Abstract

Classical mutagenesis could desensitize the feedback inhibition of L-tryptopHan biosynthesis. Among the mutants, Escherichia coli EMS4 produced 3 g/1 of L-tryptopHan within 18 hours. The feedback-resistant L-Trp operon gene prepared from E. coli EMS4 was inserted into pUC19 and pHSG576 to generate pTC701 and pTC576. When these plasmids were introduced into E. coli EMS4, chromosomal integration occurred through homologous recombination. By using Southern hybridization, we demonstrated that the integrated plasmids existed as multicopies. The strains with integrated foreign trp operon gene could produce 5.5 g/1/18h of L-tryptopHan, and this strain was named as E. coli EMS4 C25. This strain was very stable in fermentation process, even in the absance of antibiotics it could produce a high yield of L-tryptopHan.
