Abstract
POSTUPAK ZA DOBIJANJE 1,2,4-TRIAZOLON JEDINJENJA, formule: u kojoj R predstavlja vodonik, C1-4 alkil, C1-4 acil, fenil- C1-4 alkil ili fenil-C1-4 acil; n ima vrednost 2-4; Y je hloro; i Z predstavlja vodonik ili grupu , gde je R1 vodonik, fluoro, metoksi ili trifluorometil, i njihovih farmaceutski prihvatljivih soli naznač en time, što hidrazin reaguje sa jedinjenjem formule: u kojoj je P kiselo-labilna zaštitna grupa, kao što je metoksimetil, C1-4 alkil ili C1-4 alkilfenil, u metanolu, na temperaturi oko ) C, da bi se dobija hidrazidni ntermedijer formule: pa se u reakcionu smešu dodaje jedinjenje formule: u kojoj Z i n imaju značenje kao gore, na temperaturi od 0-5 C, da bi se dobilo jedinjenje formule: u kojoj su Z, n i P kao što je definisano gore; što se dobijeno jedinjenje formule (IV) dalje ciklizuje, zagrevanjem na temperaturi refluksa, u 5% rastvoru KOH, da bi se dobilo jedinjenje formule: koje se alkiluje sa 1-(3-hloropropil)-4-(3-hlorofenil) piperazinom, u rastvaraču kao što je acetonitril i na temperaturi refluksa, pri čemu se dodaje blaga kiselina, kao što je tetrabutilamonijum hidro sulfat, da bi se izvršilo cepanje zaštitne grupe P i dobilo jedinjenje formule I u kome R predstavlja vodonik; i što se, prema potrebi, dobija jedinjenje formule I u kojoj je R različito od vodonika reakcijom Na-soli jedinjenja formule I u kojoj R predstavlja vodonik, sa jedinjenjem RX, u kome R ima gornja značenja osim vodonika, a X može biti hlor, brom, jod ili tozilat; i što se, prema potrebi, dobijeno jedinjenje formule I prevelo u svoju farmaceutski prihvatljivu adicionu so sa kiselinom.
