Abstract
本发明公开了一种生物蛋白抑菌漱口水,按重量百分比计,主要由以下成分组成:卵黄生物蛋白0.03‑1%、肉豆蔻酸异丙酯0.01‑0.5%、芦荟苦素0.08‑0.1%、表面活性剂0.02‑0.1%、发泡剂0.02‑0.3%、甜味剂0.2‑0.5%、余量为去离子水;所述卵黄生物蛋白是对抗变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌五种病菌的复合抗体。本发明的生物蛋白抑菌漱口水具有安全性高、抑菌效果好的特点,可有效预防儿童乳牙龋病等口腔疾病与上呼吸道疾病。
Description
技术领域
本发明涉及漱口水技术领域,具体是涉及一种生物蛋白抑菌漱口水及其制备方法。
背景技术
随着人们生活水平的日益提高,人们对自身的健康越来越重视。其中口腔健康也是很重要的一项,口腔问题一般包括牙痛、牙龈炎、牙周炎、牙龈出血等疾病是口腔多发病症。市场上有很多的口腔清洁类产品,对相关口腔疾病有较好功效的口腔产品不多,主要是缺乏相应的功效物质基础。目前,用于口腔疾病防治的药剂如含片、口胶、膜片等在牙缝中难于透入,难以发挥应有的治疗作用,而且使用的制剂大多采用粗浸膏制作,有效成份含量低且不稳定,难以达到既定的效果。
市场上的口腔护理产品种类繁多,但在安全、高效和疾病预防上均未能满足要求,很多产品中含有不利于健康的防腐剂或者抗生素之类的的成分。另外,目前还没有将漱口水这种口腔清洁保健产品用于对一些常见的呼吸道疾病的防治。因此,本发明致力于研制处一种既能有效预防口腔疾病又能有效预防常见的呼吸道疾病的漱口水。
发明内容
本发明解决的技术问题是现有的口腔护理产品安全性不够高,效果不够好,且不能对一些常见的呼吸道疾病进行预防,进而提供一种生物蛋白抑菌漱口水及其制备方法。
本发明的技术方案是:
一种生物蛋白抑菌漱口水,按重量百分比计,主要由以下成分组成:卵黄生物蛋白0.03-1%、肉豆蔻酸异丙酯0.01-0.5%、芦荟苦素0.08-0.1%、表面活性剂0.02-0.1%、发泡剂0.02-0.3%、甜味剂0.2-0.5%、余量为去离子水;所述卵黄生物蛋白是对抗变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌五种病菌的复合抗体。也可以根据需要防治的其他口腔疾病或呼吸道疾病对所含的抗体种类进行增减调整。
进一步地,所述卵黄生物蛋白的制备方法为:
S1:选择培养出变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌;
S2:用S1培养的致病菌种按适当比例混合,一般为等比例;然后按1:1比例加入等量的福氏佐剂;再用高速匀浆器以10000-30000rpm高速匀化制备成复合抗原;
S3:用所述复合抗原对产蛋鸵鸟进行注射,检取鸵鸟所生的免疫蛋,分离抽提制得所述卵黄生物蛋白。
更进一步地,所述S3分离抽提制得所述卵黄生物蛋白的具体操作方法为:用蛋黄筛取出免疫蛋的蛋黄,搅打均匀;按1:6-7的体积比加入灭菌蒸馏水稀释混匀;调pH值至5.2;加入浓度为3%海藻酸钠至终浓度为2.2‰,搅拌直至出现浑浊沉淀;加入浓度为1.5%的CaCl2液至终浓度为1.2‰,搅拌均匀;置于3-5℃条件下静置15-20小时,然后虹吸收取上清,得到所述卵黄生物蛋白。
进一步地,所述甜味剂选自:甜菊糖苷、甜茶苷、天冬甜素、丁磺氨或蛇菊苷。
进一步地,所述表面活性剂选自:单酸甘油酯类、乙酰化单酸甘油酯、脱水山梨糖醇酯、硬脂基乳酸钠中的任意一种。
一种生物蛋白抑菌漱口水的制备方法为:按所述配比将鸵鸟卵黄抗体、肉豆蔻酸异丙酯、芦荟苦素、表面活性剂、发泡剂、甜味剂、去离子水混合,置于均质机中进行均质处理,温度为5-40℃,时间为3-10min。
一种生物蛋白抑菌漱口水的微生物含量的检测方法为:
1)取所述生物蛋白抑菌漱口水5ml,作为待测样品,按照1:10的倍比进行稀释,得到样品稀释液;
2)将上述样品稀释液装入三角瓶或试管中,置于恒温水浴中进行恒温处理10~20分钟,温度为25-35℃;同时,采用蒸馏水作为参照标准液,置于同一个水浴槽中进行恒温处理10~20分钟;
3)取出经恒温处理的样品稀释液,按照常规方法吸取并进行塗皿,然后放入温箱中进行培养;
4)2~3天后平板上长出可见的菌落,取出进行常规的菌落计数和计算,获得该样品中的菌落总数,即可代表该生物蛋白抑菌漱口水的有效微生物含量。
生物蛋白抑菌漱口水的稳定性检测标准,其特征在于,包括以下内容:
液体:均一,无沉淀,无异物;
香味:符合相应气味,无异味;
色泽:澄清透明;
pH值:5.8-9.0;
菌落总数:≦100(CFU/g);
霉菌和酵母菌总数:≦10(CFU/g);
粪大肠菌群:不得检出;
金黄色葡萄球菌:不得检出;
铜绿假单胞菌:不得检出;
卵黄生物蛋白:≧0.3g/kg
荧光剂:不得检出;
铅:不得检出;
汞:不得检出;
砷:不得检出。
符合以上标准即为合格品。
本发明的有益效果是:本发明利用口腔致病抗原与几种常见的呼吸道致病抗原免疫到产蛋鸵鸟,鸵鸟自身免疫应答产生的抗体会转移至鸵鸟蛋黄中,通过卵黄抗体(IgY)制备技术对蛋黄分离抽提制得对抗变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌的复合抗体(Igy),将制备的复合抗体用于漱口水中,制备得到的生物蛋白抑菌漱口水具有安全性高、抑菌效果好的特点,可有效预防儿童乳牙龋病等口腔疾病与上呼吸道疾病。
具体实施方式
实施例1
一种生物蛋白抑菌漱口水,按重量百分比计,主要由以下成分组成:卵黄生物蛋白0.03%、肉豆蔻酸异丙酯0.01%、芦荟苦素0.08%、表面活性剂0.02%、发泡剂0.02%、甜味剂0.2%、余量为去离子水;所述卵黄生物蛋白是对抗变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌五种病菌的复合抗体。
其中,卵黄生物蛋白的制备方法为:
S1:选择培养出变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌;

实施例2
一种生物蛋白抑菌漱口水,按重量百分比计,主要由以下成分组成:卵黄生物蛋白0.515%、肉豆蔻酸异丙酯0.255%、芦荟苦素0.09%、表面活性剂0.06%、发泡剂0.16%、甜味剂0.35%、余量为去离子水;所述卵黄生物蛋白是对抗变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌五种病菌的复合抗体。
其中,卵黄生物蛋白的制备方法为:
S1:选择培养出变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌;
S2:用S1培养的致病菌种按等比例混合;然后按1:1比例加入等量的福氏佐剂;再用高速匀浆器以20000rpm高速匀化制备成复合抗原;
S3:用所述复合抗原对产蛋鸵鸟进行注射,检取鸵鸟所生的免疫蛋,分离抽提制得所述卵黄生物蛋白。具体操作方法为:用蛋黄筛取出免疫蛋的蛋黄,搅打均匀;按1:6.5的体积比加入灭菌蒸馏水稀释混匀;调pH值至5.2;加入浓度为3%海藻酸钠至终浓度为2.2‰,搅拌直至出现浑浊沉淀;加入浓度为1.5%的CaCl2液至终浓度为1.2‰,搅拌均匀;置于4℃条件下静置18小时,然后虹吸收取上清,得到所述卵黄生物蛋白。
甜味剂为甜菊糖苷、甜茶苷、天冬甜素、丁磺氨或蛇菊苷。
表面活性剂为单酸甘油酯类、乙酰化单酸甘油酯、脱水山梨糖醇酯、硬脂基乳酸钠。
该生物蛋白抑菌漱口水的制备方法为:按所述配比将鸵鸟卵黄抗体、肉豆蔻酸异丙酯、芦荟苦素、表面活性剂、发泡剂、甜味剂、去离子水混合,置于均质机中进行均质处理,温度为22.5℃,时间为6min。
对该生物蛋白抑菌漱口水的微生物含量进行检测:
1)取所述生物蛋白抑菌漱口水5ml,作为待测样品,按照1:10的倍比进行稀释,得到样品稀释液;
2)将上述样品稀释液装入三角瓶或试管中,置于恒温水浴中进行恒温处理15分钟,温度为30℃;同时,采用蒸馏水作为参照标准液,置于同一个水浴槽中进行恒温处理15分钟;
3)取出经恒温处理的样品稀释液,按照常规方法吸取并进行塗皿,然后放入温箱中进行培养;
4)2天后平板上长出可见的菌落,取出进行常规的菌落计数和计算,获得该样品中的菌落总数,即可代表该生物蛋白抑菌漱口水的有效微生物含量。
经检测,发现该生物蛋白抑菌漱口水符合标准。
实施例3
一种生物蛋白抑菌漱口水,按重量百分比计,主要由以下成分组成:卵黄生物蛋白1%、肉豆蔻酸异丙酯0.5%、芦荟苦素0.1%、表面活性剂0.1%、发泡剂0.3%、甜味剂0.5%、余量为去离子水;所述卵黄生物蛋白是对抗变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌五种病菌的复合抗体。
其中,卵黄生物蛋白的制备方法为:
S1:选择培养出变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌;
S2:用S1培养的致病菌种按等比例混合;然后按1:1比例加入等量的福氏佐剂;再用高速匀浆器以30000rpm高速匀化制备成复合抗原;
S3:用所述复合抗原对产蛋鸵鸟进行注射,检取鸵鸟所生的免疫蛋,分离抽提制得所述卵黄生物蛋白。具体操作方法为:用蛋黄筛取出免疫蛋的蛋黄,搅打均匀;按1:7的体积比加入灭菌蒸馏水稀释混匀;调pH值至5.2;加入浓度为3%海藻酸钠至终浓度为2.2‰,搅拌直至出现浑浊沉淀;加入浓度为1.5%的CaCl2液至终浓度为1.2‰,搅拌均匀;置于5℃条件下静置20小时,然后虹吸收取上清,得到所述卵黄生物蛋白。
甜味剂为甜菊糖苷、甜茶苷、天冬甜素、丁磺氨或蛇菊苷。
表面活性剂为单酸甘油酯类、乙酰化单酸甘油酯、脱水山梨糖醇酯、硬脂基乳酸钠。
该生物蛋白抑菌漱口水的制备方法为:按所述配比将鸵鸟卵黄抗体、肉豆蔻酸异丙酯、芦荟苦素、表面活性剂、发泡剂、甜味剂、去离子水混合,置于均质机中进行均质处理,温度为40℃,时间为10min。
对该生物蛋白抑菌漱口水的微生物含量进行检测:
1)取所述生物蛋白抑菌漱口水5ml,作为待测样品,按照1:10的倍比进行稀释,得到样品稀释液;
2)将上述样品稀释液装入三角瓶或试管中,置于恒温水浴中进行恒温处理20分钟,温度为35℃;同时,采用蒸馏水作为参照标准液,置于同一个水浴槽中进行恒温处理20分钟;
3)取出经恒温处理的样品稀释液,按照常规方法吸取并进行塗皿,然后放入温箱中进行培养;
4)3天后平板上长出可见的菌落,取出进行常规的菌落计数和计算,获得该样品中的菌落总数,即可代表该生物蛋白抑菌漱口水的有效微生物含量。
经检测,发现该生物蛋白抑菌漱口水符合标准。
应用数据:
用本发明实施例1、2、3制备的漱口水与市场上销售的某品牌(李施德林)漱口水进行对比试验,选取300名儿童志愿者,随机均分成5组,每组60人,各组之间无显著差异,其中1、2、3组分别使用本发明实施例1、2、3制备的漱口水,第4组作为对照组使用李施德林牌漱口水,第5组作为空白组,使用清水,所有愿者均是每日三餐后漱口,分别于3个月后进行结果统计,统计指标包括抗变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌及儿童乳牙龋病的发病率,具体见下表1。
抗变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌的复合抗体(Igy),将制备的复合抗体用于漱口水中,制备得到的生物蛋白抑菌漱口水具有安全性高、抑菌效果好的特点,可有效预防儿童乳牙龋病等口腔疾病与上呼吸道疾病。

结果发现本发明制备的的漱口水抗菌抗病毒效果更加优异,且乳牙龋病发病人数也大大降低。

Claims (9)
1.一种生物蛋白抑菌漱口水,其特征在于,按重量百分比计,主要由以下成分组成:卵黄生物蛋白0.03-1%、肉豆蔻酸异丙酯0.01-0.5%、芦荟苦素0.08-0.1%、表面活性剂0.02-0.1%、发泡剂0.02-0.3%、甜味剂0.2-0.5%、余量为去离子水;所述卵黄生物蛋白是对抗变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌五种病菌的复合抗体。
2.如权利要求1所述的一种生物蛋白抑菌漱口水,其特征在于,所述卵黄生物蛋白的制备方法为:
S1:选择培养出变形链球茵、远缘链球菌、流感病毒、肺炎型支原体、肺炎杆菌;
S2:用S1培养的致病菌种按适当比例混合;然后按1:1比例加入等量的福氏佐剂;再用高速匀浆器以10000-30000rpm高速匀化制备成复合抗原;
S3:用所述复合抗原对产蛋鸵鸟进行注射,检取鸵鸟所生的免疫蛋,分离抽提制得所述卵黄生物蛋白。
3.如权利要求2所述的一种生物蛋白抑菌漱口水,其特征在于,所述S3分离抽提制得所述卵黄生物蛋白的具体操作方法为:用蛋黄筛取出免疫蛋的蛋黄,搅打均匀;按1:6-7的体积比加入灭菌蒸馏水稀释混匀;调pH值至5.2;加入浓度为3%海藻酸钠至终浓度为2.2‰,搅拌直至出现浑浊沉淀;加入浓度为1.5%的CaCl2液至终浓度为1.2‰,搅拌均匀;置于3-5℃条件下静置15-20小时,然后虹吸收取上清,得到所述卵黄生物蛋白。
4.如权利要求1所述的一种生物蛋白抑菌漱口水,其特征在于,所述甜味剂选自:甜菊糖苷、甜茶苷、天冬甜素、丁磺氨或蛇菊苷。
5.如权利要求1所述的一种生物蛋白抑菌漱口水,其特征在于,所述表面活性剂选自:单酸甘油酯类、乙酰化单酸甘油酯、脱水山梨糖醇酯、硬脂基乳酸钠中的任意一种。
6.如权利要求1所述的一种生物蛋白抑菌漱口水,其特征在于,所述甜味剂选自:蛇菊苷、甜茶苷、甜菊糖苷或丁磺氨。
7.如权利要求1-5任意一项所述的一种生物蛋白抑菌漱口水的制备方法,其特征在于:按所述配比将鸵鸟卵黄抗体、肉豆蔻酸异丙酯、芦荟苦素、表面活性剂、发泡剂、甜味剂、去离子水混合,置于均质机中进行均质处理,温度为5-40℃,时间为3-10min。
8.如权利要求1-5任意一项所述的一种生物蛋白抑菌漱口水的制备方法,其特征在于:按所述配比将鸵鸟卵黄抗体、肉豆蔻酸异丙酯、芦荟苦素、表面活性剂、发泡剂、甜味剂、去离子水混合,置于均质机中进行均质处理,时间为3-10min。
9.如权利要求1所述的一种生物蛋白抑菌漱口水的微生物含量的检测方法,其特征在于,包括以下步骤:
1)取所述生物蛋白抑菌漱口水5ml,作为待测样品,按照1:10的倍比进行稀释,得到样品稀释液;
