Claims (8)
- 틴달화된 (tyndallized) 락토바실러스 람노수스 (Lactobacillus rhamnosus) GG (ATCC 53103) 박테리아 세포의 샘플에서 무손상 세포막을 가진 사멸 세포의 수를 계수하는 방법으로서,
- 연속 희석 (serial dilution)에 의해, 틴달화된 락토바실러스 람노수스 (Lactobacillus rhamnosus) GG (ATCC 53103) 박테리아 세포를 105 - 107 세포/ml 범위의 농도로 포함하는 샘플을 준비하는 단계;
- 상기 샘플에 제1 시약 티아졸 오렌지와 제2 시약 요오드화프로피디움을 첨가하여, 용액을 수득하는 단계;
- 상기 용액에, 아지드화나트륨 중의 형광성 마이크로스피어 (fluorescent microsphere) 현탁액을 첨가하여, 테스트 샘플을 수득하는 단계;
- 상기 테스트 샘플에 대해, 유세포 형광측정법 (flow cytofluorometry)에 의해, 생 세포 및 사멸 세포를 포함하는 전체 세포 수의 계수 및 사멸 세포 수만의 계수를 실시하는 단계;
- 틴달화 샘플에 존재하는 사멸 세포의 수를 산출하는 단계. - 제1항에 있어서,
틴달화된 락토바실러스 람노수스 (Lactobacillus rhamnosus) GG (ATCC 53103) 박테리아 세포를 105 - 107 세포/ml 범위의 농도로 포함하는 샘플 0.5 ml에, 상기 제1 시약 2.5 ㎕과 상기 제2 시약 1.5 ㎕를 첨가하여 용액을 제조하는 단계를 포함하는, 방법. - 제1항 또는 제2항에 있어서,
상기 아지드화나트륨 중의 형광성 마이크로스피어 현탁액이 폴리스티렌 마이크로스피어를 포함하며, 바람직하게는 비드 현탁액이 0.1% 요오드화나트륨 용액 중의 형광성 폴리스티렌 마이크로스피어 현탁액인, 방법. - 제3항에 있어서,
세포 수 측정을 위해, 10 내지 100 ㎕, 바람직하게는 40 내지 60 ㎕ 범위의 기지량 (known amount)의 비드를 첨가하는, 방법. - 무손상 세포벽을 가진 틴달화된 락토바실러스 람노수스 (Lactobacillus rhamnosus) GG (ATCC 53103) 박테리아 세포의 제조 방법으로서,
- 1x106 내지 1x1010 UFC/g 범위의 농도를 가진 락토바실러스 람노수스 (Lactobacillus rhamnosus) GG (ATCC 53103) 박테리아 세포 배양물을 제조하는 단계;
- 상기 배양물에 틴달화 공정을 수행하여, 틴달화된 박테리아 세포 배양물을 수득하는 단계;
- 제1항 내지 제4항 중 어느 한 항에 따른 계수 방법을 적용하는 단계를 포함하는, 방법. - 제5항에 따른 방법으로 수득되는 면역학적으로 활성인 틴달화된 무손상 락토바실러스 람노수스 (Lactobacillus rhamnosus) GG (ATCC 53103) 박테리아 세포 배양물.
- 틴달화된 락토바실러스 람노수스 (Lactobacillus rhamnosus) GG (ATCC 53103) 박테리아 세포 샘플에서 무손상 세포막을 가진 틴달화된 락토바실러스 람노수스 (Lactobacillus rhamnosus) GG (ATCC 53103) 박테리아 사멸 세포의 수를 계수하는데 있어서의 유세포 형광측정법의 용도.
- 제7항에 있어서,
상기 무손상 세포막을 가진 틴달화된 락토바실러스 람노수스 (Lactobacillus rhamnosus) GG (ATCC 53103) 박테리아 사멸 세포는 제1항 내지 제4항 중 어느 한 항에 따른 방법에 의해 계수되는, 용도.
