Claims (16)
- 일반 화학식 III의 비닐 스탄난을 할로겐화 구리 CuX 및 알킬리튬 R1Li과 반응시켜 제조된 일반 화학식 II의 비닐 구리산의 구리산 커플링에 의한 일반 화학식 I의 화합물의 제조로서,

(화학식에서 R은 직쇄 또는 분지쇄 C1-4 알킬 기를 나타냄)

(화학식에서
R2는 H 또는, 선택적으로 규소 원자를 함유하는, 알코올 보호기, 예로서 트리메틸실릴-, 트리에틸실릴-, tert.-부틸디메틸실릴- 기, 또는 산소 원자를 함유하는 고리형 또는 열린 사슬 알킬 기, 예로서 테트라하이드로피라닐-, 메톡시메틸- 또는 에톡시메틸- 기를 나타내고;
X는 요오드 원자, 브롬 원자, CN, SCN, OSO2CF3 기를 의미하고,
R1은 C1-6 알킬 기를 나타내고
R2의 의미가 수소 원자 이외인 경우 n >2, R2의 의미가 수소 원자인 경우 n >3임),
일반 화학식 IV의 보호된 에논이

(화학식에서 R3은 THP- 또는 트리알킬실릴- 기이고, R의 의미는 위에서 정의된 바와 같음)
구리산염 반응에 참여하는데, 구리산염 반응은
다음을 특징으로 하는 것인, 일반 화학식 I의 화합물의 제조:
a) 요오드화 구리(I)와 비교하여,
R2 ≠ H인 경우 2 내지 2.4몰 비율로,
R2 = H인 경우 3 내지 3.4몰 비율로 적용되는,
과량의 알킬리튬이 일반 화학식 II와 IV의 화합물의 커플링 반응 전에 분해되고,
b) 생성된 일반 화학식 V의 화합물의 보호기

(화학식에서 R, R2 및 R3의 의미는 위에서 정의된 바와 같음)가 제거되며, 수득된 일반 화학식 I의 화합물은 크로마토그래피로 정제된다. - 제1항에 있어서, 과량의 알킬리튬의 분해는 작은 분자량의 케톤, 에스테르 또는 트리알킬실릴 할로겐화물 화합물로 수행되는 것인 제조 방법.
- 제2항에 있어서, 과량의 알킬리튬의 분해는 아세톤 또는 에틸 아세테이트로 수행되는 것인 제조 방법.
- 제1항에 있어서, 미소프로스톨의 정제는 중력 컬럼 크로마토그래피에 의한 실리카겔 컬럼 상에서 수행되는 것인 제조 방법.
- 제4항에 있어서, 실리카겔 상에서의 크로마토그래피를 위하여 다성분 단계별 구배 혼합물이 용출제로서 사용되는 것인 제조 방법.
- 제5항에 있어서, 다성분 용출제 혼합물은 소량의 산을 함유하는 것인 제조 방법.
- 제6항에 있어서, 소량의 산은 0.1 내지 0.01% 아세트산 또는 포름산인 것인 제조 방법.
- 제7항에 있어서, 실리카겔 크로마토그래피의 용출제는 0.05%의 포름산을 함유하는 것인 제조 방법.
- 제4항에 있어서, 미소프로스톨의 정제는 정제 크로마토그래피 및 여과 크로마토그래피에 의해 수행되는 것인 제조 방법.
- 제9항에 있어서, 정제 크로마토그래피는 기술적 불순물 및 관련 불순물을 제거하며, 여과 크로마토그래피는 용매의 오염을 제거하는 것인 제조 방법.
- 제10항에 있어서, 정제 크로마토그래피를 위하여 구형 YMC S75 실리카겔이 사용되거나, 또는 비정형 Kieselgel Si 60(0.063 ~ 0.200 mm) 실리카겔 및 활성탄을 통한 여과가 적용되는 것인 제조 방법.
- 제11항에 있어서, 정제 크로마토그래피의 다성분 용출제는 디이소프로필 에테르, C1-6 알코올, 선택적으로 아세톤, 메틸 에틸 케톤 및 0.05%의 포름산을 함유하는 것인 제조 방법.
- 제12항에 있어서, 정제 크로마토그래피의 용출제는 0.05%의 포름산을 함유하는, 디이소프로필 에테르: 이소프로판올 또는 디이소프로필 에테르: 아세톤: 메탄올 용매의 단계별 구배 혼합물인 것인 제조 방법.
- 제10항에 있어서, 활성탄을 통한 여과는 헥산: 아세톤 용매 혼합물을 이용하여 수행되는 것인 제조 방법.
- 제10항에 있어서, 여과 크로마토그래피는 Kieselgel Si 60(0.063 ~ 0.200 mm) 실리카겔 컬럼 상에서 수행되며, 용출제로서 0.05%의 포름산을 함유하는, 디클로로메탄: 아세톤 및 메틸 tert.-부틸 에테르: 아세톤 용매의 단계별 구배 혼합물을 적용하는 것인 제조 방법.
- 제6항에 있어서, 0.05%의 산을 함유하는 크로마토그래피의 분획을 워크업 전에 중성이 될 때까지 세척하는 것인 제조 방법.
