Abstract
Kinuklidin-derivater med formel I og farmasøytisk godtagbare salter derav hvor R1 er hydrogen eller hydroksy; R2 er hydrogen; eller R1 og R2 er bundet sammen slik at CR^CR2 danner en dobbeltbinding; Ar1 er en fenylengruppe; Ar2 er en 6-leddet heteroaryl-gruppe inneholdende ett eller to nitrogenatomer; og hvor en eller begge Ar1 og Ar2 eventuelt kan ha en eller flere substituenter som uavhengig er valgt fra halogen, hydroksy, amino, nitro, cyano, karboksy, karbamoyl, alkyl, alkenyl, alkynyl, alkoksy, alkylamino, dialkylamino, N-alkyl- karbamoyl, di-N,N-alkylkarbamoyl, alkoksykarbonyl, alkyltio, alkylsulfinyl, alkylsulfonyl, halogenalkyl, alkanoylamino, alkylendioksy, alkanoyl og oksim-derivater derav og O-alkyl- etere av oksim-derivatene, hemmer sgualen syntase og er derfor nyttige for behandling av sykdommer og medisinske lidelser hvor hemning av sgualen syntase er ønskelig, f.eks. for be- handling av hypercholesterolemi og aterosklerose. Fremgangs- måter for fremstilling av disse derivatene og farmasøytiske preparater inneholdende dem er også angitt, så vel som deres anvendelse innen medisin.Kinuclidine derivatives of formula I and pharmaceutically acceptable salts thereof wherein R 1 is hydrogen or hydroxy; R 2 is hydrogen; or R 1 and R 2 are bonded together such that CR 2 CR 2 forms a double bond; Ar1 is a phenylene group; Ar2 is a 6-membered heteroaryl group containing one or two nitrogen atoms; and wherein one or both Ar 1 and Ar 2 may optionally have one or more substituents independently selected from halogen, hydroxy, amino, nitro, cyano, carboxy, carbamoyl, alkyl, alkenyl, alkynyl, alkoxy, alkylamino, dialkylamino, N-alkyl, carbamoyl, di-N, N-alkylcarbamoyl, alkoxycarbonyl, alkylthio, alkylsulfinyl, alkylsulfonyl, haloalkyl, alkanoylamino, alkylenedioxy, alkanoyl and oxime derivatives thereof and O-alkyl ethers of the oxime derivatives, inhibit the sulfonic synthase and are therefore useful for treatment of diseases and medical disorders where inhibition of sgual synthase is desirable, e.g. for the treatment of hypercholesterolemia and atherosclerosis. Methods for preparing these derivatives and pharmaceutical compositions containing them are also disclosed, as well as their use in medicine.
