Abstract
本发明公开了一种检测牛肉中伊维菌素残留的试剂盒,包括盒式外壳及位于盒体外壳内部的免疫层析试纸条,免疫层析试纸条包括样品垫、分析膜、吸水垫、粘性底衬及包被有上转发光材料颗粒标记的抗伊维菌素单抗的结合垫,粘性底衬胶合固定在盒式外壳的底部,分析膜胶合在粘性底衬上,结合垫及吸水垫分别胶合在分析膜的两侧,样品垫胶合在结合垫远离分析膜的一侧,在分析膜上设置有包被抗伊维菌素单抗的检测带及包被羊抗鼠lgG的质控带,盒式外壳上设置有分别与样品垫、分析膜及吸水垫对应的样品添加孔、结果读取窗口及终点指示窗口。本发明采用上转发光技术与免疫技术结合,适宜在现场的快速检测,成本低,具有准确、快速、定量检测的特点。
Description
技术领域
本发明涉及医疗技术领域,具体涉及一种检测牛肉中伊维菌素残留的试剂盒。
背景技术
伊维菌素是一种半合成大环内酯类抗生素,其毒性大大低于阿维菌素,为目前兽医临床上使用最多的抗寄生虫药,对体内外寄生虫特别是线虫和节肢动物均有良好的驱杀作用,广泛地使用于猪、牛、羊等家畜中。伊维菌素具有生殖毒性,且在动物体内残留时间长,长期暴露对人体具有严重健康损害危险,且对水生环境有剧毒性。
目前对伊维菌素的检测方法主要有高效液相色谱法(HPLC)、液质联用法(LC-MS)、酶联免疫法等(ELISA)。但HPLC和LC-MS法的样品前处理复杂,操作繁琐,而且这两种方法所需的仪器昂贵,成本高,不适于对大量样本的筛查。而ELISA法易出现假阳性,造成结果的偏差。
发明内容
为了解决上述技术问题,本发明提供一种检测牛肉中伊维菌素残留的试剂盒,该试剂盒采用上转发光技术与免疫技术结合,适宜在现场的快速检测,成本低,适于对大量样本的筛查,具有准确、快速、定量检测的特点。
本发明的目的通过以下技术方案来具体实现:
一种检测牛肉中伊维菌素残留的试剂盒,包括盒式外壳及位于盒体外壳内部的免疫层析试纸条,所述免疫层析试纸条包括样品垫、分析膜、吸水垫、粘性底衬及包被有上转发光材料颗粒标记的抗伊维菌素单抗的结合垫,所述粘性底衬胶合固定在盒式外壳的底部,分析膜胶合在粘性底衬上,结合垫及吸水垫分别胶合在分析膜的两侧,所述样品垫胶合在结合垫远离分析膜的一侧,在分析膜上设置有包被抗伊维菌素单抗的检测带及包被羊抗鼠lgG的质控带,所述盒式外壳上设置有分别与样品垫、分析膜及吸水垫对应的样品添加孔、结果读取窗口及终点指示窗口。
进一步的,所述分析膜为硝酸纤维素膜。
进一步的,所述盒体外壳由透明材料制成。
本发明一种检测牛肉中伊维菌素残留的试剂盒,该试剂盒的结合垫上包被有上转发光材料颗粒标记的抗伊维菌素单抗,同时在分析膜上设置有包被抗伊维菌素单抗的检测带及包被羊抗鼠lgG的质控带,反应时样品中伊维菌素与两株抗体反应结合,在检测带形成伊维菌素蛋白抗体-伊维菌素蛋白抗原-上转发光颗粒标记的伊维菌素蛋白抗体复合物,上转发光颗粒在红外光激发下发射可见光,发射光的强度与样品中伊维菌素蛋白的浓度成正比。无论样品中是否存在伊维菌素,在质控带都会形成羊抗鼠-上转发光颗粒标记的抗伊维菌素抗体复合物,利用上转发光免疫分析仪对试纸条进行扫描分析,显示测量结果。本试剂盒采用上转发光技术与免疫技术结合,适宜在现场的快速检测,成本低,适于对大量样本的筛查,具有准确、快速、定量检测的特点。

具体实施方式
如图1-2所示,本发明实施例所述的一种检测牛肉中伊维菌素残留的试剂盒,包括盒式外壳1及位于盒体外壳1内部的免疫层析试纸条,免疫层析试纸条包括样品垫2、分析膜3、吸水垫4、粘性底衬5及包被有上转发光材料颗粒标记的抗伊维菌素单抗的结合垫6,粘性底衬5胶合固定在盒式外壳1的底部,分析膜3胶合在粘性底衬5上,结合垫6及吸水垫4分别胶合在分析膜3的两侧,样品垫2胶合在结合垫6远离分析膜3的一侧,在分析膜3上设置有包被抗伊维菌素单抗的检测带7及包被羊抗鼠lgG的质控带8,盒式外壳1上设置有分别与样品垫2、分析膜3及吸水垫4对应的样品添加孔9、结果读取窗口10及终点指示窗口11。
一种优选的实施方式,分析膜3为硝酸纤维素膜。
优选的,盒体外壳1由透明材料制成,便于观测。
本发明一种检测牛肉中伊维菌素残留的试剂盒,在使用时,添加样品中的伊维菌素与两株抗体反应结合,在检测带形成伊维菌素蛋白抗体-伊维菌素蛋白抗原-上转发光颗粒标记的伊维菌素蛋白抗体复合物,上转发光颗粒在红外光激发下发射可见光,发射光的强度与样品中伊维菌素蛋白的浓度成正比。无论样品中是否存在伊维菌素,在质控带都会形成羊抗鼠-上转发光颗粒标记的抗伊维菌素抗体复合物。利用上转发光免疫分析仪对试纸条进行扫描分析,显示测量结果,在整个检测过程中,通过虹吸作用提供液体流过整个试纸条的动力。
本试剂盒采用上转发光技术与免疫技术结合,适宜在现场的快速检测,成本低,适于对大量样本的筛查,具有准确、快速、定量检测的特点。

1.一种检测牛肉中伊维菌素残留的试剂盒,包括盒式外壳及位于盒体外壳内部的免疫层析试纸条,其特征在于:所述免疫层析试纸条包括样品垫、分析膜、吸水垫、粘性底衬及包被有上转发光材料颗粒标记的抗伊维菌素单抗的结合垫,所述粘性底衬胶合固定在盒式外壳的底部,分析膜胶合在粘性底衬上,结合垫及吸水垫分别胶合在分析膜的两侧,所述样品垫胶合在结合垫远离分析膜的一侧,在分析膜上设置有包被抗伊维菌素单抗的检测带及包被羊抗鼠lgG的质控带,所述盒式外壳上设置有分别与样品垫、分析膜及吸水垫对应的样品添加孔、结果读取窗口及终点指示窗口。
2.根据权利要求1所述的一种检测牛肉中伊维菌素残留的试剂盒,其特征在于:所述分析膜为硝酸纤维素膜。
