Description

關於保存在液體營養產品中的TGF-β生物活性的方法,與此等方法有關的參考文獻包括Cerletti之美國專利第7,057,016號或第6,057,430號。

基於由TGF-β所提供的許多利益,通常重要的是生長因子存在於或補充至各種液體營養產品中。例如,在營養產品中的某些蛋白質來源可提供TGF-β來源。或者,若營養產品本身不含有TGF-β,則生長因子可補充至產品中。然而,在任一情況中,經由液體營養產品的製造法難以保留TGF-β的生物活性。
貨架期穩定的非冷藏性液體營養產品典型地利用高熱處理加工,以破壞病原菌芽胞。常見的甑餾法可能加熱產物至130℃經2.5分鐘,其取決於營養產品的固體含量。或者,可能利用超高溫(UHT)。以UHT加熱產品至135-150℃之溫度範圍經1.5到15秒。TGF-β(不耐熱生物活性化合物)常常不可存活於製造貨架期穩定的液體營養產品所必要而常見的甑餾法。這些方法常使TGF-β的天然結構顯著變性,藉此使其成為生理惰性且不能在人的內臟中活化。

在到達任何加工廠之前,以乳製品為主之原料,諸如脫脂奶粉(亦已知為脫脂奶粉(skim milk powder))、乳清蛋白粉、煉乳、酪蛋白及其他原料的製造需要某種加熱程度以確保安全性。根據本發明,以乳製品為主之原料係以其接受的累積熱處理為基準分類,如表1及2中所示。因為難以決定熱處理類型,所以原料接受檢查,測量原料的未變性乳清蛋白氮(WPN)水平以傳達此資訊。表1顯示脫脂奶粉的一般公認之處理參數及WPN水平。

因此,如本文所使用之〝低溫加熱〞脫脂奶粉被定義成一種來到加工時含有至少約6毫克/公克之WPN的奶粉。低溫加熱脫脂奶粉有可能經歷約30秒到5分鐘暴露於低於約70℃之溫度。
〝中溫加熱〞脫脂奶粉被定義成一種來到加工時含有介於約1.51毫克/公克與約5.99毫克/公克之間的WPN之奶粉。中溫加熱脫脂奶粉有可能經歷約15到25分鐘暴露於約70℃到78℃之範圍內的溫度。
如本文所使用之〝高溫加熱〞脫脂奶粉被定義成一種來到加工時含有約1.5毫克/公克或更少的WPN之奶粉。高溫加熱脫脂奶粉有可能經歷約30分鐘或更長暴露於至少88℃之溫度。
表2顯示乳清蛋白粉的處理參數及WPN水平,包括乳清蛋白濃縮物及乳清蛋白分離物。參閱Mahmoud等人之Factors Affecting Measurement of Undenatured Whey Protein Nitrogen in Dried Whey by a Modified Harland-Ashworth Test,J. Dairy Sci. 73:1694-1699(1990)。
在乳清蛋白粉中的WPN水平大於標準的脫脂奶粉之該水平,因為在乳清蛋白粉中的乳清蛋白組成物(諸如具有約50%蛋白質之乳清蛋白濃縮物)以無水為基準計比脫脂奶粉多至少6至7倍。當以方法偵測乳清蛋白中的變性時,則預期乳清蛋白粉的結果比以脫脂奶粉所獲得的結果更高。
因此,如本文所使用之〝低溫加熱〞乳清蛋白粉被定義成一種來到加工時含有至少約14.5毫克/公克之WPN的粉。低溫加熱乳清蛋白粉有可能經歷約30秒到30分鐘暴露於少於約63℃之溫度。
〝中溫加熱〞乳清蛋白粉被定義成一種來到加工時含有介於約5.99毫克/公克與約14.49毫克/公克之間的WPN之粉。中溫加熱乳清蛋白粉有可能經歷約15到25分鐘暴露於約70℃到78℃之範圍內的溫度。
如本文所使用之〝高溫加熱〞乳清蛋白粉被定義成一種來到加工時含有約6.0毫克/公克或更少的WPN之粉。高溫加熱乳清蛋白粉有可能經歷約30分鐘或更長暴露於至少91℃之溫度。
根據本發明的成份選擇具有重要性,因為熱處理程度衝擊蛋白質來源的物理化學性質與其在食品加工中的價值。以熱誘發之乳清蛋白變性引起乳清蛋白與酪蛋白微膠粒表面交互作用。此變性提供牛奶酪蛋白在液體配方製造期間更高的熱穩定性。然而,此方法亦使許多存在於牛奶中的生物活性組份變性,諸如生長因子,如TGF-β。

在另一具體實例中,營養產品之蛋白質材料的選擇包括選自全脂奶粉、牛奶蛋白濃縮物、牛乳蛋白分離物及其組合之酪蛋白來源。在各具體實例中,酪蛋白源已以最少的熱處理發展史為基準選擇。TGF-β可藉由減至最少的加熱量施予蛋白質材料而有更好的保護且可在產品製造期間存活於UHT法。在乳清材料中的TGF-β對熱變性比存在於含酪蛋白成份中TGF-β更敏感。這通常歸因於酪蛋白致力於生物活性組份的保護效果。
在一些具體實例中,酪蛋白及乳清蛋白來源二者被用於營養產品中。
在本發明之前,一般熟習所屬技藝者可能不選擇具有本文主張之WPN水平之原料。一般熟習所屬技藝者咸信液體營養產品的熱加工可能破壞成份的任何TGF-β活性。據此,沒有選擇低溫或中溫加熱原料的動機。另外,在本發明之前,以選擇高溫加熱原料實際上較佳,由於經由高溫加熱製造技術的蛋白質變性提供比以未變性蛋白質更少起泡的最終產物之事實。本發明者驚訝地發現內源性TGF-β水平及活性可藉由選擇具有如本文所主張之WPN水平的成份且施予所主張之加工步驟而保留在最終產品中。
在本發明的具體實例中,蛋白質成份係以其製造加熱處理為基準予以選擇且與液體營養產品的所有其他組份組合,以形成漿液。在一些具體實例中,先將漿液進行加熱殺菌。加熱殺菌可在約70℃到約75℃之溫度進行約5到25秒。在其他的具體實例中,加熱殺菌可在約72℃之溫度進行約10-20秒。在又其他的具體實例中,加熱殺菌可在約72℃之溫度進行約15秒。
接著將此漿液均化。均化步驟可在介於約2500磅/平方英吋(psi)與3500psi之間的壓力下進行。在其他的具體實例中,壓力可為約3000psi。均化步驟的溫度範圍可從約55℃到約65℃。在具體實例中,均化步驟可發生約5到20秒。在特殊的具體實例中,均化步驟係在約3000psi之壓力及約55℃到約65℃之範圍內的溫度進行約5到20秒。
接著將均化乳液經UHT處理。在具體實例中,UHT處理係在約135℃到約150℃之範圍內的溫度進行。在其他的具體實例中,UHT處理係在約141℃到約145℃之範圍內的溫度進行。UHT處理可發生約1.5到15秒。在特殊的具體實例中,UHT處理係在約141℃到約145℃之範圍內的溫度進行約5秒。
此方法接著包含快速冷卻步驟。在此步驟中,可將產品冷卻到低於約8℃之溫度。冷卻步驟可發生在少於30分鐘之內。
在產物冷卻後,可將產物以無菌填充至所欲格式中,成為其中具有活性水平之TGF-β的貨架期穩定的非冷藏性乳製產品。
在此方法的替代具體實例中,可將漿液利用高壓加工技術在約400MPa到約600MPa之範圍內的壓力與約60℃到約80℃之範圍內的溫度組合下加工約1到3分鐘。若必要時,如上述所討論,接著可將此高壓加工之材料均化及甚至進一步經UHT處理。此高壓的替代具體實例允許營養產品保留其TGF-β生物活性。此高壓處理之材料亦可與上述方法中之一或全部組合,以獲得具有活性TGF-β及其他生物活性化合物的貨架期穩定產品。

在一些具體實例中,本發明的方法用於生產貨架期穩定的非冷藏性液體產品。然而,此方法亦可用於生產冷藏性產品。在具體實例中,此方法可經修改以補充在選擇之原料中的內源性TGF-β水平。

在一個具體實例中,將含有EPA之LCPUFA來源用於營養組成物中。在另一具體實例中,將實質上不含EPA之LCPUFA來源用於營養組成物中。例如,在本發明的一個具體實例中,營養組成物含有少於約16毫克EPA/100千卡。在另一具體實例中,營養組成物含有少於約10毫克EPA/100千卡。在又另一具體實例中,營養組成物含有少於約5毫克EPA/100千卡。本發明的另一具體實例包括不含甚至微量EPA之營養組成物。

Claims (8)
- 一種製備保留轉型生長因子-β(TGF-β)生物活性之液體營養產品的方法,其包含:a.選擇一或多種已接受包含中溫或較低溫加熱之熱負荷的蛋白質成分,其中該蛋白質成分包含乳清蛋白粉來源;b.將蛋白質成分與一或多種額外的液體營養產品組份組合,以形成漿液;c.使漿液在從約55℃到約65℃之溫度接受約5到約20秒從約2500磅每平方英吋(psi)到約3500psi之壓力;d.使漿液接受約1.5到約15秒從約135℃到約150℃之溫度;及e.將漿液在約30分鐘或更短之內冷卻到低於約8℃之溫度。
- 根據申請專利範圍第1項之方法,其中該熱負荷包含低溫加熱。
- 根據申請專利範圍第1項之方法,其中該乳清蛋白粉來源係選自甜乳清、去鹽乳清、乳清蛋白濃縮物、乳清蛋白分離物及其組合。
- 根據申請專利範圍第1項之方法,其中該乳清蛋白粉具有至少約6.0毫克/公克之未變性乳清蛋白氮水平。
- 根據申請專利範圍第1項之方法,其中該蛋白質成分包含酪蛋白來源及乳清蛋白來源。
- 根據申請專利範圍第1項之方法,其中該步驟(c) 係在約3000psi之壓力及在約55℃到約65℃之範圍內的溫度進行約5到20秒。
- 根據申請專利範圍第1項之方法,其中該步驟(d)係在約141℃到約145℃之範圍內的溫度進行約1.5到15秒。
