背景技术
针对疲劳机制之能源“耗竭”学说:能量主要来源于糖、脂肪、蛋白质和有氧或无氧代谢,随着运动时间的延长,能源物质被耗竭,机体不能维持预定强度,出现了疲劳。
蜂王浆是由成年工蜂上颗王浆腺分泌的一种乳白色或淡黄色的乳状体,含有三大生热营养素之外,还含有维生素、多种矿物质、多种生物活性物质;左旋肉碱主要的功能是作为载体以脂酞肉碱的形式将长链脂肪酸从线粒体膜外转运到膜内,在线粒体内进行β-氧化,产生能量;左旋肉碱能够调节线粒体内酰基比率,还可以可参加支链氨基酸代谢产物的运输,保证正常的能量代谢;左旋肉碱可排出体内过量或非理性酰基团,消除机体因酰基积累而造成的代谢毒性,还能促进乙酞乙酸的氧化,在酮体的消除和利用中起作用,防止动物体内过量氨产生的毒性;左旋肉碱可作为生物抗氧化剂清除自由基,维持膜的稳定,提高动物的免疫力以及抗病抗应激的能力;葡萄籽提取物含有原花青素、茶多酚以及天然维生素E三种成分,是一种自由基清除剂、高效生物抗氧化剂;维生素C是人体细胞内外化学反应中重的抗氧化物质,在细胞氧化防御中起重要作用,参与并促进胶原蛋白和组织间粘合质形成,是多种酶的辅因子或辅底物,可增强机体对传染病抵抗能力,提高适应能力,对体液免疫、细胞免疫均有重要调节作用;维生素E亦称“生育酚”,属脂溶性维生素。维生素E可以抑制自由基的生成,对内皮细胞具有重要的保护作用,可以维持内皮细胞内分泌功能的稳定。

目前,在现有技术中,涉及蜂王浆、左旋肉碱、葡萄籽或者维生素具有保健功能的文献很多,但涉及这几种原料的相互组合的报道较少,且尚无将上述原料优化组合发挥协同作用的研究与报道。
发明内容
本发明目的在于提供一种蜂王浆组合物,本发明另一目的在于提供该组合物的制备方法及用途。
本发明目的是通过如下技术方案实现
本发明蜂王浆组合物的原料组成为:
蜂王浆冻干粉60-100重量份,左旋肉碱60-100重量份,葡萄籽提取物30-80重量份,维生素C10-40重量份,维生素E琥珀酸酯5-30重量份。
上述原料优选重量配比如下:
蜂王浆冻干粉83.3重量份,左旋肉碱83.3重量份,葡萄籽提取物50重量份,维生素C25重量份,维生素E琥珀酸酯16.7重量份。
上述原料优选重量配比如下:
蜂王浆冻干粉72重量份,左旋肉碱90重量份,葡萄籽提取物43重量份,维生素C29重量份,维生素E琥珀酸酯12重量份。
上述原料优选重量配比如下:
蜂王浆冻干粉91重量份,左旋肉碱72重量份,葡萄籽提取物57重量份,维生素C21重量份,维生素E琥珀酸酯20重量份。
上述原料加入常规辅料,按照常规工艺制成可接受的胶囊剂、片剂、丸剂、颗粒剂、散剂、液体制剂或其他任何一种药剂学上所提到的剂型。
本发明蜂王浆组合物片剂的具体制备方法如下:
取原料左旋肉碱,葡萄籽提取物,维生素E琥珀酸酯,与80-90重量份的微晶纤维素混合,加入80%乙醇,过20目筛网制粒;沸腾干燥40-60分钟,待自然冷却至室温后,过20目筛整粒;加入蜂王浆冻干粉、维生素C,混合均匀,压片,制成片剂。
本发明蜂王浆组合物能够显著缓解慢性疲劳,增强机体耐受能力,明显减缓自由基的损伤及疲劳的发生,缓解体力疲劳。
下述实验例和实施例用于进一步说明但不限于本发明。
实验例1运动耐力实验
1.实验目的
通过运动耐力测试(小鼠游泳试验),观察本发明蜂王浆组合物的抗疲劳作用,为确定配方用量提供实验依据。
2.实验材料与试剂

组方一(G1):蜂王浆冻干粉83.3g;
组方二(G2):左旋肉碱83.3g;
组方三(G3):葡萄籽提取物50g;
组方四(G4):维生素C 25g;
组方五(G5):维生素E琥珀酸酯16.7g;
组方六(G6):蜂王浆冻干粉83.3g,左旋肉碱83.3g;
组方七(G7):蜂王浆冻干粉83.3g,葡萄籽提取物50g;
组方八(G8):蜂王浆冻干粉83.3g,左旋肉碱83.3g,葡萄籽提取物50g;
组方九(G9):蜂王浆冻干粉83.3g,左旋肉碱83.3g,维生素E琥珀酸酯16.7g
组方十(G10):左旋肉碱83.3g,葡萄籽提取物50g;
组方十一(G11):左旋肉碱83.3g,维生素C 25g;

组方十六(G16):蜂王浆冻干粉83.3g,左旋肉碱83.3g,葡萄籽提取物50g,维生素C 25g,维生素E琥珀酸酯16.7g;
组方十七(G17):蜂王浆冻干粉72g,左旋肉碱90g,葡萄籽提取物43g,维生素C29g,维生素E琥珀酸酯12g;
组方十八(G18):蜂王浆冻干粉91g,左旋肉碱72g,葡萄籽提取物57g,维生素C21g,维生素E琥珀酸酯20g;
组方十九(G19):蜂王浆冻干粉60g,左旋肉碱100g,葡萄籽提取物30g,维生素C40g,维生素E琥珀酸酯30g。
组方二十(G20):蜂王浆冻干粉100g,左旋肉碱60g,葡萄籽提取物80g,维生素C10g,维生素E琥珀酸酯5g。
2.2试剂:
肝/肌糖原测试盒,购自南京建成生物制品研究所;
浓硫酸(AR),南京化学试剂有限公司;
生理盐水,山东长富洁晶药业有限公司。
3.实验动物
ICR小鼠,♂,清洁级,体重18-22g,由扬州大学比较医学中心提供,
合格证号:SCXK(苏)2007–0001,实验期间小鼠自由饮食。
4.主要仪器
铝制游泳箱(50cm×50cm×40cm),铅丝,低温高速离心机:5804R型,Eppendrof公司;水浴锅:DK-S26型,上海精宏实验设备有限公司;电子称:BS224S型,Sartorius公司;秒表,温度计
5.实验分组
5.1剂量分组及受试样品给予时间
随机将小鼠分为21组,每组10只,第1组至第20组分别给G1~G20的药物,第21组为空白对照组,给予等体积的双蒸水,每组每日均灌胃1次,灌胃体积为0.2ml/10g,连续给予受试样品30天。
5.2样品配制
第1组至第20组:称取2.25g药物样品,用蒸馏水配至150ml;
空白对照组:双蒸水150ml。
6.实验方法
6.1负重游泳实验末次给药30min后,置小鼠于游泳箱中,水深不少于30cm,水温25±1℃,鼠尾根部负荷5%体重的铅皮,记录小鼠游泳开始至死亡的时间,作为小鼠游泳时间。
6.2小鼠血清尿素测定 末次给药30min后,在温度为30℃的水中不负重游泳90min,休息60min后摘眼球采血0.5mL(不加抗凝剂),置4℃冰箱3h,血凝固后2000r/min离心15min,取血清送连云港市第一人民医院检验科检测。
6.3肝糖原的测定末次给药30min后,在温度为25±1℃的水中不负重游泳90min,颈椎脱臼处死小鼠,用生理盐水洗净,并用滤纸吸干水分后,准确称取肝脏100mg,肝糖原检测试剂盒检测小鼠肝糖原含量。
6.4血乳酸的测定末次给药30min后采血,然后不负重在温度为30℃的水中游泳10min后停止。乳酸仪测定方法:分别在游泳前、游泳后、游泳后休息20min后各采血20μL加入40μL破膜液中,立即充分振荡破碎细胞用乳酸仪测定。(血乳酸曲线下面积=5×(游泳前血乳酸值+3×游泳后0min的血乳酸值+2×游泳后20min的血乳酸值)
7.观察指标 负重游泳时间,血乳酸、尿素、肝糖原值
8.统计方法实验数据用表示,采用t检验进行组间比较
9.实验结果
9.1本发明组合物对小鼠体重的影响
各组小鼠在给予G1~G20药物后,前、中,后期体重分别见下表所示,各组小鼠的初始体重和增重体重与对照组比较均无统计学差异(P>0.05),表明G1~G20药物均无明显的毒性。实验结果详见表1。

9.2本发明组合物对小鼠负重游泳时间的影响
经口给予小鼠G1~G20药物后,G1~G15药物与空白对照组比较,可以明显延长小鼠负重游泳时间,具有显著性差异(P<0.05),本发明组合物G16~G20药物与空白对照组比较,可以显著延长小鼠负重游泳时间,具有极显著性差异(P<0.01),且明显优于G1~G15药物。结果详见表2。

9.3本发明组合物对小鼠运动前后血乳酸的影响
经口给予小鼠本发明蜂王浆组合物后,本发明组合物G16~G20药物对小鼠运动后血乳酸曲线下面积与对照组比较有统计学差异(P<0.05),G1~G15药物组小鼠血乳酸曲线下面积与对照组比较虽有所降低,但并无统计学差异(P>0.05)。结果见表3。

9.4本发明组合物对小鼠肝糖原的影响
经口给予小鼠G1~G20药物后,G1~G15药物与空白对照组比较,小鼠肝糖原含量均有明显的升高,具有显著性差异(P<0.05),本发明组合物G16~G20药物与与空白对照组比较,小鼠肝糖原含量均有明显的升高,具有极显著性差异(P<0.01),且明显优于G1~G15药物。结果详见表4。表4本发明蜂王浆组合物对小鼠肝糖原含量的影响

9.5本发明组合物对小鼠血清尿素的影响
经口给予小鼠G1~G20药物后,G1~G15药物组与空白对照组比较,小鼠运动后血清尿素含量均有明显的降低,具有显著性差异(P<0.05),本发明组合物G16~G20药物与与空白对照组比较,小鼠运动后血清尿素含量均有明显的降低均有明显的降低,具有极显著性差异(P<0.01),且明显优于G1~G15药物。结果详见表5。

10.实验结论
本实验主要通过小鼠负重游泳实验,同时检测小鼠肝糖原的储备来观察本发明蜂王浆组合物的抗疲劳效果。初步研究结果显示如下:
1、本发明G16~G20组合物均能延长小鼠负重游泳时间(P<0.01),且效果明显优于其他G1~G15的组合物。
2、生化检测方面显示,本发明G16~G20组合物各剂量组均能减少运动后小鼠血清中葡萄糖无氧酵解所产生的乳酸含量,与对照组比较有显著性差异(P<0.05),而其他G1~G15的组合物虽然也能减少运动后小鼠血清中葡萄糖无氧酵解所产生的乳酸含量,但与对照组比较,无统计学差异(P>0.05);
3、本发明G16~G20组合物各剂量组均能显著提高小鼠肝脏中糖原的储备(P<0.01),且效果明显优于其他G1~G15的组合物;
4、高尿酸模型发现,本发明G16~G20组合物能显著降低小鼠游泳后血清中尿素的含量(P<0.01),且效果明显优于其他G1~G15的组合物;
11.讨论
上述实验证明本发明组合物能显著降低小鼠运动后血清中尿素及乳酸的含量,且能显著提高小鼠肝脏中糖原的储备,提示其可能通过减少能量代谢中两大主要能量物质蛋白质及葡萄糖的代谢而减少能量的损耗,有助于缓解运动负荷引起的疲劳;能延长小鼠负重游泳至力竭的时间。
本发明组合物主要原料为蜂王浆冻干粉,针对疲劳产生机制,还添加了左旋肉碱、葡萄籽提取物、维生素C、维生素E,具有缓解体力疲劳的保健功能。加入左旋肉碱在能量供给步骤起到促进作用,对于缓解机体疲劳起到有效地作用。加入葡萄籽提取物能够有效地缓解体力疲劳,葡萄籽原花青素能够保护和稳定维生素C,并有助于维生素C的吸收,还原维生素E自由基而使维生素E再生从而减少细胞内维生素E的损耗,增强维持机体正常的抗氧剂水平,缓疲劳的发生。能明显减缓自由基的损伤及疲劳的发生,有助缓解体力疲劳。
下述实施例均能实现上述实验例所述的效果。
实施例1 蜂王浆组合物片剂的制备
蜂王浆冻干粉83.3g 左旋肉碱83.3g 葡萄籽提取物50g
维生素C25g 维生素E琥珀酸酯16.7g
制成片剂1000片(每片重0.35g)
取上述左旋肉碱,葡萄籽提取物,维生素E琥珀酸酯,与85g的微晶纤维素混合,加入80%乙醇,过20目筛网制粒;沸腾干燥40-60分钟,待自然冷却至室温后,过20目筛整粒;加入蜂王浆冻干粉、维生素C,混合均匀,压片,制成片剂。
实施例2蜂王浆组合物片剂的制备
蜂王浆冻干粉72g,左旋肉碱90g,葡萄籽提取物43g,维生素C29g,维生素E琥珀酸酯12g。
取上述蜂王浆冻干粉、左旋肉碱、葡萄籽提取物、维生素E琥珀酸酯、维生素C,混合,加入二氧化硅、糊精、淀粉、磷酸盐、碳酸盐、甘露醇、山梨醇、山梨酸或钾盐、木糖醇、麦芽糖、葡萄糖、果糖、右旋糖苷、甘氨酸、淀粉、蔗糖、乳糖、甘露糖醇等稀释辅料中的一种或多种,干燥,粉碎,过筛,压片,制成片剂。
实施例3蜂王浆组合物片剂的制备
蜂王浆冻干粉91g,左旋肉碱72g,葡萄籽提取物57g,维生素C21g,维生素E琥珀酸酯20g。
取上述左旋肉碱,葡萄籽提取物,维生素E琥珀酸酯,与81g的微晶纤维素混合,加入适75%乙醇,混合,制成软材,制粒,干燥,整粒,加入蜂王浆冻干粉、维生素C,混合均匀,压片。
实施例4蜂王浆组合物硬胶囊的制备
蜂王浆冻干粉83.3g 左旋肉碱83.3g 葡萄籽提取物50g 维生素C25g
维生素E琥珀酸酯16.7g。
取上述原料,加入常规辅料,按照常规工艺装胶囊,即得。
实施例5蜂王浆组合物丸剂的制备
蜂王浆冻干粉72g,左旋肉碱90g,葡萄籽提取物43g,维生素C29g,维生素E琥珀酸酯12g。
取上述原料,加入常规辅料,按照常规工艺制丸,即得。
实施例6蜂王浆组合物口服液的制备
蜂王浆冻干粉91g,左旋肉碱72g,葡萄籽提取物57g,维生素C21g,维生素E琥珀酸酯20g。
取上述原料,加入常规辅料,按照常规工艺,制成口服液。
实施例7蜂王浆组合物软胶囊的制备
蜂王浆冻干粉85g,左旋肉碱65g,葡萄籽提取物70g,维生素C30g,维生素E琥珀酸酯28g。
取上述原料,加入常规辅料,按照常规工艺,制成软胶囊。
实施例8蜂王浆组合物散剂的制备
蜂王浆冻干粉60g,左旋肉碱100g,葡萄籽提取物30g,维生素C40g,维生素E琥珀酸酯30g。
取上述原料,加入常规辅料,按照常规工艺,制成散剂。
实施例9蜂王浆组合物颗粒的制备
蜂王浆冻干粉100g,左旋肉碱60g,葡萄籽提取物80g,维生素C10g,维生素E琥珀酸酯5g。
取上述原料,加入常规辅料,按照常规工艺,制成颗粒。
Claims (7)
1.一种蜂王浆组合物,其特征在于该组合物的原料组成为:
蜂王浆冻干粉60-100重量份,左旋肉碱60-100重量份,葡萄籽提取物30-80重量份,维生素C 10-40重量份,维生素E琥珀酸酯5-30重量份。
2.如权利要求1所述的蜂王浆组合物,其特征在于该组合物的原料组成为:
蜂王浆冻干粉83.3重量份,左旋肉碱83.3重量份,葡萄籽提取物50重量份,维生素C 25重量份,维生素E琥珀酸酯16.7重量份。
3.如权利要求1所述的蜂王浆组合物,其特征在于该组合物的原料组成为:
蜂王浆冻干粉72重量份,左旋肉碱90重量份,葡萄籽提取物43重量份,维生素C 29重量份,维生素E琥珀酸酯12重量份。
4.如权利要求1所述的蜂王浆组合物,其特征在于该组合物的原料组成为:
蜂王浆冻干粉91重量份,左旋肉碱72重量份,葡萄籽提取物57重量份,维生素C 21重量份,维生素E琥珀酸酯20重量份。
5.如权利要求1、2、3或4所述的蜂王浆组合物的制备方法,其特征在于该方法包括以下步骤:
取原料加入常规辅料,按照常规工艺制成可接受的胶囊剂、片剂、丸剂、颗粒剂、散剂、液体制剂或其他任何一种药剂学上所提到的剂型。
6.如权利要求1、2、3或4所述的蜂王浆组合物的片剂的制备方法,其特征在于该方法包括以下步骤:
取原料左旋肉碱,葡萄籽提取物,维生素E琥珀酸酯,与80-90重量份的微晶纤维素混合,加入80%乙醇,过20目筛网制粒;沸腾干燥40-60分钟,待自然冷却至室温后,过20目筛整粒;加入蜂王浆冻干粉、维生素C,混合均匀,压片,制成片剂。
7.如权利要求1、2、3或4所述的蜂王浆组合物在制备缓解慢性疲劳,增强机体耐受能力的药物中的应用。
