COMPLEMENT
FIXATION TEST
DEMONSTRATE PRESENCE OF ANTIBODY
(E.g. Wassermann Test - Detection of Syphilis Antibody)
STEP 1:
Natural complement inactivated
at 56°C for 30 minutes.
Patient’s serum is mixed with
known antigen. The mixture is
incubated.
STEP 2:
Complement (from guinea pig) is
added to the mixture and
incubated. This allows for any
antibodies in the patient’s serum
to bind to antigen and fix
(consume) the complement.
STEP 3:
After incubation, sheep RBCs
coated with hemolysin (anti-sheep
RBC antibodies) are added to the
mixture. The tube is incubated
again and observed for hemolysis.
STEP 4:

If no hemolysis occurs,
