Abstract
本发明公开了一种米糠壳的提取产物及其在面部护理的应用,具体公开了一种米糠壳的提取产物的提取方法,此方法效率高,成本低,易于加工生产。还公开了以该米糠壳的提取产物为原料组成的一种高保湿且修护皮肤屏障的组合物,应用于面部肌肤时,能增加皮肤角质层厚度,提高皮肤的持水能力,减少皱纹,增加皮肤弹性,延缓皮肤衰老。

技术领域
本发明涉及化妆品领域,具体涉及一种米糠壳的提取产物以及含有该提取物的一种高保湿且修护皮肤屏障的组合物。
背景技术
随着现代社会的发展,人们对于皮肤的重视程度也越来越高,也越来越追求更“天然无刺激”的化妆品原料。因为在外界环境的影响下,我们的皮肤也或多或少的受到刺激。根据调查,60%以上的亚洲女性和50%的欧洲女性都自我感觉是“敏感性皮肤”。这类人群的皮肤特征是:容易过敏,脸上的红血丝明显皮肤容易泛红,容易受坏境因素、季节变化及面部保养品刺激等等。
大部分关于敏感皮肤的文献集中于这样一个假说,即敏感性皮肤产生的原因是由于角质层屏障的受损或者化合物对皮肤渗透的提高所致。由此,皮肤屏障功能受损已经成为人们普遍接受的敏感性皮肤产生的机理。
不管活性分子是经过细胞途径,或细胞间途径渗入,分子总是需要通过第一道双层膜——填充角质层角蛋白细胞之间空间的脂质双层。构成这些脂质双层的脂质主要是由神经酰胺,胆固醇,游离脂肪酸组成。使得极性分子渗入皮肤的屏障主要位于角质层。
屏障功能的修复针对的是本身不能保持足够水分的皮肤,也就是角质层的一个恢复能力的过程。角质层是皮肤最上面的一层,也是唯一能够保持水分和挡住外界污染和颗粒入侵的一层。
那么角质层的防护力来自于三个方面:角化细胞,细胞间脂质和皮肤脂质。
屏障修复就是要让以上这三个恢复正常。使其恢复正常有以下方法:1.用内部含有的油脂停留在皮肤表面,让本来不断失去水分的角质层暂停失水,留时间恢复自己的屏障能力。2.包含细胞间脂质的重要组成成分——角鲨烷,神经酰胺,可让皮肤迅速恢复“细胞间脂质”的能力。
在现阶段的保湿乳液一般是针对肌肤表面补水的概念,很少有根据皮肤屏障损害和角质层水分流失相关的皮肤问题所发明的产品,也很少有一种特定的天然原料能有效的修护肌肤屏障。
因此,发明一个具有“天然原料”又能修复肌肤屏障的乳液,对于护理化妆品技术的发展以及提高人们的生活水平具有重要意义。
而米糠壳提取物中含有丰富的神经酰胺,来自米糠的神经酰胺主要以鞘糖脂形式存在,其酰胺连接的脂肪酸比较常见,分别是亚油酸,油酸,硬脂酸和棕榈酸。这是一种水溶性脂质物质,它和构成皮肤角质层的物质结构相近,能很快渗透进皮肤,和角质层中的水结合,形成一种网状结构,锁住水分。
神经酰胺是存在于人体皮肤最外侧的角质层细胞间类脂体的主要构成成分,起着防止水分散发及对外部刺激有防护功能的重要作用,承担着保护皮肤和滋润、保湿功能,神经酰胺具有很强的缔合水分子能力,它通过在角质层中形成的网状结构来维持皮肤的水分。神经酰胺可在表皮角质层中形成网状结构来缔合水分子,保持皮肤的水分。因此,神经酰胺对皮肤具有很强的保湿作用。
皮肤在衰老过程中,脂质合成下降,角质层中神经酰胺含量减少,表现为皮肤干燥,脱屑,粗糙,失去光泽;使用神经酰胺能使皮肤角质层中神经酰胺含量增高,可改 善皮肤干燥、脱屑、粗糙等症状;同时,神经酰胺能增加皮肤角质层厚度,提高皮肤的持水能力,减少皱纹,增加皮肤弹性,延缓皮肤衰老。
发明内容
本发明所要解决的技术问题是:为了克服很少有根据皮肤屏障损害和角质层水分流失相关的皮肤问题所发明的产品与体验感油腻,本发明分析了皮肤屏障损害和角质层水分流失的机理,目的是提供一种米糠壳提取物的提取技术以及含有该米糠壳提取物的面部护理产品,本发明公开的提取技术能够保留米糠壳提取物含有的神经酰胺,来自米糠的神经酰胺主要以鞘糖脂形式存在,它和构成皮肤角质层的物质结构相近,能很快渗透进皮肤,和角质层中的水结合,形成一种网状结构,锁住水分。
通过该米糠壳提取物的应用,并且通过对组合物体系架构的合理搭建以及对原料关键参数的限制,可修复皮肤屏障,在皮肤表面形成一层保护膜,并有效减少经皮肤水分流失率,使得面部护理产品修复皮肤屏障的同时还有优异的保湿性能。
本发明所要解决的技术问题通过以下技术方案予以实现:
一种米糠壳的提取产物, 由以下工序制作而成:
a.将米糠壳经过高速剪切粉碎成粉末状,将10-20重量份数的米糠壳粉末置于反应釜中;
b.用85℃~90℃的30-45重量份数的热纯水浸泡30-60分钟,搅拌1小时~3小时离心,用200目滤袋过滤后除去滤渣得第一滤液,转移至另外一个反应釜,;
c.向第一滤液中加入共0.1-5重量份数的纤维素酶,蛋白酶,淀粉酶进行浸泡60-90min,离心30min,过滤得第二滤液;
d. 第二滤液经循环装置流至脱色装置,循环脱色20-40分钟;
e.脱色后所得的溶液进行蒸馏浓缩,蒸馏时间为2~8h,蒸汽经冷凝装置冷凝;
f.将浓缩后的溶液放入水浴振荡器中,调节温度70~80℃,提取40~50min,调节pH至6.8-7.3;
g.在干燥箱调节温度为60℃,把上层清液放置干燥箱干燥6-8小时得成品。
一种高保湿且修护皮肤屏障的组合物,包括如下重量百分数的组分:
上述的米糠壳提取物0.5%-15%,
多元醇2%-6%,
成膜剂0.05%-0.1%,
透明质酸0.05%-0.2%,
增稠悬浮剂0.2%-3%,
乳化剂2.5%-4.5%,
烟酰胺0.1%-5%,
油脂2-11%,
余量为水。
在体系中添加增稠悬浮剂有增稠,悬浮稳定等功效,可增加体系的粘稠度,同时也提高了体系的稳定性。
所述的透明质酸是皮肤和其它组织中广泛存在的天然生物分子,具有极好的保湿作用,被国际上称为理想的天然保湿因子。它是目前自然界中发现的化妆品用保湿性能最好的物质。同时,透明质酸也是人皮肤表皮及真皮的主要基质成分之一,其生理功能是能使水分进入细胞间隙,并与蛋白质结合而形成蛋白凝胶,将细胞粘在一起,发挥正常的细胞代谢作用,起到保持细胞水分,保护细胞不受病原菌的侵害,加快恢复皮肤组织,提高创口愈合再生能力,减少疤痕,增强免疫力等作用。透明质酸还能改善皮肤生长条件,为真皮胶原蛋白和弹性纤维的合成提供优越的外部环境,加强营养物质的供给,起到护肤养颜的效果。研究表明,透明质酸HA可以阻止细胞中一些酶的产生,减少自由基的形成,在防止自由基破坏细胞结构、产生脂质过氧化和引起肌体衰老等方面起着重要作用:低分子量透明质酸具有抗炎、抑制病菌产生、保持皮肤光洁的作用;为细胞增殖与分化提供合适的场所,直接促进细胞生长、分化、重建与修复等。
所述的烟酰胺能够显著地增加糖基神经酰胺和神经鞘磷脂含量,并且能够增强丝氨酸——棕榈酰转移酶的活性,同时还可增加游离脂肪酸的合成。总之烟酰胺可以通过刺激神经酰胺合成的全过程和上调丝氨酸转移酶的活性改善皮肤的可渗透性的屏障。研究表明,人类志愿者干燥皮肤上局部应用烟酰胺增加了角质层神经酰胺和游离脂肪酸的水平,同时胆固醇水平也趋于增加。角质层细胞间脂质水平的增加,尤其是神经酰胺水平的增加,可改善表皮的受损的渗透性屏障功能。
进一步的,所述的多元醇包括乙二醇、甘油、丙二醇、聚乙二醇、聚甘油中的至少一种。
进一步的,所述的高保湿且修护皮肤屏障的组合物还包括如下重量百分数的调理剂:

所述的D-泛醇有三个羟基,生理活性较高,在皮肤泛醇转变为泛酸,泛醇以泛酸形式存在于人体活组织内,它是辅酶A关键的组分,起着乙酰化反应载体的作用。乙酸盐与辅酶A反应生成活化的乙酸盐是脂质和昼蛋白合成所必需的,呈现出维生素的生物活性。可以加强正常皮肤水合功能,有改善干燥、粗糙、脱屑、止痒以及治疗多种皮肤病相关的红斑效果。加强皮肤屏障功能外用后可以促进皮肤正常的角质化,改善皮肤对表面活性剂的耐受力。减轻化妆品中成分敏感所产生的不适像灼烧、刺痛、瘙痒和麻刺等。有助于减轻过敏症状,有助于伤口痊愈。
所述的β-葡聚糖是一种酵母细胞壁中提高免疫力的多糖,是优良的免疫激活剂,能够提高皮肤自身的免疫力,有清除自由基的功效,协助受损组织加速恢复产生细胞素,在敏感肌肤修复方面有独特的生物活性。β-1 ,3-葡聚糖在化妆品中的应用增强肌肤免疫力,可增加受损皮肤细胞的再生能力,提高角质层的再生速率,具有帮助伤口复原的特性。临床研究也表明β-葡聚糖还具有显著的消炎、抗过敏活性,并能有助于皮肤抵御外源性的各种机械和化学刺激。β-葡聚糖在老龄皮肤和皱纹皮肤上增加表皮生长因子( EGF),将促进皮肤中胶原蛋白和弹性蛋白增加,从而改善皮肤外观和祛除皱纹。紫外线照射对皮肤内的朗格罕氏细胞具有抑制作用,如肌肤长时间暴露在阳光下,朗格罕氏细胞数量会大大减少,活性降低,削弱了皮肤的防御免疫能力。而β-葡聚糖除自身具有防晒作用并能减少疼痛炎症外,它还能促进朗格罕氏细胞增殖,能增强皮肤的免疫能力。β-葡聚糖能激活免疫系统中的基础细胞---巨噬细胞,巨噬细胞产生表皮生长因子( EGF),从而促进伤口愈合所必需的胶原蛋白产生,同时血管生成因子(AF) 可促进伤口愈合必需的新血管形成。
所述的维生素E在皮肤应用于预防角质化,有美肤的功效。清除自由基的肌肤自然就健康。维生素E可中和自由基,将因日晒、污染、压力产生的自由基消除,保护肌肤组织。它可以照顾肌肤的根本,让肌肤不会早早出现细纹、松弛的状况,还能促进皮肤微血管循环。
所述的甘草酸二钾这类物质可以起到类似于肾上腺皮脂激素的作用,而又没有任何的副作用。可以抑制皮肤的炎症反应,改善毛细血管扩张,从而让皮肤的红肿得到改善 ,有抗敏消炎作用。
所述的红没药醇是存在于春黄菊花中的一种成份,不仅具有抗炎性能,还被证明有抑菌活性用,能够保护和治愈皮肤,使其免受日常张力的影响,并加速皮肤的治愈过程,尤其适用于作为敏感皮肤和身体,被广泛应用于个人护理的配方中,加上其抗炎、安全特性,使其成为一种用于皮肤护理的常用活性成分。红没药醇的稳定性很好,同时具有很好的皮肤相容性,很适合于用在化妆品中。
进一步的,所述的成膜剂包括羧甲基脱乙酰壳多糖。所述的羧甲基脱乙酰壳多糖具有良好的成膜性。涂布于皮肤表面后, 则能形成一层生物糖膜, 通过其极强的成膜性,能显著改善皮肤表皮失水问题,对角质层保持保湿作用, 防止皮肤干燥。同时可隔离pm2.5污染物以及碳颗粒,减少了pm2.5污染物引起的氧化应激反应和线粒体毒性以及减少肌肤碳颗粒附着。
其对体外表皮角质形成细胞(HaCaT)有促进生长作用,依靠分子结构中的活性羧基 也可抗氧化,清除自由基,同时与多元醇复配也有良好的防腐增效作用。
进一步的,所述的增稠悬浮剂包括丙烯酰二甲基牛磺酸铵/VP 共聚物。此悬浮稳定剂做出的乳液外观细腻,柔亮,肤感清爽。
进一步的,所述的乳化剂包括十六醇和硬脂酸甘油酯、PEG-75硬脂酸、十六醇醚-20、氢化卵磷脂其中的任意一种或任意两种。氢化卵磷脂作为一种优良的天然乳化剂,对人体皮肤具有保护作用。它是皮肤细胞的固有成分,在细胞代谢和细胞膜渗透性调节方面起着重要作用,且对皮肤还有一定的生理作用,增加皮肤的胆固醇含量,在皮肤表面形成了一层皮肤屏障,修复角质层,减少皮肤刺激。同时还具有保湿功能,与其它原料具有协同增效作用。十六醇和硬脂酸甘油酯和PEG-75硬脂酸和十六醇醚-20和氢化卵磷脂搭配肤感柔软,外观光亮。
进一步的,所述的油脂为包括天然角鲨烷、霍霍巴油、硅油的组合。所述的角鲨烷被皮肤吸收后,会变成角鲨烯,再经过一些变化成为胆固醇,角质层中有脂质,就是由“神经酰胺”,“游离脂肪酸”和“胆固醇”构成的。所以我们在使用角鲨烷一段时间后皮肤就会产生一些新的“胆固醇”,从而加强屏障功能。
所述的霍霍巴油取自西蒙得木果实。 主要成分是不饱和高级醇和脂肪酸,有良好的稳定性,极易与皮肤融合,具有超凡的抗氧化性。另外,霍霍巴油还含有丰富维生素,具有滋养软化肌肤的功效。霍霍巴油的成分和海洋中抹香鲸蜡油的成分相似,是其唯一的代用品。霍霍巴油还是一种十分优质的基础油。霍霍巴油除了可以供给皮肤所需要的营养成分,而且对于皮肤也有一定得理疗效果。霍霍巴油的具体结构为一种酒精聚合物与脂肪酸的结合,因此其结构异常的稳定,不会被高温破坏,且这种成分与皮肤表面的油脂性质极相近,所以不像一般含甘油的保养品会在皮肤表层形成油膜,霍霍巴油对皮肤提供两层作用:迅速渗透吸收与软化皮肤角质,让皮肤变得柔软有弹性。霍霍巴油非常稳定。它不会像油脂一样容易酸败,哪怕在温度很高的情况下。实际上它是一种液体的蜡酯。和如甜杏仁、葡萄籽油类相比最大的不同就是它不含有甘油三酸酯,这种东西如果分解了,油脂就会酸败。而霍霍巴油不含有这种组分,所以寿命比油脂类长很多。 因为它的质地清爽细腻,它和皮脂的组分接近,所以具有相当好的亲肤性,可以帮助清洁毛孔,可以迅速被皮肤吸收,所以油性皮肤和痘痘皮肤不需要担心,霍霍巴油绝对不会给皮肤造成负担,霍霍巴油还可以软化角质,让皮肤变得光滑有弹性。霍霍巴油的作用除了上述的抗炎以外,它也很适合特殊时期的皮肤护理。
添加硅油后具有以下优点:硅油无色无味,不影响最终制品的外观及香气;使用有轻松感,展开性好;疏水性和光滑性好;对皮肤无刺激,对人体安全性高;气体透过性好,不妨碍皮肤呼吸;化学稳定性好,与其它化妆品原料不发生反应。在皮肤护理制品中加入二甲基硅油,能提高化妆品对皮肤的展开性,起到保护皮肤的效果;目前硅油己成为许多化妆品的重要成分或助剂,硅油具有许多独特的性质,加入化妆品中可增加起舒适感并赋予光泽。
对于健康皮肤,角质层是紫外线,热或冷,细菌,机械破坏和很多化学品的屏障,然而一些油类和醇类很容易渗透入角质层。因而很多皮肤制品中含有它们,以它们作为基质,也有助于载带一些活性物质到达较深层的细胞。
进一步的,所述的硅油包括二甲基硅氧烷、聚二甲基硅氧烷醇中的至少一种。
上述的一种高保湿且修护皮肤屏障的组合物,除上述必须的原料之外,还可根据实际需要加入本领域常规辅助原料,例如酸碱类物质、防腐剂、香精等。
上述的一种高保湿且修护皮肤屏障的组合物,其配置方法为常规的配置和混合技术,在此不做详细说明。
本发明具有如下有益效果:
1.本发明所述的米糠壳提取物在面部护理产品中,通过米糠壳提取物和烟酰胺的合理搭配,在配方中协同增效,可增加皮肤中各类神经酰胺的生成。研究发现,神经酰胺在角质层中的水平在特应性皮炎和老化的皮肤中减低。其中配方中米糠壳提取物中的神经酰胺和烟酰胺的组合可在表皮屏障的稳态维持中发挥重要作用。含神经酰胺的配方能够改善受损害的皮肤状态。
2.本发明所述的米糠壳提取物在面部护理产品中,通过不同成分的比例和组合,可针对多种角质层和表皮损害相关的皮肤病变开发出新的局部治疗性配方。配方中米糠壳提取物中的神经酰胺与皮肤中的这些成分相同的脂质,富含神经酰胺、游离脂肪酸和胆固醇,是构成表皮渗透性屏障的必要成分。
3.本发明所述的米糠壳提取物在面部护理产品中,通过米糠壳提取物,油脂与消炎抗敏剂的合理搭配,在各个方面补充皮肤中所需的油脂和水分。油脂,透明质酸和成膜剂在皮肤表面形成一层保护膜,让本来不断失去水分的角质层暂停失水,留时间恢复自己的屏障能力,并修复肌肤中受损的屏障。同时这些油脂中包含细胞间脂质的重要组成成分——角鲨烷,脂肪酸,可让皮肤迅速恢复“细胞间脂质”的能力。同时以油脂作为基质,有助于载带一些消炎抗敏剂和米糠壳提取物中的神经酰胺到达较深层的细胞,修复肌肤屏障。
具体实施方式
下面结合试验例和具体实施方式对本发明做进一步的详细描述。但不应将此理解为本发明上述主题的范围仅限于以下的实施例,凡基于本发明内容所实现的技术均属于本发明的范围。
除非另外指明,所有的百分比、份数和比例均基于与所列成分相关的含量重量计,并因此不包括可能包含在可商购获得的原料中的载体或副产物。
所述的米糠壳提取物由以下工序制作而成:
a.米糠壳经过高速剪切粉碎成粉末状,将10-20重量份数的米糠壳粉末置于反应釜中;
b.用85℃~90℃的30-45重量份数的热纯水浸泡30-60分钟,搅拌1小时~3小时离心,用200目滤袋过滤后除去滤渣得第一滤液,转移至另外一个反应釜,;
c.向第一滤液中加入共0.1-5重量份数的纤维素酶,蛋白酶,淀粉酶进行浸泡60-90min,离心30min,过滤得第二滤液;
d. 第二滤液经外循环泵流至带有活性炭的脱色装置,循环脱色20-40分钟;
e.脱色后所得的溶液进行蒸馏浓缩,蒸馏时间为2~8h,蒸汽经冷凝装置冷凝;
f.将浓缩后的溶液放入水浴振荡器中,调节温度70~80℃,提取40~50min,调节pH至7;
g.在干燥箱调节温度为60℃,把上层清液放置干燥箱干燥6-8小时得成品。
用此工艺制得的米糠壳提取物成品收率为15-30%,纯度为92~96%。
下表 1 为本发明 3 个优选的实施例与3个对比例的配方组成:

制备上述3组实施例和4组对比例的制备方法如下:
将A相物质称量至烧杯,搅拌混合均匀,升温至80℃,将B相物质称量至另一烧杯,升温至80℃至呈均匀油状液;将A相物质均质至凝胶状后,将B相物质倒入A相混合均质5 min;搅拌桨搅拌至混合物温度降至40℃以下,分批次加入C相物质,并搅拌均匀,后再分批次加入D相物质,继续搅拌降温至30℃以下,停止搅拌,得成品。
对上述 8个组合物的功效进行仪器功效评估、人体感官评价两种方法进行功效评估,任意挑选 320 个志愿者,经过入组后,每组为 35 个志愿者,最终得出如表 2 所示平均结果。

对比例1不添加天然角鲨烷和氢化卵磷脂,不能和其他组分一起协同增效补充与构成表皮渗透性屏障相同的脂质,皮肤水分含量明显降低,经表皮平均水分流失率也明显增高。
对比例2不添加甘草酸二钾,红没药醇,β-葡聚糖和D-泛醇这几个消炎抗敏剂,不能在皮肤达到消炎作用,这些消炎抗敏剂在皮肤不仅具有抗炎性能,还有抑菌活性用,能够保护和治愈皮肤,使其免受日常张力的影响,并加速皮肤的治愈过程。所以对比例2的水分含量明显降低,短期经表皮平均水分流失率也在升高。
对比例3不添加米糠壳提取物,烟酰胺,壳多糖和透明质酸,烟酰胺和米糠壳提取物中的神经酰胺在配方中协同增效,可增加皮肤中各类神经酰胺的生成。米糠壳提取物和烟酰胺的组合可在表皮屏障的稳态维持中发挥重要作用。能够改善受损害的皮肤状态。羧甲基脱乙酰壳多糖和透明质酸具有良好的成膜性。涂布于皮肤表面后, 则能形成一层生物糖膜, 通过其极强的成膜性,能显著改善皮肤表皮失水问题,并能对角质层保持保湿作用,防止皮肤干燥。所以对比例3的水分含量明显降低,短期经表皮平均水分流失率也在升高。
Claims (9)
1.一种米糠壳提取产物,其特征在于,,由以下工序制作而成:
a.将米糠壳经过高速剪切粉碎成粉末状,将10-20重量份数的米糠壳粉末置于反应釜中;
b.用85℃~90℃的30-45重量份数的热纯水浸泡30-60分钟,搅拌1小时~3小时离心,用200目滤袋过滤后除去滤渣得第一滤液,转移至另外一个反应釜,;

5.根据权利要求2或3所述的高保湿且修护皮肤屏障的组合物,其特征在于,所述的成膜剂包括羧甲基脱乙酰壳多糖。
6.根据权利要求2或3所述的高保湿且修护皮肤屏障的组合物,其特征在于,所述的增稠悬浮剂包括丙烯酰二甲基牛磺酸铵/VP 共聚物。
7.根据权利要求2或3所述的高保湿且修护皮肤屏障的组合物,其特征在于,所述的乳化剂包括十六醇和硬脂酸甘油酯、PEG-75硬脂酸、十六醇醚-20、氢化卵磷脂其中的任意一种或任意两种。
8.根据权利要求2或3所述的高保湿且修护皮肤屏障的组合物,其特征在于,所述的油脂为包括天然角鲨烷、霍霍巴油、硅油的组合。
9.根据权利要求8所述的高保湿且修护皮肤屏障的组合物,其特征在于,所述的硅油包括二甲基硅氧烷、聚二甲基硅氧烷醇中的至少一种。
