化脆杆的调控基因,为水稻转基因研究提供了有力的工具,可促进高产优质水稻的育种研究。经过实验验证,该基因具有调控水稻细胞分裂的作用,为改良水稻株型提供了方向。

具体实施方式
下面通过具体实施例对本发明进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明的一部分实施例,而不是全部的实施例。基于本实用发明的实施例,本领域普通技术人员所获得的所有其他实施例,都属于本发明的保护范围。
实施例中未注明具体条件的实验方法,通常按照常规条件,例如分子克隆实验指南(第三版,J.萨姆布鲁克等著,黄培堂等译,科学出版社,2002年)中所述的条件,或按照制造厂商所建议的条件。
实施例中使用的材料:野生型缙恢10号和水稻矮化脆杆突变体DBC1由西南大学水稻研究所提供;M-MLV逆转录酶、高保真DNA聚合酶PFU、T4DNA连接酶、Trizol试剂盒、DNA凝胶回收试剂盒、质粒提取试剂盒购自TaKaRa公司;氨苄青霉素(Ampicillin,Amp)为Sigma公司产品;引物合成和DNA测序由上海英俊生物技术有限公司完成;其它化学试剂购自北京鼎国生物技术有限责任公司;大肠杆菌DH5α由西南大学水稻研究所提供;表达载体PTCK303均由西南大学水稻所提供。
在前期研究工作中,西南大学水稻研究所提供了:利用EMS诱变优良恢复系缙恢10号获得了一个遗传稳定的水稻矮化脆杆突变体(命名为DBC1),表现为株高相比野生型明显矮化,脆性明显增加,达显著差异水平,见图1A-F,其中,1E中,PL代表穗长,NL代表穗颈长,FL代表第1节间长,SL代表第2节间长,Bar=5cm,“*”表示在0.05水平上差异显著,“**”表示在0.01水平上差异显著。用表型正常的不育系中花11与突变体DBC1杂交,F1代表型正常,说明该突变体受隐性基因控制。F2代群体中出现明显的分离,分别表现双亲性状。经卡方测验,正常株﹕突变株符合3∶1分离比,表明DBC1突变体受隐性单基因控制。通过石蜡切片和电子显微镜(参照博士学位论文:水稻维管束发育相关基因AVB基因的图位克隆和功能分析,2016年,33-34页)进行细胞学观察发现,细胞的大小与野生型相比无显著差异,即通过影响细胞数目从而引起株高矮化,见图2A-F,其中2A-2D中,Bar=100μm,2E-2F中,n=50。另外,该基因影响了茎杆中维管束的结构,木质部相对面积增加,见图2G-J,其中,2G-J种,Bar=200μm,2I-J中,Bar=50μm。以中花11/DBC1杂交的F2群体作为定位群体,共获得600个突变株,用于基因定位。选用400对均匀分布于12条染色体上的SSR标记对F2代定位群体突变单株进行单株验证(参照硕士学位论文:水稻叶缘白化突变体mal的遗传分析与基因定位,2014年,22页),结果将连锁定位于2染色体上的Ind标记Indel02-2和Indel02-7与DBC1突变位点表现连锁。在两标记间进一步设计Indel引物,其中Indel02-1-Indel02-7、在两亲本间都表现出多态性,引物序列祥见表1,最终将DBC1定位在Indel标记Indel02-4和Indel02-5之间,物理距离为86.7kb,见图3A。

实施1:水稻矮化脆杆突变基因DBC1的克隆、测序及功能验证
1、水稻矮化脆杆基因DBC1的克隆及测序
在前期对突变基因DBC1精细定位的基础上,本发明通过在线基因预测(http://mendel.cs.rhul.ac.uk)、BLAST在线比对(http://blast.ncbi.nlm.nih.gov/)及基因超表达功能分析。通过图位克隆,序列比对发现,该基因是由于基因LOC_Os02g50550的第5内含子的第5碱基由正常的C突变为T。由于基因转录后需要进行修饰、剪切和拼接,即内含子突变可能会影响前体信使RNA的剪切编辑。因此,在cDNA上设计一对相距550bp左右的引物P1,P2,包含突变位点,见图3B,其中灰色盒子代表外显子,黑色连线代表内含子,下划线标示第五外显子,红色碱基代表突变位点。PCR扩增片段经凝胶电泳显示,突变体DBC1的条带相对低于野生型,可能有大片段的缺失,见图3C-D。克隆基因DBC1转入表达载体pMD19-T,挑斑,检测,送阳性菌液测序。比对结果显示,突变体相较于野生型有62bp的缺失,该片段的缺失正好位于第五外显子处,见图3E。即,突变体DBC1第5内含子的单碱基突变导致第5外显子的整体缺失。对野生型和突变体的氨基酸序列比对发现,该基因突变导致GTP结构域发生变化。
2、水稻矮脆基因DBC1的功能验证
通过实时荧光定量qPCR对野生型和突变体的LOC_Os02g50550进行表达量分析发现,(参照博士学位论文:水稻小穗发育相关基因MFS1的图位克隆与功能分析,2013年,29页)突变体DBC1中LOC_Os02g50550基因的表达量相对野生型明显下调,且LOC_Os02g50550在水稻各个部位均有表达,在植株茎秆和顶端分生组织中表达最高,见图4。然后,本发明构建了以野生型缙恢的LOC_Os02g50550基因的cDNA为模板,扩增的片段连接到表达载体pTCK303上,构建pTCK303-DBC1OE基因超表达载体,通过农杆菌介导遗传转化到突变体DBC1中。观察转基因株系表型发现,它们的表型与野生型相似,基本恢复正常。初步确定了LOC_Os02g50550为水稻矮化脆性突变基因DBC1的候选野生型基因,见图5A-C。
实施例2:水稻矮脆突变基因DBC1的激素响应实验分析
1)将突变体和野生型种子催芽,选取露白一致的种子分别播在含有0、10-10、10-8、10-6和10-4μM赤霉素(GA)的营养液中,放置于恒温培养箱中培养。每天分别添加等量的营养液和含相应浓度赤霉素的营养液。每隔24小时测量统计第二叶鞘的长度变化,至两叶一心期。并在一叶一心期(约第7天)取各处理下的叶片,提取RNA,并反转录成cDNA,保存备用。结果发现,当赤霉素浓度低于10-8μM时,野生型和突变体第二叶叶鞘的生长与空白对照组几乎无差异;当浓度上升至10-6μM,植株开始迅速增长,野生型和突变体叶鞘长度差异增大;随着浓度继续上升,野生型和突变体的第二叶叶鞘长度差也逐之增大,见图6A-C。
2)定量PCR分析
植物的生长与激素合成代谢密不可分。取一叶一心期野生型和突变体的地上部分,提取RNA,反转录成cDNA第一链,利用定量PCR分析水稻中参与赤霉素生物合成基因GA2ox-1、GA2ox-3、GA20ox-1、GA20ox-2/SD1、GA3ox-2/D18、氨甲酰磷酸合成酶(CPS1),和信号传导通路基因MYB61、NAC29、NAC31、SLR1和GID1在野生型和突变体中的表达差异。同时,检测不同浓度处理下的野生型和突变体中赤霉素灭活基因GA2ox-3的表达情况。定量引物序列见表2。结果显示:MYB61、GID1和NAC29在突变体中的表达量降低,SLR1和NAC31表达量升高,见图6D。另外,突变体DBC1中的GA2ox-3表达量显著高于野生型,除此之外,GA20ox-1和GA20ox-2/SD1在突变体中的表达量显著低于野生型,见图6E,其中,ACTIN基因作为内参基因。GA2ox-3是赤霉素灭活基因,外施赤霉素或者内源赤霉素含量增加,GA2ox-3的表达会随之升高。因此,用水和不同浓度的赤霉素处理露白一致的野生型和突变体种子,并在光照、温度及湿度适合的条件下培养。分析不同浓度下GA2ox-3在两者之间的表达差异及表达趋势。由图6F可知,与赤霉素响应实验结果趋势一致,在浓度为10-6μM时,表达量开始出现差异。随浓度上升,GA2ox-3在野生型和突变体中的表达都随之升高,且在突变体中的表达上调更快。这可能解释了外源赤霉素增加,株高变化缓慢的现象。

实施例3:突变体DBC1的细胞分裂速度实验
石蜡切片分析根尖生长点细胞,相同细胞数目下,DBC1中双核单细胞的平均数量是7.4个,野生型中为4.1个,呈显著水平,见图7A-E。这暗示着突变体细胞分裂速度减慢。RT-PCR分析野生型、突变体以及超表达株系OE-1中细胞周期蛋白相关基因的表达变化,RT-PCR定量引物序列见表3,结果表明,突变体中的相关周期蛋白基因的表达量相比野生型降低;在转基因株系中CycB,CycD,CycP和CycT的表达都有所恢复,见图7F。这就进一步说明了突变体DBC1的细胞分裂速度减缓。即,该基因的突变引起细胞分裂速度减缓。
