alpha-Synuclein is a highly conserved presynaptic protein with probable roles in normal synaptic development and plasticity as well as neurodegenerative disease, although its molecular function is not yet clear. To identify potential protein binding partners of alpha-synuclein, we performed co-immunoprecipitations using a monoclonal antibody (H3C) against its C-terminus. More than 20 detectable proteins were specifically co-immunoprecipitated from zebra finch and mouse forebrain extracts. One of these, with relative mobility of 55 kDa, was identified through microsequencing as a mixture of alpha- and beta-tubulin. Tubulin was specifically recovered from a mouse forebrain cytosolic extract by a GST/alpha-synuclein fusion protein immobilized on glutathione-Sepharose beads. In the converse experiment, alpha-synuclein bound to a column prepared from purified bovine brain tubulin immobilized upon CNBr-Sepharose. alpha-Synuclein does not appear to bind assembled microtubules, however, as alpha-synuclein did not pellet with polymerized microtubules in a standard assay for microtubule-associated proteins. Likewise, when a fusion construct of alpha-synuclein and green fluorescent protein (GFP) was expressed in African green monkey kidney epithelial (CV-1) cells, the fusion protein did not colocalize with endogenous microtubules. We conclude that alpha-synuclein may interact specifically with heterodimeric tubulin, but not microtubules, in the neuronal cytosol.

Bird vocal duets are joint displays where two individuals, generally a mated pair, produce temporally coordinated vocalizations. Duets may contribute to pair bond maintenance, mate guarding or collaborative defence of resources. The degree of coordination between mates and the variety of vocalizations, however, vary considerably. Although only 3–4.3% of bird species have been reported to duet, this may be because studies have generally focused on conspicuous duets, and more private forms of duet might have been overlooked. We investigated private vocal communication between mates in wild zebra finches, Taeniopygia guttata, a gregarious Australian songbird that forms life-long pair bonds. The partners are inseparable unless nest building, incubating or brooding. Using microphones inside nestboxes, we monitored interactive communication between partners at the nest and its variation during different stages of breeding. After periods of separation, partners performed coordinated mutual vocal displays involving specific soft vocal elements that fulfilled all the criteria used to define duets. In addition, using playback experiments, we obtained preliminary results suggesting that these soft calls could allow mate recognition. Thus, we propose that mutual displays at the nest in zebra finches represent private vocal duets and may function to mediate pair bond maintenance.

Parental care plays a key role in ontogeny, life-history trade-offs, sexual selection and intra-familial conflict. Studies focusing on understanding causes and consequences of variation in parental effort need to quantify parental behaviour accurately. The applied methods are, however, diverse even for a given species and type of parental effort, and rarely validated for accuracy. Here we focus on variability of parental behaviour from a methodological perspective to investigate the effect of different samplings on various estimates of parental effort. We used nest box cameras in a captive breeding population of zebra finches, Taenio-pygia guttata, a widely used model system of sexual selection, intra-familial dynamics and parental care. We investigated diurnal and reproductive stage-dependent variation in parental effort (including incubation, brooding, nest attendance and number of feedings) based on 12h and 3h continuous video-recordings taken at various reproductive stages. We then investigated whether shorter (1h) sampling periods provided comparable estimates of overall parental effort and division of labour to those of longer (3h) sampling periods. Our study confirmed female-biased division of labour during incubation, and showed that the difference between female and male effort diminishes with advancing reproductive stage. We found individually consistent parental behaviours within given days of incubation and nestling provisioning. Furthermore, parental behaviour was consistent over the different stages of incubation, however, only female brooding was consistent over nestling provisioning. Parental effort during incubation did not predict parental effort during nestling provisioning. Our analyses revealed that 1h sampling may be influenced heavily by stochastic and diurnal variation. We suggest using a single longer sampling period (3h) may provide a consistent and accurate estimate for overall parental effort during incubation in zebra finches. Due to the large within-individual variation, we suggest repeated longer sampling over the reproductive stage may be necessary for accurate estimates of parental effort post-hatching.

Consistent individual behavioural differences ('animal personalities') are documented across a variety of animal taxa. Sexual selection, especially assortative mating has been suggested as a possible mechanism contributing to the maintenance of different personality types within populations but little is known about non-random pair-formation with respect to personality traits in unconstrained choice tests. We here tested whether female mating preferences were non-random with respect to male and female neophobia in zebra finches (Taeniopygia guttata), an important avian model of mate choice and animal personality research. Male and female neophobia was assessed by attaching novel objects to birds' feeders. Females' mating preferences were tested with randomly assigned, unfamiliar males in a four-way choice apparatus. Females associated most with males with neophobia scores similar to their own. These results provide evidence that mating preferences and personality traits can covary, supporting evolutionary scenarios of assortative mating contributing to the maintenance of personality traits.
