Abstract

본 발명은 브랜치드 멀티펩티드 조성물 및 이를 포함하는 백신에 관한 것으로, 보다 상세하게는 종양항원 펩티드를 포함하는 브랜치드 멀티펩티드 조성물과 이를 포함하는 암 면역치료용 백신에 관한 것이다. 본 발명에 따른 브랜치드 멀티 펩티드 백신은 제조와 활용이 용이하여 쉽게 치료에 적용할 수 있으며 생체 내에서 안정적인 반응을 유지할 수 있음으로 효과적인 백신이 될 것으로 기대된다. 또한 종양항원 펩티드의 경우 본 발명에서는 악성뇌종양에서 많이 발현된 항원을 선택할 수 있으나, 향후 환자 개인의 종양 특성에 따라 발현량이 큰 종양항원을 분석하여 개인 맞춤형 브랜치드 펩티드 및 이를 이용한 백신의 제조에 활용할 수 있을 것으로 기대된다.

Description

브랜치드 멀티펩티드 조성물 및 이를 포함하는 백신{Composition comprising branched multipeptide vaccine and vaccine comprising the same}
본 발명은 브랜치드 멀티펩티드 조성물 및 이를 포함하는 백신에 관한 것으로, 보다 상세하게는 종양항원 펩티드를 포함하는 브랜치드 멀티펩티드 조성물과 이를 포함하는 암 면역치료용 백신에 관한 것이다.
뇌종양의 연간 발생 빈도는 인구 10만 명 당 5 내지 15.8 명으로, 다른 암에 비하여 발생 빈도는 낮지만 인구의 고령화, 암 진단 방법의 발전과 의료 혜택의 범위 증가로 인하여, 과거보다 발생률 자체는 증가하고 있다. 이 중 난치성 악성 뇌종양의 경우 수술, 항암요법 또는 방사선 요법 등의 다양한 치료를 수행하더라도 재발이 잘 되는 문제로 인해 생존률이 낮은 문제점이 있다. 악성 뇌종양 중 교모세포종의 경우 수술과 방사선 치료 및 항암 치료를 병행하는 표준요법을 시행하여 초기 치료에 많은 시간과 비용을 소모하더라도, 재발에 따른 평균 수명은 15 개월 정도에 불과하여 방사선 표준요법을 보완할 수 있는 새로운 치료법에 대한 개발의 필요성이 절실한 상황이다. 그러나, 효과가 입증되지 않은 신약 치료는 고비용으로 인해 환자의 경제적 부담을 증가시키는 문제가 있다.
암 치료에 사용되는 상기와 같은 일반적 방법의 한계 극복을 위해 다양한 치료 방법이 연구되고 있으며, 최근에는 암 특이적 치료효과를 보이면서 부작용이 없는 암 면역 치료 방법이 주목을 받고 있다. 암 면역 치료법 중 수지상 세포나 자연살상세포를 이용한 면역 치료는 실용 단계까지 많은 시간과 비용이 필요하나 펩티드 백신의 경우 상기 면역 치료법 대비 효율은 다소 떨어질 수 있으나 저비용으로 간단히 제조할 수 있으며, 독성이 적고 여러 가지 종양항원을 조합하여 제조할 수 있는 장점이 있다. 악성 뇌종양에서 다양하게 발현되는 종양항원을 이용하여 멀티펩티드를 제조하고 이에 대한 임상 실험을 진행한 일예 또한 알려져 있다(Dutoit et al., 2012).

본 발명에서는 상기 제시한 악성뇌종양의 종양세포 이질성 극복을 위해 다양한 종양항원을 합성하여 사용하면서 생체 내에서 보다 지속적인 효과를 낼 수 있는 브랜치드 멀티펩티드와 이를 이용한 백신을 제공하고자 하였다.
본 발명은 화학식 I의 구조를 갖는 암 면역치료용 멀티펩티드 조성물에 관한 것이다.

상기 화학식 I에서,
A, B 및 C는 각각 뇌종양항원 CD99, BIRC5, ERBB2, WT-1 및 PanDR epitope 로 이루어진 군로부터 선택되는 어느 하나의 펩티드의 잔기서열(subsequence residue)이고;
Sp는 잔기서열과 리신(lysine)의 연결을 위한 스페이서이고;
Lys은 리신(lysine)이다.
상기 A, B 및 C는 각각 서열번호 1 내지 17로부터 선택되는 어느 하나의 아미노산 서열을 갖을 수 있으며, 바람직하게 상기 A는 서열번호 1 내지 5로부터 선택되는 어느 하나의 아미노산 서열, 상기 B는 서열번호 6 내지 8로부터 선택되는 어느 하나의 아미노산 서열 및 상기 C는 서열번호 9 내지 13으로부터 선택되는 어느 하나의 아미노산 서열을 갖을 수 있으며, 더욱 바람직하게 상기 A는 서열번호 1의 아미노산 서열, 상기 B는 서열번호 8의 아미노산 서열 및 상기 C는 서열번호 9의 아미노산 서열을 갖을 수 있다.
본 발명에서 상기 Sp는 스페이서로서 제한되지는 않으나 8-amino-3,6-dioxa-octanoic acid (mini-PEG2)을 사용할 수 있다.
또한 본 발명은 상기 조성물을 포함하는 암 면역치료용 백신에 관한 것이다.
인터페론(Interferon)은 최초로 발견된 사이토카인(Cytokine)으로 처음에는 항바이러스 활성을 가진 물질로 확인되었으며, 이외에도 면역 조절 작용, 세포 증식 억제 작용 등을 가진 당단백(glycoprotein)이다. 인터페론에 의한 종양 치료의 중요 기전으로는 세포독성 T-림프구를 자극하고 주요 조직 적합 복합체 항원을 활성화 시키는 것이다. 이 외에도 자연 살해 세포와 대식 세포를 자극하는 면역 증강 효과가 있고, 내피 세포 증식과 신생 혈관 억제 효과를 나타내며, 암세포에 대한 직접적인 증식 억제 작용을 하는 것으로 알려져 있다.
인터페론은 알파(α), 베타(β), 감마(γ) 3가지로 구분되는데 알파(α)와 베타(β)는 바이러스에 감염된 대부분의 세포에서 만들어지며, 감마(γ)는 T-림프구 및 자연 살해 세포(natural killer cell, NK cell)에서만 합성된다.
본 발명의 일 실시예에 의하면 브랜치드 멀티펩티드를 투여한 경우 상기 인터페론 감마의 분비능이 현저히 증가함을 확인하였으며, 이로부터 브랜치드 멀티펩티드가 종양에 대한 면역치료를 위해 유용하게 활용하게 활용될 수 있음을 알 수 있었다.
본 발명에 따른 브랜치드 멀티 펩티드 백신은 제조와 활용이 용이하여 쉽게 치료에 적용할 수 있으며 생체 내에서 안정적인 반응을 유지할 수 있음으로 효과적인 백신이 될 것으로 기대된다. 또한 종양항원 펩티드의 경우 본 발명에서는 악성뇌종양에서 많이 발현된 항원을 선택할 수 있으나, 향후 환자 개인의 종양 특성에 따라 발현량이 큰 종양항원을 분석하여 개인 맞춤형 브랜치드 펩티드 및 이를 이용한 백신의 제조에 활용할 수 있을 것으로 기대된다.
도 1은 교모세포종조직 32개와 정상뇌조직 7개에서 종양항원의 발현 양상에 관한 것이다.
도 2는 정상뇌조직에서는 발현되지 않으면서 교모세포종조직에서만 발현되는 종양항원을 선별한 것에 관한 것이다.
도 3은 잔기서열 각각에 대하여, T-림프구와의 결합능에 관한 것이다.
도 4는 브랜치드 멀티펩티드의 T-림프구 결합능 및 결합 후 안정성에 관한 것이다.
도 5는 브랜치드 멀티펩티드와 인터페론 감마 분비능의 상관관계에 관한 것이다.
이하 본 발명을 실시예와 첨부된 도면을 참조하여 상세히 설명한다. 그러나 이들은 본 발명을 보다 상세하게 설명하기 위한 것으로, 본 발명의 권리범위가 하기의 실시예에 의해 한정되는 것은 아니다.

[시험예 1] 멀티펩티드 제조를 위한 종양항원의 선별
악성 뇌종양인 교모세포종조직 32개와 정상뇌조직 7개에서 종양항원의 발현 양상을 통상적인 RT-PCR(reverse trascription-polymerase chain reaction)과 웨스턴 블롯(western blot)을 통해 분석하여(도 1), 이 중 정상뇌조직에서는 발현되지 않으면서 교모세포종조직에서만 발현되는 종양항원을 선별하여 멀티펩티드의 제조를 위한 후보 종양항원으로 사용하였다(도 2).
선별 결과 BIRC5, ERBB2 및 CD99의 세 가지 항원이 상기 조건을 만족하였으며, 이를 이용하여 멀티펩티드를 제조하였다.
[시험예 2] 멀티펩티드 합성을 위한 종양항원 잔기서열의 T-림프구 결합능 측정
멀티펩티드 합성을 위한 종양항원의 관련 잔기서열은 하기 표 1에 정리하였다.

상기 표의 잔기서열 각각에 대하여, T-림프구와의 결합능을 측정하였다(도 3).
T-림프구와 잔기서열과의 결합능 측정
T2 세포주(한국세포주은행)는 HLA-A2를 표현하는 T세포와 B세포의 혼합형이다. T2세포에 펩티드와 Beta2-microglobulin과 함께 하룻밤 배양한 후 T2 세포를 antihuman HLA-A2-FITCmAb로 염색을 하고 FACS Caliber flow cytometer와 WinMDI software를 이용하여 분석하였다.
잔기서열의 결합능은 펩티드를 포함하는 평균 형광값(mean fluorescence with the peptide)과 펩티드를 포함하지 않은 평균 형광값(mean fluorescence without the peptide)으로부터 하기 수학식을 이용하여 계산하였다.

[실시예] 브랜치드 멀티펩티드의 합성
상기 방법을 통해 도 3과 같이 측정한 결합능으로부터, 결합능이 가장 우수한 서열번호 1의 ERBB2, 서열번호 9의 BIRC5 및 서열번호 10의 CD99의 잔기서열을 선택하였다. 또한 브랜치드 멀티펩티드 합성시 스페이서(spacer)로는 mini-PEG2(8-Amino-3,6-Dioxa-Octanoic acid)를 이용하였다. 합성은 브랜치드 멀티펩티드이 펩트론 (대전광역시 유성구 유성대로 1628번길 37-24)에 의뢰하여 수행하였다.
상기 합성을 통해 얻은 브랜치드 멀티펩티드의 구조는 아래 화학식 II와 같다.

[시험예 3] 브랜치드 멀티펩티드의 T-림프구 결합능 및 결합 후 안정성 측정
상기와 같이 합성된 화학식 II 구조의 브랜치드 멀티펩티드에 대하여 T-림프구 결합능 및 결합 후 안정성을 측정하였다.
브랜치드 멀티펩티드의 T-림프구 결합능은 T2 세포에 상기 제조한 브랜치드 멀티펩티드와 베타2-마이크로글로불린(β2-microglobulin)을 함께 하룻밤 배양한 후 T2 세포를 anti-human HLA-A2-FITCmAb로 염색을 하고 FACS Caliber flow cytometer와 WinMDI 소프트웨어를 이용하여 분석하였다. 또한 T-림프구와 브랜치드 멀티펩티드와의 안정성은 T2 세포에 상기 제조한 브랜치드 멀티펩티드와 베타2-마이크로글로불린(β2-microglobulin)을 함께 하룻밤 배양한 후 Brefeldin A를 처리하였다. 이후 0, 2, 4, 8, 24시간째에 결합능을 각각 측정하여 안정성을 확인하였다.
상기 방법을 통해 합성된 브랜치드 멀티펩티드의 T-림프구 결합능과 안정성 측정 결과와의 비교를 위해 서열번호 1의 ERBB2, 서열번호 9의 BIRC5 및 서열번호 10의 CD99의 잔기서열을 갖는 올리고펩티드의 혼합물(multipeltide-cocktail)을 비교예로 하여 브랜치드 멀티펩티드의 측정 결과와 비교하여 그 결과를 도 4에 도시하였다.
상기 결과로부터 브랜치드 펩티드의 구조가 단일 올리고펩티드의 혼합물에 비교하여 시간이 경과하더라도 T-림프구와의 결합능이 현저히 안정적으로 유지되는 것을 확인하였다.
[시험예 4] 브랜치드 멀티펩티드와 인터페론 감마 분비능의 상관관계
상기 제조한 브랜치드 펩티드로 자극된 수지상 세포(dendritic cells, DC)를 통하여 생성된 세포독성림프구는 표적세포인 T2 세포에 각각 아래 표 2의 조성으로 처리하였다.

각 실험군에 특이적 인터페론 감마의 분비능 측정은 ELISPOT(enzyme-linked immunosorbent spot, BD Biosciences)의 방법을 이용하였다. 상기 실험군의 T-림프구가 표적세포이며 Branched DC-CTLs (브랜치드 멀티펩티드를 항원으로 하여 수지상세포로 만든 세포독성 림프구)이 반응세포이다. 반응세포와 표적세포의 비율은 1:10이며 인터페론감마의 숫자는 automatic CTL immunospot analyzer에 의해서 측정하고 데이터는 Mean number of spotsㅁ standard deviation per 105 CD8+T cells로 표현하였다.
상기 각 실험군에 대한 측정 결과는 도 5에 도시하였다.
상기 결과로부터, 세포독성림프구는 실험군 2-4에 대해 인터페론 감마의 분비능이 크게 증가하였음을 확인할 수 있었으며, 특히 브랜치드 멀티펩티드를 발현시킨 경우 분비능이 가장 많이 증가한 놀라운 결과를 확인할 수 있었다. 이를 통해, 본원 발명의 브랜치드 멀티펩티드를 이용하여 면역 치료를 위한 백신으로 활용이 가능할 것으로 기대된다.
TAA: tumor-assoicated antigen, 종양항원, multipeptide-cocktail: BIRC5 96 methionine, CD99 9, ERBB2 369의 세가지 싱글 펩티드들의 혼합, branched-multipeptide: BIRC5 96 methionine, CD99 9, ERBB2 369의 세가지 펩티드들을 mini-PEG2을 이용하여 브랜치드 펩티드 구조로 만듦, branched DC-CTLs: 브랜치드 펩티드로 자극된 수지상세포(Dendritic cell, DC)를 통하여 만들어진 세포독성 림프구들, unpulased T2: 펩티드가 발현되어있지 않은 T림프구, A,B,C T2: 각각의 BIRC5 96 methionine, CD99 9, ERBB2 369의 펩타이들을 나누어서 T2에 발현시킨 T림프구, A/B/C T2: BIRC5 96 methionine, CD99 9, ERBB2 369의 펩타이들을 혼합한 후 T2에 발현시킨 T림프구, Branched T2: 브랜치드 펩티드를 T2에 발현시킨 T림프구, HIV T2: HIV가 발현되어진 T림프구.

Claims (6)

  1. 화학식 I의 구조를 갖는 암 면역치료용 멀티펩티드 조성물:

    상기 화학식 I에서,
    A, B 및 C는 각각 뇌종양항원 CD99, BIRC5, ERBB2, WT-1 및 PanDR epitope 로 이루어진 군로부터 선택되는 어느 하나의 펩티드의 잔기서열(subsequence residue)이고;
    Sp는 잔기서열과 리신(lysine)의 연결을 위한 스페이서이고;
    Lys은 리신(lysine)이다.
  2. 제 1항에 있어서,
    상기 A, B 및 C는 각각 서열번호 1 내지 17로부터 선택되는 어느 하나의 아미노산 서열을 갖는 것을 특징으로 하는 조성물.
  3. 제 2항에 있어서,
    상기 A는 서열번호 1 내지 5로부터 선택되는 어느 하나의 아미노산 서열, 상기 B는 서열번호 6 내지 8로부터 선택되는 어느 하나의 아미노산 서열 및 상기 C는 서열번호 9 내지 13으로부터 선택되는 어느 하나의 아미노산 서열을 갖는 것을 특징으로 하는 조성물.
  4. 제 3항에 있어서,
    상기 A는 서열번호 1의 아미노산 서열, 상기 B는 서열번호 8의 아미노산 서열 및 상기 C는 서열번호 9의 아미노산 서열을 갖는 것을 특징으로 하는 조성물.
  5. 제 1항에 있어서,
    상기 Sp는 8-amino-3,6-dioxa-octanoic acid (mini-PEG2)인 것을 특징으로 하는 조성물.
  6. 제 1항 내지 제 5항 중 어느 한 항의 조성물을 포함하는 암 면역치료용 백신.
