Abstract
一种含氨基酸衍生物的杀芽孢制剂,属消毒剂领域。含有以下量的两种必要活性组分:双癸基二甲基氧合脯氨酸铵DAPC(Proline,5‑oxo‑,ion(1‑),N‑decyl‑N,N‑dimethyl‑1‑decanaminium(1:1)),或双癸基二甲基N‑乙酰基丙氨酸铵DAAC(1‑Decanaminium,N‑decyl‑N,N‑dimethyl‑,salt with N‑acetylalanine(1:1))两者中的任一种,其重量占制剂重量的0.1%~2.0%;增效剂戊二醛,其重量占制剂重量的0.05%~1.00%;制剂中的其余量为相应剂型可以接受的非活性组分。发明制剂可高效杀灭芽孢,无毒、无刺激,性质稳定,可以长期保存。

技术领域
本发明属消毒剂领域,具体为一种含氨基酸衍生物的杀芽孢制剂。
背景技术
芽孢是代谢性休眠的微生物,其在多种环境条件下保持有生活力,是整个生物界中抗逆性最强的生命体,在抗热,抗化学药物和抗辐射等方面十分突出。由于芽孢在结构和化学成分上均有别于营养细胞,所以芽孢也就具有了许多不同于营养细胞的特性。芽孢最主要的特点就是抗性强,对高温、紫外线、干燥、电离辐射和很多有毒的化学物质,芽孢都有很强的抗性。肉毒梭菌的芽孢在沸水中要经过5至9.5小时才被杀死;巨大芽孢杆菌芽孢的抗辐射能力比E.coli细胞要强36倍。芽孢的休眠能力更为突出,在常规条件下,一般可以保持几年至几十年而不死。据文献记载,有的芽孢甚至可以休眠数百至数千年。是否能消灭芽孢是衡量各种消毒灭菌手段的最重要的指标。
由于其稳定性,在被芽孢污染的环境,尤其是医院、影院、商城等人流量大且密集的公共场所,芽孢极易传播,常常需要进行消毒。此外,随着经济发展,货物进出口贸易激增,大量货物需流通,这就使得货物在进出口时需要进行消毒,特别是货物进口,海关为防止病菌随货物侵入形成疫情,需要对货物进行有效的消毒处理。
艰难梭菌(Clostridium difficile),是一种革兰氏阳性厌氧菌,它所形成的芽孢可以导致严重并发症,表现为引起重度伪膜性结肠炎。艰难梭菌广泛分布于自然环境中,如土壤、干草、沙、以及一些大型动物(牛、驴和马)的粪便,狗、猫、啮齿动物和人的粪便中。此外,艰难梭菌还大量存在于水中和动物的肠道中。婴儿的粪便中也常有艰难梭菌,为新生儿肠道中正常菌群,大约50%的12月龄婴儿的肠道中有艰难梭菌,2岁以上儿童的带菌率大约为3%。但艰难梭菌在健康成人中出现频率较低,无症状带菌的成人在瑞典是1.9%,在日本为15.4%。艰难梭菌往往感染住院病人,并在体内定植,常导致住院病人交叉感染。
医护人员使用的乙醇消毒液是不能有效杀灭芽孢的。对于一些公共场所的消毒,大多采用含氧消毒剂、含氯消毒剂、醛类消毒剂或酚类消毒剂。含氧消毒剂如过氧乙酸、过氧化氢、过氧戊二酸、二氧化氯等。过氧乙酸,过氧化氢、过氧戊二酸不稳定、刺激性强,长期使用对人和动物眼睛、呼吸道黏膜有害,对环境有强力的破坏。含氯消毒剂,指在水中能产生具有杀菌活性的次氯酸的消毒剂,其代谢物三氯甲烷有高致癌性、绝大多数刺激性强。醛类消毒剂如甲醛、戊二醛、聚甲醛等,能产生自由醛基、在适当条件下与微生物的蛋白质及某些其他成分发生反应,其缺点是存在有机污染物时消毒效果很差,且甲醛、聚甲醛具有高度刺激性、高致癌性。酚类消毒剂如卤化酚(氯甲酚)、甲酚(煤酚皂液又称来苏儿)、二甲苯酚和双酚类、复合酚等,具有强致癌及累积毒性,酚臭味重,且对芽孢无效。
中国发明专利ZL 201210166719.3提供了一种杀芽孢制剂,但其配方需同时使用两种氨基酸双链氨基羧酸盐,添加量大,总量占制剂质量的2.4%,本发明只需使用一种氨基酸双链氨基羧酸盐,且添加量很小,仅占制剂质量的0.4%,已授权专利(专利号:ZL201210166719.3)配方中的活性物质使用量是本发明的6倍。
发明内容
本发明的目的在于提供一种由氨基酸衍生物组成的杀芽孢制剂,该制剂只需使用一种氨基酸双链氨基羧酸盐,且添加量很小就能表现出很好的杀芽孢效果。由于大幅度降低了制剂中活性物质的使用量,能进一步提高制剂的使用安全性、性质稳定性和经济性,不引起环境污染,可长期保存。
本发明制剂的特征在于:
(1)含有以下量的两种必要活性组分:
双癸基二甲基氧合脯氨酸铵DAPC(Proline,5-oxo-,ion(1-),N-decyl-N,N-dimethyl-1-decanaminium(1:1)),或双癸基二甲基N-乙酰基丙氨酸铵DAAC(1-Decanaminium,N-decyl-N,N-dimethyl-,salt with N-acetylalanine(1:1))两者中的任一种,其重量占制剂重量的0.1%~2.0%;
增效剂戊二醛,其重量占制剂重量的0.05%~1.00%;
(2)制剂中的其余量为相应剂型可以接受的非活性组分。
本发明制剂可以为液体,载体可以是水,也可以是甘油、丙烯醇、聚乙二醇、乙醇,或它们中两种或两种以上的混合物。换言之,其中的水可用去离子水、蒸馏水或纯化水,也可采用其它溶剂,如甘油、明胶甘油、聚乙二醇、乙醇、水与其它溶剂按不同比例的混合。
本发明制剂也可为膏剂或凝胶剂,载体可以是鲸蜡酯、硬脂酸、石蜡、凡士林、亲水凡士林油、白凡士林、矿物油、羊毛脂、无水羊毛脂、甘油硬脂酸或鲸蜡醇。可以是乙二醇醚及其衍生物、聚乙二醇、聚乙二醇/甘油羟基硬脂酸酯40、聚山梨醇酯或它们中两种或两种以上的混合物。
本发明制剂中可含有缓蚀剂,用以调节制剂的pH值,正常值6.4~7.7。
本发明制剂中可加入0.5%~1%重量的磷酸氢二钠和磷酸二氢钠用于降低对金属的腐蚀性。
本发明制剂中可含有氯化铵类的稳定剂。
上面所述的载体、缓蚀剂、稳定剂等均为所述的相应剂型可以接受的非活性组分,也称为辅料。
本发明制剂可以与产品组合使用。所述杀芽孢制剂可以与一种或多种常规的载体材料一起配制成可供使用的产品,具体地杀芽孢制剂可以加入到基材中或基材上,其中所述的基材如织物、吸收性基材或布料基材或纸巾基材。
DAPC(Proline,5-oxo-,ion(1-),N-decyl-N,N-dimethyl-1-decanaminium(1:1))的结构式为:

所述的氨基酸双链季氨基羧酸盐由带负电荷的氨基酸阴离子和不含卤族元素的双链季氨基阳离子构成。
所述的氨基酸双链季氨基羧酸盐,其中包含了C1-C30烷基或芳香基取代的烷。适用于本发明的脂肪族季铵盐包括但不限于:双十八烷基二甲基卤化氨(dioctadecyldimethyl ammonium halide),双癸基双辛基卤化氨(didecyldioctylammonium halide),双癸基双己基卤化氨(didecyldihexyl ammonium halide),以及癸基十八烷基十二烷基卤化氨(hexyloctyldecyldodecyl ammonium halide)。N,N,N'-四乙基双十八烷基乙烯卤化二氨(N,N,N'-tetraethyl-N,n"-di-octadecyl-1,2-ethylenediammonium halide),四乙基双十六烷基丙烯卤化二氨(N,N,N',N'-tetraethyl-N,N'-dihexadecyl-1,4-butylene diammonium halide)。
适用于本发明的氨基酸包括:亮氨酸,缬氨酸,苯丙氨酸,脯氨酸,丙氨酸,异亮氨酸,酪氨酸,甘氨酸,蛋氨酸,赖氨酸,丝氨酸,天门冬酰胺,天门冬氨酸,甲基半胱氨酸,焦谷氨酸。
例如,本发明制剂可以加入下面一种或多种辅料(其量为占制剂总重量的百分比):
缓腐蚀剂:0.5~2.0%,
双十烷基二甲基氯化铵:0.1~2.0%,
十四烷基二甲基氧化铵:0.1~1.0%,
十四烷基二甲基苄基氯化铵:0.1~2.0%,
乙醇:10.0~80.0%,
增稠剂:0.01~0.5%。
本发明的芽孢杀灭试验及相关试验。
一、枯草杆菌黑色变种芽孢(Bacillus subtilis)杀灭试验
1.主要材料
1)菌株:枯草杆菌黑色变种芽孢ATCC9372,第5代,由中国微生物菌种保藏管理委员会普通微生物中心提供。
2)中和剂:含1%卵磷脂、0.5%Na2SO3、3%吐温-80的PBS溶液。
3)待测样品:预先按实施例5中的配方1、配方2配制待测样品,若有杀菌效果,则继续往下测试下一稀释梯度待测样品的杀菌效果。
4)有机干扰物:3%牛血清白蛋白(溶解后用孔径为0.45um的微孔滤膜滤过除菌)。
5)标准硬水:见《消毒技术规范》2008版附录A
6)作用菌浓度:1×107cuf/ml~5×107cuf/ml。
2.方法
1)试验按《消毒技术规范》(2008年版),第2.1.2.2、2.1.2.3、2.1.2.5及2.1.2.7项“悬液定量杀灭试验法”进行。
2)悬液定量杀灭试验操作程序
取消毒试验用无菌大试管,先加入0.5ml试验用菌悬液,再加入0.5ml有机干扰物质,混匀,置20℃±1℃水浴中5min后,用无菌吸管吸取上述浓度消毒液4.0ml注入其中,迅速混匀并立即记时。
待试验菌与消毒剂相互作用至各预定时间,分别吸取0.5ml试验菌与消毒剂混合液加入4.5ml经灭菌的中和剂中,混匀。
各管试验菌与消毒剂混合液经加中和剂作用10min后,分别吸取1.0ml样液,按活菌培养计数方法测定存活菌数,每管样液接种2个平皿即可。如平板上生长的菌落数较多时,可进行系列10倍稀释后,再进行活菌培养计数。
同时用标准硬水代替消毒液,进行平行试验,作为阳性对照。
所有试验样本均在37℃温箱中培养,对细菌繁殖体培养48h观察最终结果。
试验重复1次,计算各组的活菌浓度(cfu/ml),并换算为对数值(N),然后按下式计算杀灭对数值:
杀菌对数值(KL)=对照组平均活菌浓度的对数值(No)-试验组活菌浓度对数值(Nx)。
3)“杀菌试验”用各待测样品,分别对枯草芽孢杆菌作用2min、10min,试验在20±1℃条件下重复1次。
3.结果
表1.对枯草杆菌黑色变种芽孢的检测结果
实验在相同条件下重复一次,结果为:配方1及配方2的样品分别对枯草杆菌黑色变种芽孢ATCC9372作用2、10min,平均杀菌对数值(KL)>5。
4.试验结论
在有机干扰物存在的条件下,试验样品与枯草杆菌黑色变种芽孢作用2、10min.平均杀菌对数值(KL)值大于5。
讨论:中国发明专利ZL 201210166719.3的杀芽孢的配方,需要同时使用两种氨基酸双链氨基羧酸盐,添加量占制剂质量的2.4%,作用2min,对枯草杆菌黑色变种芽孢的平均杀菌对数值(KL)>5。本发明制剂只需使用一种氨基酸双链氨基羧酸盐,添加量仅占制剂质量的0.4%,作用2min,同样对枯草杆菌黑色变种芽孢的平均杀菌对数值(KL)>5。说明在同等杀菌效果的前提下,本发明的配方更为有效,本发明在低浓度下就达到了很好的杀芽孢效果。
二、有机物干扰试验
评价在有机物影响的情况下,样品的抗菌效果
1.主要材料
1)菌株:白色念珠菌ATCC10231,第6代,由中国微生物菌种保藏管理委员会普通微生物中心提供。
2)中和剂:含1%卵磷脂、0.5%Na2SO3、3%吐温-80的PBS溶液。
3)试验样品:将“原液”(实施例5中的配方3)用蒸馏水或去离子水稀释至“原液”的80%(实施例5中的配方4)待测样品
4)有机干扰物:3%牛血清白蛋白(溶解后用孔径为0.45um的微孔滤膜滤过除菌)。
5)作用菌浓度:5×105~5×106cuf/ml。
2.方法
1)试验按《消毒技术规范》(2008年版),第2.1.2.2、2.1.2.3、2.1.2.5及2.1.2.7项项“悬液定量杀灭试验法”进行。
2)悬液定量杀灭试验操作程序
取消毒试验用无菌大试管,先加入0.5ml试验用菌悬液,再加入0.5ml有机干扰物质,混匀,置20℃±1℃水浴中5min后,用无菌吸管吸取上述浓度消毒液4.0ml注入其中,迅速混匀并立即记时。
待试验菌与消毒剂相互作用至各预定时间,分别吸取0.5ml试验菌与消毒剂混合液加入4.5ml经灭菌的中和剂中,混匀。
各管试验菌与消毒剂混合液经加中和剂作用10min后,分别吸取1.0ml样液,按活菌培养计数方法测定存活菌数,每管样液接种2个平皿即可。如平板上生长的菌落数较多时,可进行系列10倍稀释后,再进行活菌培养计数。
同时用稀释液代替消毒液,进行平行试验,作为阳性对照。
所有试验样本均在37℃温箱中培养,对细菌繁殖体培养48h观察最终结果。
杀菌对数值(KL)=对照组平均活菌浓度的对数值-试验组活菌浓度对数值。
3)“杀菌试验”用样品原液(作用浓度为样品的80%稀释液,配置浓度为待测浓度1.25倍)分别对白色念珠菌作用2min、5min、10min、20min,试验在20±1℃条件下重复3次。
3.结果
在20±1℃条件下,三次重复试验结果表明:作用浓度为样品的80%稀释液对白色念珠菌作用2min,平均杀菌对数值>4,见下表:

4.试验结论
实验样品(作用浓度为样品的80%稀释液)在有机干扰物存在时,对白色念珠菌作用2min,平均杀菌对数值>4,有机干扰物不影响杀菌效果。
三、稳定性试验
1.试验器材
1)菌株:白色念珠菌ATCC10231,第6代,由中国微生物菌种保藏管理委员会普通微生物中心提供。
2)中和剂:含1%卵磷脂、0.5%Na2SO3、3%吐温-80的PBS溶液。
3)试验样品:实施例5中的配方5。样品在37℃温箱保存90天后再行检测。
2.试验方法
1.试验按GB15979-2002《一次性使用卫生用品卫生标准》附录C3项“杀菌性能试验方法”进行。
2.按实施例5中的配方5制备水溶液,分别对白色念珠菌作用2min、5min、10min、20min,试验在20±1℃条件下重复2次。
3.试验结果
在20±1℃条件下,两次重复试验结果表明:37℃温箱保存90天后的样品对白色念珠菌作用2min,平均杀菌率分别为100.00%,结果见下表:

4.试验结论:试验样品在37℃温箱保存90天后,检测对白色念珠菌ATCC10231作用2min.,平均杀菌率为100.00%。
四、冻融试验
1.试验器材
1)50mL带塞透明玻璃瓶20个
2)试验样品:实施例5中的配方1,2,3,4,5,6,7,8,9,10各60mL.
2.试验方法
制备配方1,2,3,4,5,6,7,8,9,10溶液,每种溶液分装在3个玻璃瓶中,每瓶20mL一瓶,共30瓶。将30个玻璃瓶放在-4℃的冰箱冻格内冰冻,24小时后取出,放置在室温下12小时,让其完全解冻。观察样品有无分离、沉淀、浑浊等现象。然后再放入冰箱冻格内冰冻。此冻、融试验重复5次。

4.试验结论
所有30个样品经过5个周期的冻融试验,无分离、沉淀、浑浊等情况出现。
本发明的有益效果:发明制剂可高效杀灭芽孢,无毒、无刺激,性质稳定,可以长期保存。该氨基酸双链羧酸盐带正电荷,可聚集在芽孢的胞膜上,与其带的负电荷作用,产生室阻效应,导致芽孢生长受抑而死亡;同时由于芽孢的胞膜上的电荷发生改变,可使芽孢蛋白变性而沉淀,导致芽孢死亡。制剂中的DAPC或DAAC能与芽孢壁上的多磷脂反应发挥本身所具有的杀菌作用外,也能使芽孢壁上的肽变性而破坏芽孢表面的蛋白受体。戊二醛在本发明中仅仅作为辅助成分,因为戊二醛只有在高浓度(3.2%)及碱性条件下,经2~4小时才能杀灭芽孢。

下面结合实例对本发明作进一步解释,但本发明不局限于以下实施例。其他人根据专业知识对以下实施例及配方范围所做的改动,做成其它的剂型或用于其它用途,都属于本发明的范围。
具体实施方式
实施例1:产品为液态,用下列原料按重量百分比配制:DAPC为1.0%,戊二醛为0.4%,去离子水水为余量。
制备:①.按质量配比取上述原料,将它们混匀,在30℃制得均质溶液;②.用灌装机分装至合适的容器中得成品。
还可按占一种杀芽孢的制剂的质量百分比再加入下面一种或多种辅料:
缓蚀剂:0.5~2.0%,
双十烷基二甲基氯化铵:0.1~2.0%,
十四烷基二甲基氧化铵:0.1~1.0%,
十四烷基二甲基苄基氯化铵:0.1~2.0%,
乙醇:10.0~80.0%,
增稠剂:0.01~0.5%。
实施例2:
液态发明产品用下列原料按重量百分比配制:DAAC为2.0%,戊二醛为0.8%,蒸馏水水为余量。制备方法同实施例1。
实施例3:
发明产品用下列原料按重量百分比配制:DAAC为0.5%,戊二醛为0.07%,缓蚀剂磷酸氢二钠和磷酸二氢钠为1.0%,双十烷基二甲基氯化铵为0.5%,增稠剂羟丙基甲基纤维素为0.3%,蒸馏水水为余量。
实施例4:
发明产品用下列原料按重量百分比配制:DAPC为1.0%,戊二醛为0.4%,十四烷基二甲基苄基氯化铵为0.5%,乙醇为20.0%,蒸馏水水为余量。
实施例5:下表中列出了10个配方的原料及其质量配比。对每一个配方分别按质量配比取其原料,按实施例1的制备方法制得均质溶液或无色透明溶液。

Claims (3)
1.一种含氨基酸衍生物的杀芽孢制剂,其特征在于:
(1)含有以下量的两种必要活性组分:
双癸基二甲基氧合脯氨酸铵DAPC(Proline,5-oxo-,ion(1-),N-decyl-N,N-dimethyl-1-decanaminium(1:1)),或双癸基二甲基N-乙酰基丙氨酸铵DAAC(1-Decanaminium,N-decyl-N,N-dimethyl-,salt with N-acetylalanine(1:1))两者中的任一种,其重量占制剂重量的0.1%~2.0%;
增效剂戊二醛,其重量占制剂重量的0.05%~1.00%;
(2)制剂中的其余量为相应剂型可以接受的非活性组分。
2.如权利要求1所述的含氨基酸衍生物的杀芽孢制剂,其特征在于:制剂为液体、膏剂或凝胶剂。
3.如权利要求1~2所述的含氨基酸衍生物的杀芽孢制剂,其特征在于:制剂中含有缓蚀剂,用于降低对金属的腐蚀性并调节制剂的pH值为6.4~7.7。
