Abstract
본 발명은 트록세루틴(Troxerutin)을 유효성분으로 포함하는 피부 미백 화장료 조성물에 관한 것이다. 본 발명에 따른 트록세루틴을 포함하는 화장료 조성물은 멜라닌 생성 억제 효과 및 티로시나아제 활성 저해 효과가 우수하다.
Description
본 발명은 트록세루틴을 함유하는 피부 미백 화장료 조성물에 관한 것이다.
인간의 피부는 외부 자극으로부터 보호해주는 기본적인 특징 외에 미용과도 깊은 연관을 가지고 있다. 특히 피부색은 피부 미용의 가장 근본적인 현상을 나타내고 있어 피부색을 조절하는 기작에 대한 연구가 화장품업계에서는 활발히 진행되고 있다. 피부가 검게 되는 것은 내적, 외적 요인에 대한 피부세포의 반응에 의하여 발생하는데, 현상적으로는 티로시나제라는 효소가 작용하고 뒤따르는 일련의 산화과정을 통해 멜라닌이라는 중합체가 과량으로 만들어져 피부에 축적되는 것이다. 이 멜라닌의 생합성에 영향을 미치는 가장 큰 요인은 자외선과 체내 호르몬 분비이다. 멜라닌은 자외선을 흡수 또는 산란시켜 자외선으로부터 피부가 손상되는 것을 방지하는데 큰 역할을 하는데, 특별한 최대흡수파장이 없으며 전 영역의 빛을 흡수한다. 또한 활성 산소를 제거하는 기능이 탁월하나, 때로는 멜라닌 자체가 활성산소를 발생시키기도 하며, 멜라닌 구조내의 카테콜이나 퀴논에 의하여 다른 물질을 환원시키거나 산화시키고, 멜라닌 자체가 자유라디칼의 성질을 나타내기도 한다. 최근의 노화 예방에 대한 기술은 주로 콜라겐 합성을 촉진해주거나 멜라닌 생성을 억제하여 피부 색소 침착을 예방하는 방향으로 진행되고 있다. 흑화 피부나 기미, 주근깨 등의 색소성 피부질환을 개선, 치료하고자 하는 욕구는 특히 여성에게 있어서는 아주 크다. 종래에 비타민 C 및 그의 유도체를 유효 성분으로 함유한 피부 미백용 조성물을 많이 사용하여 왔으나, 이들 물질들은 수용액 중에서 매우 불안정하고 쉽게 산화되어 미백 효과가 지속적이지 못하였다.
트록세루틴[Vitamin P4; 3', 4', 7' - Tris(O-(2-hydroxyethyl))]은 괴화나무(Sophora Japonica)로부터 얻어지는 천연 플라보노이드이다. 가장 안정적인 바이오 플라보노이드 중 하나인 트록세루틴은 염증과 알러지를 유발하는 리폭시게나아제를 억제하고, 염증 매개 물질인 프로스타글란딘의 형성을 억제하는 것으로 알려져 있다(Blasig et al., 1988; Elangovan and Pari, 2013; Fan et al.,2009; Lu et al., 2010a; Lu et al., 2010b; Lu et al., 2011; Zhang et al., 2009). 또한 트록세루틴은 모세혈관 저항력을 조절함으로써 건강한 혈액과 림프의 미세순환을 도움으로써 피부를 안정화시키는 역할을 한다(Budzianowski et al., 1999). 또한 트록세루틴은 피부 세포에서 자외선에 의해 유되되는 세포사, 세포 이동 제한, 성장 중지, DNA 손상으로부터 피부 세포를 보호하는 효과가 있다(Lu et al., 2011; Ping et al., 2012).
한국특허출원 제2007-7000934호는 플라보노이드 화합물, 비타민 B3 화합물 및 피부학적으로 허용가능한 오일 연속 상 담체를 함유하는 피부케어 조성물에 관한 것으로, 플라보노이드 화합물과 비타민 B3 화합물의 병용에 의해 피부미백, 노화 방지의 효과가 있음이 기술되어 있으나, 트록세루틴 자체에 대하여 화장료에 사용하는 경우 피부 미백효과가 있음에 대해서는 기재되어 있지 않다.
이에, 본 발명자들은 괴화나무로부터 단일 성분을 추출하여 피부 미백 효과를 연구하던 중, 트록세루틴(Troxerutin)이 특히 피부 미백 효능이 뛰어난 것을 발견하였다. 또한, 이러한 트록세루틴을 화장료에 유효성분으로 첨가할 경우 멜라닌 과생성으로 인해 나타나는 피부 색소 침착을 억제해주는 효과가 매우 좋을 것으로 보고, 트록세루틴을 화장료에 첨가하여 확인한 결과, 실제로도 티로시나아제 활성 억제 효과 및 멜라닌 생성 억제 효과, 피부 색소 침착 등을 억제하여 피부 미백을 효과적으로 개선할 수 있음을 확인함으로써 본 발명을 완성하였다.
상기의 목적을 달성하기 위하여, 본 발명은 트록세루틴(Troxerutin)을 유효성분으로 함유하는 피부 미백용 화장료 조성물을 제공한다.
바람직하게는, 트록세루틴은 화장료의 총 중량에 대하여 0.00001 내지 10.0 중량%를 함유한다.
바람직하게는, 상기 화장료 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이로 구성된 군으로부터 선택되는 제형을 갖는다.
본 발명에 따르면, 트록세루틴(Troxerutin)은 티로시나아제 활성 억제 효과(실험예 1) 및 멜라닌 생합성 억제 효과(실험예 2)가 탁월하며, 이를 화장료에 함유시킬 경우, 미백효과(실험예 3)가 우수하다.
본 발명은 트록세루틴(Troxerutin)을 함유하는 피부 미백용 화장료 조성물에 관한 것이다.
본 발명의 트록세루틴(Troxerutin)은 이미 공지된 물질로서 시판되는 것을 구입하여 사용하거나, 괴화나무(Sophora japonica) 등의 식물 등에서 추출하여 얻어낼 수도 있다. 트록세루틴은 아래의 화학식을 갖는다.

본 발명의 바람직한 실시예에 따르면, 당업계 공지된 통상의 방법, 즉, 통상적인 용매를 사용하여, 통상적으로 이용되는 열수추출, 실온추출, 가온추출, 초음파추출, 초임계추출 등의 방법으로 제조될 수 있다.
바람직하게는 물, 탄소수 1 내지 6의 저급 글리콜, 탄소수 1 내지 4의 저급알코올 및 이들의 혼합물로 이루어진 군으로부터 선택되는 용매를 사용하여 추출한다. 가장 바람직하게는 1,3-부틸렌글리콜을 추출용매로 추출한다. 또한 상기 추출물은 추가적으로 여과 또는 정제하는 과정을 거칠 수 있다.
또한, 시판되는 트록세루틴의 경우 구입 후 정제하여 사용하는 것이 바람직하며, 이때 사용되는 정제방법은 이 분야에서 통상적으로 사용되는 방법이 가능하다. 각종 유기용매를 혼합하여 사용한 액-액 추출, 실리카겔이나 활성 알루미나 등의 각종 합성수지를 충진한 컬럼크로마토그래피 및 고속액체크로마토그래피(HPLC) 등의 정제방법이 가능하나. 상기에서 서술한 정제 방법에 한정된 것은 아니다.
본 발명의 보다 바람직한 실시예에 따르면, 본 발명의 트록세루틴은 티로시나제 활성 저해 효과와 멜라닌 생합성 억제 효과가 우수하다.
또한, 본 발명의 바람직한 실시예에 따르면, 본 발명의 트록세루틴의 함량은 화장료 조성물의 총 중량에 대하여 0.00001 ~ 10.0중량%, 바람직하게는 0.0001 ~ 10.0중량%, 보다 바람직하게는 0.001 ~ 5중량%, 보다 바람직하게는 0.01 ~ 1중량%이다. 이때 트록세루틴의 함량이 상기 범위 미만일 경우 뚜렷한 효과가 나타나기 어렵고 10.0중량%를 초과할 때에는 피부에 자극을 유발할 가능성이 높으며 제형의 안정화에도 큰 영향을 미칠 수 있다.
본 발명의 화장품 조성물에 포함되는 성분은 유효 성분으로서의 트록세루틴 이외에 화장품 조성물에 통상적으로 이용되는 성분들을 포함하며, 예컨대 항산화제, 안정화제, 용해화제, 비타민, 안료 및 향료와 같은 통상적인 보조제, 담체 등을 포함한다.
본 발명의 화장료 조성물은 당 업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 예를 들어, 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 오일, 분말 파운데이션, 유탁액 파운데이션 등으로 제형화 될 수 있으나, 이에 한정되는 것은 아니다. 보다 상세하게는, 유연 화장수(스킨), 영양 화장수(밀크로션), 영양 크림, 마사지 크림, 에센스, 아이 크림, 클렌징 크림, 클렌징 폼, 클렌징 워터, 팩, 스프레이 또는 파우더의 제형으로 제조될 수 있다.
본 발명의 제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 셀룰로오스 유도체, 폴리에틸렌 글라이콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명의 제형이 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용되고, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌 글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다.
본 발명의 제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상의 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실험예
1:
트록세루틴의
티로시나아제 활성 저해 실험
발명자들은 본 발명의 트록세루틴에 대한 티로시나아제 활성 억제 정도를 조사하기 위하여 다음과 같이 실험을 실시하였다. 본 실험에 사용한 트록세루틴은 JIAHERB사(중국)로부터 구입하여 사용하였다. 기질로는 인산 나트륨 완충액 (pH 6.8)에 용해한 1.5 mmol/ℓ의 티로신 (시그마사, 미합중국) 40 ㎕를 사용하였다. 구입한 트록세루틴을 각각 0.05 M 인산나트륨 완충용액 (pH 6.8)으로 희석하여 농도 100ppm, 10ppm, 5ppm, 1ppm의 시료액을 제조하였다. 티로신 40 ㎕에 상기 농도별 시료액 240 ㎕을 각각 첨가하고, 상기 용액에 티로시나아제 (시그마사, 1500 U/㎖) 20 ㎕을 첨가한 후 37℃에서 15분 동안 반응시킨 다음, 490 nm에서 흡광도를 측정하였다. 양성 대조군으로는 트록세루틴 대신 미백효능이 알려진 알부틴 1%를 사용하였다. 하기 수학식 1에 의하여 티로시나아제의 활성 저해도를 계산하였으며, 실험 결과를 하기 표 1에 기재하였다.

표 1의 결과에서도 알 수 있듯이, 본 발명의 트록세루틴은 1ppm부터 100ppm까지 넓은 영역에서 티로시나아제 활성 억제효과가 우수하였으며, 약 5ppm 이상일 경우 오히려 양성대조군에 비하여 더 우수하였다.
실험예
2 :
트록세루틴의
멜라닌 생합성 억제 실험
발명자들은 본 발명의 트록세루틴에 대한 멜라닌 생합성 억제정도를 조사하기 위하여 다음과 같이 실험을 실시하였다. 상세히 설명하면, B-16 세포 (쥐 멜라노마 세포, 한국세포주은행)를 6-웰 플레이트 (well plate)에 1× 105 cell/well이 되도록 접종한 후에 24시간 배양하였다. 각 웰에 트록세루틴을 각각 100ppm, 10ppm, 5ppm, 1ppm을 처리하고 양성 대조군으로 알부틴을 1.0중량% 처치한 후 3-4일 동안 배양한 후 각 웰의 배양액을 원심 분리하였다. 분리된 세포는 디메틸설폭사이드 (DMSO)에 용해한 1N NaOH 용액 500 ㎕로 용해하여 10분간 90℃ 수욕 (water bath)에서 가열하였다. 이 상등액을 490 ㎚에서 흡광도를 측정하였다. 음성 대조군으로는 트록세루틴을 처리하지 않은 것을 사용하였다. 멜라닌 생성 저해도는 하기 수학식 2에 따라 계산하였으며, 그 결과를 표 2에 나타내었다.

표 2의 결과에서도 알 수 있듯이, 본 발명의 트록세루틴은 5ppm 내지 100ppm의 넓은 영역의 함량에서 양성대조군과 비교하여 멜라닌 생합성 억제효과가 유사하거나 오히려 훨씬 우수하였으며, 또한 이러한 결과로부터 트록세루틴이 멜라닌 생성을 억제하여 피부 색소 문제를 해결할 수 있음을 알 수 있었다.
이하, 상기한 실험예의 결과를 근거로 하여 본 발명의 트록세루틴을 포함하는 화장료를 조성하여 제시한다. 그러나 본 발명의 조성물을 하기의 처방예들로 한정하고자 하는 것은 아니다.

트록세루틴을
함유한
화장료의
미백 효과 실험
20∼40세의 특히 피부가 검은 한국 여성 20명을 대상으로 본 발명의 화장료 조성물의 미백 효과 실험을 실시하였다. 실험제품은 먼저 트록세루틴이 0.1중량% 함유한 크림 제품(실시예 1)과 대조제품으로는 트록세루틴이 들어 있지 않은 크림 제품(대조실시예 1)을 사용하였다. 실험은 먼저 대조제품과 트록세루틴이 함유된 제품을 20명의 피검자에게 각각 얼굴의 양쪽 면을 사용하였다. 측정 장비로는 Minolta CR 300을 사용하였으며 8주 동안 측정하였다. 실험 결과는 8주에 찍은 L값과 0주에 찍은 L값을 비교하였으며 그 결과는 하기 표 6에 나타나있다.

상기 표 6에 나타난 바와 같이, 트록세루틴을 함유한 제품(실시예 1)이 대조 제품(대조실시예 1)에 비하여 미백 효과가 훨씬 높게 나타났다. 따라서 본 발명의 화장료 조성물은 매우 우수한 미백 효과를 갖고 있음을 알 수 있었다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
Claims (3)
- 트록세루틴을 함유하는 피부 미백용 화장료 조성물.
- 제1항에 있어서, 상기 트록세루틴은 화장료의 총 중량에 대하여 0.00001 내지 10.0중량%를 함유하는 것인 화장료 조성물.
- 제1항에 있어서, 상기 화장료 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이로 구성된 군으로부터 선택되는 제형을 갖는 것인 피부 미백용 화장료 조성물.
