用於口臭(bad breath)或口腔異味(malodor)的專有名詞halitosis係一種被視為最常見及有時可能造成尷尬和影響生活品質的嚴重問題。患有口臭的人們經常仍是渾然不覺。口臭係來自能產生含硫化物(VSC)亦即硫化氫、甲基硫醇和二甲硫醚之口腔細菌的過度生長。治療方法為減少口腔內微生物數量、中和VSC化合物或遮蔽口臭。達成此目標最大的困難為不良傳遞和口腔內抑臭分子的滯留。通常活性劑由於易被洗脫及無法結合至牙齒或黏膜表面而因此大部分無法到達標的位置。該活性劑因而無法長期駐留並且大大地減低其發揮作用的能力。
用於控制口臭之組成物的美學訴求極為重要,並且明顯影響消費者的接受度和使用率。已證實美學效應在許多產品的消費者接受度上扮演重要角色。雖然此類產品已被消費者所認同,但是技術上仍尋求美學效應的進一步改善以及這些產品在化粧和治療上的效益。事實上,已知技術中許多此類組成物仍缺少上述一或多種的優點。
雖然一些市售產品已被消費者所認同,但是由於控制臭味之活性劑易被洗脫而無法長期駐留於口腔內,因此許多產品仍無有效地控制口臭。因此,仍亟需一種能使控制臭味之活性劑長期駐留於口腔內以減少或消除口臭的產品。
本發明的一些具體實施例提供能延長、延遲或控制釋放控制臭味之活性劑的組成物。在一些具體實施例中,控制臭味之活性劑能結合至牙齒或口腔黏膜。在一些具體實施例中,控制臭味之活性劑能捕捉揮發性硫化物及其他造成口臭的產物。
本發明的一些具體實施例提供一種口腔保健組成物,其包含一口腔可接受載劑;及一薄膜包含:一控制臭味之活性劑;一黏膜附著聚合物;一調節釋放聚合物;一聚合基質;以及一任選調味劑。在一些具體實施例中,該控制臭味之活性劑以能使控制臭味之活性劑長期駐留於口腔內的方式被釋出。
在一些具體實施例中,該薄膜包含複數個薄膜碎片。在其他具體實施例中,本發明提供一種包含複數個載體內板層狀碎片的組成物。
在一些具體實施例中,該口腔保健組成物包括一夾帶入口腔可接受載劑(vehicle)內的多層薄膜,該多層薄膜包含至少一黏膜附著層和一重疊層,該黏膜附著層包含一黏膜附著聚合物、第一聚合基質和一控制臭味之活性劑以及該重疊層包含一調節釋放聚合物、第二聚合基質和一任選的調味劑。
本發明之一些具體實例係提供一種用於治療或預防口臭之方法,其包含:投與需其之個體一種口腔保健組成物,其包含用於延長或延遲釋放控制臭味之活性劑之一薄膜,其中該薄膜包含一黏膜附著聚合物、一調節釋放聚合物、一聚合基質、及控制臭味之活性劑。
在另一具體實施例中,本發明提供製備一種夾帶入控制釋放載劑內之黏膜附著薄膜之口腔保健組成物的方法,其中該薄膜具有控制臭味之活性劑,其包括:
a. 提供一口腔可接受載劑;
b. 加入含有至少一黏膜附著層和一重疊層的多層薄膜,該黏膜附著層包含一黏膜附著聚合物、第一聚合基質和一控制臭味之活性劑,以及該重疊層包含一調節釋放聚合物、第二聚合基質和含量從約35至約60%重量比之載劑的調味劑以形成一混合物;以及
c. 均質化該混合物。
此處所述的"口腔保健組成物"意指具有口腔保健、口腔衛生或口腔美觀用途,或其使用方法包括投與至口腔的組成物。
此處"調節釋放聚合物"指適用於能延遲控制臭味之活性劑釋出之口腔保健配製物內,或來自此處所述薄膜及/或組成物之其他活性劑的聚合物。適當調節釋放聚合物的實例可見於雷明登製藥科學第18版,Gennaro編輯,賓州,Easton市Mack出版公司,1990年,藉由引述將其完整併入於此。
本發明的一些具體實施例提供一種口腔保健組成物,其包含:一口腔可接受載劑;及一薄膜,其包含:一控制臭味之活性劑;一黏膜附著聚合物;一調節釋放聚合物;一聚合基質;以及任選的一調味劑,其中該薄膜可控制釋放該控制臭味之活性劑。在一些具體實施例中,該控制臭味之活性劑係選自由下列組成的群組:一除臭劑;一吸臭劑;以及一堵臭劑。在一些具體實施例中,該除臭劑係一種選自由下列組成的群組之含鋅化合物:氧化鋅、檸檬酸鋅、乳酸鋅、亞氯酸鋅、氯化鋅、鋅沸石,及其二或多種的組合。
在一些具體實施例中,該黏膜附著聚合物係選自下列組成的群組:聚丙烯酸或聚丙烯酸酯、交聯聚丙烯酸或聚丙烯酸酯、聚丙烯酸烷基酯、交聯聚丙烯酸烷基酯、聚乙烯吡咯啶酮、聚乙烯吡咯啶酮和聚丙烯酸酯的共聚物、幾丁聚糖或化學改性幾丁聚糖,及/或其二或多種的組合。
在一些具體實施例中,該調節釋放聚合物係聚醋酸乙烯酯或羥乙基纖維素。在一些具體實施例中,該調節釋放聚合物係聚醋酸乙烯酯。
在一些具體實施例中,該聚合基質係選自由下列組成的群組:羥丙基甲基纖維素、甲基纖維素、羥丙基纖維素,及其共聚物。

本發明的進一步具體實施例提供一種用於治療或預防口臭的方法,其包含:將包含能延長或延釋控制臭味之活性劑之薄膜的口腔保健組成物投與至需其的個體,其中該薄膜包含:一黏膜附著聚合物、一調節釋放聚合物、一聚合基質,及一控制臭味之活性劑。在一些具體實施例中,該薄膜係一種包含至少一黏膜附著層和一重疊層的多層薄膜,該黏膜附著層包含:一黏膜附著聚合物、第一聚合基質和一控制臭味之活性劑;及該重疊層包含:一調節釋放聚合物、第二聚合基質,以及任選的一調味劑。

各種具體實施例的口腔保健組成物較佳為被製成潔齒劑的形態。全文中所述的"潔齒劑"一詞意指牙膏、凝膠、舌錠、牙膠,或液體配製物。該潔齒劑可呈任何所欲的形式,例如深條紋、淺條紋、多層、具有凝膠包覆牙膏,或其任何組合。口腔保健組成物內所含的薄膜可為任何所欲的形狀或構造,包括多重小條紋或一連續條紋。
全文說明中所述"載劑"或"水性載劑"的陳述係指用於此處的任何安全及有效材料。此類材料包括,例如增稠劑、濕潤劑、離子活性成分、緩衝劑、抗牙石劑、拋光材料、過氧化物源、鹼金屬重碳酸鹽、表面活性劑、二氧化鈦、著色劑、調味系統、甜味劑、抗微生物劑、草藥劑、減敏劑、去污劑,及其混合物。
除非另有明述,否則全部用於此處的百分比和比例為該口腔組成物的重量比。除非另有明述,否則全部均在25℃之下進行測量。此處所引述的一特定值,不論指成分的各別含量或該具體實施例的其他特徵,擬用於表示計入測量誤差之加減一變異度的值。例如,10%的量為計入熟習本領域之技術者所能判斷和瞭解的已知誤差,包括9.5%或10.5%。
此處"薄膜"指具有實質上層狀結構的材料。一"層狀"結構具有或能具有實質上較大於第三維(如z-方向)構造之厚度的一或二維(如x-或y-維)尺寸。用於此處的層狀結構其中一些包括實質上為平面、層狀或薄片形者。在一具體實施例中,該層狀結構為具有實質上大於z-方向之x-和y-維尺寸的實質平面。在其他具體實施例中,該層狀結構係非平面。在一具體實施例中,本發明的一薄膜包含一實質上連續之呈現實質上完整平面的表面,但是在一些具體實施例中該薄膜可能變形。在此類具體實施例中,該薄膜可為任何的許多形狀,包括具有一平滑變曲表面。該碎片可為一所欲尺寸,以及可具有規則或不規則周界。
本發明的一些具體實施例包含用於延遲或延長控制臭味之活性劑釋出的一薄膜。結合至口腔內牙齒及/或黏膜表面的薄膜可延長殺菌劑於細菌細胞的駐留時間,因而可殺死產生揮發性硫化物的細菌。此外,薄膜內的多層結構將可產生較大的表面積而可傳遞高濃度的控制臭味之活性劑至標的位置。
此處"即釋"或"即釋組成物"一詞指可快速而非緩慢地從組成物釋出活性劑的組成物。此類即釋組成物缺乏調節釋放聚合物或能延緩從該組成物釋出活性劑的其他物質,或含有相對無此類聚合物或物質之相同配製物不延緩組成物釋出活性劑的極少量此類聚合物或物質。
此處"控制釋放"或"控制釋放口腔保健組成物"等名詞指含有相對上述的"即釋"組成物能延緩從組成物釋出活性劑之物質的口腔保健組成物。因此,"控制釋放"一詞適用於任何長效性劑型,以及其實質上與延遲釋放、時間釋放、延長釋放、緩釋、時程釋放、順時釋放、時塗釋放、持續釋放、緩效、長效、延遲作用、間隔釋放、時隔釋放、延長作用、延緩釋放、延緩作用等同義。
此處"控制臭味之活性劑"或"活性劑"指較佳為經由牙膏或漂洗配製物傳遞至口腔以及其被用於三種主要防口臭領域亦即除臭、吸臭和阻臭的全部功能性化合物。
適用於除臭的控制臭味之活性劑包括,但不侷限於含鋅如氧化鋅、檸檬酸鋅和鋅沸石之述於美國專利案2006/0045860中的化合物。此外,此處所述的組成物亦可加入其他含金屬例如銅、亞錫、鉍、鍶的化合物;多汁物或其他可增加唾液流動以洗除臭味的成分。所宣稱的組成物亦可使用某些濃橙基香料;可捕捉或吸附臭氣分子的吸臭複合物,例如Bell Air香料公司所製造的。具有包覆臭氣分子例如硫醇、硫化物及其構造內胺的能力,其已述於例如美國專利案6,664,254。
適用於阻臭或作為堵臭劑的控制臭味之活性劑包括,但不侷限於能干擾產生臭味之過程的酵素。例如,黏膜附著薄膜內可有效地配製入堵臭酵素如精胺酸脫亞胺酶。同樣,可有效抑制臭氣分子產生菌的分子亦可用於控制口臭,例如可干擾胱胺酸脫硫基酶及/或甲硫胺酸γ-裂解酶之細菌酵素的物質。
適用於阻臭或作為堵臭劑的控制臭味之活性劑包括,但不侷限於可氧化或與臭氣分子反應的物質,包括過氧化物、過氯酸鹽,以及與活化雙鍵反應的分子。
此處"黏膜附著劑"指具有水溶或可澎脹性及能黏附至黏膜表面的天然或合成親水性聚合物。此類黏附劑可使含控制臭味之活性劑配製物黏附至黏膜組織及/或牙齒以及可經由接觸黏膜被溶解或吸收之作為其和其他活性劑的容器。
用於本發明的黏膜附著聚合物包括,但不侷限於供應自市售商品CP94ONF、CP971NF、CP974PNF的羧酸聚甲烯;市售商品Plasdone K-90 USP的聚乙烯吡咯啶酮(PVP);以及的市售商品Noveon AA-1 USP(PC);羥丙基纖維素;羧甲基纖維素鈉;羥丙基甲基纖維素;乙基纖維素;聚乙烯醇;聚(異丁烯)、聚(異戊二烯);聚(丙烯酸);天然樹膠,例如三仙膠和刺槐豆膠;幾丁聚糖及幾丁聚糖、果膠、矽膠的化學衍生物,以及其混合物和共聚物。
940聚合物係一種已知作為生物黏著劑的交聯聚丙烯聚合物,以及亦作為一種能提供組成物高黏度的高效流變改性劑。為丙烯酸交聯二乙烯醇的聚合物,可作為生物黏著劑及亦用於設計控制釋放配製物。
較佳地,該黏膜附著聚合物包含從約0.5至約20%重量比的薄膜,較佳為從約1至約10%重量比,以及更佳為從約2至約4%重量比。
在一些具體實施例中,該薄膜包含一聚合基質。在一些具體實施例中,該聚合基質係選自一水溶性聚合物例如羥丙基甲基纖維素、甲基纖維素、羥丙基纖維素,及其共聚物的混合物。在其他具體實施例中,可使用薄膜形成聚合物例如聚乙烯醇、聚乙烯吡咯啶酮等。
因此本發明提供口腔保健配製物,其包括口腔可接受和黏膜附著之水可崩解單或雙層薄膜,其以延緩或延長方式釋放控制臭味之活性劑,且藉由唾液恒流從口腔被移除。此處"水可崩解"意指不溶解於水或唾液,然而暴露於水或唾液之下可被完全崩解的材料或物質。
將更詳細描述單層和多層薄膜,藉由這些薄膜溶解或分散於口腔液體環境內的速率以及控制臭味之活性劑從薄膜的擴散速率測定此類薄膜釋出控制臭味之活性劑的速率。藉由調節薄膜基質內調節釋放聚合物及/或聚合基質的釋出量可輕易地裁製範圍從所欲分鐘或小時之控制臭味之活性劑釋出自薄膜基質的釋放速率。例如,增加聚醋酸乙烯酯的數量可減少薄膜的膨脹但增加薄膜於口腔黏膜和牙齒的駐留時間因而延遲控制臭味之活性劑從薄膜的釋出。
在一較佳具體實施例中,本發明薄膜內調節釋放聚合物的含量為從0.1至30%,更佳為從0.5至20%,以及最佳為從5至20%。此外,聚合基質的含量較佳為從5至80%,更佳為從10至60%,以及最佳為從40至50%。
調節存在於本發明薄膜內的控制臭味之活性劑含量亦可被用於控制、延長或延遲該控制臭味之活性劑的釋出。理想上,儘可能加入高量的控制臭味之活性劑。然而,實務上必需在最終薄膜的製造和處理之間取得平衡。因此,控制臭味之活性劑的含量較佳為從10至70%,較佳為從10至60%,以及最佳為從40至60%。在一些具體實施例中,該控制臭味之活性劑的含量為從約15至約25%重量比的薄膜。
利用技術中習知的方法例如傳統擠壓法、含水和溶劑鑄造法可製備本發明的延釋或遲釋薄膜。
在一些具體實施例中,提供一種包含黏膜附著聚合物、調節釋放聚合物、聚合基質和控制臭味之活性劑的單層薄膜。於製備本發明的單層薄膜中,將黏膜附著聚合物、調節釋放聚合物、聚合基質、控制臭味之活性劑和任選的著色劑、調味劑、甜味劑及/或治療劑和其他薄膜形成成分溶解於相容溶劑內以形成一薄膜形成組成物。
適當溶劑的實例包括水、醇、丙酮、醋酸乙酯,或其混合物。溶液形成之後,可在攪拌下加入增塑劑以及需要時加熱以助溶解直至形成透明而均質的溶液為止。該溶液可被塗佈於一適當載劑材料上然後乾燥形成薄膜。該載劑材料必需具有容許薄膜溶液均勻散佈於載劑上而不浸漬形成薄膜載劑基質間破壞鍵的表面張力。適當載劑材料的實例包括玻璃、不銹鋼、鐵氟龍和聚乙烯浸漬紙。可利用不破壞構成薄膜之成分的乾燥箱、乾燥端子、真空乾燥器,或任何其他適當乾燥設備於高溫下進行薄膜的乾燥。
在一些具體實施例中,該薄膜的厚度為從0.5至10 mils(1 mil=0.001吋),較佳為從1至5 mils,更佳為從1.5至2.5 mils的單層薄膜厚度,以及從2至4 mils的雙層或三層薄膜厚度。在一些具體實施例中,該乾燥薄膜被切割或沖壓成0.01至0.50吋,較佳為0.08至0.25吋粒徑的薄片狀。
形成薄片之前可將保護性阻隔包衣例如食品級蟲膠或乙基纖維素塗佈至該薄膜以強化該形化自乾燥薄膜的額外安定性。
該薄膜若需裝飾效應時,可將該薄膜切割或沖壓成例如心形、星形、稜形和圓形的各種醒目薄片狀。在一些具體實施例中,併入本發明潔齒劑基質內之薄片的濃度為0.05至1.0%重量比,較佳為0.1至0.5%重量比。在一些具體實施例中,其含有較低濃度的第一裝飾用薄膜,以及被製成透明或不醒目之較高濃度的另一薄膜。
在一些具體實施例中,提供一種雙層薄膜。藉由各別黏漿的鑄製及分別乾燥各層以形成連續複合層(A和B)的方法形成該雙層薄膜。例如,將含有黏膜附著聚合物、第一聚合基質和控制臭味之活性劑的黏漿組成物鑄製成0.010吋包膜,然後在100℃烘箱內乾燥10分鐘而形成第一層(A)。接著以含有調節釋放聚合物、第二聚合基質和一調味劑的黏漿組成物在第一層上鑄製0.020吋包膜,然後在100℃烘箱內乾燥10分鐘而形成第二層(B)。含有調節釋放聚合物及/或其他調節釋放化合物的層被認為有助於含黏膜附著劑層之控制臭味之活性劑的延釋或遲釋。因此消費者在使用併入本發明薄膜的潔齒劑或潄口液時,其口腔保健配製物可將控制臭味之活性劑傳遞至標的位置及/或不易到達的口腔內區域。在一些具體實施例中,藉由薄膜的含層可使其選擇性地結合至沾有唾液的表面上。在一些具體實施例中,該含有聚醋酸乙烯層的雙層薄膜並不黏附至口腔黏膜或牙齒表面但可延釋及/或遲釋該控制臭味之活性劑。因此,該雙層薄膜易黏附於口腔軟組織的內側並且在緩慢崩解含調節釋放劑層及快速崩解含黏膜附著聚合物層之下延釋及/或遲釋出該控制臭味之活性劑。
一些具體實施例提供一種三層薄膜。藉由各別黏漿的鑄製及分別乾燥各層以形成連續複合層(C、D和C’)的方法形成該三層薄膜。例如,將含有黏膜附著聚合物、第一聚合基質和控制臭味之活性劑的黏漿組成物鑄製成0.010吋包膜,然後在100℃烘箱內乾燥10分鐘而形成第一層(C)。接著以含有調節釋放聚合物、第二聚合基質和一調味劑的黏漿組成物在第一層上鑄製0.020吋包膜,然後在100℃烘箱內乾燥10分鐘而形成第二層(D)。最後,將含有黏膜附著聚合物、第一聚合基質和控制臭味之活性劑的黏漿組成物在第二層上鑄製0.010吋包膜,然後在100℃烘箱內乾燥10分鐘而形成第三層(C)。可連續地依上述方法,或直接從黏漿組分形成多層疊合黏漿的方法製造該薄膜。然後依照相同方法乾燥該疊合黏漿。亦可使用各種方法的組合。例如,於層壓之後藉由溶劑進行不同層的鑄製。
在一些具體實施例中,本發明提供一種三層薄膜其外表層為水可崩解及含有黏膜附著聚合物、第一聚合基質和一調節釋放聚合物,同時中心層併入調味劑、控制臭味之活性劑和第二聚合基質。

在某些具體實施例中,該薄膜及/或口腔保健組成物含有防腐劑。該防腐劑的含量為從約0.001至約5 w/w%,較佳為從約0.01至約1 w/w%的薄膜。防腐劑的非限制性實例包括苯甲酸鈉和山梨酸鉀。
本發明的一些具體實施例提供一種包含夾帶一薄膜或一或多個薄膜碎片之載劑的組成物。此處所稱的"載劑"係可夾帶薄膜並且適合投與或塗佈至人或動物體的任何物質或組成物。

該載劑可包含任何各種的材料,包括乳化劑、增稠劑、充填劑和防腐劑。在一些具體實施例中,該載劑可含有如上所述的一功能性或活性劑。在一些具體實施例中,該載劑包含作為薄膜的相同功能性材料。

在某些潔齒劑中,該載劑包含一表面活性劑或表面活性劑的混合物。用於此處的表面活性劑其中一些包括至少一種高級脂肪酸單硫酸甘油單酯的水溶性鹽,例如氫化椰油脂肪酸之單硫酸甘油單酯的鈉鹽;椰油醯胺丙基甜菜鹼;高級烷基硫酸鹽例如月桂基硫酸鈉;烷基芳基磺酸鹽例如十二烷基苯磺酸鈉;高級烷基磺基乙酸鹽;月桂基磺基乙酸鈉;1,2-二羥基丙磺酸鹽的高級脂肪酸;以及低級脂族胺基羧酸的實質上飽和高級脂肪醯胺,例如脂肪酸、烷基或醯基內具有12至16個碳原子者;及其混合物。醯胺可為例如N-月桂醯基肌胺酸,以及N-月桂醯基、N-肉豆蔻醯基或N-棕櫚醯基肌胺酸的鈉、鉀和乙醇胺鹽。在各種具體實施例中,該載劑的表面活性劑含量為從約0.3至約5%,或0.5至3%重量比的組成物。
本發明亦提供製造潔齒劑載劑的方法。在一具體實施例中,將水和至少一種濕潤劑分散於習知混合機內直至形成第一均質凝膠相為止。然後將拋光劑加入該第一均質凝膠相內。混合該第一均質凝膠相和拋光劑直至形成第二均質凝膠相為止。在20至100毫米汞的真空高速下混合這些成分。
本發明組成物較佳為可穩定地被儲存於正常條件下。此處所稱"穩定"意指對構成例如外觀、風味、流變性和化學組成的任一或較佳為全部組成物無明顯不良效應。本發明的穩定性較佳為包括組成物內薄膜(包括碎片,若有時)的組合及物理穩定性。在各種具體實施例中,包含一薄膜的組成物可於室溫下被穩定地儲存至少約兩年。然而,已瞭解在一些具體實施例中,該穩定薄膜可崩解於使用過程(如上所述)中例如使用潔齒劑組成物刷牙時。
在某些具體實施例中,一組成物除了上述的薄膜碎片之外可包含二或多種分別構成組成物一相的載劑。此類組成物不易滲色。例如,一組成物可包括薄膜碎片和一條紋潔齒劑如揭示於Wong等人的美國專利案號6,315,986中者。在某些具體實施例中,該薄膜碎片可具有與條紋不同的顏色以增加美觀。
習知的潔齒劑組成物含有增稠劑以提供潔齒劑所需的流變性而使潔齒劑可被長期儲存於容器內並且易於被消費者所使用。潔齒劑不僅因為易於分散而需具有適當黏度而且在刷牙時可使其更呈滑順感。典型的增稠劑包括改性纖維素,例如羧甲基纖維素(CMC)及其他多糖或膠質成分。
該多糖增稠劑包含至少一種三仙膠和羥乙基纖維素。該多糖增稠劑一般為由至少一種三仙膠和羥乙基纖維素所組成。該多糖增稠劑的三仙膠含量較佳為從0.1至1.5wt%,更佳為從0.5至1 wt%的組成物。然而,可存在少量的其他增稠劑例如卡拉膠、黃蓍樹膠、澱粉、聚乙烯吡咯啶酮、羥乙基丙基纖維素、羥丁基甲基纖維素、羥丙基甲基纖維素、羥乙基纖維素、羧甲基纖維素鈉(CMC鈉)和矽膠。在一具體實施例中,增稠劑的濃度根據組成物的重量比可為0.1至5 wt%。在另一具體實施例中,該增稠劑的濃度根據組成物的重量比可為0.5至2 wt%。
本發明的一些具體實施例亦提供製備包含載劑內夾帶薄膜之組成物的方法。在各種具體實施例中,將含複數個碎片的薄膜基質與一載劑相混合。在一些具體實施例中,可混合一載劑與複數個薄膜碎片。在一些具體實施例中,該混合包括緩慢地攪拌。在其他具體實施例中,該製備含有複數個本發明薄膜層合碎片分散於其中之載劑的組成物包含:提供該口腔可接受載劑;加入單或雙層薄膜的層合碎片以緩釋或延釋一控制臭味之活性劑至口腔可接受載劑而形成一混合物;以及均質化該混合物。
此處所述"均質化"一詞係指碎片和載劑經混合之後可使碎片實質上均質地分佈於載劑內之意。然而,應注意所形成的組成物仍保持兩相的組成物性質。均質化時可使用任何各種習知的均質機。
在一些具體實施例中,該薄膜被加至一口腔可接受載劑的成分(例如用於潔齒劑的濕潤劑)。接著製造其餘的載劑,然後將薄膜混合物加至該載劑。
此處所述的某些具體實施例亦提供投與一控制臭味之活性劑至人或動物體的方法。此處所稱"投與"意指將一組成物塗抹於或投與至該個體的任何方法。在各種具體實施例中,該投與可為局部性,其中該組成物被塗抹至個體的外表面,例如口腔內(如牙齒、齒齦和舌頭)的表面。當投與的特定途徑和方法當然視該組成物的用途而定。
在一些具體實施例中,本發明提供投與一控制臭味之活性劑至需其之人或動物個體的方法,包括將含夾帶控制臭味之活性劑於載劑內之控制釋放薄膜基質的組成物局部塗佈至個體。在一些具體實施例中,該方法進一步包括於局部塗抹組成物之後破壞該薄膜基質。在一些具體實施例中,該破壞可藉由任何方法包括化學及/或機械方法被完成。化學方法包括於投與部位接觸水或物質(例如口腔內的唾液)而破壞該薄膜。物理方法包括使用時(例如使用潔齒劑刷牙)將物理能如攪動、磨擦和剪力施予至該組成物。
在各種具體實施例中,本發明提供用於治療口腔疾病或病症的方法。此處所述"口腔保健病症"指可藉由投與一組成物至口腔被預防或治療的疾病或病症,包括牙齒、黏膜、齒齦和舌頭的疾病或病症。此類病症包括齲齒、齒齦炎、牙周病,及美觀上病症例如黃牙病和口臭。
將藉由特定實例更詳細說明本發明。提供下列實例作為說明之目的而在任何情況下非擬限制本發明的範圍。熟習本技術者將輕易地瞭解改變或改良各種非決定性的參數基本上將可產生相同的結果。
此實例說明含有如下表1所示CP971作為黏性聚合物、聚醋酸乙烯作為調節釋放聚合物、羥丙基甲基纖維素(HPMC)作為聚合基質及氧化鋅作為控制臭味之活性劑之組合的單層薄膜。
此實例說明含有如下表2所示CP971作為黏性聚合物、聚醋酸乙烯作為調節釋放聚合物、HPMC作為聚合基質及作為控制臭味之活性劑之組合的單層薄膜。
其他具體實施例包含其底薄膜層含黏性聚合物、聚合基質和控制臭味之活性劑或活性劑與調味劑之組合的雙層薄膜系統。其頂層含有作為調節釋放劑的聚醋酸乙烯以及聚合基質及/或調味劑之組合。下列實例3和4為雙層聚合物薄膜的具體實施例。
此實例說明含有CP971作為黏性聚合物、聚醋酸乙烯作為調節釋放劑、HPMC作為聚合基質以及氧化鋅及/或鋅沸石作為如述於美國專利申請案2006/0045860中之制菌劑或殺菌劑的雙層薄膜。此雙層配製物包括以如下表3A所示CP971作為黏性聚合物、HPMC作為聚合基質以及制菌劑如氧化鋅及/或鋅沸石的一黏性底層。
該雙層配製物亦包括具有如下表3B所示聚醋酸乙烯作為調節釋放聚合物、HPMC作為聚合基質以及調味劑的頂層。
此實例說明含有CP971作為黏性聚合物、聚醋酸乙烯作為調節釋放劑、HPMC作為聚合基質以及作為控制臭味之活性劑之組合的雙層薄膜。此雙層配製物包括以如下表4A所示CP971作為黏性聚合物、HPMC作為聚合基質以及作為控制臭味之活性劑的一黏性底層。
此雙層配製物亦包括具有如下表4B所示聚醋酸乙烯作為調節釋放聚合物、HPMC作為聚合基質以及調味劑的緩溶頂層。
此實例說明一種含有被夾於兩層唾液可溶解表面層間之中心層的三層薄膜。下表5說明用於製備本發明三層薄膜的黏漿組成物。
表面層1被鑄成5 mils,然後在95℃烘箱內乾燥15分鐘。中心層2於表面層1上被鑄成15 mils,然後以相同方法乾燥。表面層3被鑄成5 mils,以及該最終組成物於95℃再乾燥15分鐘。該乾燥薄膜的厚度為約2.75 mils,其具有1.65 mils的中心層及兩層各為0.55 mils的表面層。在實驗室內藉由IKA-WERKE(MF 10型)將該薄膜研磨成小片。其被製成用於製造產品之介於12和20大小的碎片。

Claims (13)
- 一種口腔保健組成物,其包含:一口腔可接受載劑;及一薄膜,其中該薄膜包含:一控制臭味之活性劑,其選自由下列組成的群組:氧化鋅、檸檬酸鋅、乳酸鋅、亞氯酸鋅、氯化鋅、鋅沸石,及其二或多種的組合;一黏膜附著聚合物,其選自下列組成的群組:聚丙烯酸或聚丙烯酸酯、交聯聚丙烯酸或聚丙烯酸酯、聚丙烯酸烷基酯、交聯聚丙烯酸烷基酯、聚乙烯吡咯啶酮、聚乙烯吡咯啶酮和聚丙烯酸酯的共聚物、幾丁聚糖或化學改性幾丁聚糖,及其二或多種的組合;一或多種調節釋放聚合物,其選自下列組成的群組:聚醋酸乙烯酯及羥乙基纖維素;一聚合基質,其選自由下列組成的群組:羥丙基甲基纖維素、甲基纖維素、羥丙基纖維素,及其共聚物;以及一調味劑;其中該薄膜被夾帶入該口腔可接受載劑內。
- 如申請專利範圍第1項之口腔保健組成物,其中該黏膜附著聚合物含薄膜重量之2至5%。
- 如申請專利範圍第1項之口腔保健組成物,其中該調節釋放聚合物含薄膜重量之20至30%。

- 如申請專利範圍第1項之口腔保健組成物,其中該控制臭味之活性劑含薄膜重量之30至40%。
- 如申請專利範圍第1項之口腔保健口腔組成物,其中該薄膜含口腔可接受載劑重量之0.1至5%。
- 一種口腔保健組成物,其包含:一夾帶入口腔可接受載劑內的多層薄膜,其中該多層薄膜包含至少一種黏膜附著層和一重疊層,以及其中該黏膜附著層包含:一黏膜附著聚合物,其選自下列組成的群組:聚丙烯酸或聚丙烯酸酯、交聯聚丙烯酸或聚丙烯酸酯、聚丙烯酸烷基酯、交聯聚丙烯酸烷基酯、聚乙烯吡咯啶酮、聚乙烯吡咯啶酮和聚丙烯酸酯的共聚物、幾丁聚糖或化學改性幾丁聚糖,及其二或多種的組合;第一聚合基質,其選自由下列組成的群組:羥丙基甲基纖維素、甲基纖維素、羥丙基纖維素,及其共聚物;和一控制臭味之活性劑,其選自由下列組成的群組:氧化鋅、檸檬酸鋅、乳酸鋅、亞氯酸鋅、氯化鋅、鋅沸石,及其二或多種的組合;以及其中該重疊層包含:一調節釋放聚合物,其選自由下列組成的群組:聚醋酸乙烯酯及羥乙基纖維素;第二聚合基質,其選自由下列組成的群組:羥丙基甲基纖維素、甲基纖維素、羥丙基纖維素,及其共聚物;和一任選調味劑。
