Abstract
[식 I]
상기 식에서, - E1 및 E2는, 동일하거나 상이한, 각각 독립적으로 분석물의 적어도 하나의 에피토프를 포함하는 펩타이드 서열을 나타내고, - X 및 Y는, 동일하거나 상이할 수 있고, 각각 독립적으로 링커 암(linker arm)을 나타내며, - 캐리어 분자는 가용성이고, - Z는 캐리어 분자와의 결합 이전에 티올 작용기를 지닌 아미노산 유도체를 나타낸다. 본 발명은 또한 상기 화합물을 함유하는 조성물, 및 면역검정에서 대조군 또는 표준물로서 이러한 화합물 또는 당해 화합물을 함유하는 조성물의 용도, 대조군 또는 표준물로서 상기 화합물 또는 당해 화합물을 함유하는 조성물을 사용하는 방법, 및 최종적으로 상기 화합물 또는 당해 화합물을 함유하는 조성물을 포함하는 면역검정을 시행하기 위한 키트에 관한 것이다.
Description
- 도 1은 선행 기술에 따른 이중-에피토프 화합물(REF 화합물), 본 발명에 따른 이중-에피토프 화합물(화합물 1) 및 본 발명의 이중-에피토프 화합물 1에 상응하지만 캐리어 분자에 커플링되지 않는 이중-에피토프 펩타이드(커플링되지 않은 펩타이드 1)에 의해 방사된, 이들 농도의 함수로서, VIDAS® 자동화된 장치에 의해 측정된, 형광성 시그날 RFV를 제공하는 그래프이다.
- 도 2는 선행 기술에 따른 이중-에피토프 화합물(REF 화합물) 및 본 발명에 따른 이중-에피토프 화합물(화합물 1 및 3)에 의해 방사된, 이들의 농도의 함수로서, VIDAS® 자동화된 장치에 의해 측정된, 형광성 시그날 RFV를 제공하는 그래프이다.

Claims (17)
- 청구항 1에 있어서,
링커 암 X 및 Y가 한편으로는 E1 또는 E2와, 또 다른 한편으로는 Z와, 2개의 펩타이드 결합 -CO-NH-를 각각 형성하는 아미노산 유도체인, 식 I의 이중-에피토프 화합물. - 청구항 2에 있어서,
링커 암 X 및 Y가, 동일하거나 상이할 수 있고, 각각 독립적으로 하나 이상의 아미노산 유도체를 포함하는, 식 I의 이중-에피토프 화합물. - 청구항 1 내지 청구항 3 중의 어느 한 항에 있어서,
Z가 시스테인 유도체, 호모시스테인 유도체 및 페니실라민 유도체로부터 선택되는, 식 I의 이중-에피토프 화합물. - 청구항 1 내지 청구항 4 중의 어느 한 항에 있어서,
상기 캐리어 분자가, 분자량이 20kDa 내지 700kDa, 바람직하게는 60kDa 내지 250kDa인 단백질인, 식 I의 이중-에피토프 화합물. - 청구항 5에 있어서,
상기 캐리어 분자가 소 혈청 알부민(bovine serum albumin)인, 식 I의 이중-에피토프 화합물. - 청구항 1 내지 청구항 6 중의 어느 한 항에 있어서,
E1 및 E2가 적어도 하나의 심장 트로포닌 I 에피토프(cardiac troponin I epitope) 또는 프로디펜신-A6 에피토프(prodefensin-A6 epitope)를 포함하는 펩타이드 서열인, 식 I의 이중-에피토프 화합물. - 물, 완충액 또는 생물학적 유체 내 용액 중에, 청구항 1 내지 청구항 7 중의 어느 한 항에 따른 식 I의 이중-에피토프 화합물을 포함하는 조성물.
- 면역검정(immunoassay)에서의 대조군 또는 표준물로서의, 청구항 1 내지 청구항 7 중의 어느 한 항에 따른 화합물 또는 청구항 8에 따른 조성물의 용도.
- 심장 트로포닌 I 면역검정 또는 프로디펜신-A6 면역검정에서의 대조군 또는 표준으로서의, 청구항 1 내지 청구항 7 중의 어느 한 항에 따른 화합물 또는 청구항 8에 따른 조성물의 용도.
- 하기를 포함하는, 분석물을 함유할 수 있는 시험 샘플에서 면역검정(immunoassay)에 의해 상기 분석물을 검출하는 방법:
i. 시험 샘플을 분석물에 대한 하나 이상의 결합 파트너와 접촉시킴에 의한 면역검정 시험의 단계,
ii. 양성 대조군으로서 청구항 1 내지 청구항 7 중의 어느 한 항에 따른 식 I의 이중-에피토프 화합물 또는 청구항 8에 따른 조성물을 분석물에 대한 하나 이상의 결합 파트너와 접촉시킴으로써 면역검정 시험의 유효성(validity)을 입증하기 위한 시험의 단계,
iii. 유효성 입증 시험이 양성인지에 대한, 면역검정 시험의 판독 단계,
iv. 단계 i의 면역검정 시험에 의해 수득된 시그날이 면역검정 시험의 검출 한계점(threshold)보다 큰 경우, 시험 샘플 내 상기 분석물의 존재의 측정 단계. - 청구항 11에 있어서,
분석물이 심장 트로포닌 I 또는 프로디펜신-A6인, 면역검정에 의해 분석물을 검출하는 방법. - 하기를 포함하는, 분석물을 함유할 수 있는 시험 샘플 속에서, 면역검정에 의해 상기 분석물을 정량화하는 방법:
i. 상기 시험 샘플을, 분석물에 대한 하나 이상의 결합 파트너와 접촉시킴에 의한 면역검정 시험 단계,
ii. 양성 대조군을 상기 분석물에 대한 상기 하나 이상의 결합 파트너와 접촉시킴으로써 면역검정 시험의 유효성을 입증하기 위한 시험 단계,
iii. 유효성 입증 시험이 양성인지에 대한 면역검정 시험의 판독 단계,
iv. 상기 면역검정 시험에 의해 수득된 시그날을 청구항 1 내지 청구항 7 중의 어느 한 항에 따른 식 I의 이중-에피토프 화합물 또는 청구항 8에 따른 조성물을 사용하여 사전에 수득한 표준 곡선과 비교함에 의한 시험 샘플 속에서의 상기 분석물의 존재의 측정 단계. - 청구항 13에 있어서,
양성 대조군이 청구항 1 내지 청구항 7 중의 어느 한 항에 따른 식 I의 이중-에피토프 화합물이거나 또는 청구항 8에 따른 조성물인, 면역검정에 의해 정량화하는 방법. - 하기를 포함하는, 분석물을 함유할 수 있는 시험 샘플 속에서 면역검정에 의해 상기 분석물을 정량화하는 방법:
i. 시험 샘플을 분석물에 대한 하나 이상의 결합 파트너와 접촉시킴에 의한 면역검정 시험 단계,
ii. 양성 대조군으로서, 청구항 1 내지 청구항 7 중의 어느 한 항에 따른 식 I의 이중-에피토프 화합물 또는 청구항 8에 따른 조성물을, 분석물에 대한 하나 이상의 결합 파트너와 접촉시킴에 의한 면역검정 시험의 유효성을 입증하기 위한 시험 단계,
iii. 유효성 입증 시험이 양성인 경우, 면역검정 시험을 판독하는 단계,
iv. 면역검정 시험을 표준 곡선과 비교함에 의한 시험 샘플 속에서의 상기 분석물의 양의 측정 단계. - 청구항 13 내지 청구항 15 중의 어느 한 항에 있어서,
상기 분석물이 심장 트로포닌 I 또는 프로디펜신-A6인, 면역검정에 의해 분석물을 정량화하는 방법. - 청구항 1 내지 청구항 7 중의 어느 한 항에 따른 식 I의 이중-에피토프 화합물, 또는 청구항 8에 따른 조성물을 포함하는, 면역검정을 시행하기 위한 키트(kit).
