Abstract
本发明涉及一种中药保健食品云芝咀嚼片及其制备方法,它含有云芝菌丝粉30-40份,乳糖30-40份,木糖醇10-15份,甘露醇10-15份,淀粉5-10份,阿斯巴甜0.5-1份,硬脂酸镁0.8-1.5份。该咀嚼片每片质量约为0.5g,可直接口服、含化、咀嚼。

技术领域
本发明涉及一种中药保健食品云芝咀嚼片及其制备方法,属于中药制药领域。
背景技术
云芝为多孔菌科云芝属植物,是我国传统名贵真菌类药材之一,现代科学证明其有广泛的药理作用和医疗保健价值,其主要生理活性物质为云芝糖肽、多糖、三萜、甾醇、氨基酸、维生素、蛋白质等,此外还含有挥发油、树脂、生物碱等物质,具有清热、解毒、消炎、抗癌、保肝等功效,其中云芝糖肽和多糖具有良好的免疫调节作用,在调节血糖,控制血压,辅助肿瘤放化疗,促进睡眠等方面均具有显著疗效。市售云芝药材包括云芝孢子和云芝子实体,其中子实体是云芝糖肽、多糖等活性物质提取的主要来源,但云芝野生资源稀缺,人工栽培周期长(1~2个月),产量低,受气候影响显著,严重影响了云芝产品的开发生产,导致其市场价格高居不下。深层液态发酵以其周期短(10~12d),产量大等优势,大大缩减了云芝的生产成本,是深入研究云芝及其药用成分的有效方式。发酵所得菌丝体含有丰富的云芝糖肽、多糖、三萜、甾醇等有效成分,药用价值等同与云芝子实体。本发明以发酵云芝菌丝体为主要原料,制成一种能直接口服、含化、咀嚼的片状补品。
发明内容
本发明的目的是提供一种以云芝菌丝体为原料的咀嚼片及其制备方法,其主要配方如下:云芝菌丝粉30-40份,乳糖30-40份,木糖醇10-15份,甘露醇10-15份,淀粉5-10份,阿斯巴甜0.5-1份,硬脂酸镁0.8-1.5份。以上组分除云芝菌丝粉外其余均为市售现成产品。
云芝菌丝体的培养方式如下:
斜面培养基(g/L):马铃薯200,葡萄糖20,琼脂20,pH自然。
发酵培养基(g/L):葡萄糖20-30,蛋白胨8-10,MgSO4 1.5-2.0,KH2PO4 3.0-4.0,pH自然。
培养条件为:以孢子接种斜面培养基,25-30℃培养至孢子成熟,制得孢子悬液,以1.0-1.5*105 个孢子/mL的接种量接种至摇瓶发酵培养基,120-150 rpm摇床振荡培养5-7天,以10%接种量接种至装有3L发酵培养基的5L发酵罐中,通风量0.8-1.0 vvm,搅拌转速50-100 rpm,温度25-30℃,发酵6-8天。
菌丝体处理方式如下:
1、 发酵液经4℃,5000 rpm离心10min,沉淀菌丝体以去离子水重悬洗涤三遍去除培养基成分等杂质,离心后得到湿菌丝体;
2、 菌丝体经-80℃预冻,冷冻干燥机干燥得到干菌丝,保留了多糖、维生素、蛋白质等活性物质生物活性;
3、 干菌丝经研磨,过100目筛得到菌丝粉。
咀嚼片制备方式如下:菌丝粉及上述辅料按比例混匀,过40目筛,以硬脂酸镁为润滑剂,采用粉末直接压片法压制成型,每片质量约为0.5 g。
具体实施方式
实施例1 云芝孢子的液态深层发酵
取云芝孢子划线接种至装有斜面培养基的茄子瓶中,25-30℃恒温培养至大量孢子成熟,在超净工作台上用无菌生理盐水将孢子荡洗下来,制成1.0-1.5*106个/mL的孢子悬液,以10%的接种量接种至装有50mL发酵培养基的500 mL三角瓶中,25-30℃,120-150 rpm摇床振荡培养5-7天,以10%接种量接种至装有3L发酵培养基的5L发酵罐中,通风量0.8-1.0 vvm,搅拌转速50-100 rpm,温度25-30℃,发酵6-8天。
实施例2 云芝菌丝粉的制备
取上述发酵液于4℃,5000 rpm离心10min,去上清,沉淀为菌丝体及少量孢子。以去离子水重悬洗涤,4℃,5000 rpm离心10min,去上清,重复上述操作2次以洗净菌丝体中残余培养基成分,得到湿菌丝体。为了保持菌丝体的生物活性,采用冷冻干燥的方法进行干燥,得到的干菌丝经研磨,过100目筛得到菌丝粉。
实施例3 云芝咀嚼片的制备
取上述云芝菌丝粉30-40份,市售乳糖30-40份,木糖醇10-15份,甘露醇10-15份,淀粉5-10份,份阿斯巴甜0.5-1份,充分混匀,过40目筛,以0.8-1.5份硬脂酸镁为润滑剂,采用粉末直接压片法压制成型,每片质量约为0.5 g。
实施例4 本发明云芝咀嚼片的食用典型例证
例1:本发明产品经50例病人口服试验:50例认为本品口感较好,清凉、微甜,适合咀嚼、口含。
例2:李××,男,65岁,机械厂退休工人,有10年糖尿病史,空腹血糖浓度为9.6±0.3 mmol/l。服用本发明云芝咀嚼片,1日3次,每次2片,每片0.5 g,30天后血糖浓度明显降低,60天后,空腹血糖降为6.7 mmol/l,接近正常血糖浓度。
例3:王×,男,37岁,软件开发公司员工,工作压力大,常年加班,失眠多梦,导致身体免疫力下降,精力匮乏,工作效率低,易疲惫,易感冒,皮肤暗淡,生活质量低。服用本发明云芝咀嚼片,1日3次,每次2片,每片0.5 g,60天后,皮肤重现光泽,精力充沛,身体素质变好,睡眠渐佳。
Claims (3)
1.一种云芝咀嚼片,其特征在于它含有(按质量份计):云芝菌丝粉30-40份,乳糖30-40份,木糖醇10-15份,甘露醇10-15份,淀粉5-10份,阿斯巴甜0.5-1份,硬脂酸镁0.8-1.5份。
2.一种云芝咀嚼片的制备方法,其特征在于:将云芝菌丝粉30-40份,乳糖30-40份,木糖醇10-15份,甘露醇10-15份,淀粉5-10份,阿斯巴甜0.5-1份,硬脂酸镁0.8-1.5份混合制成;所用云芝菌丝粉是由云芝孢子经深层液态发酵制得菌丝体,经离心、洗涤、冷冻干燥、研磨、过筛得到菌丝粉。
3.如权利要求2所述的深层液态发酵,其特征在于:培养基组成为(g/L)葡萄糖20-30,蛋白胨8-10,MgSO4 1.5-2.0,KH2PO4 3.0-4.0,pH自然;培养条件为:5L发酵罐,通风量0.8-1.0 vvm,搅拌转速50-100 rpm,温度25-30℃,发酵6-8天。
