Abstract
"MEIO DE CULTURA DIFERENCIAL E SELETIVO PARA AS LEVEDURAS ZYGOSACCHAROMYCES BAILII E ZYGOSACCHAROMYCES BISPORUS, UTILIZAçãO DO MESMO, E, PROCESSO DE DETECçãO DE LEVEDURAS ZYGOSACCHAROMYCES BAILII E ZYGOSACCHAROMYCES BISPORUS". A invenção refere-se a um meio de cultura diferencial e seletivo para detectar leveduras das espécies Zygosaccharomyces bailii e Zygosaccharomyces bisporus, que permite reduzir drasticamente o tempo e trabalho envolvidos na identificação convencional destas espécies. De acordo com a presente invenção, a detecção de Zygosaccharomyces bailii e Zygosaccharomyces bisporus resume-se a um único teste que exige apenas a elaboração de um meio de cultura sólido ou líquido e a sua inoculação. Este meio de cultura é constituído por um meio mineral base, suplementado com oligoelementos e vitaminas, por glucose e ácido fórmico como únicas fontes de carbono e energia, e por um indicador ácido-base. O indicador ácido-base, em particular verde de bromocresol, no meio confere-lhe uma coloração verde que é convertida a azul por estas leveduras. Adicionalmente, as colónias apresentam uma coloração azul que é uma característica específica destas espécies e pode ser visível no meio entre as 48 e 96 horas de incubação, consoante a metodologia de inoculação utilizada. A invenção pode-se aplicar quer a estirpes de leveduras previamente isoladas e purificadas, quer a suspensões celulares de populações mistas de leveduras, para além de Zygosaccharomyces bailii e Zygosaccharomyces bisporus, para detecção destas espécies na indústria alimentar, nomeadamente em vinhos e outras bebidas. The invention relates to a differential and selective culture medium to detect yeasts of the species Zygosaccharomyces bailii and Zygosaccharomyces bisporus, which allows to drastically reduce the time and work involved in the conventional identification of these species. According to the present invention, the detection of Zygosaccharomyces bailii and Zygosaccharomyces bisporus is limited to a single test that requires only the elaboration of a solid or liquid culture medium and its inoculation. This culture medium consists of a base mineral medium, supplemented with trace elements and vitamins, glucose and formic acid as the sole sources of carbon and energy, and an acid-base indicator. The acid-base indicator, in particular bromocresol green, in the middle gives it a green color that is converted to blue by these yeasts. In addition, the colonies have a blue color that is a specific characteristic of these species and can be visible in the medium between 48 and 96 hours of incubation, depending on the inoculation methodology used. The invention can be applied either to strains of previously isolated and purified yeasts, or to cell suspensions of mixed yeast populations, in addition to Zygosaccharomyces bailii and Zygosaccharomyces bisporus, for the detection of these species in the food industry, namely in wines and other drinks. The medium may also be integrated into yeast identification test galleries.
