Description
본 설명은 첨부 도면에 비추어 다음의 상세한 설명을 읽음으로써 더 잘 이해될 것이며, 이들 도면에서,
도 1A 내지 1C는 각각 본 개시의 일 실시양태에 따라 (A) CD90, (B) CD 105에 대해 양성이고 (C) CD34에 대해 음성인 실시예 1의 hMSC를 도시하고;
도 2A는 본 개시의 일 실시양태에 따라 알칼리성 포스파타제 염색으로 확인된 실시예 1의 hMSC로부터 분화된 연골 세포를 나타내는 사진이며;
도 2B는 본 개시의 일 실시양태에 따라 알시안 블루 (Alcian Blue)에 의해 염색된 실시예 1의 hMSC로부터 분화된 연골을 나타내는 사진이고;
도 2C는 본 개시의 일 실시양태에 따라 오일 레드 O로 염색된 실시예 1의 hMSC로부터 분화된 지방 세포를 나타내는 사진이다.
Claims (15)
- 미분화된 인간 중간엽 줄기 세포 (hMSC)의 집단을 제공하는 방법으로서,
(a) 인간 대상으로부터 말초 혈액을 수득하는 단계;
(b) 단계 (a)에서의 말초 혈액에 글리세린을 첨가하는 단계;
(c) 단계 (b)의 말초 혈액에서 다른 체세포의 줄기 세포에 대해 hMSC를 분리하는 단계; 및
(d) 단계 (c)의 분리된 hMSC를 글리세린 함유 배지에서 배양하여 hMSC 집단을 생성하는 단계;를 포함하고,
상기 배지는 과립구 콜로니 자극 인자 (G-CSF), 과립구-대식세포 콜로니 자극 인자 (GM-CSF) 및 이들의 조합으로 이루어진 군으로부터 선택된 어떤 작용제도 함유하지 않는,
미분화된 인간 중간엽 줄기 세포 (hMSC)의 집단을 제공하는 방법. - 제1항에 있어서, 단계 (a)에서, 말초 혈액이 어떤 용혈제도 함유하지 않는 튜브에서 수집되는, 방법.
- 제1항에 있어서, 단계 (b)에서, 글리세린이 말초 혈액에 약 1 내지 10 mg/mL의 농도로 존재하는, 방법.
- 제1항에 있어서, 단계 (d)에서, 배지가 적어도 20% (v/v) 혈청을 포함하고, 글리세린은 배지에 약 0.1 내지 1.0 mg/mL의 농도로 존재하며, hMSC는 37 ℃에서 적어도 16 시간 동안 글리세린 함유 배지를 포함하는 배양 플레이트에서 배양되는, 방법.
- 제4항에 있어서,
(e) 배양된 hMSC를 단계 (d)에서의 것보다 표면적이 적어도 20 배 더 큰 다른 배양 플레이트로 옮기고, 미분화된 hMSC 집단을 생성하기 위해 글리세린 함유 배지에서 배양을 계속하는 단계를 추가로 포함하는 방법. - 제5항에 있어서, 단계 (e)에서, hMSC가 미분화된 hMSC의 집단을 생성하기 위해 적어도 4 일 동안 배양되는, 방법.
- 제4항 내지 제6항 중 어느 한 항에 있어서, 생성된 미분화된 hMSC의 집단이 CD34-, CD45-, 및 HLA-DR 세포 표면 마커에 대해 음성이고, CD73+, CD90+, 및 CD 105+ 세포 표면 마커에 대해 양성인, 방법.
- 제5항 또는 제6항의 방법에 따라 생성된 실질적으로 순수한 hMSC 집단을 포함하는 클론 세포주.
- 치료를 필요로 하는 대상의 질환 또는 장애의 치료를 위한, 미분화된 hMSC 또는 그의 분화된 자손의 집단의 용도로서, 여기서 미분화된 hMSC의 집단은 제4항 내지 제6항 중 어느 한 항의 방법에 따라 생성되고, 미분화된 hMSC 또는 그의 분화된 자손의 집단이 질환 또는 장애의 치료를 위해 대상에게 투여되는, 용도.

- 제4항 내지 제6항 중 어느 한 항의 방법에 따라 생성된 미분화된 hMSC 집단의 유효량을 질환 또는 장애를 치료하기 위해 대상에게 투여하는 단계를 포함하는, 대상의 질환 또는 장애를 치료하는 방법.
- 제12항에 있어서, 제4항 내지 제6항 중 어느 한 항의 방법에 따라 생성된 미분화된 hMSC 집단이 실질적으로 순수한 hMSC 집단인, 방법.
- 제13항에 있어서, 미분화된 hMSC 집단이 연골 세포, 연골 또는 지방 세포로 분화될 수 있는 방법.
