' De immuniserande B-lymfocyterna utvinnes företrädesvis sid. ur den lymfatiska vävnaden hos möss.
Nedan beskrives såsom ett utföringsexempel ett sätt för framställning av anti-tymocytglobulin under utnyttjande av T-lymfoblaster från cellinjen JM. vilket sätt omfattar följande Steg: l. Centrifugering av T-lymfoblastercellerna. 2. Inställning av cellsuspensionen med ett fysiologiskt cell- medium (CL 15) till en cellkoncentration av 30 x 106 celler _ per ml. 3. Immunisering av kaniner genom intravenös tillförsel av 3 ml cellsuspension i örvenen. 4. Efter fyra veckors immunseringstid avtappas blod från djuren.
. Genom centrifugering av blodet utvinnes serumet. 6. Efter bestämning av antikropptitern och sterilitetsundersök- ning präglas serumet. 7. Absorption av serumet med olika humanceller. 8. Upprening av antiserumet för globulinfraktionen medelst ammoniumsulfatfäïlnind och efterföljande kolonnkrømatografi. 9. Immunglobulinfraktionen inställes till en bestämd äggvitom halt (20 mg/nl), 460 826 3 . Anti~tymocytglobulinet ultracenfirifugeras och steril- filtreras. ll. Tappning av immunglobulinet i ampuller øch genomförande av slutkontroller (kvalitets-, sterilitets- och pyrogen- -testning).
