Description
本发明涉及防晒配合物,具体说来是提高了UVA吸收作用的防晒金属配合物。这些配合物包括与阳离子类配合的二甲苯甲酰甲烷吸收UVA防晒剂(优选与金属阳离子配合)。这些配合物用来保护皮肤免受紫外线照射的伤害。本发明还涉及含有这些配合物的组合物。
阳光对于皮肤的有害作用已有大量报道。长期暴露于阳光下的主要的短期伤害是红斑(即晒斑)。290~320毫微米波长紫外线范围,称作“UVB”波长范围,是引起红斑的主要原因。320~400毫微米波长紫外线范围,称作“UVA”波长范围,也引起红斑。
除红斑的短期伤害外,还有与UV射线相关的长期伤害。长期伤害中的一种是皮肤表面的恶变。许多流行病学研究证明太阳照射与人类皮肤癌之间存在密切关系。
紫外线的另一长期伤害是皮肤过早衰老。这种状况的特征是皮肤产生皱纹和变黄,并伴随有其它物理变化例如裂纹,毛细管扩张(蜘蛛状脉管),光化性角质病(赘生物),瘀斑(皮下出血损伤),缺乏弹性(下垂)。在De Simone先生的“防晒与晒黑产品”(Handbook of Nonpres-cription Drugs,7th Ed.,第26章,499~511页American Pharmaceutcal Association,Washington,D.C.,1982)一文中,Grove和Forbes先生的“评价防晒剂对UV-A射线的保护作用的方法”(International Journal of Cosmetic Science,4,第15~24页1982))一文中,以及1983年6月7日公开的Depolo先生的专利U.S.4387089中都详细讨论了与暴露于UVA和UVB波长射线相关的有害作用。这些文献均列入本申请作为参考。
防晒剂和物理遮阳物在商业上都可用于保护皮肤免受UV照射。物理遮阳物可散射,反射和吸收紫外线,见Soyre.R.M等人的“物理防晒物”(J.Soc.Cosmet,Chem.,Vol.41,no.2,103~109页(1990))一文。物理遮阳物的例子包括二氧化肽和氧化锌。然而有高含量这些试剂的组合物是不透明的,通常色泽不美观,被认为仅仅除鼻子或耳尖外别处是不能使用的。并且这些试剂很容易擦掉或抹去而结果很少或几乎没有什么保护作用。
相反,防晒剂是通过化学作用来显示其效果的,即它们吸收紫外线使其不能透过皮肤。防晒剂对于使用者来说存在几个问题。比如,在暴露时间里它们必须在皮肤表面方才有效。防晒剂是预防措施,因此人们必须要事先予知自己将暴露于阳光才行。为使其最有效,一定要将防晒剂涂于皮肤上呈连续的均匀的膜,在皮肤表面形成这样的一层膜是很困难的。
大多数商业上可用的防晒剂主要吸收UVB。UVA吸收剂数量有限,已知的有苯酮和二苯甲酰甲烷,引入本申请作参考的Welters等人1983年12月18日公开的美国专利U.S.4489057和Depolo先生1983年6月7日分开的专利U.S.4387089中公开了二苯甲酰甲烷防晒剂。
本发明发现当二苯甲酰甲烷生色团与某些阳离子类物质,例如金属阳离子配合时能显著增加它对UVA的吸收。因此它有可能用来制备具有增加UVA吸收,保护皮肤免受UV线伤害的防晒剂。
因此本发明的一个目的是提供能加强UVA吸收的防晒配合物。
本发明的另一目的是提供加强UVA吸收的防晒金属配合物。
本发明的再一目的是提供预防暴露于阳光或其它UV线源引起的急性(红斑)和慢性(光化衰老,癌)症状的防晒金属配合体。
本发明再一目的是提供制备防晒金属配合物的方法。
本发明的另一目的是提供含有防晒金属配合物,能保护皮肤不受UV线有害影响的表面用防晒组合物。
本发明的另一目的是提供含有防晒金属配合物防晒组合物,它不易被皮肤吸收;它可提高防晒保护性能并减少于日常或几乎日常应用中产生过敏,刺激,或毒性问题等弊病;它不易擦掉,在化妆美容上是可接受的。
由下面详细的描述,以上这些目的和其它目的将变得显而易见。

防晒配合物
本发明中所使用的防晒金属配合物具有通式:
(S)m代表二苯甲酰甲烷防晒基团,其中m代表配合物中该基团的数目。
(M)n代表阳离子类物质例如金属阳离子,或者铵或取代的铵,其中n代表配合物中该类物质的数目。

本发明防晒金属配合物优选对于可见波长范围(即大约400nm以上)光的吸收很弱或几乎没有。因此这些配合物或者只具有浅颜色(例如浅黄或奶油色)或者是基本白色。这对美容来说是期望的。因此该防晒金属配合物优选对大于约400nm的任何波长不具有大于约500的ε值,最优选对于大于约400nm的任何波长ε基本为零。
(S)m防晒基团
构成本发明防晒配合物的防晒基团是那些含有二苯甲酰甲烷生色的基团。这种生色团的特征是因强吸收UVA范围的射线而有效。这些构成本发明配合物的二苯甲酰甲烷防晒基团在上文所提到的引作参考的美国专利U.S.4489057和U.S.4387089中已详细公开过。
本发明防晒配合物中防晒基团的数目以m表示,其中m是选自1~8的整数,优选m是选自1~4的整数,更优选m是选自1至3的整数,最优选m是2。并且由此可知本发明配合物可以作为m值不同的各种种类的混合物存在。因此可能得到防晒基团的平均m值是非整数的配合物。

在上面的结构中,A是选自H,-OR或-NR2位于芳环上不同位置的取代基,其中每个R各自是H,含有约1~约20个碳原子的直链或支链烷基,(CH2CH2O)q-H,或(CH2CH(CH3)O)q-H,其中q是1~约8的整数;且B是芳环上不同位置上的取代基,选自H,具有大约1~20个碳原子的直链或支链烷基,(CH2CH2O)q-H,或(CH2CH(CH3)O)q-H,其中q是1至约8的整数。在本发明中A优选对-OCH3,且B是对-叔丁基,即4,4′-甲氧基-叔丁基二苯甲酰甲烷(CTFA采纳名:丁基二苯甲酰甲烷,以ParsolR1789注册商标可从Givaudan买到),或者A是H且B是对-异丙基,即4-异丙基-二苯甲酰甲烷(CTFA采纳名:异丙基二苯甲酰甲烷,以EusolexR8020注册商标,从Merck买到)。
在防晒基团中,尽管二苯甲酰甲烷生色团以1,3-二酮为代表,但应明白其代表并不排除功能基的其它互变异构形式,如烯醇形式。因此无论什么时候提到1,3-二酮,应明白它也涉及和包含所有适当烯醇的互变异构形式。这些二苯甲酰甲烷生色团的互变异构烯醇形式可用下面互变异构结构代表。
并且也应理解二苯甲酰甲烷生色团可以失去一个氢原子形成相应的阴离子之类基团。这种现象常发生在高pH值(即碱性值)及当二苯甲酰甲烷生色团与阳离子类物质例如金属配合时。因此每当提到1,3-二酮形式时,应明白所有适当的阴离子形式也被考虑和包括在内。这些二苯甲酰甲烷生色团的阴离子形式可用下面共振结构表示。
这里所使用的术语“摩尔吸光度”是一个分子吸收特定波长紫外光能力的定量单位。特定波长摩尔光吸收值表示为摩尔吸收系数(这里以“ε”代表,它以单位liter/mole cm表示),用下面等式计算:

特定波长光摩尔吸光度的计算是本领域公知的,在Atlas of Spectral Data and Phys ical Constants for Organic Compounds,2nd Ed.,Vol.I.pp.399-408(Grasselli and Ritchey,Editors;CRC Press,Inc.,Cleveland,Ohio,1975)中有详细论述,该文引入本发明作为参考。为计算摩尔吸光度而测定光强度使用的仪器在本领域也是公知的(例如,Varion DMS-100,Beckman DU-7,and Philips PU 8800)。下文实施例中提供了本发明代表化合物的摩尔吸光度。
这里所使用的术语“最大吸收”是指射线的某一波长,在该波长处含生色团分子较之其紧紧相邻的上下各波长处具最大摩尔吸光度。因此在UV-射线吸收的典型光谱中,最大吸收值作为由测量UV吸收仪器得到的光谱的峰值而很易识别。下文实施例中提供了本发明代表性防晒化合物的最大吸收(这里以λmax表示)。
(M)n
本发明防晒配合物含有阳离子类物质,优选金属,或者铵或取代的铵,以通式结构(M)n表示。用于本发明配合物的金属阳离子的例子包括碱金属(例如钠和钾),碱土金属(例如钙和镁),和过渡金属和重金属(例如铝和锶)。优选用于本发明配合物的金属选自下组:铝,锌,钙,镁,铜,铁,钡,锶,锆,钛,锡,铍,镓,铟,镧,锰,锑,铋,铈,钍,铌,钽,钼,钨,锂,钠,钾及其混合物。这些金属阳离子可以任何可能的价态和以这些价态的混合物应用(即例如某些金属阳离子是以这种阳离子可允许的价态中的一种存在,而某些金属阳离子是以这种阳离子的可允许价态中的另一种存在等等)。
或者,本发明防晒配合物也可包括与其它阳离子例如铵,取代的铵(例如单,二,三和四烷基和烷氧基取代的铵),和二胺阳离子(例如,四烷基和烷氧基取代的亚乙基二胺)相配合的配合物。
本发明防晒配合物优选包含选自下列金属铝,钛,铜,铁和锌的阳离子类物质。
本发明防晒配合物中3+价铝阳离子是最为优选的。
在本发明防晒配合物中,配合物出现的M类物质的数目用n表示,其中n是选自1~4的整数,优选n是选自1~3的整数,更优选n是选自1和2的整数,最优选n是1。并且应该明白本发明配合物可以以具有不同n值的不同种类的混合物存在。因此,有可能得到n值的平均值是非整数的配合物种类。
可从任何适当来源得到本发明组合物中的M类物质。例如当配合物中的M类物质是金属阳离子时,该阳离子可以从很宽范围的盐中得到。盐的例子包括金属氧化物,氢氧化物,氟化物,氯化物,溴化物,碘化物,碳酸盐,碳酸氢盐,磷酸盐,磷酸氢盐,磷酸二氢盐,烷氧化物(例如异丙氧化物),硫酸盐,硫酸氢盐,硝酸盐,亚硫酸盐,亚硝酸盐,硼酸盐,氯酸盐,溴酸盐,过氯酸盐,过溴酸盐,二磷酸盐,多磷酸盐,硫氰酸盐,羧酸盐(优选乙酸盐和硬脂酸盐)及其混合物。或者,当M类物质是铵或取代的铵时,铵和取代的铵类物质可以从很宽范围的铵或取代铵源例如盐(如氯化物,溴化物,氢氧化物等等)中衍生而来。
本发明组合物中优选的阳离子类物质源包括单醋酸铝,二醋酸铝,硬脂酸铝及其混合物。尤其优选单醋酸铝。
(L)p配位体
本发明防晒配合物可任意地包含以通式结构(L)p表示的有机或无机配位体。本发明配合体可应用中性和阴离子配位体。有用的有机配位体包括(但不局限于)选自下组的配位体:羧酸,二羧酸,和多羧酸及其阴离子;胺,二胺,和多胺;醇,二醇,和多醇及其阴离子;硫醇,二硫醇,和多硫醇及其阴离子;氨基酸及其阴离子;任何其它药学上可接受的有机配位体及其混合物。有用的无机配位体包括(但不局限于)水和羧基阴离子,卤化物(例如氟化物,氯化物,溴化物和碘化物),碳酸盐,碳酸氢盐,磷酸盐,磷酸氢盐,磷酸二氢盐,硫酸盐,硫酸氢盐,硝酸盐,亚硫酸盐,亚硝酸盐,硼酸盐,氯酸盐,溴酸盐,过氯酸盐,过溴酸盐,二磷酸盐,多磷酸盐,硫氰酸盐,任何其它药学上可接受的无机配位体及其混合物。这里所用术语“药学上可接受的”有机和无机配位体是指那些从毒性角度出发可接受的有机和无机配位体。
本发明防晒配合物配位体优先选自水,羟基阴离子,和具有约2至约22个碳原子的羧酸及其阴离子。其它优选的配位体包括乙氧化物和异丙氧化物。特别优选的羧酸及其阴离子是乙酸和乙酸根阴离子,辛酸及辛酸根阴离子,和硬脂酸和硬脂酸根阴离子。
本发明防晒配合物中出现在配合物中的配位体的数目用p来表示,其中p是选自0~4的整数,优选p是选自0~3的整数,更优选是选自0~2的整数,更好的p是选自0和1的整数,最优选p是0。并且应明白本发明化合物的配合物可以作为具有不同p值的不同种类的混合物的配合物存在。因此有可能获得平均p值是非整数重量平均p值的配合物。
本发明防晒金属配合物优选实例包括例如:

防晒配合物的制备
本发明防晒金属配合物可按下文实施例的描述制备。在进一步的实施方案中,这些配合物可直接与所需载体配方。或者,在与所需载体配方之前将这些防晒配合物离析出来。或者还可以在所需载体中直接制备这些防晒配合物。
通常,可在适当溶剂中使防晒化合物与金属盐结合而制备防晒金属配合物,这些适当的溶剂选自下组:水,丙酮,乙酸乙酯,甲基叔丁基醚,C1~C6醇,二醇,三醇,苯甲酸C12-15醇酯,氯代溶剂(例如二氯甲烷),二甲基异山梨酐(dimethyl isosrbide),新戊酸异癸酯,己二酸二异丙酯,及其混合物。优选的溶剂包括水,丙酮,乙醇,及其混合物。优选的金属盐包括那些选自下组金属的盐:铝,锌,钙,镁,铜,铁,钡,锶,锆,钛,锡,铍,镓,铟,镧,锰,锑,铋,铈,钍,铌,钽,钼,钨,锂,钠,钾,及其混合物。更优选铝,钛,铜,铁,锌的盐,及其混合物。特别优选的是下面所列的铝盐:醋酸铝,二酯酸铝,硬脂酸铝,二硬脂酸铝,辛酸铝,乙氧基铝,异丙氧基铝,及其混合物,而最优选的是二乙酸铝。或者可选用铵和取代铵和氯化物,溴化物和氢氧化物。
另外,为了促进防晒化合物的溶解,可至少加入一当量的碱。最好是在加入金属盐之前使防晒化合物和碱预先在溶剂体系中反应。优选的碱包括氢氧化钠,氢氧化钾,氢氧化铵,碳酸氢钠,碳酸钠,及其混合物。另外,如果需要,可将溶剂系统加热至其沸点。若要分离和提纯防晒金属配合物,最好是通过过滤和蒸发滤液来完成。另外,可从适当溶剂重结晶进一步纯化配合物。下面给出的实施例提供了防晒金属配合物和含它们的组合物的代表性制备方法。
含有防晒金属配合物的组合物
一种或几种本发明防晒配合物可以加入各种载体,包括药学和化妆载体,颜料,涂料,高分子基质,纤维基质,等等。优选将配位体与药学和化妆载体相结合。
典型的本发明防晒金属配合物的组合物含有大约0.1%至大约30.0%重量的本发明防晒金属配合物,优选大约1%至大约20%,最优选大约5%至大约15%。本发明组合物可含有下面成份。
药学上可接受的载体
本发明组合物可含有安全有效剂量的多种不同形式的表面用药学上可接受的载体或稀释剂。所谓:“安全有效”是指其量足以起到防晒金属配合物和任何其它成份的适当赋形剂的作用,而又不至于引起任何副作用或皮肤反应。“药学上可接受的”是指该载体适于皮肤表面施用而不会引起任何安全和毒性困忧,换言之,这些载体是适用于人和较低级动物的。这些表面用载体可以以乳液的形式存在,包括(但不局限于)水包油,油包水,水包油再包水,及硅氧烷包水再包油乳状液。这些乳状液可涉及很宽稠度范围,包括稀薄洗剂(它也适用于喷雾或气雾施放),乳膏状洗剂,稀乳膏剂,稠乳膏剂,等等。其它适当的表面用载体包括无水液体溶剂例如油和醇;以水为基础的单相液体溶剂(例如水-醇溶剂体系);无水固体和半固体(例如胶和粘剂);和水基质的胶和摩丝体系。用于本发明表面用载体体系的例子在下面四篇文献中所有描述,它们的全文均引入本文作为参考:“防晒产品配方”Cosmetics & Toiletries,Vol.105,PP122-139(1990年12月):“防晒产品配方”,Cosmetics & Toiletries,Vol.102,PP.117-136(1987年3月);1990年10月2日公开的Figueroa等人的美国专利U.S.4960764;1981年3月3日公开的Fukuda等人的美国专利U.S.4254105。
总的来说,药学上可接受的表面载体一般占本发明防晒组合物的大约0.1%至大约99.8%重量,优选大约80%至大约99%,更优选85%至大约95%。
本发明组合物优选的表面用载体是水包油型乳状液。这些水包油乳状组合物优选pH值范围是大约3.5至约9。另外,分散在油相介质中的平均颗粒度的范围是约1至约10微米,且约75%以上的颗粒小于约12微米。
附加防晒剂
大量的一种或几种附加防晒剂适用于本发明,Segarin等人在Cosmetics Science and Technology,第Ⅷ章,pages189 et seq.中公开了很多适宜制剂。特别适用的防晒剂包括(但不局限于)例如:乙基己基-P-甲氧基肉桂酸酯(以Parsol MCX注册商标由Givaudan公司可购买),对-氨基苯甲酸,其盐及其衍生物(乙酯、异丁酯、甘油酯);对-二甲氨基苯甲酸;N,N-二甲氨基苯甲酸2-乙基己酯;对-甲氧基肉桂酸二乙醇胺盐(由Bernel化学公司购得商标为Bernel Hydro的产品);氨茴酸酯(即邻-氨基苯甲酸酯;其甲酯,辛酯,戊酯,

酯和苄酯;苯基肉桂腈:肉桂酰丙酮丁酯);二羟基肉桂酸衍生物(7-羟基香豆素,甲基-7-羟基香豆素,甲基乙酰-7-羟基香豆素);三羟基肉桂酸衍生物(七叶亭,甲基七叶亭、瑞香素,和其糖苷,七叶苷和瑞香苷);烃(二苯基丁二烯,芪);二亚苄基丙酮和亚苄基乙酰苯酮;2-苯基苯并咪唑-5-磺酸及其盐;萘酚磺酸盐(2-萘酚基3,6-二磺酸钠和2-萘酚基-6,8-二磺酸钠盐);二羟基萘甲酸及其盐;邻位和对位羟基二苯基二磺酸盐;香豆素衍生物(7-羟基,7-甲基,3-苯基衍生物);二唑(2-乙酰基-3-溴代吲唑,苯基苯并噁唑,甲基萘并噁唑,各种芳基苯并噻唑);金鸡纳盐(硫酸氢盐,硫酸盐,氯化物,油酸盐,和单宁酸盐);喹啉衍生物(8-羟基喹啉盐,2-苯基喹啉);羟基或甲氧基取代的苯酮,尿酸和Vilouric acid;单宁酸及其衍生物(例如,六乙基醚);(丁基卡必基)(6-丙基胡椒基)醚;对苯二酚;苯酮类(苯酚,Sulisobenzone,Dioxybenzone,苯并间苯二酚,2,2′,4,4′-四羟基苯酮,2,2′-二羟基-4,4′-二甲氧基苯酮,Octabenzone);4-异丙基二苯甲酰甲烷;丁基甲氧基二苯甲酰甲烷;Octocrylene;4-异丙基二苯甲酰甲烷;及樟脑衍生物例如甲基亚苄基或亚苄基樟脑;三乙醇胺水杨酸盐;及其混合物。其它防晒剂包括固体物理遮阳物例如二氧化钛(微粒化的二氧化钛,0.03微米,0.035微米,0.050微米,和其它适当尺寸),氧化锌,二氧化硅,氧化铁等等。不受理论上的限制,人们确信这些无机材料通过反射,散射和吸收有害的UV,可见光和红外线而提供防晒功能。
在同一分子中既具有UVA又具有UVB吸收生色团的其它有用的防晒剂,公开于美国专利U.S.5041282,U.S.4999186和U.S.4937370,及欧洲专利申请EP416837,本发明将其作为参考。
通常,这些附加防晒剂可占组合物的0~大约20%,优选约0.5%至约10%。确切含量将取决于所选择的具体防晒剂和所期望的避阳系数(Sun Protection Factor)(SPF)。SPF通常用于度量防红斑防晒光保护效果。见Federal Register,Vol.43,No.166,pp.38206~38269,1978年8月25日。
除这些防晒剂外,组合物还可含有一种或几种人造晒黑成份例如二羟基丙酮,酪氨酸,氨基酸,及氨基酸衍生物,一般可加入到本发明组合物中的人造晒黑成份的含量大约为0.1%至约10%,优选约0.1%至大约5%。
增稠剂
本发明组合物中另外任意成份是增稠剂。可使用的增稠剂的例子包括(但不局限于)黄原胶,硅酸铝镁,瓜耳胶,阳离子瓜耳胶,Rhamson Gum(由Kelco化学公司购得),海草灰,藻酸铵,藻酸盐,淀粉和淀粉衍生物,羟丙基纤维素,羟乙基纤维素,羧甲基纤维素。甲基纤维素,乙基纤维素,绿土粘土增稠剂例如水辉石和膨润土,硅酸钠镁及其混合物。适当增稠剂的例子公开在Lochhead,R.Y.的“聚合物和增稠剂百科全书”,Cosmetics & Toiletries,Vol.103,no.12,pp.99-129(1988);Meer,G.的“用作化妆品成份的天然胶聚合物”,Cosmetics & Toiletries,Vol.99,no.6,pp.61-64(1984);以及Freeland M.S.的“阳离子瓜耳胶”,Cosmetics & Toiletries,Vol.99,no.6,83~87(1984)这些文献中;这三篇文章全文均作本发明参考。优选的增稠剂包括硅酸铝镁和黄原胶及其混合物。本发明组合物含有大约0.1%至大约5%的增稠剂,优选大约0.25%至大约2%,最优选约0.5%至约1%。
润湿剂/增水剂
本发明组合物还可任意含有一种或几种润湿剂/增水剂。可使用多种润湿剂/增水剂其含量范围大约0.5%至约30%,优选约2%至大约8%,最优选约3%至大约5%。这些物质包括脲;胍;甘醇酸和甘醇酸盐(例如铵和四烷基铵盐);乳酸和乳酸盐(例如铵和四烷基铵盐);多羟基醇例如山梨醇,甘油,己三醇,丙二醇,己二醇等等;聚乙二醇;糖和淀粉;糖和淀粉衍生物(例如烷氧基化的葡萄糖);panthenol;透明质酸、乳酰胺单乙醇胺;乙酰胺单乙醇胺;及其混合物。
用于本发明组合物的优选湿润剂/增水剂是C3-C5二醇和三醇。特别优选三醇,甘油。
润肤剂
本发明组合物还可任意包含至少一种润肤剂。适当润肤剂的例子包括(但不局限于)挥发性和非挥发性硅油,高支化烃,和非极性脂肪酸的脂肪醇酯,及其混合物。用于本发明的润肤剂在1990年4月24日公开的Deckner等人的专利U.S.4919934中有更详细描述,本发明将其全文作为参考。
一般含有润肤剂的总量是大约1%至50%,优选约1%至大约25%,更优选占本发明组合物重量的大约1%至大约10%。
乳化剂
本发明组合物的另一任意成份是至少一种乳化剂。适当乳化剂包括任何公开于先前专利和其它参考文献中的大量非离子,阳离子,阴离子和两性离子乳化剂,见由Allured出版公司出版的McCutcheon的Detergents and Emulsifiers,北美版(1986);1991年4月30日公开的Ciotti等人的专利U.S.5011681;1983年12月20日公开的Dixon等人的专利U.S.4421769;1973年8月28日公开的Dickert等人的U.S.3755560;本申请均将该四篇文献全文作为参考。
适当的乳化剂类型包括甘油酯,丙二醇酯,聚乙二醇脂肪酸酯,聚丙二醇脂肪酸酯,山梨醇酯,脱水山梨醇酐的酯,羧酸共聚物,葡萄糖的酯和醚,乙氧基化醚,乙氧基化醇,烷基磷酸酯,聚氧亚乙基脂肪醚磷酸酯,脂肪酸酰胺,酰基乳酸酯,皂及其混合物。
适当的乳化剂可以包括(但不局限于)聚乙二醇20脱水山梨醇单月桂酸酯(Polysorbate 20)、聚乙二醇5大豆甾醇、Steareth-20、Ceteareth-20、PPG-2、甲基葡萄糖醚二硬脂酸酯、Ceteth-10、Polysorbate 80、鲸蜡醇磷酸酯,鲸蜡醇磷酸酯钾,鲸蜡醇磷酸酯二乙醇胺、Polysorbate 60、硬脂酸甘油酯、PEG-100硬脂酸酯、及其混合物。
乳化剂可单独使用或以一种或几种的混合物使用,其含量为本发明组合物的大约0.1%至大约10%,优选约1%至大约7%,最优选大约1%至大约5%。
维它命
本发明组合物也可任意含有各种维生素。可应用的非限制性例子包括维生素A,及其衍生物,抗坏血酸,维生素B,维生素H,维生素D,维生素E及其衍生物例如乙酸生育酚酯,遍多酸(Panthothenic acid)及其混合物。
羧酸共聚物
本发明组合物中另一任意成份是羧酸共聚物(丙烯酸共聚物)。最优选Carbomer1342(以Carbopol 1342商标由B.F.Goodrich购买)。这些聚合物在1985年四月五日公开的Huang等人的U.S.4509949和1957年7月2日公开的Brown的专利U.S.2798053中有更全面的描述,本发明将这些专利作为参考。还可应用丙烯酸酯/烷基丙烯酸酯交联聚合物例如Acrylates/C10-C30Alkyl Acrylate Crosspolymer(以Pemnlen TR-1和Pemulen TR-2商标由Goodrich可购买到)。参考Include Pemulens。
这些聚合物的含量是大约0.025%至大约0.75%,优选0.05%至大约0.25%,最优选大约0.075%至大约0.175%。
其它任意成份
本发明乳液组合物可以包含各种附加成份,这些附加成份的非限制性例子包括各种有助于组合物的成膜性和牢固性的聚合物(例如由GAF化学公司购买的商品名为Ganex V-200R的二十碳烯与乙烯吡咯烷酮的共聚物);胶,树脂,和增稠剂;为保持组合物的抗微生物完善性的防腐剂,抗氧化剂;螯合剂和多价螯合剂;抗痤疮剂;溶角蛋白剂;及适于审美目的的试剂例如香味,颜料和彩色。其它有用材料包括酸性物质例如水杨酸,乳酸,乙醇酸,苯甲酸,柠檬酸等等,不受理论上限制,人们相邻这些酸类物质可用来维持组合物的pH值并增强产品性能。
防止晒斑的方法
本发明还进一步涉及防止人类或较低级动物的皮肤受UVA和UVB波长射线有害作用(例如晒斑和皮肤过早衰老)的方法。该方法通常包括在人或较低级动物皮肤上施以一层防晒剂或本发明组合物有效涂层。这里所用术语“有效涂层”是指足以实质上减少到达皮肤表面的UVA和UVB波长光的量的防晒剂膜。一般皮肤有效涂层是大约0.5mg防晒剂或本发明组合物/cm2皮肤至大约5mg防晒剂或本发明组合物/cm2皮肤。见Federal Register,Vol.43,No.166,pp.38206-38269,1978年8月25日。
下面的实施例进一步描述和演示本发明范围内的优选实施方案。这些实施例只是为了描述的目的,而并不构成对本发明限制。因为在不违背其精神和其范围内可进行许多改变。

UV-可见光谱:λmax366nm,A=1.3502(10ppm于50∶50 DMSO:氯仿中)
实施例2
使用单醋酸铝制备4,4′-甲氧基-叔-丁基-二苯甲酰甲烷(Ⅲ)配合物
在1000ml烧瓶中,于大约300ml乙醇中加入33.75克(0.240moles)单醋酸铝(碱性,用硼酸稳定,Aldrich化学公司,Milwaukee,WI),并加热至沸点保持大约10分钟。在另外一500ml烧瓶中,在大约200ml丙酮中加入106.33g(0.343moles)4,4′-甲氧基-叔-丁基-二苯甲酰甲烷酸与4-羟基二苯甲酰甲烷的酯,加热至沸点并保持大约10分钟。然后向该防晒剂丙酮溶液中加入80ml11%(W/W)氢氧化钠水溶液,将混合物剧烈搅拌至均匀。然后边搅拌边向单醋酸铝溶液中缓慢加入该防晒剂溶液,混合物回流大约10分钟。旋转蒸发去除溶剂得到防晒剂铝配合物粗产物。将此粗产物配合物重新溶于大约300ml二氯甲烷中并加热,然后用粗孔烧结玻璃滤器真空抽滤去除任何不溶物,用二氯甲烷洗涤(大约2×50ml)。用水萃取汇集的滤液,用硫酸钠干燥。用旋转蒸发器蒸发得到防晒剂铝配合物,为浅黄色玻璃样固体。该固体适用于配制防晒组合物。
实施例3
使用二硬脂酸铝制备4,4′-甲氧基-叔-丁基二苯甲酰甲烷铝(Ⅲ)配合物
在500ml烧瓶中,于100ml苯甲酸C12-15醇酯中加入2.0克(3.28moles)二硬脂酸铝(Aldrich化学公司,Milwaukee,WI),将混合物加热至90℃直到溶解。然后边搅拌边加入1.1克(3.78mmoles)4,4′-甲氧基-叔-丁基-二苯甲酰甲烷生成防晒铝配合物。该防晒金属配合物的油相溶液适于配制乳状或凝胶状组合物。(另外可用于该制备过程的其它溶剂包括二甲基异山梨酐,新戊酸异癸酯,和己二酸二异丙酯。)
实施例4
用二辛酸铝制备4,4′-甲氧基-叔-丁基-二苯甲酰甲烷铝
(Ⅲ)配合物
用实施例3中的方法,以1.0克二辛酸铝(Aldrich化学公司,Milwaukee,WI)为起始物制备防晒铝配合物。所得配合物油相溶液适于配制乳状或凝胶状组合物。
实施例5
用单乙酸铝制备4-异丙基二苯甲酰甲烷铝(Ⅲ)配合物
用实施例2的方法,用33.75克(0.340moles)单乙酸铝和91.24克(0.343moles)4-异丙基二苯甲酰甲烷制备防晒铝配合物。得到适合配制防晒组合物的浅黄色固体。
实施例6
含有4,4′-甲氧基-叔-丁基二苯甲酰甲烷的油基质胶状组合物。
在加热至90℃时,将4.0克实施例1中制备的防晒铝配合物加至100克苯甲酸C12-15醇酯中。将混合物冷却至80℃并加入1克二丁基月桂酰谷氨酰胺(商品名Coagulen GP-1,由Ajinomoto Co.Inc.,Tokyo,Japan得到)使混合物在冷却时凝胶化。这种胶状组合物施用于皮肤表面,保护皮肤不受紫外线的有害作用。
另外,可用本发明其它防晒金属配合物和其它溶剂例如新戊酸异癸酯,己二酸二异丙酯,和二甲基异山梨酐制备类似胶状组合物。
实施例7
以4,4′-甲氧基-叔-丁基二苯甲酰甲烷铝(Ⅲ)配合物通过原位制备防晒乳剂。
用标准方法由下面组份制备乳状组合物。

这种乳状液组合物可于皮肤表面施用提供防止紫外线有害影响的保护作用。
或者可使用0.50克二辛酸铝代替二硬脂酸铝制备上述防晒乳剂。
实施例8
用离析出的4,4′-甲氧基-叔-丁基二苯甲酰甲烷铝(Ⅲ)配合物制备防晒乳剂。
用标准方法由下面组份制备乳剂组合物。

用离析的4,4′-甲氧基-叔-丁基二苯甲酰甲烷制备防晒乳剂。

甲基异噻唑啉酮 0.100
将A相各成份结合并在搅拌下加热至80℃形成油相。然后,将B相各成份结合并在搅拌下加热至80℃。然而将A相加入至B相并经匀浆处理生成乳状液,然后搅拌下将其冷却至室温。
该乳状组合物表面施与皮肤提供防止紫外线有害损伤的保护作用。
或者,可应用本发明其它离析的防晒金属配合物制备类似乳剂。
Claims (20)
1、具有如下通式结构的防晒金属配合物:
其中:
(A)(S)是具有下面通式结构的防晒基团:
其中A是选自H,-OR,或-NR2的取代基,其中每个R各自独立地为H,具有大约1至约20个碳原子的直链或支链烷基,
(CH2CH2O)q-H,或(CH2CH(CH3)O)q-H,其中q是1至大约8的整数;而B是选自H,具有大约1至大约20个碳原子的直链或支链烷基,(CH2CH2O)q-H,或
(CH2CH(CH3)O)q-H,其中q是1至大约8的整数;
(B)m是选自1,2,3或4的整数;
(C)M是金属阳离子,铵阳离子,或取代的铵阳离子;
(D)n是选自1,2,3或4的整数;
(E)L是具有中性或负电荷的有机或无机基基团的配位体;且
(F)p是选自0,1,2,3或4的整数。
2、根据权利要求1的防晒配合物,其中A是对-OCH3,而B是对-叔丁基,或其中A是H而B是对-异丙基。
3、权利要求2中列举的防晒配合物,其中M是选自下组的金属阳离子:铝,锌,钙,镁,铜,铁,钡,锶,锆,钛,锡,铍,镓,铟,镧,锰,锑,铋,铈,钍,铌,钽,钼,钨,锂,钠,钾及其混合物。
4、根据权利要求3的防晒配合物,其中所述金属阳离子具有1,2,3或4的价态。
5、根据权利要求4的防晒配合物,其中所述金属阳离子选自铝,钛,铜,铁,锌,及其混合物。
6、根据权利要求5的防晒配合物,其中所述金属阳离子是价态为3+的铝。
7、权利要求6列举的防晒配合物,其中所述配位体L选自下组配位体:羧酸,二羧酸,和多羧酸及其阴离子;胺,二胺,和多胺;醇,二醇,和多醇及其阴离子;硫醇,二硫醇,和多硫醇及其阴离子;氨基酸及其阴离子;或选自下组无机配位体:水和羟基阴离子,氟化物、氯化物、溴化物、碘化物、碳酸盐、碳酸氢盐、磷酸盐、磷酸氢盐、磷酸二氢盐、硫酸盐、硫酸氢盐、硝酸盐、亚硫酸盐、亚硝酸盐、硼酸盐、氯酸盐、溴酸盐、过氯酸盐、过溴酸盐、二磷酸盐、多磷酸盐、硫氰酸盐,及其混合物。
8、根据权利要求7的防晒配合物,其中所述配位体L选自下组:水,羟基阴离子,羧酸,羧酸阴离子,及其混合物。
9、根据权利要求8的防晒配合物,其中所述羧酸或羧酸阴离子具有大约2至大约22个碳原子。
10、根据权利要求9的防晒配合物,其中所述羧酸或羧酸阴离子选自乙酸或乙酸根阴离子,辛酸或辛酸根阴离子,硬脂酸或硬脂酸根阴离子,及其混合物。
11、根据权利要求10的防晒配合物,其中m是选自1,2,或3的整数,而n是1。
12、根据权利要求11的防晒配合物,其中m是2。
13、选自下组的防晒配合物:4,4′-甲氧基-叔-丁基二苯甲酰甲烷铝(Ⅲ)配合物、4-异丙基二苯甲酰甲烷铝(Ⅲ)配合物、4,4′-甲氧基-叔-丁基二苯甲酰甲烷铁(Ⅲ)配合物、4-异丙基二苯甲酰甲烷铁(Ⅲ)配合物、4,4′-甲氧基-叔-丁基二苯甲酰甲烷铜(Ⅱ)配合物、4-异丙基二苯甲酰甲烷铜(Ⅱ)配合物,及其混合物。
14、一种防晒组合物,含有:
(a)大约0.1%至大约30%的一种或几种权利要求1的防晒配合物;
(b)0%至大约20%选自下组的第二种防晒剂或防晒剂混合物:P-甲氧基肉桂酸2-乙基己酯、N,N-二甲基-P-氨基苯甲酸2-乙基己酯、P-氨基苯甲酸、octocrylene、2-苯基苯并咪唑-5-磺酸、oxybenzone、水杨酸高薄荷酯、水杨酸辛酯、Sulisobenzone、P-甲氧基肉桂酸戊酯、N,N-二甲基-P-氨基苯甲酸戊酯、丁基甲氧基二苯甲酰甲烷、4-异丙基二苯甲酰甲烷、2,4-二羟基苯酮的4-N,N-(2-乙基己基)甲基氨基苯甲酸酯;4-羟基二苯甲酰甲烷的N,N-二-(2-乙基己基)-4-氨基苯甲酸酯;4-羟基二苯甲酰甲烷的4-N,N-(2-乙基己基)甲基-氨基苯甲酸酯;2-羟基-4-(2-羟乙氧基)苯酮的4-N,N-(2-乙基己基)甲基氨基苯甲酸酯;4-(2-羟乙氧基)二苯甲酰甲烷的4-N,N-(2-乙基己基)甲基氨基苯甲酸酯;2-羟基-4-(2-羟乙氧基)苯酮的N,N-二-(2-乙基己基)-4-氨基苯甲酸酯和4-(2-羟乙氧基)二苯甲酰甲烷的N,N-二-(2-乙基己基)-4-氨基-苯甲酸酯;二氧化钛、氧化锌、氧化铁,及其混合物;和
(c)药学上可接受的防晒载体。
15、根据权利要求14的防晒组合物,其中所述载体是水包油乳液。
16、根据权利要求14的防晒组合物,其中所述载体是油包水乳液。
17、根据权利要求14的防晒组合物,其中所述载体是油。
18、根据权利要求14的防晒组合物,其中所述载体是水基质的洗剂或胶。
19、根据权利要求14的防晒组合物,其中所述载体是无水胶。
20、保护人或较低级动物皮肤免受UVA和UVB波长射线伤害的方法,所述方法包括大体上日常基础上表面施与人或较低级动物皮肤权利要求14防晒组合物的有效涂层。
