실험은 본 연구원에서 구축한 후각 실험실(4.8 m × 3 m × 2.4 m)에서 수행되었다. 상기 실험실 내부에는 후각의 순응을 줄이기 위한 목적으로 흡기 및 배기 시설이 되어 있으며, 피험자의 실험 참여도를 극대화하고 실험시 외부 환경의 영향을 배제하기 위하여 방음장치가 설치되어 있다. 실험시 내부 온도(25 ℃), 습도(40-50%), 조도(150-200 Lux)는 일정하게 유지되도록 하였다. 한 조건에 대한 실험이 끝난 후에는 피험자가 향에 대한 순응과 피로에 대한 영향이 없도록 충분히 쉬도록 하고, 실험실 내부를 환기시킨 후, 실험을 실시하였다. 향 조건으로는 무향(향을 제시하지 않음), 복합향(네롤리 오일 + 일랑 일랑 오일), 네롤리 오일향, 일랑 일랑 오일향을 제시하여 총 4가지 조건으로 실시하였다. 향은 피험자가 눈을 감은 상태에서 제시하였고, 생리신호를 측정하였다. 무향 조건에서는 물이 든 시약병을 제시하였고, 각 향 자극시 피험자의 코 밑 1-2 cm 거리에서 30초간 제시하였다. 향에 대한 순서 효과를 배제하기 위하여 피험자마다 향 제시 순서는 무작위로 실시하였다. 구체적으로는 하기와 같은 순서로 진행되었다. 뇌파는 국제 10-20 전극 배치법에 따라 두정엽 부위에 대하여 GRASS(USA) 장비와 Acknowledge 프로그램을 이용하여 측정하였다. 측정 부위의 저항치는 10 ㏀ 이하로 하였고, Sampling rate는 512 Hz로 설정하였다. EEG was measured using GRASS (USA) equipment and Acknowledge program for parietal lobe in accordance with international 10-20 electrode placement method. 뇌파는 FFT법에 의한 전력 스펙트럼 분석과 주파수 대역에 따라 β/(α+β)를 구하였고, 뇌파 맵핑 프로그램(BrainMap 3D, (주)Neuromedi, 한국)을 이용하여 시간에 따른 뇌파 이미지를 맵핑하였다. 피부전기저항(Galvanic skin response)은 각각 왼손 소지, 왼손 중지와 약지에 센서를 부착하여 측정하였다. 생리신호 측정은 Biopac 100 system과 Acqknowledge 프로그램을 이용하였으며 Sampling rate는 512 Hz로 설정하였다. 주관적 기분감 평가는 Profile of Moods States(POMS: 일본어판, 65문항)를 번역하여 이용하였다. 65문항 중 긴장(9문항), 우울(15문항), 성냄(12문항), 활기(8문항), 피로(7문항), 혼란(7문항)으로 구분되며 점수화시켜 평가하였다. 조건에 따른 주관적 기분감에 대한 평가항목에 대하여 평균과 표준편차를 구하였다. 통계적 분석은 SPSS 프로그램을 이용하여 ANOVA, t-test로 검증하였다. 향 조건에 따른 30초 동안의 뇌파의 상대적 β/(α+β) 변화량은 도 1과 같다. 무향 조건 수치를 기준으로 하여 복합향(네롤리 오일 + 일랑 일랑 오일)(A), 네롤리 오일(B), 일랑 일랑 오일(C) 향의 피험자별 β/(α+β) 변화량을 비교하였다. 주로 긴장되거나 각성과 관련된 향에서 α파에 대한 β파의 비가 증가하는데, 두정엽 부위에서 복합향의 β/(α+β)는 네롤리 오일(B)과 일랑 일랑 오일(C)에 비해 훨씬 높았다. 도 2는 향 자극시 피험자들의 시간에 따른 β/(α+β)의 변화를 뇌파 맵핑 이미지로 나타낸 것이다. 시간에 따른 β/(α+β)는 맵핑 이미지에서 β/(α+β)의 파워 스펙트럼 값이 클수록 붉은 색을 나타내고, 작을수록 푸른 색을 나타낸다. 무향이나 네롤리 오일(B), 일랑 일랑 오일(C)보다 복합향(네롤리 오일 + 일랑 일랑 오일)(A)을 제시하였을 때 뇌의 거의 모든 부위에서 더 활성화되었다. 복합향을 제시한 후 10초~15초 사이에 활성화도가 높았다. 향 자극에 따른 피부전기저항은 도 3에 나타내었다. 각성향을 제시할 경우 피부전기저항이 증가하는 경향을 보이는데, 본 실험에서 무향과 비교하였을 때 복합향(네롤리 오일 + 일랑 일랑 오일)(A), 네롤리 오일(B) 및 일랑 일랑 오일(C)을 제시한 경우에 피부전기저항 수치가 통계적으로 유의하게 높았다(p<0.05).
