Description
Claims (25)
- Fc-함유 단백질을 포함하는 유체를 양이온 교환 크로마토그래피로 처리하는 것을 포함하여, 상기 유체에서 유리 Fc-부분의 농도를 감소시키는 방법.
- 제 1항에 있어서, 상기 유체를 상기 Fc-함유 단백질의 등전점(pI) 보다 적어도 1 단위(one unit) 낮은 pH에서 양이온 교환 수지 상에 로딩시키는 단계를 포함하는 방법.
- 제 1항 또는 제 2항에 있어서, 양이온 교환 수지를 6 내지 10 mS/cm의 전도성 및 5.5 내지 7.5의 pH를 지니는 완충액으로 세척하는 단계를 포함하는 방법.
- 제 3항에 있어서, 세척 단계가 8.2 내지 8.6 mS/cm의 전도성에서 수행되는 방법.
- 제 3항 또는 제 4항에 있어서, 세척 단계가 6.0 내지 7.0의 pH에서 수행되는 방법.
- 제 3항 내지 제 5항 중의 어느 한 항에 있어서, 세척 단계가 75 내지 125 mM의 인산나트륨을 포함하는 완충액에서 수행되는 방법.
- 제 1항 내지 제 6항 중의 어느 한 항에 있어서, Fc-함유 단백질을 7.0 내지 8.5 범위의 pH에서 용리시키는 단계를 포함하는 방법.
- 제 7항에 있어서, 용리 단계가 15 내지 22 mS/cm의 전도성에서 수행되는 방법.
- 제 1항 내지 제 8항 중의 어느 한 항에 있어서, 양이온 교환 크로마토그래피가 강한 양이온 교환 수지 상에서 수행되는 방법.
- 제 9항에 있어서, 양이온 교환 수지가 SO3 - 기를 포함하는 방법.
- 제 9항 또는 제 10항에 있어서, 수지가 가교된 메타크릴레이트 매트릭스를 포함하는 방법.
- 제 1항 내지 제 11항 중의 어느 한 항에 있어서, 친화력 크로마토그래피, 음이온 교환 크로마토그래피 및 히드록시아파타이트 크로마토그래피로부터 선택된 정제 단계를 추가로 포함하는 방법.
- 제 1항 내지 제 12항 중의 어느 한 항에 있어서, Fc-함유 단백질이 면역글로불린(Ig) 불변 영역을 포함하는 방법.
- 제 13항에 있어서, 불변 영역이 인간 불변 영역인 방법.
- 제 13항 또는 제 14항에 있어서, 면역글로불린이 IgG1인 방법.
- 제 13항 내지 제 15항 중의 어느 한 항에 있어서, 불변 영역이 CH2 및 CH3 도메인을 포함하는 방법.
- 제 1항 내지 제 16항 중의 어느 한 항에 있어서, Fc-함유 단백질이 면역글로불린 가변 영역을 포함하는 방법.
- 제 17항에 있어서, Fc-함유 단백질이 항체인 방법.
- 제 1항 내지 제 16항 중의 어느 한 항에 있어서, Fc-함유 단백질이 Fc-융합 단백질인 방법.
- 제 19항에 있어서, Fc-융합 단백질이 종양 괴사 인자 수용체(TNFR) 수퍼패밀 리의 멤버의 리간드 결합용 부분을 포함하는 방법.
- 제 20항에 있어서, 리간드 결합용 부분이 TNFR1 또는 TNFR2의 세포외 도메인, 또는 이의 TNF 결합 단편으로부터 선택되는 방법.
- 제 20항에 있어서, 리간드 결합용 부분이 BAFF-R, BCMA 또는 TACI의 세포외 도메인, 또는 Blys 또는 APRIL 중 적어도 하나와 결합되는 이의 단편으로부터 선택되는 방법.
- 제 22항에 있어서, Fc-융합 단백질이a. 서열번호 2의 아미노산 34 내지 66;b. 서열번호 2의 아미노산 71 내지 104;c. 서열번호 2의 아미노산 34 내지 104;d. 서열번호 2의 아미노산 30 내지 110;e. 서열번호 3;f. 서열번호 4;g. 고도로 엄격한 조건하에 서열번호 5 또는 6 또는 7의 상보서열(complement)에 하이브리드화되는 폴리누클레오티드에 의해 엔코딩되는 폴리펩티드; 및h. (c), (d), (e) 또는 (f)의 폴리펩티드에 적어도 80% 또는 85% 또는 90% 또는 95%의 서열 동일성을 지니는 (c), (d), (e) 또는 (f) 중 임의의 것의 뮤테인으로부터 선택된 폴리펩티드를 포함하고, 여기서 상기 폴리펩티드는 Blys 또는 APRIL 중 적어도 하나에 결합되는 방법.
- Fc-함유 단백질을 포함하는 조성물에서 유리 Fc-부분의 농도를 감소시키기 위한 양이온 교환 크로마토그래피의 용도.
