本发明提供了一种穿心莲内酯的生产方法,包括如下步骤:S1、取穿心莲茎叶100~150g,用400mL的乙醇浸泡18-24小时;S2、加入400mL的乙醇浸泡18-24小时,加入15g活性炭脱色30min,回收乙醇;S3、加入100mL的水,去除无机盐,加入500mL~800mL乙醇,加热溶解;S4、加入5~8g活性炭回流30min,滤液放置结晶,得到穿心莲内酯粗品;S5、加入200mL~400mL氯仿,放置40~60min,再加入200mL~400mL氯仿放置40~60min,提取不溶部分;S6、不溶部分于50℃干燥,加入400mL的乙醇浸泡18-24小时,加入15g活性炭脱色30min,回收乙醇,得到穿心莲内酯。本发明收集率提高了穿心莲内酯的收集纯度,纯度达98%,并且优化了工艺参数。
Description
技术领域
本发明涉及穿心莲内酯的生产方法,具体涉及利用穿心莲茎叶进行一种穿心莲内酯的生产方法。
背景技术
穿心莲内酯为天然植物穿心莲的主要有效成份,具有祛热解毒,消炎止痛之功效,对细菌性与病毒性上呼吸道感染及痢疾有特殊疗效,被誉为天然抗生素药物。本品为二萜类内酯化合物,均难溶于水,通常仅能口服给药。穿心莲有明显的中止妊娠作用。穿心莲口服能使小鼠避孕、孕兔流产。水煎剂腹腔注射对小鼠的着床、早孕、中孕、晚孕等不同时期均有显著的中止作用。肌注、皮下注射、灌服、腹腔注射、静脉注射及宫腔内注射均能产生抗早孕效果,其中以腹腔、静脉和宫内注射效果最好。宫腔内注射,用量既小,效果又佳。若将外源性孕酮或黄体生成素释放、激素(LH-RH)与穿心莲同时注射,则可防止流产,这提示,穿心莲可能有对抗体内孕酮的作用,从而引起流产。有人认为,穿心莲对体外培养的胎盘绒毛滋养层细胞的生长有一定的抑制作用。研究表明:穿心莲内酯的一些半合成衍生物亦有不同程度的抗早孕效果,脱水穿心莲内酯衍生物-穿-4对小鼠和大鼠均有良好的抗早孕作用,是值得进一步研究的一个避孕新药,该药对胎盘滋养叶细胞有显著的影响,可使胎盘绒毛萎缩、细胞数目减少,合体细胞和朗罕细胞消失、间质水肿出血。穿心莲内酯可增加肾上腺皮质功能,穿心莲甲、乙、丙、丁素在较大剂量时可使幼年小鼠胸腺萎缩。目前的穿心莲内酯的制法由于工艺的粗糙,穿心莲内酯的收集比例较低,造成大量的浪费。
发明内容
本发明的目的是提供一种穿心莲内酯的生产方法,解决上述现有技术中一个或者多个问题。
根据本发明的一种穿心莲内酯的生产方法,包括如下步骤:
S1、取穿心莲茎叶100~150g,用400mL的乙醇浸泡18-24小时;
S2、加入400mL的乙醇浸泡18-24小时,加入15g活性炭脱色30min,回收乙醇;
S3、加入100mL的水,去除无机盐,加入500mL~800mL乙醇,加热溶解。
S4、加入5~8g活性炭回流30min,滤液放置结晶,得到穿心莲内酯粗品;
S5、加入200mL~400mL氯仿,放置40~60min,再加入200mL~400mL氯仿放置40~60min,提取不溶部分;
S6、不溶部分于50℃干燥,加入400mL的乙醇浸泡18-24小时,加入15g活性炭脱色30min,回收乙醇,得到穿心莲内酯。
本发明收集率提高了穿心莲内酯的收集纯度,纯度达98%。
具体实施方式
实施例1:
本发明提供了一种穿心莲内酯的生产方法,包括如下步骤:
S1、取穿心莲茎叶100g,用400mL的乙醇浸泡18小时;
S2、加入400mL的乙醇浸泡18小时,加入15g活性炭脱色30min,回收乙醇;
S3、加入100mL的水,放置结晶,抽滤,用少许水洗涤,去除无机盐,加入500mL乙醇,加热溶解。
S4、加入5g活性炭回流30min,滤液放置结晶,得到穿心莲内酯粗品;
S5、加入200mL氯仿,放置40min,再加入200mL氯仿放置40min,提取不溶部分;
S6、不溶部分于50℃干燥,加入400mL的乙醇浸泡18小时,加入15g活性炭脱色30min,回收乙醇至200mL,放置结晶,抽取滤液,得到穿心莲内酯。
本发明收集率提高了穿心莲内酯的收集纯度,纯度达95%。
实施例2:
本发明提供了一种穿心莲内酯的生产方法,包括如下步骤:
S1、取穿心莲茎叶125g,用400mL的乙醇浸泡21小时;
S2、加入400mL的乙醇浸泡21小时,加入15g活性炭脱色30min,回收乙醇;
S3、加入100mL的水,放置结晶,抽滤,用少许水洗涤,去除无机盐,加入650mL乙醇,加热溶解。
S4、加入6.5g活性炭回流30min,滤液放置结晶,得到穿心莲内酯粗品;
S5、加入300mL氯仿,放置50min,再加入300mL氯仿放置50min,提取不溶部分;
S6、不溶部分于50℃干燥,加入400mL的乙醇浸泡21小时,加入15g活性炭脱色30min,回收乙醇至200mL,放置结晶,抽取滤液,得到穿心莲内酯。
本发明收集率提高了穿心莲内酯的收集纯度,纯度达97%。
实施例3:
本发明提供了一种穿心莲内酯的生产方法,包括如下步骤:
S1、取穿心莲茎叶150g,用400mL的乙醇浸泡24小时;
S2、加入400mL的乙醇浸泡24小时,加入15g活性炭脱色30min,回收乙醇;
S3、加入100mL的水,放置结晶,抽滤,用少许水洗涤,去除无机盐,加入800mL乙醇,加热溶解。
S4、加入8g活性炭回流30min,滤液放置结晶,得到穿心莲内酯粗品;
S5、加入400mL氯仿,放置60min,再加入400mL氯仿放置60min,提取不溶部分;
S6、不溶部分于50℃干燥,加入400mL的乙醇浸泡24小时,加入15g活性炭脱色30min,回收乙醇至200mL,放置结晶,抽取滤液,得到穿心莲内酯。
本发明收集率提高了穿心莲内酯的收集纯度,纯度达98%。

1.一种穿心莲内酯的生产方法,其特征在于,包括如下步骤:
S1、取穿心莲茎叶100~150g,用400mL的乙醇浸泡18-24小时;
S2、加入400mL的乙醇浸泡18-24小时,加入15g活性炭脱色30min,回收乙醇;
S3、加入100mL的水,去除无机盐,加入500mL~800mL乙醇,加热溶解。
2.根据权利要求1所述的一种穿心莲内酯的生产方法,其特征在于,还包括S3后的如下步骤:
S4、加入5~8g活性炭回流30min,滤液放置结晶,得到穿心莲内酯粗品;
S5、加入200mL~400mL氯仿,放置40~60min,再加入200mL~400mL氯仿放置40~60min,提取不溶部分;
S6、不溶部分于50℃干燥,加入400mL的乙醇浸泡18-24小时,加入15g活性炭脱色30min,回收乙醇,得到穿心莲内酯。
