그림 2는 미담청결안이 Propionibacterium acnes의 성장에 미치는 영향을 시간대별로 OD값을 측정 조사한 것이다. 미담청결안의 추출물을 0.1%~1.0% (w/v)까지 P.acnes 배양배지에 첨가하여 1×105cfu/ml가 되게 균을 접종하였다. 미담청결안 추출물을 함유하고 있지 않은 배지의 경우 균의 유도기는 14시간이 었으며, 그 후 급격한 균체량의 증가를 나타내는 대수 증식기에 들어갔다. 48시간이 지난 후 균은 정지기에 들어감을 관찰할 수 있었다. 그렇지만 S4 추출물 0.1%(w/v)를 첨가한 배지의 경우 60시간까지 유도기에 있었으며 점차적인 균체량 증가를 보였지만 급격한 균의 증가를 보이지는 않았다. 이는 미담청결안이 저 농도에서도 P.acnes의 성장을 억제한다는 것을 말해준다. 0.5% 이상의 미담청결안 첨가 배지에서는 균의 성장이 관찰되지 않았다. 이는 미담청결안을 0.5%첨가 시 P.acnes의 성장을 완전 억제할 수 있다는 것으로 의미한다. 또한, 미담청결안 추출물은 이 균에 대해 상당히 높은 성장 억제력을 가지고 있으므로 P.acnes에 대한 성장억제물질로 사용 가능하리라 여겨진다. 그림 3은 미담청결안이 Stapylococcus aureus의 성장에 미치는 영향을 시간대별로 OD값을 측정하여 조사한 것이다. Stapylococcus aureus는 실험에 사용한 다른 미생물에 비해 성장속도가 빠르다는 것을 알 수 있으며 1×106cfu/ml 접종시 1시간 유도기를 거쳐 12시간 후까지 대수 증식기를 나타냈다. 미담청결안이 0.1%~1.0% 첨가된 배지의 경우 특이적인 저해능이 관찰되지 않았지만, 미담청결안의 처리 농도가 증가할수록 미생물이 유도기에서 대수 증식기로 들어가는 시간을 늦추어주며 배양 시 최대 균체량을 낮추어 주었다. 이는 미담청결안의 첨가농도가 높을수록 Stapylococcus aureus의 성장 억제 효과가 있음을 보여주고 있다. 또한, 미담청결안이 Stapylococcus aureus의 성장을 억제하지만 효과를 나타내기 위해서는 높은 농도 처리가 필요하다는 것을 말해주고 있다.

제조예1의 미담청결안의 세포독성실험을 위해 세포주는 생쥐 섬유모세포의 일종인 L929 세포주를 사용하였다. 세포 배양은 10% fetal bovine serum(FBS)과 100unit/ml의 penicillin/streptomycin을 1% 첨가한 Dulbecco's Modified Eagle's Medium(DEME) 배지를 사용하며, 37℃, 5% CO2로 조정된 배양기내에서 배양하며, 배양액은 3일마다 교환한다. 배양된 세포는 1× trypsin-EDTA로 부유시킨 후 0.4% trypan blue로 염색하여 혈구계측기 (Hemocytometer)로 세포수를 계산한다. 세포독성 시험은 배양된 세포를 96 well plate에 5×103 cell/well이 되도록 세포 부유액을 120㎕씩 분주하여 배양한 후 실험에 사용한다. 상기 제조예 2, 비교제조예 1에 따른 화장료용 조성물의 안전성을 확인하기 위하여 인체 피부 누적자극 시험을 실시하였다. 구체적으로, 건강한 20명의 성인 남여를 대상으로 상기 제조예 2, 비교제조예 1에 따른 화장료용 조성물 각각에 대해, 윗 팔뚝 부위에 격일로 총 9회의 24시간 누적 첩포를 수행하였다. 상기 첩포 방법으로는 핀 챔버(Finn chamber) 방법이 이용되었으며, 챔버당 15㎕의 화장료용 조성물을 적하한 후 첩포를 실시하였다. 매회 피부에 나타난 반응도를 그 정도에 따라 ±, +, ++로 나누어 나타냈으며, 각각에 대하여 1점, 2점, 및 4점(최고점수)을 부여하였다. 또한, 하기 수학식 6에 따라 평균 반응도를 계산하였으며, 상기 반응도의 결과와 함께 하기 표 3에 나타내었다. 하기 표 3에서 평균 반응도가 3 미만일 때 안전한 조성물로 판정된다.
