Abstract

Det er beskrevet en fremgangsmåte for valgt en kandidatmedikament-forbindelse, idet fremgangsmåten omfatter: i) identifisering av et utgangsoligopeptid som har en bindingsaffinitet for en celleoverflate eller annet reseptorsete; ii) dannelse av en mengde homologe oligopeptider som har enkeltsete aminosyre-modifikasjoner relativt til nevnte utgangs oligopeptid; iii) identifisering av komponentene til nevnte utgangsoligopeptid ansvarlig forden sekundære strukturen derav; iv) for modifisering av seter identifisert i trinn (ii) selektering av et sett aminosyrer; v) for komponenter identifisert i trinn (iii) selektering av minst en ikke-naturlig aminosyre som etterligner den sekundære strukturen til komponenten; vi) dannelse av et første kombinatorisk bibliotek av oligopeptider som har det nevnte identifiserte modifikasjonssete aminosyreresidier som korresponderer med nevnte selekterte sett av aminosyrer; vii) identifisering av medlemmer av nevnte første bibliotek som har en bindingsaffinitetverdi over et forutbestemt minimum og dannelse av et ytterligere kombinatorisk bibliotek av oligopeptider som har medlemmer som ikke er tilstede i første bibliotek, og identifisering åv medlemmene til nevnte ytterligere bibliotek som har forbedrede bindingsaffinitetsverdier i forhold til de identifiserte medlemmene av nevnte første bibliotek og med den forutsetningen at minst av nevnte første og ytterligere bibliotek inneholder oligopeptider inneholdende residier av nevnte ikke-naturlige aminosyre eller syrer; og (viii) selektering fra de identifiserte medlemmene av nevnte bibliotek en medikamentkandidat.A method of selecting a candidate drug compound is described, the method comprising: i) identifying a parent oligopeptide having a binding affinity for a cell surface or other receptor site; ii) forming a plurality of homologous oligopeptides having single-site amino acid modifications relative to said parent oligopeptide; iii) identifying the components of said parent oligopeptide responsible for its secondary structure; iv) for modifying sites identified in step (ii) selecting a set of amino acids; v) for components identified in step (iii) selecting at least one non-natural amino acid that mimics the secondary structure of the component; vi) forming a first combinatorial library of oligopeptides having said identified site of modification amino acid residues corresponding to said selected set of amino acids; vii) identifying members of said first library having a binding affinity value above a predetermined minimum and forming an additional combinatorial library of oligopeptides having members not present in the first library, and identifying members of said additional library having enhanced binding affinity values in relative to the identified members of said first library and provided that at least one of said first and further libraries contains oligopeptides containing residues of said unnatural amino acid or acids; and (viii) selecting from the identified members of said library a drug candidate.
