Description

약제학적 수성제제내에서 거품을 억제하는 방법
본 발명은 약제학적 액상 제제내에서 거품을 억제하는 방법에 관한 것으로 구체적으로는 약제학적 액상제제의 거품 억제용으로서 에틸렌 옥사이드/프로필렌 옥사이드 블록 공중합체의 용도, 그를 함유하는 약제학적 제제 및 그의 제조방법에 관한 것이다.
거품을 형성하기 쉬운 비경구투여용 약제학적 액상 제제의 제조 및 투약은 어려우며 항상 시간 소모적이다. 또한 거품이 소멸될 때까지 기다려야 하므로 투여준비가 지체된다. 용액의 표면이 거품으로 덮여있는 주사액은 투여할 수 없다.
실리콘 오일 또는 옥탄올과 같은, 경구 투여용 약제학적 액상 제제에 사용되는 통상적인 소포제는, 그의 독성으로 인하여 비경구투여용 제제에 대한 첨가제로서는 문제점이 있다. 또한, 이러한 첨가제는 주사용액을 혼탁시킬 수 있다. 미량의 에틸렌 옥사이드/프로필렌 옥사이드 블록 공중합체형 계면활성제가 거품을 신속히 소멸시킨다는 놀라운 사실이 밝혀졌다.
에틸렌 옥사이드/프로필렌 옥사이드 블록 공중합체는 거품을 약간 일으키는 원료로서 식기 세척 및 세탁용 세제에 사용된다. 유화제, 해유화제, 및 습윤제로서의 그의 특성은 공지되어 있다. 그러나, 그의 소포제로서의 사용가능성은 공지되어 있지 않다.
따라서, 본 발명은 에틸렌 옥사이드/프로필렌 옥사이드 블록 공중합체의, 약제학적 액상제제, 특히 비경구투여용 액상 제제의 거품 억제용 용도에 관한 것이다.
특히 바림직한 에틸렌 사이드/프로필렌 옥사이드 블록 공중합체는 폴리에틸렌/폴리프로필렌 글리콜1800(PPG1800)이다. 이는 비경구투여용 제제에 사용하기에 특히 바람직하다. 소포제는 약제학적 보조제에 대해 통상적으로 요구되는 순도 기준에 부합되어야 한다.
원칙적으로, 에틸렌 옥사이드/프로필렌 옥사이드 블록 공중합체는 약제의 모든 액상 제제, 특히 수성 제제의 거품을 억제하는데 적절하다. 물론, 액제는 사용되는 에틸렌 옥사이드/프로필렌 옥사이드 블록 공중합체와 혼화성이어야 한다. 적절한 액제는 예는 세프리롬(cefpirom; HR 810)과 같은 세팔로스포린 유도체 및 프로카인 벤질페니실린(procaine benzylpinicillin)과 같은 퍼닐실린 유도체이다.
본 발명은 또한 거품을 억제하기 위한 에틸렌 옥사이드/프로필렌 옥사이드 블록 공중합체를 함유하는 약제학적 제제 및 이를 제조하는 방법에 관한 것이다.
액상 제제는, 예를 들어, PPG 1800과 같은 에틸렌 옥사이드/프로필렌 옥사이드 블록 공중합체를 약 0.1내지 0.00001중량%함유하며, 바람직하게는 0.01 내지 0.0001중량%의 PPG1800을 함유한다.
제제의 제조방법은 에틸렌 옥사이드/프로필렌 옥사이드 블록 공중합체를 고체 활성 화합물에 도포하거나, 고체 활성 화합물을 중합체로 함침시키거나, 중합체를 용매에 가하는 것을 포함한다.
실시예 1
1.23ｇ의 HR 810 설페이트 및 0.22ｇ의 NaCl3(무수)를 함유하는 주사용 제제는,10㎖의 물에 용해된 후, 약 5분 동안 안정한 거품을 나타냈다.
물 대신에 폴리에틸렌/폴리프로필렌 글리콜 1800의 0.0005%수용액에 용해시킨 경우, 거품은 용해 직후 소멸되었다.
대략 등량의 PPG 1800을 고체 성분에 적용시키는 경우, 단독으로 또는 함께, 예를 들어, PPG 1800의 존재하에 침전시키거나, 침전된 고체를 세척하는 데 사용되는 용액에 PPG 1800을 가하거나 고체상에 PPG 1800용액을 분무한 후 건조시킴으로써 동일한 효과를 얻을 수 있다. 또한, 1차 포장(예를 들어, 주사용 바이알 및 /또는 주사용 바이알 스토퍼)을 PPG1800으로 함침시켜 PPG 1800의 소포효과를 수득할 수 있다.
실시예 2
300,000IU/㎖프로카인 벤질페닐실린 현탁액은 거품이 과도하여 투약이 어렵다. 현탁액은 거품에 의해 부피가 증가되었으므로, 1차 포장을 위한 주사용 바이알에 충전하지 못한다. 단지 0.001%의 PPG1800은 하기 표에 나타난 바와 같이 거품형성을 억제한다.

Claims (6)

  1. 폴리에틸렌/폴리프로필렌 글리콜 1800을 고형의 페니실린 유도체 또는 세팔로스포린 유도체에 도포하거나, 고형의 페니실린 유도체 또는 세팔로스포린 유도체를 폴리에틸렌/폴리프로필렌 글리콜 1800으로 함침시킨 다음 이것을 적절한 용매에 용해시키는 단계를 포함하거나; 페니실린 유도체 또는 세팔로스포린 유도체를 폴리에틸렌/폴리프로필렌 글리콜 1800의 존재하에 용매에 용해시키는 단계를 포함함을 특징으로 하여, 활성 화합물로서 페니실린 유도체 또는 세팔로스포린 유도체를 함유하는 약제학적 수성제제내에서 거품을 억제하는 방법.
  2. 제1항에 있어서, 약제학적 수성제제가 비경구투여용 제제인 방법.
  3. 제1항에 있어서, 활성 화합물로서 세프피롬을 사용하는 방법.
  4. 제1항에 있어서, 폴리에틸렌/폴리프로필렌 글리콜 1800을 0.1내지 0.00001중량%의 농도로 사용하는 방법.
  5. 제4항에 있어서, 폴리에틸렌/폴리프로필렌 글리콜 1800을 0.01내지 0.0001중량%의 농도로 사용하는 방법.
  6. 제1항에 있어서, 활성화합물로서 프로카인 벤질페니실린을 사용하는 방법.
