Abstract
Description
도 2는 실시예 2의 탯줄 유래 줄기세포에 있어서, 배아줄기세포의 마커가 RNA 수준에서 발현됨을 보여 주는 결과이다.
도 3은 실시예 3의 탯줄 유래 줄기세포의 계대배양에 따른 중간엽줄기세포 마커를 분석한 결과이다. 도 3에 나타나 있듯이, 중간엽줄기세포의 특성인 CD29, CD73, CD105, CD166 등이 계대배양 20회까지 유지됨을 확인할 수 있고, 25번째 계대배양부터는 줄기세포의 마커를 상실하였다.
이때, x축은 강도(intensity)이며, Y축은 세포갯수(카운트)이다. x축 및 y축의 변화로 위에 기재되어 있는 CD 마커를 식별할 수 있다.
도 4는 실시예 4의 탯줄 유래 줄기세포의 분화능(연골, 뼈 및 지방으로의 분화)을 보여 주는 그림이다.
도 5는 실시예 5 및 실시예 7에서 제조한 매트릭스에 있어서, 콜라겐과 히알루론산 유도체의 비율에 따른 형태 유지를 관찰한 결과이다.
도 6 및 도 7은 실시예 7에서 제조한 매트릭스에 탯줄 유래 줄기세포를 혼합하여 배양했을 때, 매트릭스 내에서의 세포 증식 및 생존률을 나타낸다. 대조군인 콜라겐으로만 이루어진 지지체보다 히알루론산 유도체와 탯줄 유래 콜라겐을 혼합한 실험군에서 증식력과 생존률이 보다 높게 유지되는 것을 보여 준다.
도 8은 실시예 8에 따른 탯줄 유래 줄기세포의 생체내 분화(마우스 피하)를 보여 준다.
도 9는 실시예 8에 따른 따른 탯줄 유래 줄기세포의 생체내 분화(마우스 피하)를 다양한 염색법을 통하여 보여 준다.
도 10 및 도 11은 토끼 무릎 관절연골에 연골하골 부위까지 손상을 준 후 아무 처리하지 않은 것(non-treatment)과 지지체만 넣어준 것(대조군) 및 탯줄 유래 줄기세포와 지지체를 혼합한 것(실험군)을 이식하고 8주 후 및 16주 후에 관찰한 결과로서 실험군에서 연골이 완벽하게 재생된 결과를 보여준다.
도 12는 실시예 9에 따른 따른 탯줄 유래 줄기세포와 지지체 혼합물의 손상된 연골 재생효과(토끼 관절)를 H&E 염색법을 통하여 보여 준다.
도 13은 실시예 9에 따른 따른 탯줄 유래 줄기세포와 지지체 혼합물의 손상된 연골 재생효과(토끼 관절)를 II형 콜라겐 염색법을 통하여 보여 준다.

Claims (21)
- 콜라겐 및 히알루론산 또는 그의 유도체를 포함하는 조성물.
- 제1항에 있어서, 상기 콜라겐은 포유류 유래 콜라겐인 것인 조성물.
- 제2항에 있어서, 상기 포유류 유래 콜라겐은 인간 탯줄 유래 콜라겐인 것인 조성물.
- 제3항에 있어서, 상기 인간 탯줄 유래 콜라겐은 I형 콜라겐인 것인 조성물.
- 제1항에 있어서, 상기 히알루론산 유도체는 히알루론산을 BDDE를 사용하여 가교화하여 제조한 것인 조성물.
- 제1항의 조성물을 포함하는 의료용 복합 생체소재.
- 줄기세포, 콜라겐 및 히알루론산 또는 그의 유도체를 포함하는 연골세포치료제.
- 제7항에 있어서, 상기 줄기세포는 포유류 유래의 줄기세포인 것인 연골세포치료제.
- 제8항에 있어서, 상기 포유류 유래의 줄기세포는 탯줄 유래 줄기세포인 것인 연골세포치료제.
- 제7항에 있어서, 상기 콜라겐은 포유류의 탯줄에서 수득한 것인 연골세포치료제.
- 제7항에 있어서, 상기 줄기세포는 포유류의 탯줄 추출물을 포함한 줄기세포 배양배지를 이용하여, 세포 부착 단백질로 코팅된 용기에서 배양하여 제조된 것인 연골세포치료제.
- 제11항에 있어서, 상기 줄기세포 배양배지는 혈청을 포함하지 않는 것인 연골세포치료제.
- 제11항에 있어서, 상기 세포 부착 단백질은 포유류 탯줄 유래 콜라겐, 젤라틴(gelatin), 피브로넥틴(fibronectin), 라미닌(laminin) 및 폴리-D-라이신(poly-Dlysin)으로 이루어진 군으로부터 선택되는 것임을 특징으로 하는 연골세포치료제.
- 제7항에 있어서, 상기 히알루론산 유도체는 히알루론산 또는 BDDE를 사용하여 가교화하여 제조된 것인 연골세포치료제.
- 제7항에 있어서, 상기 히알루론산 또는 그의 유도체와 상기 콜라겐의 조합 비율은 3:1 내지 1:3인 것임을 특징으로 하는 연골세포치료제.
- 제15항에 있어서, 상기 히알루론산 또는 그의 유도체는 1% 내지 3%인 것인 연골세포치료제.
- 제15항에 있어서, 상기 콜라겐은 1% 내지 3%인 것인 연골세포치료제.
- 제7항에 있어서, 상기 포유류의 탯줄 유래 줄기세포는 1.0×106 내지 1×108 cells/ml의 농도로 히알루론산 유도체와 포유류의 탯줄 유래 콜라겐 젤과 혼합되는 것임을 특징으로 하는 연골세포치료제.
- 제7항에 있어서, 상기 연골세포치료제는 하이드로젤 형태인 것임을 특징으로 하는 연골세포치료제.
