Abstract

Un metodo mejorado para la identificacion simultanea especifica en secuencia de ARNm's en una poblacion de ARNm permite la visualizacion de casi cada ARNm expresado por un tejido como una banda distinta en un gel cuya intensidad corresponde aproximadamente a la concentracion del ARNm. En general, el metodo comprende la formacion de ADNc usando cebadores de anclaje para fijar un punto de extremo 3, produciendo insertos clonados a partir del ADNc en un vector que contiene un promotor especifico de bacteriofago para sintesis subsecuente de ARN, generando fragmentos linealizados de insertos clonados, preparando ARNc, transcribiendo ADNc a partir del ARNc usando un conjunto de cebadores 5-RT, y llevando a cabo PCR usando un cebador 3-PCR cuya secuencia es derivada del vector y un conjunto de cebadores 5-PCR que se deriva de los cebadores 5-RT usados para la transcripcion de ADNc a partir de ARNc. El metodo puede identificar cambios en la expresion de ARNm asociados con la administracion de farmacos o con condiciones fisiologicas o patologicas.
