Description
Claims (14)
- (a) 검출대상 시료가 수용되는 반응 챔버와, 상기 반응 챔버 내에 위치하고 상기 검출대상 시료 내의 생리활성물질의 검출을 위한 복수개의 센싱 전극을 구비하는 전기적 검출장치를 제공하는 단계와,(b) 상기 반응 챔버 내로 유전상수가 큰 레퍼런스 유체를 도입한 후, 상기 센싱 전극들 간의 임피던스값 또는 커패시턴스값을 측정하는 단계와,(c) 상기 반응 챔버로부터 상기 유전상수가 큰 레퍼런스 유체를 제거한 후, 상기 반응 챔버 내에 반응용액 및 검출대상 시료를 제공하는 단계와,(d) 상기 반응용액 하에서 상기 검출대상 시료를 상기 반응 챔버 내에서 반응시키는 단계와,(e) 상기 반응용액을 상기 반응 챔버로부터 제거하는 단계와,(f) 상기 반응 챔버 내로 상기 유전상수가 큰 레퍼런스 유체를 다시 도입한 후, 상기 센싱 전극들 간의 임피던스값 또는 커패시턴스값을 측정하는 단계와,(g) 상기 (b)단계에서 측정된 임피던스값 또는 커패시턴스값과 상기 (f)단계에서 측정된 임피던스값 또는 커패시턴스값을 비교하는 단계를 포함하는 생리활성물질을 전기적으로 검출하는 방법.
- 제1항에 있어서,상기 (b)단계 및 (f)단계에서는 교류전원을 이용하는 측정장치에 의해 각각의 주파수별로 임피던스값 또는 커패시턴스값을 측정하는 것을 특징으로 하는 생리활성물질을 전기적으로 검출하는 방법.
- 제2항에 있어서,상기 측정된 임피던스값 또는 커패시턴스값은 주파수값에 대한 곡선으로 표현되고, 상기 (b)단계에서 측정된 임피던스값 또는 커패시턴스값의 곡선이 상기 (f)단계에서 측정된 임피던스값 또는 커패시턴스값의 곡선과 구분되는 경우 상기 검출대상 시료 내에 생리활성물질이 존재하거나 생리활성물질이 상기 센싱 전극과 반응하는 것으로 판단하는 것을 특징으로 하는 생리활성물질을 전기적으로 검출하는 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서,상기 유전상수가 큰 레퍼런스 유체는 유전상수가 4 이상인 유체인 것을 특징으로 하는 생리활성물질을 전기적으로 검출하는 방법.
- 제4항에 있어서,상기 유전상수가 큰 레퍼런스 유체는 증류수 또는 탈이온수(deionized water)인 것을 특징으로 하는 생리활성물질을 전기적으로 검출하는 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서,상기 센싱 전극은 IDE(interdigitated electrode)로서 형성되는 것을 특징으로 하는 생리활성물질을 전기적으로 검출하는 방법.
- 제6항에 있어서,상기 센싱 전극의 각각의 전극 사이의 간격은 1-4μm인 것을 특징으로 하는 생리활성물질을 전기적으로 검출하는 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서,상기 센싱 전극에는 상기 생리활성물질과 상호작용을 하는 수용체가 결합되는 것을 특징으로 하는 생리활성물질을 전기적으로 검출하는 방법.
- 제8항에 있어서,상기 수용체는 프로브 또는 프라이머인 것을 특징으로 하는 생리활성물질을 전기적으로 검출하는 방법.
- 복수개의 센싱 전극이 형성된 제1기판과,상기 제1기판 상에 일정한 거리를 두고 상기 제1기판과 결합하여 반응 챔버 공간을 형성하는 제2기판을 포함하고,상기 반응 챔버 공간을 형성하는 상기 제2기판에는 상기 제2기판의 두께 방향으로 유체 유입구가 관통되어 형성되어 있고 상기 유체 유입구에 대향하는 위치에 상기 제2기판의 두께 방향으로 유출구가 관통되어 형성되어 있으며,상기 유입구와 상기 반응 챔버 공간, 그리고 상기 유출구와 상기 반응 챔버공간은 유체가 유동될 수 있도록 연통되고,상기 유입구를 통해 유전상수가 큰 레퍼런스 유체, 반응용액 및 검출대상 시료가 유입되고 상기 유출구를 통해 유전상수가 큰 레퍼런스 유체, 반응용액 및 검출대상 시료가 유출되는 것을 특징으로 하는 생리활성물질을 전기적으로 검출하기 위한 바이오 칩.
- 제10항에 있어서,상기 제2기판 상에는 제3기판이 적층되어 상기 유입구로 유체를 도입안내하는 유입 마이크로 채널과, 상기 유출구로부터 유체를 배출안내하는 유출 마이크로 채널을 형성하고,상기 유입 마이크로 채널에는 상기 유전상수가 큰 레퍼런스 유체, 반응용액 및 검출대상 시료가 주입되는 주입구가 연통되어 상기 제3기판을 관통하여 형성되어 있으며,상기 유출 마이크로 채널에는 상기 유전상수가 큰 레퍼런스 유체, 반응용액 및 검출대상 시료가 배출되는 배출구가 연통되어 상기 제3기판을 관통하여 형성되어 있는 것을 특징으로 하는 생리활성물질을 전기적으로 검출하기 위한 바이오 칩.
- 제11항에 있어서,상기 유입 마이크로 채널 또는 상기 유출 마이크로 채널에는 상기 반응 챔버 공간 내로의 유체의 유입 또는 상기 반응 챔버 공간으로부터 유체의 유출을 제한할 수 있는 밸브 구조가 제공되는 것을 특징으로 하는 생리활성물질을 전기적으로 검출하기 위한 바이오 칩.
- 제12항에 있어서,상기 밸브구조는 상기 주입구와 상기 반응 챔버 공간 사이에서, 또는 상기 배출구와 상기 반응 챔버 공간 사이에서 상기 제3기판의 두께 방향으로 관통된 후 연장된 통로로서, 이러한 연장된 통로에는 오일이 내장되는 것을 특징으로 하는 생리활성물질을 전기적으로 검출하기 위한 바이오 칩.
- 제11항 내지 제13항 중 어느 한 항에 있어서,상기 제3기판은 PDMS(polydimethylsiloxane) 중합체로 형성되는 것을 특징으로 하는 생리활성물질을 전기적으로 검출하기 위한 바이오 칩.
