Abstract
本发明公开了一种含有多种功效成分的全效修复牙膏,该牙膏含有四种功效成分及基质成分,其中功效成分包括具有再矿化、脱敏及修补牙齿功能的羟基磷灰石,可防龋齿和牙髓炎的氟化钠,可抑菌和预防牙周病的橘皮提取物,及可清热去火的板蓝根提取物。本发明所述的牙膏可预防口腔疾病,全面修护牙齿。而且所含成分易于采购,成本较低,制备工艺简便易行,适合大规模生产。
Description
技术领域
本发明属于日用化学品技术领域,具体涉及一种含有多种功效成分的全效修复牙膏。
背景技术
世界卫生组织早在1984年提出的现代人十项健康标准就已经将牙周健康列入其中:第8条为“牙齿无洞,无疼痛,牙龈不出血”是牙齿健康的体现。所以牙齿的疾病主要集中在牙周、牙体和牙髓。
一、牙周病
根据最新的亚洲健康调查显示,30岁以上人群的牙周病患病率为7%~99%不等,如果以折中的牙周附着丧失3mm为度,则30岁以上人群的牙周附着丧失率也达53.1%,而且随年龄而递增。牙周病主要表现为牙龈红肿、出血、牙周局部肿痛不适或溢脓,牙齿松动、早起脱落、咀嚼效能降低或丧失等。
菌斑是牙周病发生的始动因子,细菌及其毒性产物可引发牙龈的炎症和肿胀,并可进一步发展成为牙周炎,使牙周组织破坏,菌斑即使除去,也还会不断地在牙面重新形成。因此,牙周炎患者必须重视菌斑的控制,应当每天持之以恒地彻底清除菌斑。才能消除牙周炎症及其所导致的不适、出血等症状,使牙周破坏停止,并能防止治疗后的复发,始终保持牙周的健康状态。
二、龋病
龋病的临床表现可以概括为牙齿色、形、质的变化和患者感觉的变化。正常的釉质表面呈半透明状,牙本质的颜色为淡黄色,透过釉质显现出来。釉质表面应该光滑、无色素沉着。牙齿表面色泽的变化是临床最早可以注意到的龋的变化。当龋发生在平滑面时,擦去表面的菌斑,吹干后可见病变部位表面光泽消失,呈白垩色,即粉笔样的颜色。当龋发生在窝沟时,早起可见窝沟呈白垩色。窝沟内脱矿严重时,窝沟可呈墨浸状,即透过半透明的完整的釉质所反映出的牙本质的脱矿。
导致龋病有很多因素,其中影响较大的为口腔环境和致龋菌斑。龋病必须在菌斑这样一个微生态环境中产生,菌斑能否致龋,关键在于菌斑的性质。致龋的菌斑都具有较强的产酸力,如乳杆菌、变异性链球菌、口腔放线菌等,细菌在易感的牙面代谢形成牙菌斑,牙菌斑使细菌发酵糖产生的酸在牙面达到一定的浓度(在临界ph以下)和维持相当长的时间就会导致龋齿。
细菌在牙菌斑生物膜中的生长及其致龋力与口腔环境条件密切相关。细菌细胞感受环境刺激做出相应反应的调节机制对其生存和致龋性非常重要,其致龋毒力因子在不同的环境条件下,特别是不利生长条件下的表达可能促进其在牙菌斑中的生存,增强其致龋性。
三、牙髓病
牙髓病是指发生于牙髓组织的疾病。牙髓组织对通过牙体硬组织和牙周病传入的各种伤害都有敏锐的反应,可导致牙髓病。牙髓组织中含有丰富的自律神经和躯体感觉神经。前者主要分布于血管壁,调节牙髓血管的收缩和舒张。牙髓的感觉神经来源于三叉神经的第二支或第三支的分支,其低级中枢位于三叉神经节。在牙髓组织中,有髓鞘和无髓鞘神经均可见到,有髓鞘神经成束通过根尖孔,进入牙髓,行至接近层牙本质细胞层处,失去髓鞘,成为单独的纤维而密布于无细胞层。这些失去髓鞘的游离神经末梢即为痛觉感受器,对刺激的种类缺乏辨别能力,冷、热、触、压或化学刺激传到中枢均为痛觉,而且不能准确定位,所以,在临床上,疼痛的症状成为诊断牙髓炎的主要依据。
其中龋病是人类口腔多发病、常见病、如未及时治疗往往发展为牙髓病,所以有人称牙髓病是龋病的继发病。
中国专利CN201210584585.7,一种同时清洁美白修复牙齿的牙膏及其制备方法,公开了一种含有羟基磷灰石、珍珠岩的牙膏,通过羟基磷灰石和珍珠岩复配起协同作用,主要集中在修复牙釉质,打磨牙齿表面凹凸面,达到修复清洁的目的。
本发明通过抑菌、清火、防龋,再矿化等组方设计针对牙周病、龋齿、牙髓病等牙齿的疾病,开发一款防治和修护的全效牙膏。
四、组方设计
羟基磷灰石具有再矿化性能和脱敏性能,可有效防护龋齿。羟基磷灰石是人及动物骨骼、牙齿的主要无机成分,羟基磷灰石具有优良的生物相容性和生物活性,具有吸附蛋白质,氨基酸、脂质和葡聚糖的作用,它可以直接用于牙齿表面,和牙齿中的羟基磷灰石融合。还可以修复刀齿表面的细微裂痕,促进牙釉质表面再矿化、增强釉质的抗龋能力。
牙釉质的再矿化是龋坏的釉质表面重新形成羟基磷灰石或氟羟磷灰石晶体的过程,实际上釉质表面存在磷酸钙盐的溶解—沉淀平衡,即处于脱矿和再脱矿的频繁交替过程中,如再矿化强于脱矿,则龋病有愈合的可能。再矿化过程受唾液中Ca2+、PO4 3-、F-以及菌斑的pH值等因素的影响,唾液中Ca2+、PO4 3-是矿化的物质基础,它们的浓度越高,越有利于再矿化的发生。因此,从溶解—沉淀平衡的角度看,预防牙齿脱矿的关键是增加口腔唾液中的Ca、P离子的浓度,推动羟基磷灰石的溶解—沉淀平衡向形成羟基磷灰石结晶方向移动。
羟基磷灰石特别是高活性的纳米级羟基磷灰石不仅可以中和口腔中的酸性物质,其较高的溶解反应可极大地提高唾液中Ca、P离子的浓度,从根本上预防脱矿的发生,同时纳米级的羟基磷灰石可直接封闭因脱矿而形成的釉面空隙,具有优良的再矿化效果。
羟基磷灰石对牙齿敏感也有很好的效果,国内外许多学者研究表明纳米羟基磷灰石微粒能够进入牙小管内,有效的封闭了牙小管,使牙小管的渗透性下降96%,从而能够治疗牙本质过敏。
氟化钠可以增强牙齿的防龋能力。有资料表明:用含1100-1500ppm氟牙膏可降低龋发率20%-40%,局部涂氟可降低龋发率30%-40%;而使用透氟法可降低龋发率50%。研究认为,氟化物用于防龋机制是在低pH环境中,氟离子有可能结合磷灰石替代羟基团形成氟磷灰石,使牙齿更不易收到酸的侵蚀;在高pH环境中氟离子可能促进磷灰石沉淀到牙齿内,有利于促进牙齿的再矿化。
鉴于氟的摄入剂量安全问题,各个国家对在牙膏中加入氟的量都有规定,我国强制性国家标准《牙膏》GB8372-2001中规定,含氟牙膏的总氟含量应在0.04%~0.15%之间;美国食品与药品管理局规定含氟牙膏的氟含量为0.11%到0.15%;欧盟规定含氟牙膏的氟含量应小于0.15%;拉丁美洲规定含氟牙膏的氟含量为0.11%到0.15%;亚洲一些国家规定含氟牙膏的氟含量为0.10%到0.15%。由此可见,我国与世界主要地区对含氟牙膏中氟含量上限值的规定是一致的,而且我国对含氟牙膏中氟含量的下限值规定是偏低的。因此,只要是经检测合格的含氟牙膏产品,消费者可以根据自己实际需要放心使用。
本发明所述的牙膏中氟的含量为0.10%-0.15%。
龋病、牙周病是牙菌斑生物膜相关的细菌感染性疾病。龋病的发病原因主要是菌斑中的细菌利用碳水化合物产酸、导致牙齿脱矿,继发侵入牙髓和根尖周围组织;牙周病的发病时菌斑中的致炎因子导致的牙周组织直接损害和间接免疫病理损害。变形链球菌是龋病和牙周病的始动因子,它首先附着在牙齿硬组织表面,产酸引起牙齿应组织脱矿,继发其它细菌黏附,形成致病菌斑;金黄色葡萄球菌、白色葡萄球菌是口腔化脓感染及手术感染的主要致病因素;白假丝酵母菌是引发口腔内交叉感染的重要原因,也是口腔修复体周围炎、口腔黏膜疾病、颌面部感染的主要原因,经口内途径的外科手术后感染等都是由口腔病原生物感染所引起。口腔中的微生物除细菌、真菌外,由唾液中或可检出HIV、HSV、EBV以及CMV等多种病毒。
橘皮提取物对口腔常见病原微生物变形链球菌、金黄色葡萄球菌、白色葡萄球菌、白假丝酵母菌的生长有抑制作用;可以抑制变形链球菌等细菌对光滑玻璃表面的粘附;对单纯疱疹病毒有灭活作用,橘皮提取物在有效抑菌及抗病毒浓度未见明显细胞毒性反应,而且使用低浓度橘皮提取物对细菌仅有抑制其生长增殖和附着的作用,而不会全面杀死细菌,从而能够有效维持口腔微生态的平衡。因此橘皮提取物可作为一种天然、高效、安全的新型口腔局部抑菌剂、在口腔疾病特别是龋病和牙周病的预防和控制中发挥重要作用。
《本草纲目》记载:板蓝根主治“时气头痛,火热口疮,热病发斑,热毒下痢,喉痹、丹毒、黄疸、痄腮……”。现代药理实验证明:板蓝根具有抗炎、抗病毒、解热和增强免疫等作用。近年来,随着我国社会和经济的快速发展,由于生活节奏的加快和工作压力的加大,很多人生活不规律,长期在外就餐,饮食习惯不佳,再加上吸烟、饮酒、情绪紧张、环境污染等因素,很容易出现上火引发的口腔问题,如口腔溃疡、牙周炎、智齿冠周炎等,板蓝根提取物具有清热、去火、解毒的功效,有助于缓解上述问题。
发明内容
本发明公开了一种含有多种功效成分的全效修复牙膏,可预防口腔疾病,全面修护牙齿。
本发明的技术方案概述如下:
一种含有多种功效成分的全效修复牙膏,该牙膏含有四种功效成分及基质成分,其中功效成分包括具有再矿化、脱敏及修补牙齿功能的羟基磷灰石,可防龋齿和牙髓炎的氟化钠,可抑菌和预防牙周病的橘皮提取物,及可清热去火的板蓝根提取物。
本发明所述的一种含有多种功效成分的全效修复牙膏,其特征在于所含纳米羟基磷灰石占牙膏膏体质量的3-6%,氟化钠占膏体质量的0.4-1.0%,橘皮提取物占膏体质量的5-15%,板蓝根提取物占膏体质量的5-15%。
本发明所述的一种含有多种功效成分的全效修复牙膏,除了上述的功效成分以外,其基质成分由摩擦剂、保湿剂、粘合剂、调味剂、表面活性剂和香精等成分组成。
本发明所述的一种含有多种功效成分的全效修复牙膏,其制备方法如下:
(1)称取粘合剂、橘皮提取物、板蓝根提取物,加适量水慢慢溶胀成胶液备用;
(2)称取摩擦剂、保湿剂、氟化钠、纳米羟基磷灰石、表面活性剂,混匀,加入适量水搅拌、研磨;
(3)在混合物中加入制备好的胶液,边加边搅拌研磨混合,加入香精搅匀,放置一段时间;
(4)脱气,灌装。

依实施例1-4制备的牙膏均能够达到:膏体外观均匀,无异物;pH在7.5-8.0之间;色泽正常,有特征香味;无过硬颗粒。
选择40例受试者,每名受试者至少有2颗牙被临床确诊为龋齿或牙周病。将这40例受试者分为5组,每组8人,分别使用依实施例1-4制备的牙膏和普通含氟牙膏治疗。试验期间,受试者每天刷牙2次,每次3min,刷牙时将2.0cm的牙膏段挤在湿润的牙刷上,共治疗4周。禁止受试者使用日常使用的牙膏,但可以继续日常采取的口腔卫生措施。采用冷空气刺激法对受试者的牙本质敏感性进行测定。结果发现,使用实施例1-4制备的牙膏,有效率可达到68%以上,与普通含氟牙膏的31%存在显著性差异。
Claims (4)
1.一种含有多种功效成分的全效修复牙膏,该牙膏含有四种功效成分及基质成分,其中功效成分包括具有再矿化、脱敏及修补牙齿功能的羟基磷灰石,可防龋齿和牙髓炎的氟化钠,可抑菌和预防牙周病的橘皮提取物,及可清热去火的板蓝根提取物。
2.根据权利要求1所述的一种含有多种功效成分的全效修复牙膏,其特征在于所含纳米羟基磷灰石占牙膏膏体质量的3-6%,氟化钠占膏体质量的0.4-1.0%,橘皮提取物占膏体质量的5-15%,板蓝根提取物占膏体质量的5-15%。
3.根据权利要求1所述的一种含有多种功效成分的全效修复牙膏,除了上述的功效成分以外,其基质成分由摩擦剂、保湿剂、粘合剂、调味剂、表面活性剂和香精等成分组成。
4.根据权利要求1所述的一种含有多种功效成分的全效修复牙膏,其制备方法如下:
(1)称取粘合剂、橘皮提取物、板蓝根提取物,加适量水慢慢溶胀成胶液备用;
(2)称取摩擦剂、保湿剂、氟化钠、纳米羟基磷灰石、表面活性剂,混匀,加入适量水搅拌、研磨;
(3)在混合物中加入制备好的胶液,边加边搅拌研磨混合,加入香精搅匀,放置一段时间;
(4)脱气,灌装。
