Abstract
Description
そこで、経口投与時におけるCoQ10の吸収性を改善し簡便で、効果的かつ経済的な技術の提供が求められている。
CoQ10の吸収性を改善するため、例えば、CoQ10を油に加熱溶解する方法(例えば、特許文献1参照)、水溶性化する技術を用いる方法(例えば、特許文献2参照)、シクロデキストリンで包接化する技術を用いる方法(例えば、特許文献3参照)等が提案されている。
しかしながら、特許文献1に記載のCoQ10を油に加熱溶解することで得られるCoQ10含有素材では、経口投与における体内吸収性が充分でないという問題がある。特許文献2に記載のCoQ10を水溶性化する技術では、カゼインNa、デキストリン、およびCoQ10を混合、撹拌後、凍結乾燥するという工程を経なければならない。また、CoQ10に対し、カゼインNaおよびデキストリンを少なくとも25倍以上と多量に配合しないと充分な効果が得られないという問題がある。一方、特許文献3に記載のCoQ10をシクロデキストリンで包接する技術では、包接化するための煩雑な工程を経なければならず、生産コストが高価になるという問題がある。
すなわち、本発明は、以下の(1)~(18)を提供する。
(1)種子処理物と、コエンザイムQ10とを含む経口摂取用組成物。
(2)種子処理物が大豆タンパクまたは大豆タンパク加水分解物から選ばれる少なくとも1種類である、上記(1)に記載の経口摂取用組成物。
(3)大豆タンパクが、酸処理物、アルカリ処理物またはアルコール処理物である、上記(2)に記載の経口摂取組成物。
(4)大豆タンパク加水分解物が酵素加水分解物である、上記(2)に記載の経口摂取組成物。
(5)種子処理物が、種子の粉砕粉末および種子の抽出物より得られた粉末から選ばれる一種以上の粉末である、上記(1)に記載の経口摂取用組成物。
(6)種子の粉砕粉末が、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分を粉砕することによって得られた粉末である、上記(5)に記載の経口摂取用組成物。
(7)種子の種皮、胚乳および胚芽が、焙煎した種子の種皮、胚乳および胚芽、または脱脂した種子の種皮、胚乳および胚芽である、上記(6)に記載の経口摂取用組成物。

CoQ10はソラネソールとベンゾキノンを用いた合成法、酵母やバクテリア等の微生物を用いた発酵法により製造することができる。
大豆タンパク粉には抽出大豆タンパク粉、濃縮大豆タンパク粉、分離大豆タンパク粉等がある。これらの元になる原料豆乳の一般的な製造工程は、以下のような工程からなる。すなわち、原料大豆の脱皮工程→脱胚軸工程(土壌細菌、不快味成分の除去)→浸漬工程(膨潤、少糖類の除去)→磨砕(呉)工程(微細化)→加熱工程(85~95℃、トリプシンインヒビター、リポキシゲナーゼ等の失活)→タンパク質の抽出分離工程(おからの除去)→調整工程(植物油、糖等の添加)→殺菌工程→均質化工程→充填工程である。近年、製造技術の改良がなされ、青臭さのない高品質の豆乳製品が製造されるようになっている。
一方、大豆特有の青臭みやえぐみの原因となる成分を低減した大豆の品種改良が進められており、青臭みの原因であるリポキシゲナーゼを無くし、強いえぐみを呈するグループAアセチルサポニンを欠失させた大豆品種キヌサヤカ(だいず農林130号)等が育出され青臭さやえぐみが少ないより良質な豆乳が生産されるようになって来ている。
乾燥豆乳粉末中のタンパク含量は40~44%程度、これに、部分精製を加えた抽出大豆タンパク粉中のタンパク含量は60%程度である。
分離大豆タンパクは、大豆タンパクの中では最も精製度が高いもので、脱脂大豆粉から希アルカリ溶液(pH8~9)でタンパク質を抽出後、不溶成分を遠心分離によって沈殿したタンパク質を回収し、水洗後水に懸濁して噴霧乾燥することによって得られる。
本発明で使用する大豆タンパクやその加水分解物に含まれるタンパク含有量は40%以上が好ましく、より好ましくは60%以上である。
本発明はCoQ10の生体吸収性を高めることができるだけではなく、同時に栄養学上の優れた特性を活かした経口摂取用組成物を製造することが可能となる。
大豆タンパクやその加水分解物は水やアルコールに均一の分散を作りやすく、それを使って混合してもよい。配合割合が質量比で大豆タンパクやその加水分解物が、CoQ10に対して好ましくは0.01倍以上であり、より好ましくは0.05倍以上である。
CoQ10と大豆由来粉の粉体の大きさは、どの種類でもそれぞれ0.1から500μmが好ましく、より好ましくは5から180μmである。
種子の粉砕粉末の具体例としては、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分を粉砕することによって得られた粉末、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分を焙煎したものを粉砕することによって得られた粉末、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分を脱脂したものを粉砕することによって得られた粉末等が挙げられる。
種子の抽出物から得られた粉砕粉末としては、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分から、圧搾または水若しくはアルコール類を含む水系溶媒を用いて抽出された抽出液を乾燥させることによって得られた粉末、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分を加熱処理したものから抽出された抽出液を乾燥させることによって得られた粉末、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分から抽出された抽出液のタンパク画分を乾燥させた粉末等が挙げられる。
一般的な大豆の粉砕粉末としては、黄粉、全脂大豆粉、脱脂大豆粉等がある。本発明に用いる大豆の種子から得られた粉末としては、丸大豆粉、丸大豆黄粉、脱脂大豆粉、脱脂大豆黄粉が好ましい。
黄粉は、大豆を焙煎して製粉した食品であり、焙煎することによってリポキシゲナーゼが不飽和脂肪酸に接することなく加熱失活する。それによって、不快臭がなく香ばしい味わいを有するものを作ることができる。また、さらに不快味を除くため脱皮、脱胚軸してもよい。
大豆の焙煎は、丸大豆または脱脂大豆を平釜で100~200℃、10~30分の条件で加熱することによって行われ、通常は150℃の温度で15分間程度行なわれる。現在では、さらに外観および食味に優れた黄粉を得る目的で、焙煎砂を用いた回転式焙煎機によって、220℃前後で30秒程度と高温短時間焙煎する方法が多く行われている。この焙煎処理によって、消化吸収阻害を起こすトリプシン阻害因子等の生大豆に存在する生理上好ましくない成分を失活させ除くことができる。なお、黄粉や大豆粉は全粒大豆を破砕することによって製造されるが、品質向上のため、粉砕の前に粗砕して、脱皮する工程を入れてもよい。
また、大豆から得られた粉末としては、上述した豆乳粉末、大豆タンパク粉末または大豆タンパク加水分解物粉末等の大豆の種子抽出物から得られた粉末を用いることができる。
このようにして得られた、粉砕、磨砕物は腐敗等による変質を防止するために乾燥する。乾燥方法としては特に限定されないが、例えば棚段式の通風乾燥器、通風加熱乾燥器、凍結乾燥器、ドラム乾燥機、噴霧乾燥機等を用いて乾燥する。乾燥温度、乾燥時間は、乾燥粉体の食味性や保存性を考慮しつつ適宜決めればよい。
このように、大豆の粉砕粉末や抽出物より得られた粉末を用いることによって、CoQ10の生体吸収性を高めることができるだけではなく、同時に種々の大豆品種が持ち合わせている農産製造上または栄養学上の優れた特性を活かした経口摂取用組成物を製造することが可能となる。
CoQ10と大豆粉を混合する場合、CoQ10の結晶をそのまま混和する方法、予めCoQ10を溶融したり、油脂等に溶解して液状となした後、大豆黄粉や大豆タンパク粉に注加しまぶすような形で混合することができる。還元型CoQ10と大豆粉の混合物は、例えば、酸化型CoQ10をハイドロサルファイトナトリウムや水素化ホウ素ナトリウム等の還元剤によって還元し、酸化型CoQ10と同様に大豆黄粉や大豆タンパク粉に混合することによって製造することができる。なお、その際、窒素気流下等の無酸素状態下で処理することにより、還元型Q10の酸化型Q10への変化を防ぐことができる。
一方、機能性食品として製品化する場合には、食品に用いられる添加剤、例えば甘味料、着色料、保存料、増粘安定剤、酸化防止剤、発色剤、漂白剤、防菌防黴剤、ガムベース、苦味料、酵素、光沢剤、酸味料、調味料、乳化剤、強化剤、製造用剤、香料、香辛料抽出物等を用いることができる。
(摂餌方法)
8週齢のSD系ラット(オス、一群3匹、体重250~300g、試験前日夕方より試験終了まで絶食させた)に対し、実施例および比較例で調製した経口摂取用組成物をCoQ10量として100mg/kgラット重となるように経口的に強制摂取させた後、投与3時間後に尾静脈よりヘパリン加真空採血管を用いて採血し、血漿を分離した。
得られた血漿を1ml分取し、血漿中のCoQ10の全量が酸化型になるように、2%FeCl3-エタノール溶液を2.5ml加えて混合し、還元型CoQ10を酸化型CoQ10に変換した。次いで、ヘキサンを5ml加えて激しく混合し、血漿中の酸化型CoQ10を抽出した後、遠心分離し、ヘキサン層を別の試験管に採取した。この操作を3回繰り返し得られたヘキサン抽出液15mlをエバポレーターにて減圧濃縮〔1.3~13.3kPa(10~100torr)〕し、その後、イソプロパノールを0.2ml加えて溶解した。得られたイソプロパノール溶液0.2mlを、下記の条件でHPLCにより分析し、血漿1ml中の濃度を算出した。結果を表1及び表2に示す。

酸化型CoQ10(三菱瓦斯化学株式会社製)8g、種子処理物として大豆から大豆油を抽出分離後の脱脂大豆をアルコール洗浄し可溶性の糖類を除いた大豆タンパクを2gの割合でポリびんに入れ、良く振って混合した後、オリーブ油で分散させ、濃度12.5質量%の経口摂取用組成物を調製した。得られた経口摂取用組成物を用いてCoQ10生体吸収性試験を行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)8g、種子処理物として大豆から大豆油を抽出分離した脱脂大豆をアルコール洗浄し可溶性の糖類を除いた後、酵素加水分解処理した大豆タンパク2gを用いたこと以外は実施例1と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)8g、種子処理物として大豆から大豆油を抽出分離した脱脂大豆から不溶分を除いた後、アルカリ処理した大豆タンパク2gを用いたこと以外は実施例1と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)8g、種子処理物として大豆の皮を取り除いた脱皮大豆を加熱後粉砕した大豆粉末2gを用いたこと以外は実施例1と同様に行った。

酸化型CoQ10(三菱瓦斯化学株式会社製)9.5g、種子処理物として大豆から大豆油を抽出分離した脱脂大豆をアルコール洗浄し可溶性の糖類を除いた後、酵素加水分解処理した大豆タンパク0.5gを用いたこと以外は実施例1と同様に行った。
酸化型CoQ10を10g使用し、種子処理物を使用しなかったこと以外は実施例1と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を20質量部、種子処理物として豆乳粉(不二製油株式会社製)を80質量部の割合で乳鉢に加えよく混合した後、これを水に分散させ、濃度12.5質量%の経口摂取用組成物を調製した。得られた経口摂取用組成物を用いてCoQ10の生体吸収性試験を行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を20質量部、種子処理物として豆乳粉(不二製油株式会社製)を80質量部の割合で乳鉢に加えよく混合した後、これをオリーブ油に分散させ、濃度12.5質量%の経口摂取用組成物を調製し用いたこと以外は、実施例7と同様に行った。
豆乳粉のかわりにリポキシゲナーゼ欠失大豆である「すずさやか(だいず農林125号)」より調製された大豆粉(池田屋製)を用いたこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を80質量部、大豆タンパク粉(日清オイリオ株式会社製)を20質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を80質量部、豆乳粉(不二製油株式会社製)を20質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を20質量部、ゴマ粉(株式会社真誠製)を80質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を80質量部、ゴマ粉(株式会社真誠製)を20質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を90質量部、大豆タンパク加水分解物(HSP310タツア・ジャパン社製)を10質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を95質量部、大豆タンパク粉(日清オイリオ株式会社製)を5質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
豆乳粉のかわりに黄粉(株式会社真誠製)を用いたこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を20質量部、焙煎したコーヒー豆(コロンビア産:キーコーヒー株式会社製)の粉砕品を80質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を80質量部、焙煎したコーヒー豆(コロンビア産:キーコーヒー株式会社製)の粉砕品を20質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を99質量部、大豆タンパク粉(日清オイリオ株式会社製)を1質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)を20質量部、小麦粉(強力粉:日清製粉株式会社製)を80質量部の割合で混合したものを使用したこと以外は実施例8と同様に行った。
酸化型CoQ10(三菱瓦斯化学株式会社製)より、還元剤にハイドロサルファイトナトリウムを用いて調製した還元型CoQ10を20質量部、黄粉(株式会社真誠製)を80質量部の割合で混合したものを使用したこと以外は実施例7と同様に行った。
還元型CoQ10を100質量部使用し、種子処理物を使用しなかったこと以外は、実施例8と同様に行った。
Claims (18)
- 種子処理物と、コエンザイムQ10とを含む経口摂取用組成物。
- 種子処理物が大豆タンパクまたは大豆タンパク加水分解物から選ばれる少なくとも1種類である、請求項1に記載の経口摂取用組成物。
- 大豆タンパクが、酸処理物、アルカリ処理物またはアルコール処理物である、請求項2に記載の経口摂取組成物。
- 大豆タンパク加水分解物が酵素加水分解物である、請求項2に記載の経口摂取組成物。
- 種子処理物が、種子の粉砕粉末および種子の抽出物より得られた粉末から選ばれる一種以上の粉末である、請求項1に記載の経口摂取用組成物。
- 種子の粉砕粉末が、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分を粉砕することによって得られた粉末である、請求項5に記載の経口摂取用組成物。
- 種子の種皮、胚乳および胚芽が、焙煎した種子の種皮、胚乳および胚芽、または脱脂した種子の種皮、胚乳および胚芽である、請求項6に記載の経口摂取用組成物。
- 種子の抽出物から得られた粉末が、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分から、圧搾または水若しくはアルコール類を含む水系溶媒を用いて抽出された抽出液を乾燥させることによって得られた粉末である、請求項5に記載の経口摂取用組成物。
- 種子の種皮、胚乳および胚芽が、加熱処理した種子の種皮、胚乳および胚芽、または脱脂した種子の種皮、胚乳および胚芽である、請求項8に記載の経口摂取用組成物。
- 種子の抽出物から得られた粉末が、種子の種皮、胚乳および胚芽から選ばれる一種以上の部分から、圧搾または水若しくはアルコール類を含む水系溶媒を用いて抽出された抽出液のタンパク画分を乾燥させた粉末である、請求項8または9に記載の経口摂取用組成物。
- 種子の粉砕粉末および種子の抽出物から得られた粉末より選ばれる一種以上の粉末が、豆科植物(Fabaceae)の種子から得られた粉末である、請求項5に記載の経口摂取用組成物。
- 種子の粉砕粉末および種子の抽出物から得られた粉末より選ばれる一種以上の粉末が、大豆(Glycine max)の種子から得られた粉末である、請求項11に記載の経口摂取用組成物。
- 大豆の種子から得られた粉末が、丸大豆粉若しくは丸大豆黄粉または脱脂大豆粉若しくは脱脂大豆黄粉である、請求項12に記載の経口摂取用組成物。
- 大豆の種子から得られた粉末が、豆乳粉末、大豆タンパク粉末または大豆タンパク加水分解物粉末である、請求項12に記載の経口摂取用組成物。
- 種子処理物のコエンザイムQ10に対する配合割合が、質量比で0.01倍以上である、請求項1~14のいずれかに記載の経口摂取用組成物。
- コエンザイムQ10が、酸化型および還元型コエンザイムQ10から選ばれる一種以上である、請求項1~15のいずれかに記載の経口摂取用組成物。
- 請求項1~16のいずれかに記載のコエンザイムQ10の経口摂取用組成物を用いた、医薬品。
- 請求項1~16のいずれかに記載のコエンザイムQ10の経口摂取用組成物を用いた、機能性食品。
