本发明公开了一种轻量化的空心胶囊,胶囊壁的密度为0.2~1.0g/cm3。本发明的空心胶囊的胶囊壁的密度比市售的胶囊的密度低,但胶囊的其他性能却没有大幅的变化,满足使用性能的同时,方便胶囊的运输,节约成本。

技术领域
本发明涉及空心胶囊技术领域,具体涉及一种轻量化的空心胶囊。
背景技术
空心胶囊在医药上指用特种成膜材料(如明胶、纤维素、多糖等)制成的囊状物,把内容物(如粉状、液体状各类药物等)装入其中,便于吞服。用胶囊装的药物,一般都是对食道和胃粘膜有刺激性的粉末或颗粒,或口感不好、易于挥发、在口腔中易被唾液分解,以及易吸入气管的药。这些药装入胶囊,既保护了药物药性不被破坏,也保护了消化器官和呼吸道。去掉胶囊壳可能会造成药物流失、药物浪费、药效降低。
空心胶囊作为特定物品的包装材料,虽然具有上述的保护胶囊内容物等功能,但其本身并没有特别的药效,并且胶囊制备原料必须达到食品级和药品级。但是胶囊的原料及其生产过程中可能含有杂质或受到污染,因此会存在危害人体健康的风险。为此,需要一种在保证空心胶囊使用效果的情况下,减少空心胶囊质量,从而降低胶囊带来的风险,同时,也能够降低胶囊的运输成本和能源消耗。
发明内容
为此,本发明提供了一种空心胶囊,其胶囊壁的密度为0.2~1.0g/cm3。所述的胶囊壁的密度是指胶囊壁的表观密度。相比市售胶囊,本发明的胶囊具有密度更小,质量更轻,在体内降解速度更快,药物见效更快等特点。当胶囊壁的密度小于0.2g/cm3,胶囊的强度(比如脆碎度)不足,影响胶囊的填装和胶囊的运输,降低胶囊剂的使用寿命和保存。优选的,本发明的空心胶囊的胶囊壁的密度为0.4~0.8g/cm3。
进一步,所述的胶囊壁的孔隙率为30%~80%。胶囊壁的孔隙率越高,胶囊的密度越小,但是,胶囊的强度可能会降低,不利于胶囊的填装和运输。当所述的胶囊壁的孔隙率大于80%,胶囊的强度较低,胶囊在填装和运输时容易破碎,实用性下降。当所述的胶囊壁的孔隙率小于30%,胶囊的密度难以大幅降低。优选的,所述的胶囊壁的孔隙率为40%~70%。
进一步的,所述的胶囊壁中含有直径为100~800nm的孔。当胶囊壁中含有上述直径的孔时,对胶囊的密度降低效果较明显,且胶囊的强度(比如脆碎度)也能保证较高的水平,具有很好的实用性。优选的,所述的胶囊壁中含有直径300~600nm的孔。
进一步的,所述的直径为100~800nm的孔的孔径分布<1.5。孔径分布越大,胶囊的强度分布越不均匀,会出现缺陷,在运输和填装胶囊剂过程中容易破损而导致空心胶囊和胶囊内容物的浪费。优选的,所述的直径为100~800nm的孔的孔径分布<1.3。
进一步的,所述的胶囊壁中还含有直径小于100nm的孔。这些孔对降低胶囊的密度有一定贡献,且能保证胶囊具有更高的强度。
进一步的,所述的直径小于100nm的孔的孔径分布为1.5~2.0。当直径小于100nm的孔的孔径分布在上述范围内时,胶囊的强度较高,综合性能优异。优选的,所述的直径小于100nm的孔的孔径分布为1.6~1.8。
进一步的,所述的直径小于100nm的孔与直径为100~800nm的孔的数量比为1:1~1:2。当胶囊壁中上述两种孔的数量比在这个比例范围内时,胶囊的密度和强度等综合性能更好。优选的,所述的直径小于100nm的孔与直径为100~800nm的孔的数量比为1:1~1:1.5。
进一步的,所述的胶囊的外表面上直径为100~800nm的表面孔的数目小于10个/mm2。胶囊的外表面是指胶囊填装内容物后不接触内容物的一侧表面。当所述的胶囊的外表面上直径为100~800nm的表面孔的数目大于10个/mm2,胶囊外表面粗糙度增加,胶囊外观不佳且容易黏附杂质。优选的,所述的胶囊的外表面上直径为100~800nm的表面孔的数目小于5个/mm2。
进一步的,所述的胶囊的内表面上直径为100~800nm的表面孔的数目小于10个/mm2。胶囊的内表面是指胶囊填装内容物后接触内容物的一侧表面。当所述的胶囊的内表面上直径为100~800nm的表面孔的数目大于10个/mm2,胶囊内表面粗糙度增加,胶囊内表面容易黏附内容物,影响胶囊的填装。优选的,所述的胶囊的内表面上直径为100~800nm的表面孔的数目小于5个/mm2。
本发明中所述的空心胶囊的胶囊中的孔可以用已知的任何一种方法制备,具体包括化学发泡法、物理发泡法、粒子淋洗法等其中的任何一种。所述的化学发泡法是指通过化学反应产生气泡而使物质发泡的方法,化学发泡剂可以是碳酸氢钠、碳酸氢钙、碳酸铵或氯化铵中的一种或多种。以碳酸氢钠作为发泡剂为例,制备方法可以是:将胶囊的原料和碳酸氢钠混合,制成水溶液,按一般的方法制备得到无孔胶囊,制备温度始终控制在70℃以下,优选60℃以下。最后,在110℃以上,优选130℃以上加热,升温速率为5℃/s以上,加热时间为10s~1min,继而快速冷却至室温,得到含有上述孔的胶囊。
物理发泡法是指通过产品内组分挥发使产品产生泡孔的方法,物理发泡剂可以是水、正己烷、正戊烷或正庚烷中的一种或多种。以水作为发泡剂为例,制备方法可以是:按一般的方法制备得到无孔胶囊,制备温度始终控制在70℃以下,优选60℃以下。最后,在110℃以上,优选130℃以上加热,升温速率为5℃/s以上,加热时间为10s~5min,继而快速冷却至室温。得到含有上述孔的胶囊。
粒子淋洗法是用特定溶剂(比如水)淋洗产品,使产品中的组分中产品分离出来,在产品的表面和内部形成微孔的方法,所述的粒子(和淋洗液)可以是氯化钠、氯化镁、氯化钙、醋酸钙、醋酸钠或醋酸钙中的一种或多种。
优选的,所述的孔使用化学发泡法或物理发泡法制备得到。
进一步的,所述的空心胶囊含有如下性能:
脆碎度:50粒中破碎胶囊不超过5粒;
崩解时限:<10min;
松紧度:10粒中漏粉胶囊不超过1粒;
重金属含量:<40ppm。
胶囊的脆碎度反应胶囊的力学强度,保证空心胶囊及其胶囊剂在包装和运输过程中具有很低的破损率。
崩解是指固体制剂在规定条件下全部崩解溶散或成碎粒,除不溶性包衣材料或破碎的胶囊壳外,应全部通过筛网。崩解时限是指固体制剂在规定的介质中,以规定的方法进行检查全部崩解溶散或成碎粒并通过筛网所需时间的限度。一般而言,崩解时限越短,胶囊剂的治疗效果越好。
松紧度是指胶囊体和胶囊帽套合的松紧程度,如果胶囊体和胶囊帽套合太松,在胶囊剂的搬运过程中,胶囊内容物洒出,造成原料的浪费,并影响胶囊剂的使用。
重金属含量是指空心胶囊中重金属的百分含量,重金属对人体有危害,其含量越低,对人体的危害越小。
以上性能可以根据中国药典记载的方法进行测定。
优选的,所述的白色空心胶囊具有如下性能:
脆碎度:50粒中破碎胶囊不超过3粒;
崩解时限:<8min;
重金属含量:<20ppm。
进一步的,本发明胶囊含有明胶、淀粉、羟丙甲基纤维素或普鲁兰中的一种或多种。
进一步的,胶囊中还可以含有颜料、胶凝剂、促凝剂、增塑剂、粘合剂、PH调节剂等添加剂。
所述的颜料是指能为胶囊上色的物质,为已知的任何一种,具体举例如姜黄、核黄素、喹啉黄、叶绿素、二氧化钛、靛蓝、番茄红、辣椒红、叶黄素、花青素、甜菜红、果红、丹宁酸或氧化铁中的一种或多种。
所述的胶凝剂是指在胶囊制备过程中,能使胶囊原料水溶液逐渐转变成均匀的半刚性固体凝胶、并保持原有形状的添加剂。可以是乙酰化结冷胶、去乙酰化结冷胶、κ型卡拉胶、β型卡拉胶、ι型卡拉胶、琼脂、果胶、海藻酸钠、黄原胶、西黄芪胶、刺梧桐胶、刺槐豆胶、红藻胶、罗望子胶、他拉胶、菌核葡聚胶、微生物藻酸盐或卡波姆中的一种或多种。
所述的促凝剂是指在胶囊制备过程中,能提高胶囊原料水溶液逐渐转变成均匀的半刚性固体凝胶、并保持原有形状速率的添加剂。可以是氯化钾、氯化钙、磷酸钾或柠檬酸钾中的一种或多种。
所述的增塑剂是可以提高胶囊原料加工性能的添加剂,可以是甘油、山梨糖醇、聚乙二醇、乙二醇、乙酸乙酯、1,3-丁二醇、1,4丁二醇、木糖醇、甘油二乙酯、甘油三醋酸酯、甘油单醋酸酯、甘露醇、肌醇、麦芽糖醇、葡萄糖或聚丙二醇等中的一种或多种。
所述的粘合剂可以是淀粉、糊精、聚乙烯吡络烷酮、羧甲基甲壳素、聚乙烯醇、田菁胶、阿拉伯胶、肉硅胶、普鲁兰、爱生兰、印度胶、葡萄糖或乙烯吡络烷酮和醋酸乙烯酯共聚物中的一种或多种。
所述的PH调节剂用于调节空心胶囊制备过程中的原料水溶液的PH值,可以是碳酸钠、碳酸钾、氢氧化钠、氢氧化钾、氢氧化钙、碳酸氢钠、碳酸氢钾、碳酸铵、磷酸二氢钠、磷酸二氢钾或磷酸氢二钾中的一种或多种。
本发明的空心胶囊的胶囊壁的密度比市售的胶囊的密度更低,但胶囊的其他性能却没有大幅的变化,满足使用性能的同时,方便胶囊的运输,节约成本。

1、表观密度(ρ):通过样品的质量除以其排水体积计算得到。
2、孔隙率(P):以式1计算胶囊壁的孔隙率,
式中,ρ1是胶囊壁的真密度,通过将多孔样品用热水溶解,浇注成薄膜状,再干燥到原样品的含水量后,通过测定该样品的质量和体积,并以质量除以体积计算得到;ρ是胶囊壁的表观密度。
3、孔直径(d):用扫描电子显微镜(SEM)观察样品的平整断面,随机拍摄不同位置放大倍率10000倍的照片5张,用笔勾出孔的轮廓后,用图像处理软件计算各表面孔的面积S,再按式1计算各孔的直径d(一种等价的直径,即与孔面积相等的圆的直径):
4、孔的数量比:利用上述照片,分别统计直径d小于100nm的孔与直径d为100~800nm的孔的数目,并计算这两种直径范围的孔的数量比,记为Φ。
5、孔径分布(SD):利用上述照片,按式2计算各直径范围的孔的孔径分布,
式中,∑d4是孔径d的4次方的加和;∑d3是孔径d的3次方的加和;∑d是孔径d的加和。
将直径为100nm~800nm的孔的孔径分布记为SDA,将直径小于100nm的孔的孔径分布记为SDB。
6、表面孔数量:用SEM分别观察外表面和内表面上随机选取的面积为1mm2的区域,计算此区域的表面孔数,在相同表面测定10个不同区域的表面孔数并取平均值。
7、脆碎度:按2015年版《中国药典》四部明胶空心胶囊脆碎度的测试方法进行测试。
8、崩解时限:按2015年版《中国药典》四部明胶空心胶囊崩解时限的测试方法进行测试。
9、松紧度:按2015年版《中国药典》四部明胶空心胶囊松紧度测试方法测试。
10、重金属含量:按2015年版《中国药典》四部明胶空心胶囊重金属的测试方法进行测试。
本发明实施例所用原料:
明胶,浙江绿洲生物技术有限公司产。
玉米淀粉,浙江绿洲生物技术有限公司产。
羟丙甲基纤维素,韩国乐天产。
普鲁兰多糖,浙江绿洲生物技术有限公司产。
实施例1~18
使用表1所示的原料,通过以下方法中的任一种制备出具有表1所示结构和性质的0#空心胶囊。
方法一:将胶囊的原料和碳酸氢钠混合,制成水溶液,按一般的方法制备胶囊,但温度始终控制在70℃以下,优选60℃以下。最后,在110℃以上,优选130℃以上加热,升温速率为5℃/s以上,加热时间为10s~1min,继而快速冷却至室温,得到含有上述孔的胶囊。
方法二:按一般的方法制备胶囊,最后,在110℃以上,优选130℃以上加热,升温速率为5℃/s以上,加热时间为10s~5min,继而快速冷却至室温。得到含有上述孔的胶囊。
可以通过调节发泡剂或淋洗粒子的含量、加热温度、加热时间、冷却速率等方式来调节胶囊孔的直径、孔隙率及孔径分布等性能。

表1中,实施例1~4的胶囊壁中孔的孔径都大于800nm;实施例5~18的胶囊壁中含有直径为100~800nm的孔;实施例7~18的胶囊壁中除了含有直径为100~800nm的孔以外,还含有直径小于100nm的孔。
对比例1
市售0#明胶空心胶囊,经检测,胶囊的密度为1.2g/cm3。
对比例2
市售0#普鲁兰空心胶囊,经检测,胶囊的密度为1.2g/cm3。
对比例3
市售0#羟丙甲基纤维素空心胶囊,经检测,胶囊的密度为1.2g/cm3。
将实施例制备的0#空心胶囊和对比例的市售0#空心胶囊按照本发明提供的检测方法进行性能测定,部分测定结果如表2所示。

将实施例和对比的胶囊进行性能检测得到,实施例1~18的胶囊内表面和外表面上没有孔,松紧度测试时10粒胶囊中没有漏粉胶囊,胶囊的重金属含量也都小于20ppm,脆碎度也不超过5粒,崩解时限小于10min。本发明的空心胶囊的性能满足中国药典(2015版)的标准,具有实用性;并且,本发明胶囊的密度比市售胶囊小,单个胶囊质量轻,方便胶囊的运输,节约成本。
Claims (10)
1.一种空心胶囊,其特征在于,胶囊壁的密度为0.2~1.0g/cm3。
2.根据权利要求1所述的空心胶囊,其特征在于,胶囊壁的孔隙率为30%~80%。
3.根据权利要求2所述的空心胶囊,其特征在于,所述的胶囊壁中含有直径为100~800nm的孔。
4.根据权利要求3所述的空心胶囊,其特征在于,所述的直径为100~800nm的孔的孔径分布<1.5。
5.根据权利要求3所述的空心胶囊,其特征在于,所述的胶囊壁中还含有直径小于100nm的孔。
6.根据权利要求5所述的空心胶囊,其特征在于,所述的直径小于100nm的孔的孔径分布为1.5~2.0。
7.根据权利要求5所述的空心胶囊,其特征在于,所述的直径小于100nm的孔与直径为100~800nm的孔的数量比为1:1~1:2。
8.根据权利要求3所述的空心胶囊,其特征在于,所述的胶囊的外表面上直径为100~800nm的表面孔的数目小于10个/mm2。
9.根据权利要求3所述的空心胶囊,其特征在于,所述的胶囊的内表面上直径为100~800nm的表面孔的数目小于10个/mm2。
10.根据权利要求1~9任一项所述的空心胶囊,其特征在于,含有如下性能:脆碎度:50粒中破碎胶囊不超过5粒;
崩解时限:<10min;
松紧度:10粒中漏粉胶囊不超过1粒;
重金属含量:<40ppm。
