Abstract

본 발명은 현미, 볶은현미, 보리, 보리분 또는 볶은 통보리중 적어도 1종 40~60wt%와 지하수 40~60wt%로 구성되어지되 종균배양용 병에 병입한 후 고압멸균기를 이용하여 15lb/㎤의 증기압(120~121℃)에서 50~60분간 가열하여 멸균하고 냉각한 배지에 자연산 상황의 자실체에서 조직을 분리하고 뽕나무 추출 배지에서 계대배양한 후 상기 배지와 같은 배지에서 종배양한 종배양액을 접종하여 25℃±1의 온도에서 25~30일간 배양함을 특징으로 하는 상황균사체 배양용 배지 조성물 및 이를 이용한 상황균사체의 배양방법에 관한 것으로, 쉽게 얻을 수 있는 곡물류를 배지성분으로 사용하므로서 상황균사체를 고밀도를 용이하게 얻을 수 있을 뿐만아니라 배양완료 후 배지성분과 더불어 상황균사체를 모두 이용할 수 있으므로 상황균사체의 추출정제가 필요없이 경제적이며 균사체 배양공정이 간단한 효과가 있다.

Claims (2)

  1. 현미, 볶은현미, 보리, 보리분 또는 볶은 통조리중 적어도 1종 40~60wt%와 지하수 40~60wt%로 구성되어지되 종균배양용 병에 병입한 후 고압멸균한 것을 특징으로 하는 상황균사체 배양용 배지 조성물.
  2. 자연산 상황의 자실체에서 조직을 분리한 다음, 뽕나무 고목을 재료로 하는 고형배지에서 정지배양의 방법으로 계대배양하고, 250㎖의 삼각플라스크에 현미50g과 50cc의 물을 주입후 고압살균(121℃/1.2㎏/50분)을 실시하고 냉각시킨 다음, 상기 계대배양액된 균사체를 무균상자 또는 무균실에서 접종하여 종배양한 종배양액을 제1항의 조성물로 조성된 배지에 접종하여 배양함을 특징으로 하는 상황균사체의 배양방법.
