Abstract
Yeast cell lysis buffer for enhancing the recovery of subcellular particles comprises a buffered aq. soln. of a non-ionic detergent (I) at 0.5-1 vol.%. The buffer pref. comprises 150mM NaCl, 10% glycerol and 50mM tris buffer at pH8. Also EDTA may be present at 1-10mM. The buffer is pref. used to break yeast cells at 3-8 deg.C The cells may be mixed with the buffer and with a glass bead suspn. The prod. contg. the lysed cells may be centrifuged and the supernatant is removed from pelleted debris for further purificn. The (I) is esp. "Triton" X-100 (RTM), and in a buffer it typically causes a 2-fold greater release of antigen.
