背景技术
可吸收止血材料是当前外科领域的热点,外科及创伤手术或者战伤等过程中出血和渗血是手术中经常遇到的难题,相较于传统的电烙,缝合,结扎以及传统纱布等方法,可吸收止血材料具有对组织伤害小、操作简便、止血迅速等特点,但现有技术中的可吸收止血材料的生物相容性仍然不够理想。
壳聚糖是由甲壳素经脱乙酰化后制得的天然聚阳离子多糖,羧甲基壳聚糖是甲壳素经化学改性后得到的水溶性衍生物。壳聚糖、羧甲基壳聚糖均具有凝血、抑菌、促进创伤愈合、抑制瘢痕形成等多种作用,是一类生物相容性好、易生物降解、无免疫原性、无刺激性的多功能高分子生物材料,近年来在快速止血领域的应用日益广泛。但是目前单纯壳聚糖止血材料存在韧性差和止血效果不显著的问题。
发明内容
本发明主要解决的技术问题:针对目前单纯壳聚糖止血材料存在韧性差和止血效果不显著的问题,本发明首先通过对壳聚糖进行酰基化,与亮氨酸结合,再添加抗菌性物质与其混合反应,使结合后的壳聚糖具有疏水性,然后再将其与地榆提取物混合,进行接枝反应,增强其止血愈合能力,从而制备得改性壳聚糖基止血海绵。
为了解决上述技术问题,本发明所采用的技术方案是:
(1)称取700~800g壳聚糖放入2L烧杯中,向其中加入质量分数为12%的乙酸溶液,以转速180r/min进行搅拌,直至壳聚糖完全溶解,停止加入乙酸溶液,再称取110~130g亮氨酸放入烧杯中,搅拌均匀,随后把烧杯移至超声清洗仪中,在22~25KHz下振荡30~50min;
(2)待上述振荡结束后,将烧杯移至磁力加热搅拌器中,分别量取320~340mL甘油和称取180~210g丁二酸酐放入烧杯,设定温度为70~80℃,搅拌加热反应6~9h,再分别称取50~70g磺胺间二甲氧嘧啶和20~30g1-乙基-(3-二甲基氨基丙基)碳酰二亚胺盐酸盐放入烧杯中,搅拌均匀;
(3)在上述搅拌均匀后,将烧杯移至水浴锅中,在60~70℃下,改性反应20~22h,待反应结束后停止加热,自然冷却至室温,再把烧杯中的混合物通过陶瓷膜进行过滤,收集过滤物放入透析袋中,使用去离子水透析27~32h后得混浊液,将所得的混浊液放入喷雾干燥机中喷雾干燥,收集干燥物,得改性壳聚糖,备用;
(4)取500~600g地榆放入粉碎机中进行粉碎,过200目标准筛,将过筛后的颗粒放入带有搅拌器和温度计的2L三口烧瓶中,向其中加入900~1200mL质量分数为5%的醋酸溶液,将烧瓶移至电加热炉中进行加热,设定温度为80~90℃,转速为200r/min,保持温度2~3h后停止加热,自然冷却至室温,再向烧瓶中加入300~400mL质量分数为60%的乙醇溶液,搅拌40~50min后过滤;
(5)待上述过滤结束后收集过滤液,将所得的过滤液与步骤(3)所得的改性壳聚糖按质量比2:3混合均匀放入容器中,向其中加入改性壳聚糖质量2~4%的过硫酸钾,将容器移至紫外灯下,搅拌4~6h后,将容器内的混合液放入浓缩罐中浓缩至原有体积的40~50%,将所得的浓缩液倒入模具中,再将模具放置于真空冷冻干燥机内在冷冻干燥20~25h,即可得到改性壳聚糖基止血海绵。
本发明的应用方法:用手将本发明所制得的改性壳聚糖基止血海绵按在伤口处,在8~110s内伤口处不流血,达到止血效果,检测止血海绵得每克止血海绵吸血4.6~5.2g,且伤口愈合比传统止血海绵止血伤口愈合时间缩短40~52%,且无任何感染情况出现,不沾粘伤口。
本发明的有益效果:
(1)本发明所制得的止血海绵可任意折叠,止血效果显著;
(2)本发明所制得的止血海绵不沾粘伤口,可防止伤口感染,促进愈合。
具体实施方式

用手将本发明所制得的改性壳聚糖基止血海绵按在伤口处,在20s内伤口处不流血,达到止血效果,检测止血海绵得每克止血海绵吸血4.6g,且伤口愈合比传统止血海绵止血伤口愈合时间缩短40%,且无任何感染情况出现,不沾粘伤口。
实例2
称取800g壳聚糖放入2L烧杯中,向其中加入质量分数为12%的乙酸溶液,以转速180r/min进行搅拌,直至壳聚糖完全溶解,停止加入乙酸溶液,再称取130g亮氨酸放入烧杯中,搅拌均匀,随后把烧杯移至超声清洗仪中,在25KHz下振荡50min;待上述振荡结束后,将烧杯移至磁力加热搅拌器中,分别量取340mL甘油和称取210g丁二酸酐放入烧杯,设定温度为80℃,搅拌加热反应9h,再分别称取70g磺胺间二甲氧嘧啶和30g1-乙基-(3-二甲基氨基丙基)碳酰二亚胺盐酸盐放入烧杯中,搅拌均匀;在上述搅拌均匀后,将烧杯移至水浴锅中,在70℃下,改性反应22h,待反应结束后停止加热,自然冷却至室温,再把烧杯中的混合物通过陶瓷膜进行过滤,收集过滤物放入透析袋中,使用去离子水透析32h后得混浊液,将所得的混浊液放入喷雾干燥机中喷雾干燥,收集干燥物,得改性壳聚糖,备用;取600g地榆放入粉碎机中进行粉碎,过200目标准筛,将过筛后的颗粒放入带有搅拌器和温度计的2L三口烧瓶中,向其中加入1200mL质量分数为5%的醋酸溶液,将烧瓶移至电加热炉中进行加热,设定温度为90℃,转速为200r/min,保持温度3h后停止加热,自然冷却至室温,再向烧瓶中加入400mL质量分数为60%的乙醇溶液,搅拌50min后过滤;待上述过滤结束后收集过滤液,将所得的过滤液与所得的改性壳聚糖按质量比2:3混合均匀放入容器中,向其中加入改性壳聚糖质量4%的过硫酸钾,将容器移至紫外灯下,搅拌6h后,将容器内的混合液放入浓缩罐中浓缩至原有体积的50%,将所得的浓缩液倒入模具中,再将模具放置于真空冷冻干燥机内在冷冻干燥25h,即可得到改性壳聚糖基止血海绵。

实例3
称取750g壳聚糖放入2L烧杯中,向其中加入质量分数为12%的乙酸溶液,以转速180r/min进行搅拌,直至壳聚糖完全溶解,停止加入乙酸溶液,再称取120g亮氨酸放入烧杯中,搅拌均匀,随后把烧杯移至超声清洗仪中,在24KHz下振荡40min;待上述振荡结束后,将烧杯移至磁力加热搅拌器中,分别量取330mL甘油和称取200g丁二酸酐放入烧杯,设定温度为75℃,搅拌加热反应8h,再分别称取60g磺胺间二甲氧嘧啶和25g1-乙基-(3-二甲基氨基丙基)碳酰二亚胺盐酸盐放入烧杯中,搅拌均匀;在上述搅拌均匀后,将烧杯移至水浴锅中,在60~70℃下,改性反应21h,待反应结束后停止加热,自然冷却至室温,再把烧杯中的混合物通过陶瓷膜进行过滤,收集过滤物放入透析袋中,使用去离子水透析30h后得混浊液,将所得的混浊液放入喷雾干燥机中喷雾干燥,收集干燥物,得改性壳聚糖,备用;取550g地榆放入粉碎机中进行粉碎,过200目标准筛,将过筛后的颗粒放入带有搅拌器和温度计的2L三口烧瓶中,向其中加入1100mL质量分数为5%的醋酸溶液,将烧瓶移至电加热炉中进行加热,设定温度为85℃,转速为200r/min,保持温度2h后停止加热,自然冷却至室温,再向烧瓶中加入350mL质量分数为60%的乙醇溶液,搅拌45min后过滤;待上述过滤结束后收集过滤液,将所得的过滤液与所得的改性壳聚糖按质量比2:3混合均匀放入容器中,向其中加入改性壳聚糖质量3%的过硫酸钾,将容器移至紫外灯下,搅拌5h后,将容器内的混合液放入浓缩罐中浓缩至原有体积的45%,将所得的浓缩液倒入模具中,再将模具放置于真空冷冻干燥机内在冷冻干燥24h,即可得到改性壳聚糖基止血海绵。

Claims (1)
1.一种改性壳聚糖基止血海绵的制备方法,其特征在于具体制备步骤为:
(1)称取700~800g壳聚糖放入2L烧杯中,向其中加入质量分数为12%的乙酸溶液,以转速180r/min进行搅拌,直至壳聚糖完全溶解,停止加入乙酸溶液,再称取110~130g亮氨酸放入烧杯中,搅拌均匀,随后把烧杯移至超声清洗仪中,在22~25KHz下振荡30~50min;
(2)待上述振荡结束后,将烧杯移至磁力加热搅拌器中,分别量取320~340mL甘油和称取180~210g丁二酸酐放入烧杯,设定温度为70~80℃,搅拌加热反应6~9h,再分别称取50~70g磺胺间二甲氧嘧啶和20~30g1-乙基-(3-二甲基氨基丙基)碳酰二亚胺盐酸盐放入烧杯中,搅拌均匀;
(3)在上述搅拌均匀后,将烧杯移至水浴锅中,在60~70℃下,改性反应20~22h,待反应结束后停止加热,自然冷却至室温,再把烧杯中的混合物通过陶瓷膜进行过滤,收集过滤物放入透析袋中,使用去离子水透析27~32h后得混浊液,将所得的混浊液放入喷雾干燥机中喷雾干燥,收集干燥物,得改性壳聚糖,备用;
(4)取500~600g地榆放入粉碎机中进行粉碎,过200目标准筛,将过筛后的颗粒放入带有搅拌器和温度计的2L三口烧瓶中,向其中加入900~1200mL质量分数为5%的醋酸溶液,将烧瓶移至电加热炉中进行加热,设定温度为80~90℃,转速为200r/min,保持温度2~3h后停止加热,自然冷却至室温,再向烧瓶中加入300~400mL质量分数为60%的乙醇溶液,搅拌40~50min后过滤;
(5)待上述过滤结束后收集过滤液,将所得的过滤液与步骤(3)所得的改性壳聚糖按质量比2:3混合均匀放入容器中,向其中加入改性壳聚糖质量2~4%的过硫酸钾,将容器移至紫外灯下,搅拌4~6h后,将容器内的混合液放入浓缩罐中浓缩至原有体积的40~50%,将所得的浓缩液倒入模具中,再将模具放置于真空冷冻干燥机内在冷冻干燥20~25h,即可得到改性壳聚糖基止血海绵。
