Claims (22)
1. Автоматизированный автономный многомодульный инструмент для редактирования клеток для осуществления направляемого нуклеазой редактирования в клетках млекопитающих, включающий:
корпус, выполненный с возможностью размещения всех или некоторых модулей; емкость, выполненную с возможностью приема клеток млекопитающих; одну или более емкостей, выполненных с возможностью приема векторов экспрессии, где векторы экспрессии остов вирусного вектора и редактирующую кассету; модуль для выращивания клеток для трансфекции; модуль для трансфекции, выполненный с возможностью введения векторов в клетки млекопитающих; модуль для направляемого нуклеазой редактирования, выполненный с возможностью для введенных векторов экспрессии редактировать нуклеиновые кислоты в клетках млекопитающих; процессор, выполненный с возможностью обеспечения работы автоматизированного многомодульного инструмента для редактирования клеток, основываясь на вводе данных пользователем и/или выборе пользователем предварительно подготовленного скрипта; и автоматизированную систему подачи жидкости для перемещения жидкостей из емкости с клетками в модуль для выращивания, из модуля для выращивания в модуль для трансформации и из модуля для трансформации в модуль для направляемого нуклеазой редактирования без участия пользователя.
2. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 1, где автоматизированная система подачи жидкости содержит сиппер или дозатор.
3. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 1, где модуль для направляемого нуклеазой редактирования является также модулем восстановления.
4. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 1, дополнительно включающий модуль восстановления, отдельный от модуля редактирования.
5. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 1, где модуль для выращивания отделен от модуля для трансфекции.
6. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 1, где модуль для выращивания и модуль для трансфекции являются одним и тем же модулем.
7. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 1, дополнительно содержащий модуль для отбора.
8. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 7, где модуль для отбора является тем же модулем, что и модуль для направляемого нуклеазой редактирования.
9. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 1, где модуль для трансфекции обеспечивает условия для вирусной трансдукции.
10. Автоматизированный автономный многомодульный инструмент для редактирования клеток для осуществления направляемого нуклеазой редактирования в клетках млекопитающих, включающий:
корпус, выполненный с возможностью размещения всех или некоторых модулей; емкость, выполненную с возможностью приема клеток млекопитающих; одну или более емкостей, выполненных с возможностью приема векторов экспрессии, где векторы экспрессии содержат остов вирусного вектора и редактирующую кассету; модуль для выращивания клеток для трансфекции; модуль для трансфекции, выполненный с возможностью введения векторов в клетки млекопитающих; модуль для направляемого нуклеазой редактирования, выполненный с возможностью для введенных векторов экспрессии редактировать нуклеиновые кислоты в клетках млекопитающих; процессор, выполненный с возможностью обеспечения работы автоматизированного многомодульного инструмента для редактирования клеток, основываясь на вводе данных пользователем и/или выборе пользователем предварительно подготовленного скрипта; и автоматизированную систему подачи жидкости для перемещения жидкостей из емкости с клетками в модуль для выращивания, из модуля для выращивания в модуль для трансформации и из модуля для трансформации в модуль для направляемого нуклеазой редактирования без участия пользователя.
11. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 10, где автоматизированная система подачи жидкости содержит сиппер или дозатор.
12. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 10, где модуль для направляемого нуклеазой редактирования является также модулем восстановления.
13. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 10, дополнительно включающий модуль восстановления, отдельный от модуля редактирования.
14. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 10, где модуль для выращивания отделен от модуля для трансфекции.
15. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 10, где модуль для выращивания и модуль для трансфекции являются одним и тем же модулем.
16. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 10, дополнительно содержащий модуль для отбора.
17. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 16, где модуль для отбора является тем же модулем, что и модуль для направляемого нуклеазой редактирования.
18. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 10, где модуль для трансфекции обеспечивает условия для липофекции.
19. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 10, где модуль для трансфекции обеспечивает условия для магнитофекции.
20. Автоматизированный автономный многомодульный инструмент для редактирования клеток по п. 10, где модуль для трансфекции обеспечивает условия для микроинъекции.
