    Although scientific evidence is limited, clopidogrel is frequently used as prophylaxis for arterial thromboembolism in cats with hypertrophic cardiomyopathy (HCM). Evaluating effects of clopidogrel therapy in asymptomatic cats with HCM on (1) conventional whole blood aggregation (WBA), (2) alternative platelet aggregation assessed with tubes of the Plateletworks® assay and (3) standard coagulation parameters.     In this report two dogs with essential thrombocythaemia (ET) are described. Both dogs were presented more or less at the same time with a combination of reduced exercise tolerance and pale mucous membranes without any report of blood loss. Moderate-to-severe, Coomb's-negative anaemia and thrombocytosis (> 1249 x 10'/l) were present. In addition, the peripheral blood smear revealed the presence of basophilia and large numbers of abnormally shaped megakaryocytes in the bone marrow of both dogs. Treatment with vincristine (0.7 mg/m2 once intravenously) and hydroxyurea (500 mg/m2 p.o. per day) was started. Because of insufficient response to treatment after 3 weeks, the dosage of hydroxyurea was increased in both dogs to 2000 mg/m2 p.o. per day. The dogs deteriorated further, however, and were euthanized at 6 weeks after the start of treatment.     In this prospective randomized double-blind clinical study the anti-tumour activity of liposome-encapsulated muramyl tripeptide phosphatidylethanolamine (L-MTP-PE) was evaluated as an adjuvant immunotherapy in dogs with mammary tumours of the simple carcinoma type. Dogs were randomized after surgery to one of two treatment groups, in which they were treated with either L-MTP-PE (2 mg/m2 i.v.; Ciba Geigy Basel, Switzerland) twice weekly for eight weeks, or with empty liposomes according to the same protocol. The minimal follow-up period was one year. Thirteen dogs were entered in the L-MTP-PE group and fourteen dogs in the placebo control group. Only minor toxicities (fever and shivering during 10-24 hours) were seen in six dogs treated with L-MTP-PE, these being mainly of the smaller breeds.     The Bouvier des Flandres (BdF) dog is predisposed to develop prostate carcinoma (PCA). In humans, ethnic groups with higher prevalence of PCA have higher serum androgens concentrations and shorter polyglutamine (CAG) repeat lengths in the androgen receptor (AR) gene. In dogs, shorter CAG-I lengths are associated with increased PCA risk. To compare serum androgens concentrations in the BdF with other breeds and to determine whether CAG repeats length and plasma androgens concentrations are correlated. Androgens were measured in 46 BdF and in 53 other dog breed. Length of CAG-I and CAG-III repeats were measured in 41 BdF and correlated with androgen levels. In the BdF group shorter CAG-I and longer CAG-III repeat lengths were associated with lower androgen concentrations. As the shorter CAG-I repeat is associated with an increased PCA risk, higher androgen concentrations may protect against the development and progression of PCA.

    SummaryObjective: To evaluate the diagnostic value of cytology of endoscopically obtained biopsies in cats presented for chronic gastrointestinal complaints with emphasis on the diagnosis of low-grade alimentary lymphoma (LGAL). Materials and methods: Data of endoscopically obtained duodenal biopsies from 137 cats were evaluated retrospectively. Cytology was performed using the squash smear technique with subsequent Diff Quick® staining. Pathological findings were categorized according to type and grade of the inflammatory infiltrate. Moreover, reports were reviewed with regard to diagnosis of LGAL. Histopathology reports were analysed correspondingly and compared to cytology results. Histopathological samples were further evaluated by immunohistochemistry (IHC) if a diagnosis of LGAL had been expressed.     In dogs with primary hypoadrenocorticism, hypocortisolism and hypoaldosteronism usually are present, but these deficiencies also may occur in isolated forms. The diagnosis is commonly made by measuring plasma cortisol concentration before and after stimulation with ACTH, thereby ignoring aldosterone. In search of an alternative approach that would include assessment of glucocorticoid and mineralocorticoid production, 2 pairs of endocrine variables were measured: (1) plasma concentration of cortisol and ACTH, and (2) plasma aldosterone concentration and plasma renin activity. In addition, the cortisol-to-ACTH ratio (CAR) and the aldosterone-to-renin ratio (ARR) were calculated. Reference intervals were established in a population of 60 healthy dogs. In these dogs, CAR ranged from 1.1 to 26.1 and ARR ranged from 0.1 to 1.5. The variables were compared with those of 22 dogs with spontaneous primary hypoadrenocorticism. Plasma concentration of cortisol and ACTH in both groups of dogs overlapped, whereas CAR did not. Similarly, plasma aldosterone concentration and plasma renin activity overlapped, whereas ARR did not. These observations indicate that measurement of these endogenous variables (in one blood sample) allows the specific diagnoses of primary hypocortisolism and primary hypoaldosteronism.

    Objective—To assess intra- and interobserver repeatability of ocular biometric measurements obtained by means of high-resolution B-mode ultrasonography in dogs. Animals—6 Beagles without ocular abnormalities. Procedures—B-mode ultrasonography was performed bilaterally with a 10.5-MHz broadband compact linear array transducer. All measurements were made on 2 different occasions by 2 observers. The Bland-Altman method was used to assess agreement between measurements obtained by the 2 observers and between the 2 sets of measurements obtained by each observer. Results—Intra- and interobserver repeatability was highest for larger measurements, such as depth of the eye and depth of the anterior chamber. When repeatability was examined, bias was significantly different from 0 for only a few measurements, but the percentage difference between observations was as high as 180% for some measurements. Material und Methoden: Das Patientengut umfasste 129 Hunde und 19 Katzen, bei denen intraoperativ zytologische Abklatschbioptate unterschiedlicher Lokalisationen gewonnen wurden.     The reliability of vaginal cytology for determining the optimal mating period is controversial. The importance of the site for obtaining the smear (the vestibule or the vagina) and the best staining method are also unclear. Therefore, we investigated the reliability of vaginal cytology as compared to plasma progesterone concentrations for determining the optimal mating period. Additionally, the influence of different locations for obtaining the smears and of the staining methods, i.e. May-Grünwald-Giemsa or Papanicolaou, a method that stains keratin in superficial cells, was examined. In total, the vaginal cytology of 35 bitches during the follicular phase was examined three times a week, until the optimal time for mating was established, based on plasma progesterone concentrations measured with a 125Iodine radio-immunoassay.     For the first time cases of babesiosis have been diagnosed in dogs that had not been abroad. Three cases were seen in the province of Gelderland (Bennekom, Elspeet), two in North-Holland (Koog aan de Zaan). A female Dermacentor reticulatus tick, the main vector of the disease in Europe, was found on one of the dogs at Koog; previously specimens of this tick species had only occasionally been seen on dogs returning from abroad. Serological evidence was obtained that in at least four of the cases European Babesia canis, transmitted by Dermacentor ticks, was involved.

    Modern laser-based haematology analysers such as the ADVIA 120 have species-specific software and offer the possibility of assessing new haematological parameters. These parameters have yet to be evaluated, and as these analysers are often used in referral laboratories, it is important to know whether the values of haematological parameters change during sample transport. Therefore, samples of EDTA-anticoagulated blood from nine healthy dogs and EDTA- and citrate-anticoagulated blood from six healthy horses were collected and stored at room temperature for 72 and 48 hours, respectively. In canine samples, WBC and the red blood cell parameters Hb, Hb(cell), Ht, MCV, and MCHC changed significantly after only 24 hours of storage. Thus if canine blood samples need to be stored for 24 hours or longer, Hb, RBC, and MCH would appear to be more reliable parameters than Ht, Hb(cell), MCV, and MCHC.     Abstract. Liver cytology was evaluated in 28 healthy dogs 1–14 years of age with normal liver structure and function. Smears were stained with May–Grünwald–Giemsa. Hepatocytes had distinct cell borders, and cells did not overlap. Cells with two nuclei and cells with intranuclear crystalloid structures were observed regularly. Cytoplasm contained small numbers of vacuoles characteristic of glycogen and lipid and small amounts of pigment consistent with ceroid or bile. Nuclei were uniform. Small numbers of biliary epithelial cells were seen in most samples. Lymphocytes and neutrophils occurred in small numbers, with lipocytes, mast cells, fibrocytes, mesothelial cells, eosinophils, and Kupffer macrophages seen less frequently. Mean parenchy-mal cell sizes were significantly greater in older dogs, but no age-related differences were observed in nuclear size.     This retrospective study determined disease free survival (DFS) and progression free survival (PFS) in chemo-naïve dogs with multicentric or cranial mediastinal high-grade T-cell lymphoma, treated with a first-line CCNU-L(-chlorambucil)-CHOP protocol. Of thirteen dogs with multicentric lymphoma, 92.3% achieved a complete remission (CR), and the median DFS and PFS was 317 and 256 days, respectively. Three dogs had cranial mediastinal lymphoma, and achieved a CR with a median DFS and PFS of 978 and 1007 days, respectively. The oneand two-year DFS/PFS probability estimate for dogs with multicentric lymphoma was 0.50/0.46 and 0.42/0.38, respectively, for dogs with cranial mediastinal lymphoma 0.67/0.67. Neutropenia and thrombocytopenia were reported in 52.9% and 50% of the dogs, respectively, while 56.3% experienced grade 1-4 nephrotoxicity, hypothesized to be lomustine-induced.     The aims of this study were to establish the prevalence of innocent cardiac murmurs in clinically healthy puppies, to investigate a possible correlation between the presence of an innocent murmur and hematocrit, and to describe the auscultation characteristics of innocent murmurs. Lower hematocrit contributes to the genesis of innocent murmurs. Five hundred and eighty-four client-owned clinically healthy puppies, between 20 and 108 days old. Two cross-sectional surveys with a 1-year (n = 389 pups) pilot and a half-year (n = 195 pups) principal study periods. Cardiac auscultation was performed by a single, board-certified cardiologist. The majority of the dogs had visited an endemic area (most often the Mediterranean area or the Dutch Antilles), but two dogs became infected with Babesia in the Netherlands. Babesia infections were associated with a stay in an endemic area and an incubation period that are both significantly shorter (less than 3 months) than those for Ehrlichia and co-infections (more than 3 months).     Canine osteosarcoma, the most common bone tumor in dogs, is a well-established, naturally-occurring animal model for human OS. The aim of this study was to evaluate the clinical and hematological side-effects and to assess the efficacy of lobaplatin chemotherapy in dogs with appendicular osteosarcoma as an adjuvant therapy to surgical resection. Twenty-eight dogs without systemic signs of disease were treated with surgical resection of the tumor and adjuvant lobaplatin chemotherapy at a dose of 35 mg/m2, i.v., once every three weeks, for a maximum of 4 doses. Clinical signs of toxicosis were uncommon and consisted mainly of vomiting and depression. Hematological signs of toxicoses were common 7 to 10 days after lobaplatin chemotherapy and consisted of thrombocytopenia, leukopenia and neutropenia.     In this case report we describe an atypical clinical manifestation of malignant lymphoma in a horse. The most obvious clinical symptoms were hyperaemic mucosae and skin lesions. The skin and mucosal lesions appeared to be caused by a leukemic form of malignant lymphoma. The lymphocytosis consisted mainly of different populations of T-lymphocytes. Histopathology and immunohistochemistry identified the malignant lymphoma as a T-cell rich B-cell lymphoma.

    In a retrospective study (1995) cytological diagnoses of 3987 specimen of different origin in the dog were evaluated. Biopsies were performed at the Department of Clinical Sciences of Companion animals at the University of Utrecht or sent in by external practitioners. Biopsies were obtained from different regions including skin/subcutis, mammary gland, lymph node, external ear canal, kidney, urinary bladder, urethra, prostatic gland, testes, bone, bone marrow, synovia, nose, oral cavity, cerebrospinal fluid, bronchial lavage, lung, spleen, liver, conjunctiva, and retrobulbar region. The majority of biopsies were obtained from skin/subcutis (n = 1815), lymph node (n = 706), and mammary gland (n = 415). A high percentage of non-diagnostic biopsies were observed with biopsies of lung, mammary gland, cerebrospinal fluid, external ear canal, and oral cavity. The frequencies of diagnoses in the different areas will be presented and the diagnostic indications of cytology are discussed.

    The blood analyser Vet ABC was evaluated using the blood of dogs and cats. The correlation of results with those obtained with standard reference methods, reproducibility, and ease of operation were investigated. Good accuracy reproducibility were found in both species for leucocyte and erythrocyte counts, haematocrit, haemoglobin level, MCV, MCH, and MCHC. However, for some parameters new reference values will have to be established. The Vet ABC is not reliable for the measurement of platelets in the cat, and in the dog it does not give reproducible measurements of low numbers of platelets. In general, the Vet ABC is easy to use and does not need much maintenance.

    Malignant lymphoma in the dog is frequently postulated and used as a therapeutic model for non-Hodgkin's lymphoma (NHL) in humans. In this study DNA ploidy and the cell kinetic characteristics of canine malignant lymphoma were studied by flow cytometric (FCM) nuclear DNA measurements on fresh frozen tumor tissue from 94 dogs with NHL and on material from non-neoplastic lymph nodes from 20 dogs. The results were correlated with histomorphology, immunophenotype and survival. All non-neoplastic tissues were diploid, whereas of the 94 lymphomas 74 were diploid or near-diploid and 20 aneuploid. Of the aneuploid lymphomas, 1 contained a hypoploid cell population. DNA-indices of the aneuploid peaks ranged from 0.87 to 1.21 (mean 1.11). The mean S-phase fraction (8.2%, SD 4.8) was significantly lower in the non-neoplastic tissues than in the lymphomas (11.4%, SD 5.1).     Epidermal growth factor (EGFR), oestrogen (ER), and progestin (PR) receptor concentrations were determined by radioligand binding assay in non-affected mammary tissues (n = 13) and benign (n = 11) and primary/locally recurrent malignant proliferative mammary lesions (n = 45) and metastases (n = 19) in 65 female dogs. The number of specimens expressing EGFR was not significantly different among these tissues, but EGFR concentration was lower in metastases (P = 0.02) than in benign or primary/locally recurrent malignant lesions not mixed with non-affected mammary tissue. The presence of non-affected mammary tissue in primary cancer specimens was noticed as a factor that may influence results of receptor measurements. No relation was found between the expression of EGFR and that of ER or PR in non-affected or in tumorous mammary tissues.     Mast cell tumours (MCT) are common skin tumours in dogs. If complete surgical removal of the tumours is not possible, then another therapy is needed. In the current study we tested the therapeutic effect of intratumoural injection of interleukin-2 (IL-2). Seven dogs had non-resectable cutaneous MCT. The tumours were injected with 4.5×10(6) IU IL-2. The early clinical effects in the seven dogs with cutaneous MCT were: complete regression (CR) in two dogs; partial regression (PR) in four, and stable disease (SD) in one dog. The final clinical effects were CR in three dogs, PR in two dogs, and PD in two dogs. This pilot study shows that intratumoural IL-2 application can exert an anti-MCT effect. A larger study would be required to precisely establish the magnitude of the therapeutic effect against MCT. A single application of IL-2 in cases of non-resectable MCT has no observable side-effects.
